Feed costs comprise the majority of variable expenses in beef cattle systems making feed efficiency an important economic consideration within the beef industry. Due to the expense of recording individual feed-intake phenotypes, a genomic-enabled approach could be advantageous toward improving this economically relevant trait complex. A genome-wide association study (GWAS) was performed using 748 crossbred steers and heifers representing seven sire breeds with phenotypes for ADG and ADFI. Animals were genotyped with the BovineSNP50v2 BeadChip containing approximately 54,000 SNP. Both traits were analyzed using univariate SNPbased (BayesC) and haplotype-based (BayesIM) models and jointly using BayesIM to perform a bivariate GWAS. …

The rumen has several important physiological functions, including absorption, transport, metabolic activity, and host protection (Roh et al., 2007). The rumen is extensively researched due to the importance of ruminants in agriculture and the major role played by rumen microbes in nutrient utilization and health of the ruminant animal. The microbial community of the rumen is altered by diet (McCann et al., 2014), age (Jami et al., 2013), and environment.

Intrauterine growth restriction (IUGR) greatly increases perinatal mortality and morbidity rates, and leads to much greater risk for metabolic complications later in life. One such complication is the development of glucose intolerance or diabetes, which typically develops concurrently with abhorrent patterns of insulin secretions due to diminished β-cell mass and impaired function as well as an overall reduction in pancreatic endocrine tissue. The mechanisms by which IUGR causes problems with health and function of the pancreatic islets are not well understood. Therefore, our goal for this study was to determine how materno-fetal inflammation (MI) affects β-cell growth and function. To …

Research Note Prediction Model For Manure Zinc Excretion In Laying Hens, Kelli M. Martin, N. Vargas-Jurado, S. E. Purdum

Department of Animal Science: Faculty Publications

The objective of this research trial was to compare 2 sources of zinc and different levels of supplementation on manure zinc excretion in laying hens. Bovan White Leghorn hens were used in this study and fed one of 6 dietary treatments. Treatments were in a 2 × 3 factorial arrangement with 2 sources of zinc (zinc sulfate or Availa R < Zn) and 3 levels (40, 80, or 120 ppm) and were randomly assigned to 48 cages with 5 hens/cage and 8 replicates/treatment. Hens were housed in a tiered manure-belt housing system providing 627.1 cm2/hen and were given access to 110 g/hen/d of feed. Manure samples were collected, and manure zinc content was calculated at 10- week intervals. Data were analyzed using the Glimmix procedure in SAS. There was a significant overall effect of source (P < 0.0001) for zinc content, such that hens fed zinc sulfate had lower amounts of zinc excretion. There was also a significant level effect (P < 0.0001) in which hens fed 120 ppm zinc excreted the greatest amount of zinc. A significant source by level interaction was observed (P < 0.0001) for both the overall and individual analyses, such that 120 ppm Availa R < Zn showed the highest zinc excretion, and both 40 ppm zinc sulfate and Availa R < Zn showed the lowest zinc excretion. Based on these results, a prediction equation was written for an estimated amount of zinc excretion based on the amount of zinc provided in the diet.

Owning to advancements in sensor‐based, non‐destructive phenotyping platforms, researchers are increasingly collecting data with higher temporal resolution. These phenotypes collected over several time points are cataloged as longitudinal traits and used for genome‐wide association studies (GWAS). Longitudinal GWAS typically yield a large number of output files, posing a significant challenge to data interpretation and visualization. Efficient, dynamic, and integrative data visualization tools are essential for the interpretation of longitudinal GWAS results for biologists; however, these tools are not widely available to the community. We have developed a flexible and user‐friendly Shiny‐based online application, ShinyAIM, to dynamically view and interpret temporal …

Effects Of Carbon Dioxide On Turkey Poult Performance And Behavior, M G.L Candido, Y Xiong, R S. Gates, I F.F. Tinoco, K W. Koelkebeck

Department of Animal Science: Faculty Publications

Appropriate ventilation of poultry facilities is critical for achieving optimum performance. Ventilation promotes good air exchange to remove harmful gases, excessive heat, moisture, and particulate matter. In a turkey brooder barn, carbon dioxide (CO2) may be present at higher levels during the winter due to reduced ventilation rates to maintain high temperatures. This higher CO2 may negatively affect turkey poult performance. Therefore, the objective of this study was to evaluate the effects of subjecting tom turkey poults (commercial Large White Hybrid Converters) to different constant levels of atmospheric CO2 on their growth performance and behavior. In …

Expression And Role Of Gonadotropin-Releasing Hormone 2 And Its Receptor In Mammals, Amy Desaulniers, Rebecca A. Cederberg, Clay A. Lents, Brett R. White Dec 2017

Expression And Role Of Gonadotropin-Releasing Hormone 2 And Its Receptor In Mammals, Amy Desaulniers, Rebecca A. Cederberg, Clay A. Lents, Brett R. White

Department of Animal Science: Faculty Publications

Gonadotropin-releasing hormone 1 (GnRH1) and its receptor (GnRHR1) drive mammalian reproduction via regulation of the gonadotropins. Yet, a second form of GnRH (GnRH2) and its receptor (GnRHR2) also exist in mammals. GnRH2 has been completely conserved throughout 500 million years of evolution, signifying high selection pressure and a critical biological role. However, the GnRH2 gene is absent (e.g., rat) or inactivated (e.g., cow and sheep) in some species but retained in others (e.g., human, horse, and pig). Likewise, many species (e.g., human, chimpanzee, cow, and sheep) retain the GnRHR2 gene but lack the appropriate coding sequence to produce a full-length …

Islet Adaptations In Fetal Sheep Persist Following Chronic Exposure To High Norepinephrine, Xiaochaun Chen, Amy C. Kelly, Dustin T. Yates, Antoni R. Macko, Ronald M. Lynch, Sean W. Limesand

Department of Animal Science: Faculty Publications

Complications in pregnancy elevate fetal norepinephrine (NE) concentrations. Previous studies in NE-infused sheep fetuses revealed that sustained exposure to high NE resulted in lower expression of α2-adrenergic receptors in islets and increased insulin secretion responsiveness after acutely terminating the NE infusion. In this study, we determined if the compensatory increase in insulin secretion following chronic elevation of NE is independent of hyperglycemia in sheep fetuses and whether it is persistent in conjunction with islet desensitization to NE. Following an initial assessment of glucose-stimulated insulin secretion (GSIS) at 129±1 days of gestation, fetuses were continuously infused for seven days with NE …

After ovulation, somatic cells of the ovarian follicle (theca and granulosa cells) become the small and large luteal cells of the corpus luteum. Aside from known cell type-specific receptors and steroidogenic enzymes, little is known about the differences in the gene expression profiles of these four cell types. Analysis of the RNA present in each bovine cell type using Affymetrix microarrays yielded new cell-specific genetic markers, functional insight into the behavior of each cell type via Gene Ontology Annotations and Ingenuity Pathway Analysis, and evidence of small and large luteal cell lineages using Principle Component Analysis. Enriched expression of select …

Congenital or juvenile cataract is a disease condition in which opacification of the lenses is present at birth or manifests early in life. It has been attributed to different monogenic factors with a high degree of heterogeneity and is often studied using mouse models. A spontaneous mutation was identified in a mouse line selected for heat loss that influenced lens formation and resulted in juvenile cataracts in mice homozygous for the recessive allele. Genetic dissection of this selection line by combining high-density genotypes and homozygosity mapping uncovered a 906 kb fragment on MMU7 encompassing 21 SNPs split into two groups …

Department of Animal Science: Faculty Publications

In ruminants, prostaglandin F2alpha (PGF2α)-mediated luteolysis is essential prior to estrous cycle resumption, and is a target for improving fertility. To deduce early PGF2α-provoked changes in the corpus luteum a short time-course (0.5-4 h) was performed on cows at midcycle. A microarraydetermined transcriptome was established and examined by bioinformatic pathway analysis. Classic PGF2α effects were evident by changes in early response genes (FOS, JUN, ATF3) and prediction of active pathways (PKC, MAPK). Several cytokine transcripts were elevated and NF-kB and STAT activation were predicted by pathway analysis. Self-organizing map analysis grouped differentially expressed transcripts into ten mRNA expression patterns indicative …

Dietary Energy Drives The Dynamic Response Of Bovine Rumen Viral Communities, Christopher L. Anderson, Matthew B. Sullivan, Samodha C. Fernando

Department of Animal Science: Faculty Publications

Background: Rumen microbes play a greater role in host energy acquisition than that of gut-associated microbes in monogastric animals. Although genome-enabled advancements are providing access to the vast diversity of uncultivated microbes, our understanding of variables shaping rumen microbial communities is in its infancy. Viruses have been shown to impact microbial populations through a myriad of processes, including cell lysis and reprogramming of host metabolism. However, little is known about the processes shaping the distribution of rumen viruses or how viruses may modulate microbial-driven processes in the rumen. To this end, we investigated how rumen bacterial and viral community structure …

Background: High-density marker panels and/or whole-genome sequencing, coupled with advanced phenotyping pipelines and sophisticated statistical methods, have dramatically increased our ability to generate lists of candidate genes or regions that are putatively associated with phenotypes or processes of interest. However, the speed with which we can validate genes, or even make reasonable biological interpretations about the principles underlying them, has not kept pace. A promising approach that runs parallel to explicitly validating individual genes is analyzing a set of genes together and assessing the biological similarities among them. This is often achieved via gene ontology analysis, a powerful tool that …

Swine are the only livestock species that produce both the second mammalian isoform of gonadotropin-releasing hormone (GNRH2) and its receptor (GNRHR2). Previously, we reported that GNRH2 and GNRHR2 mediate LH-independent testosterone secretion from porcine testes. To further explore this ligand-receptor complex, a pig model with reduced GNRHR2 expression was developed. Small hairpin RNA sequences targeting porcine GNRHR2 were subcloned into a lentiviral-based vector, lentiviral particles were generated and microinjected into the perivitelline space of zygotes, and embryos were transferred into a recipient. One GNRHR2 knockdown (KD) female was born that subsequently produced 80 piglets from 6 litters with 46 hemizygous …

Physically effective neutral detergent fiber (peNDF) is the fraction of neutral detergent fiber (NDF) that stimulates chewing activity and contributes to the floating mat of large particles in the rumen. Multiplying dietary NDF by particle size has been used as an estimate of peNDF. In re-evaluating the concept of peNDF, we compared the use of peNDF as dietary NDF × particle size with the use of individual NDF and particle size descriptors (physically adjusted NDF; paNDF) when used with other physical and chemical diet descriptors to predict dry matter (DM) intake (DMI), rumination time, and ruminal pH in lactating dairy …

Previous work examining the transcriptome of steer tissue samples from animals with divergent gain have shown a relationship with the expression of genes with functions in immune and inflammatory pathways. The process of mounting an immune or inflammatory response is energetically expensive and variation in cytokine responses may affect cattle production traits. In addition, a previous study has identified variation in the transcript abundance of numerous genes, including the cytokine gene IL6ST, in the circulating white blood cells of pigs associated with high and low residual feed intake (RFI) lines. The aim of this study was to determine whether …
