Validation Of Pcr-Based Assays And Laboratory Accreditation For Environmental Detection Of Aquatic Invasive Species, Invasive Species Advisory Committee

This white paper provides:

a) Background information on the use, accuracy and reliability of PCR-based assays such as environmentally sampled DNA (eDNA) for early detection of aquatic invasive species (AIS) and;

b) Recommendations for establishing a system for validating assays and accrediting laboratories that report on the presence or absence of AIS.

This white paper was developed by the members of ISAC and discusses the need for developing validation requirements for Polymerase Chain Reaction (PCR) and other DNA-based molecular assays that are increasingly being used to detect AIS. Nantucket’s shellfish resources are an important part of the Island’s history, culture, and economy. Nantucket waters support one of the country’s last wild-caught bay scallop fisheries. Elsewhere along the Atlantic coast, fishing pressure, habitat loss, and disease have severely depleted bay scallop populations. Although Nantucketers continue to make a living harvesting shellfish from the Island’s waters, many do so with concern for the future of the resources and the habitats that support them. In the village of Jambiani, Unguja, a Swiss NGO called Marine Cultures has established an experimental sponge farming initiative to improve local employment and support economic development. Currently, the main farming site is only accessible through SCUBA diving. This is consistent with experimental farm sites in other oceanic regions, as most are located in depths of at least five meters with sponges suspended vertically to maximize water flow exposure. Farms located in shallower water may not be conducive to sponge growth due to highly variable environmental conditions; however, no conclusive research has been conducted on this matter (Duckworth 2009). Considerable research on the documention of the bycatch of cetaceans, seals, turtles, and sea lions during trawl fishing operations has been undertaken worldwide. These include the Argentinean squid and hake trawl fisheries (Crespo et al., 1997; Fertl & Leatherwood, 1997), the Dutch pelagic trawl fishery (Couperus, 1997), the NE Atlantic pelagic trawl fishery (Tregenza & Collet, 1998) and the New Zealand squid fishery (Fishing News International, 2005).
