Improving production parameters and controlling foodborne pathogens have been challenges to the poultry industry. Salmonella has been the most common bacterial pathogen in laboratory confirmed foodborne illness cases, and contaminated poultry and poultry products have been identified as the most important source of transmission of Salmonella to humans. Therefore, research on effective interventions to reduce Salmonella transmission at the poultry production level has gained attention. The importance of poultry production ultimately hinges on the demands of the consumers and the perceptions that drive their purchases. With this in mind, this study surveyed consumers to assess their perceptions of poultry production in Arkansas. The instrument used to survey consumers was created by the researcher and an expert committee at the University of Arkansas. It is estimated that 8% of broiler deaths annually can be attributed to ascites, an economically important disease that has been challenging the industry for the past 2 decades. This dissertation consists of three research parts: 1) development of rapid detection methods for foodborne pathogens; 2) immune response of chicken cells against Salmonella and bacteriophage P22; 3) evaluation of novel control measures for poultry productions. In order to develop rapid and accurate detection methods for foodborne pathogens, two types of PCR assays were utilized. The purpose of this project was to create a video designed to educate the public about chicken egg storage and refrigeration. The information in the video was based on a research experiment performed by the author, which was designed to measure the chronological decline of internal egg quality over a ten-week period. By creating an educational video about the longevity of chicken eggs, it is hoped that consumers will learn to keep their eggs past their sell-by dates. This project includes an educational video as well as an outline for a research project.

Six experiments were conducted to evaluate the response of broilers fed canola meal. Experiment 1 assessed performance and carcass yields when broilers were fed various combinations of canola and soybean meal in nutritionally balanced diets based on digestible amino acid values. The results suggested that canola meal can be used in isocaloric diets as a partial replacement for Soybean meal.

There has been increasing concern on the emergence of multidrug-resistant foodborne pathogens from foods of animal origin, including poultry. The current study aimed to evaluate antibiotic-resistant Enterobacteriaceae from raw retail chicken/turkey parts (thigh, wings, breast, and ground) and beef meat (ground and chunks) in Middle Tennessee. Resistance of the collected Enterobacteriaceae to a panel of antibiotics was determined by the Kirby-Bauer disk diffusion test. Retail meats were also assayed for the presence of Salmonella spp. and Escherichia coli O157:H7. Bioenergetic mechanisms responsible for ATP production are essential in carrying out maintenance and cell-specific functions. In this study, hepatocytes (liver cells) were used to test both endogenous and exogenous stress on cellular respiration. The secondary lipid peroxide, 4-hydroxynonenal (HNE), was used because it alters bioenergetics by increasing mitochondrial proton leak that attenuates mitochondrial radical production and, therefore, endogenous oxidative stress. Three studies were conducted to evaluate how the broiler has changed over the years due to intense genetic selection. Four different broiler genotypes, one unselected since the 1950's (HER) representing the old meat-type bird and three current strains (Ross 308, Ross 708, and a test product (TY)) were studied. Differences in growth, morphometry, and response in performance and processing yield to different nutrient levels were studied.

Campylobacter spp. has been identified as one of the leading causative agents of food borne diarrheal illness. Epidemiological evidence has shown that poultry is the main source for human infection. Poultry are asymptomatic carriers of Campylobacter within their gastrointestinal tract, with colonization levels reaching 106-108 cfu/g cecal content. Surveys of domestic poultry flocks have estimated that approximately 90% of flocks are positive for Campylobacter colonization. Risk assessment studies have determined that by reducing levels of Campylobacter colonization during rearing, incidences of human infection will be significantly reduced. Currently there are no consistently effective treatments to eliminate Campylobacter from poultry flocks. …

In 1935, the National Poultry Improvement Plan (NPIP) was created to control Salmonella gallinarum and Salmonella pullorum . These two pathogens were devastating economically for poultry producers. Through cooperative efforts using vaccination and strict biosecurity, these two pathogens were eradicated from the United States. Currently, the NPIP program is targeting two other poultry pathogens, Salmonella enterica serovar Enteritidis and Mycoplasma . In the broiler industry it targets 2 specific Mycoplasma species (synoviae, gallisepticum). Vaccinations for these bacteria are available, but are not fully effective at controlling all strains and serovars. Thus, constant monitoring systems and strict biosecurity measures are necessary …

A study was conducted to examine broiler performance and carcass yield across 5 consecutive commercial broiler flocks after the removal of roxarsone (ROX) and growth-promoting antibiotics (GPA) from the feed. Over a 1-yr period, approximately 552,000 broilers were reared in 4 solid-walled, tunnel-ventilated houses, divided into 2 paired-house facilities, and were assigned 1 of 2 dietary treatments. The treated group received basal diets containing salinomycin, ROX, and GPA, whereas the control group received the same diets without ROX and GPA. This research was conducted with broilers to investigate the interactive effects of P and Ca on P requirement, digestibility, utilization, and phytase efficacy. Each treatment had a minimum of 6 replications. One experiment with 3 trials was conducted to identify the nonphytate P (nPP) requirement of 0 to 14 d old broilers. The nPP levels fed ranged from 0.30 to 0.60% with a Ca:nPP of 2.22:1 or 1.9:1. Requirement estimates for 0 to 14 d ranged from 0.52 to 0.60% nPP. The purpose of this research was to determine if increasing levels of RBC would affect growth performance and carcass characteristics of finishing pigs and growth performance of broilers. Three experiments were conducted to determine the effect of incremental levels of red blood cells (RBC; 0 to 4% and 0 to 2%), and thus increasing levels of dietary Leu on growth performance and linear carcass measurements of finishing pigs. Our results suggest that feeding 3 or 4% RBC causes a decrease in growth performance. However, feeding 1 or 2% RBC in the diets of finishing pigs had no detrimental effects on …

In western Algeria, 251 Escherichia coli isolates were recovered from broilers with clinical signs and lesions of colibacillosis. Serotyping showed that 82% of the isolates belong to one of the following serotypes: O78, O2, and O1. Antibiograms revealed a high level of resistance to oxytetracycline (82%), amoxicilline and ampicilline (47%), trimethoprim-sulfmethoxazole (42%), oxolinic acid and flumequine (31%). However, only a low percentage of strains were resistant to enrofloxacine (6%) and colistine (3%). Although multiresistance was common, low association was found between the serotype and antibioresistance.

The efficacy of killed vaccines generally is not equal to live vaccines. However, due to safety and ease of production, they remain a vital part of controlling and preventing diseases. In this study, the immune response to four different vaccination preparation techniques for the agent of bordetellosis of turkeys, Bordetella avium (BA), was compared. Preparation/inactivation techniques included (1) formalin inactivation, (2) opsonization of formalin-inactivated BA, (3) buffered acetic-acid BA inactivation, or (4) opsonization of buffered acetic-acid-inactivated BA. Respiratory diseases are very common in poultry. Bordetella avium and Ornithobacterium rhinotracheale are among the major microorganisms responsible for these diseases. The study was conducted to investigate the seroprevalence of O. rhinotracheale and B. avium in Aydın. Two hundred seventy-six chicken and 360 turkey serum samples were collected from poultry with respiratory disease suspicion in poultry enterprises around the city and samples were then examined for the presence of B. avium and O. rhinotracheale by commercial ELISA kits. The aims of this study were the isolation of Ornithobacterium rhinotracheale from poultry and serotyping of the isolates and field sera by agar gel precipitation (AGP) test. For the isolation 257 chickens and 214 turkeys were used and for the AGP test 333 chickens and 250 turkeys were used. O. rhinotracheale was isolated from the sinus discharge of 3 broiler breeders at 37, 42 and 46 weeks of age. An experiment was conducted to assess daily egg production and Se deposition in eggs of hens fed diets supplemented with inorganic or organic Se. This research was conducted to determine the non-phosphorus effects of phytase in diets for swine and poultry. An experiment was conducted to determine the effect of phytase addition on energy availability and protein and fat deposition in growing pigs. Results from this experiment indicated that phytase had small positive effects on energy availability, protein deposition, and fat deposition. The possible use of a genus-specific polymerase chain reaction (PCR) for the identification of Salmonella colonies was tested on 14 reference strains and 38 clinical Salmonella strains isolated from chickens by the standard method of the National Poultry Improvement Plan and Auxiliary Provisions (NPIP), USDA, U.S.A. All the PCRs using primer pairs InvA1 and InvA2 (5) gave 281 bp amplification product specific for the Salmonella genus and produced results identical (100%) to those obtained with biochemical and serological methods.
