Abstract

Ionoregulation and nitrogenous waste (N-waste) metabolism in fishes has been extensively documented in teleosts and elasmobranchs, but less so in lampreys (Petromizontiformes). All lamprey species undergo metamorphosis which drastically changes their physiology and modes of feeding. However, differences in ionoregulatory mechanisms due to anadromous versus non anadromous life history strategies emerge following metamorphosis in different lamprey species. Furthermore, some species, such as anadromous juvenile sea lamprey (Petromyzon marinus), are parasitic and migrate to seawater (SW) to ingest blood protein from host fishes, while juveniles of freshwater (FW), non-parasitic species, such as the American brook lamprey …

Invertebrate drift is a key process that potentially affects multiple levels of food web organization within stream environments. However, our understanding of the mechanistic drivers of drift in high latitude streams and subsequent bottom-up control that drift may have on fish predators in these environments remains understudied. The parasitic and invasive land-locked sea lamprey Petromyzon marinus populations contributed to the collapse of the Great Lakes fisheries in the mid-1900s, while native anadromous populations are currently under threat and require conservation efforts for protection. Despite the increasing concern for both populations of this species, the juvenile stage of the sea lamprey’s life cycle has not been well studied. This is due, in part, to ethical concerns raised around holding live host fish with juveniles and the lack of an alternative feeding method to maintain these juveniles in the laboratory. Parasitic sea lamprey (Petromyzon marinus) ingest large quantities of blood from fishes using their oral disc and rasping tongue, most often killing the host. In the early 1900s, sea lamprey invaded the Laurentian Great Lakes, decimating sport, commercial and culturally significant fisheries. Since the early 1960s, chemical control using the lampricides 3- trifluoromethyl-4-nitrophenol (TFM) and niclosamide have helped to reduce sea lamprey populations by 90%. Lampricides are applied to larval lamprey nursery streams targeting many generations of lamprey at once. Biological pigments, also called biochromes, are coloured compounds which are displayed by a variety of life forms, including animals, due to selective colour absorption. The combination of light absorption and reflection enables each pigment to portray a distinct colour which results in the broad spectrum of colours we observe in our surroundings. Carotenoids are a large group of yellow, orange, and red biological pigments found in living organisms. Our current biomolecular knowledge of carotenoids is heavily derived from studying the pathway in photosynthetic prokaryotes, bacteria, fungi, and plants. The electron transport systems in mitochondria of many organisms contain alternative respiratory enzymes distinct from those of the canonical respiratory system depicted in textbooks. Two of these enzymes, the alternative NADH dehydrogenase and the alternative oxidase, were of interest to a limited circle of researchers until they were envisioned as gene therapy tools for mitochondrial disease treatment. Recently, these enzymes were discovered in several animals. Here, we analyse the functioning of alternative NADH dehydrogenases and oxidases in different organisms. In addition to the typical electron transport system in animal mitochondria responsible for oxidative phosphorylation, some species possess an alternative oxidase (AOX) pathway, which causes electrons to bypass proton pumping complexes. Although AOX appears to be energetically wasteful, studies have revealed its wide taxonomic distribution, and indicate it plays a role in environmental stress tolerance. AOX discovery in animals is recent, and further research into its expression, regulation, and physiological role has been impeded by the lack of an experimental model organism. The fruit fly Drosophila melanogaster has a long history as a research model for studies on behaviour and group dynamics. When individuals are grouped in an environment and resources decrease, then they may behave co-operatively with one another or antagonistically compete. Hamilton’s Law states that if the benefit of helping a related individual out-weighs the cost to its direct fitness, then the individual’s indirect fitness will increase when they help their kin compared to helping an unrelated stranger. It is well known that stomach acid secretion by oxynticopeptic cells of the gastric mucosa is accomplished by the H+/ K+-ATPase (HKA), which is comprised of the HKα1 (gene: atp4a) and HKβ (gene: atp4b) subunits. Lamprey (Petromyzontiformes) are a phylogenetically ancient group of jawless fishes that begin their lives as filter-feeding larvae (ammocoetes) before undergoing a complex metamorphosis into juvenile lamprey that involves major changes to their internal and external body plan. The lampricide 3-trifluoromethyl-4-nitrophenol (TFM) has been used for decades to control invasive sea lamprey (Petromyzon marinus) populations in the Great Lakes, normally with little harm to other fish populations. However, due to the nature of lampricide treatments and the chemical properties of TFM, adverse effects to non-target fishes and non-target mortality occasionally occur. In fishes, hyperammonemia may occur following feeding or exposure to abnormally high concentrations of environmental ammonia due to sewage effluents, agricultural run-off and in crowded aquaculture pens. Increased internal ammonia can result in hyperactivity, convulsions, coma and death. In mammals, it is also associated with potentially fatal brain edema, in which the accumulation of intracellular water results in swelling, increased intracranical pressure and herniation leading to death. Fishes living in freshwater need to actively compensate for the diffusive loss of ions and osmotic gain of water. The gill is the primary organ of ion regulation and contains an array of ion transport proteins to help maintain homeostasis. Two of the more well studied ion pumps are the Na+/K+-ATPase (NKA) and vacuolar type proton ATPase (V-ATPase). This thesis focuses on another ion pump known as the non-gastric H+/K+-ATPase (ngHKA). The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), is widely used in the Great Lakes to control invasive sea lampreys (Petromyzon marinus) populations, but much about its sub-lethal effects remains unknown. A better understanding of its toxicity is needed to improve TFM effectiveness and to protect non-target organisms from its potential adverse effects. The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), has been highly successful in the control of sea lamprey (Petromyzon marinus) populations in the Great Lakes. Treatments with TFM involve applying it to streams, where it targets larval sea lamprey which live burrowed in the stream substrate. While the toxic mechanism of TFM has been elucidated, and its effects on sea lamprey described, its effects on non-target fish species such as rainbow trout (Oncorhynchus mykiss) and lake sturgeon (Acipenser fulvescens) are not as well understood. There has been recent discussion within the field of environmental toxicology as to the viability of using the bioaccumulation of a chemical toxicant to predict toxic effects. Current literature has shown this concept to carry some merit, although there is a major flaw in that with many cases, namely those involving sensitive species, body burdens cannot be reliably linked to effects (Adams et al. The primary method of controlling invasive sea lamprey (Petromyzon marinus) populations in the Great Lakes is the application of the pesticide 3-trifluoromethyl-4-nitrophenol (TFM) to larval lamprey nursery streams. Recent evidence suggests that the mode of toxic action of TFM involves the inhibition of oxidative ATP production in the mitochondria. As a result, there is increased reliance on the anaerobic pathways of ATP production which involves the hydrolysis of phosphocreatine (PCr) and anaerobic glycolysis. Aquatic organisms respond to metal exposures by modulating uptake, elimination and detoxification capacity and, therefore, it is difficult to link accumulation within fish to adverse chronic effects. When fish are exposed to sublethal concentrations of metal, physiological responses follow a damage-repair scenario with rapid uptake into the gills followed by transfer via the circulatory system and accumulation into key organs such as the liver and kidney. A particularly striking example is that of the relationships between testosterone, social behavior, and social context. The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), has been extensively used over the last 50 years to control invasive sea lamprey (P. marinus) populations in the Great Lakes, but its mechanism of toxicity is unresolved. Due to its structural similarity to phenolic compounds known to inhibit mitochondrial ATP production, one hypothesis on the mode of action of TFM is that it impairs mitochondrial oxidative phosphorylation. A second hypothesis is that TFM targets the gill, interfering with gill ion uptake. This dissertation aims to understand the history of, and possible alternatives to, managerial responses to socio-ecological issues by examining one of the largest natural resource management failures of the twentieth century—the collapse of the Northern cod fisheries off Newfoundland and Labrador, Canada. In 1992, the Northern cod fishery off Newfoundland and Labrador (the world's largest ground fishery) was shut down. The Northern cod had been reduced to 1% of their historic spawning biomass and cod fishing as a way of life had come to an end after a 500 year history. The intent of this study is to examine the present institutional arrangements of the Newfoundland and Labrador Northern shrimp fishery, to identify the roles of cooperative fisheries economic development corporations and make recommendations for institutional improvements. There are a number of institutional and organizational arrangements more favorable for fisheries management and development than present ones (Bishop, 1981; Bromley, 1982; McCay and Acheson, 1987; Pinkerton, 1989).
