Genetic Determinants Associated With Persistence Of Listeria Species And Background Microflora From A Dairy Processing Environment, Vaishali Poswal, Sanjeev Anand, Jose L. Gonzalez-Hernandez, Brian Kraus

Listeria monocytogenes is a persistent foodborne pathogen capable of surviving in food processing environments, often in association with diverse environmental microflora. This study examines genomic determinants of persistence, specifically stress adaptation and biofilm-associated traits, in environmental Listeria species and other environmental microflora from a dairy processing facility by analyzing whole-genome sequences of 6 environmental Listeria isolates, 4 ATCC reference strains, and 22 air and floor swab cultures, annotated using the RAST platform. Subsystem analysis revealed that Listeria isolates carried a defined set of genes linked to biofilm formation, antimicrobial resistance, and stress response, though in lower abundance than environmental cultures. …

The continued expansion of the American Plains Bison (Bison bison) industry has increased the need for science-based nutritional recommendations tailored specifically to the species, due to limited species-specific research. Because trace minerals influence immune function, growth, and reproductive performance, establishing baseline mineral status and evaluating responses to supplementation are critical for advancing bison production and conservation efforts. This thesis integrates two complementary investigations to evaluate trace mineral status in American Plains bison at both regional and herd management levels. Winter-Feeding High Forage Vs. High Concentrate Diets To Beef Cows: Effects On Placental Transcriptomics, Lactation Capacity, And Enteric Methane Emissions, Megan Ann Wehrbein

Nutritional management and lactation performance are critical factors influencing productivity in cattle. This research includes two complementary studies. The first study was a meta-analysis to develop a lactation curve for Bos taurus cattle in North America using data from machine or hand milked cows. Data from 7 studies were used resulting in 1340 cow-day observations. Milk yield was analyzed by parity (primiparous or multiparous) and feeding management (non-nutrient restricted or nutrient restricted), revealing that primiparous cattle peaked earlier and at lower yields than multiparous cattle, and nutrient restriction reduced peak milk yield. Riverine systems are highly dynamic, and changes in flow conditions can impact the population dynamics of fishes. Increased flows from historical averages in eastern South Dakota rivers have likely impacted fish populations. The goals of this study were to 1) better understand the changes in resource use by Blue Suckers Cycleptus elongatus through retrospective eye lens stable isotope analysis, 2) determine the effects of record flooding in the James River on complex woody habitat and Flathead Catfish Pylodictis olivaris population structure, and 3) assess the influence of river discharge and temperature on Channel Catfish Ictalurus punctatus and Flathead Catfish condition …

Enhancing The Biopreservative Effect Of Non-Starter Lactic Acid Bacteria Using Soluble Fiber During Cheese Ripening, Rakesh Kaushik, Sanjeev Anand

Cheddar cheese ripening creates favorable conditions for desired microbial changes but also allows survival and outgrowth of spores like Clostridium tyrobutyricum and Bacillus licheniformis, leading to late-blowing defects. In the first phase of the study, NSLAB dynamics were evaluated in the presence of spores, where pilot-scale cheeses (110 L) were produced in four treatments: control, T1 (BL), T2 (CT), and T3 (BL+CT), each inoculated at 2.0 Log10 CFU/mL with spores. Results showed that SLAB declined from 8.0 to 0.2 Log10 CFU/g, while NSLAB increased from 2.0 to 8.5 Log10 CFU/g by the third month and remained stable thereafter. Spore counts …

This meta-analysis aimed to evaluate the efficacy of automated activity monitoring (AAM) in detecting estrous expression and ovulatory status in cows during the voluntary waiting period (VWP). A comprehensive literature search was conducted in PubMed, ScienceDirect, and Google Scholar using specific search terms. Inclusion criteria focused on studies that assessed estrous expression within the VWP using modern AAM systems alongside blood progesterone (P4) measurements. Four manuscripts involving 2,198 cows were included. Data extraction was performed by a single investigator and validated by a coauthor. The analysis considered estrous expression and ovulatory status determined through serial blood P4 measurements. Cows were …

This study evaluated the effectiveness of individual clean-in-place (CIP) steps in removing biofilms from reverse osmosis (RO) membranes under dynamic flow conditions using the Centers for Disease Control (CDC) biofilm reactor. Biofilms were developed in the laboratory under continuous flow, using mixed-species bacterial isolates obtained from 10-month-old RO membrane biofilms from a commercial facility. Individual CIP chemicals, representative of those used in commercial protocols, were tested against 24 h-old biofilms. Additionally, a complete six-step sequential CIP process was conducted under dynamic conditions, consisting of treatments with alkali, surfactant, acid, enzyme, a secondary surfactant, and sanitizer. Cheese ripening involves microbial changes, with starter lactic acid bacteria (SLAB) initiat- ing fermentation and nonstarter lactic acid bacteria (NSLAB) driving flavor and texture development. However, heat-resistant spores of Clostridium and Bacillus can survive pas- teurization and cause spoilage during ripening. This study evaluated NSLAB dynamics in the presence of spores during cheese ripening. Cheddar cheese samples at pilot-scale level (110 L) with four treatments, namely control, with spores of B. licheniformis (T1), with spores of Cl. tyrobutyricum (T2), and both spores (T3) at 2.0 Log10 CFU/mL, were ripened at 7 ◦C for six months. Characterizing Environmental Background Microflora And Assessing Their Influence On Listeria Persistence In Dairy Processing Environment, Vaishali Poswal, Sanjeev Anand, Brian Kraus

Listeria monocytogenes (Lm) may persist in food processing environments (FPEs) alongside diverse background microflora. While microbial communities in FPEs can in- fluence Lm survival, their role in supporting or suppressing its growth remains unclear. This study aimed to characterize the microflora in floor swabs and air samples collected from a dairy processing facility across three seasons and assess their potential impact on the growth of a Lm test strain previously isolated from a dairy processing environment. A total of 167 environmental isolates, representing 30 bacterial genera, were identified. Pseudomonas was consistently prevalent across all sample types. Seasonal shifts in bacterial …

Maintaining optimal fertility in dairy cattle herds is a global challenge that is typically addressed through the genetic selection of fertility indicator traits. However, many of the traits currently implemented in breeding programs are heavily influenced by environmental factors, resulting in a slow rate of genetic improvement. Anogenital distance (AGD) has recently emerged as a promising fertility indicator trait due to its association with favorable reproductive outcomes and its higher heritability estimates compared with currently evaluated traits. Pre-weaning mortality (PWM) remains a significant challenge in modern swine production systems, particularly in the context of hyper prolific sows. Although genetic selection has successfully increased litter sizes, these advancements have also intensified physiological stress on sows and increased the vulnerability of neonatal piglets. The majority of PWM occurs within the first 72 hours postpartum, with crushing by the sow as the leading cause of death. Additionally, iron deficiency anemia remains one of the most widespread nutritional disorders in piglets, further compromising survival, immune function, and growth. Invasive species disrupt ecosystems by altering habitats, competing with native species, and disrupting food webs. The northern Great Plains is an ecologically important region with many wetlands, lakes, and rivers that are under threat from two invasive species, Bighead (Hypophthalmichthys nobilis) and Silver Carp (H. molitrix; bigheaded carp). These large planktivores change the abundance and composition of phytoplankton and zooplankton communities which alters prey for native mussels, fishes, and, potentially, waterfowl. Bigheaded carps have invaded a portion of the Northern Great Plains, and preventing further spread is a primary management goal. My work assists these efforts by providing tools to …

This Review aims to provide an overview of the bovine estrous cycle and pregnancy establishment, beginning with follicular development, luteolysis, maternal recognition of pregnancy and early embryonic development. Manipulating the estrus cycle with synchronization protocols is a critical aspect of the beef industry and has greatly improved the efficiency of artificial insemination. This review will evaluate various strategies to synchronize estrus from the very basic to more advanced synchronization protocols. The objective of this study was to evaluate the influence of maternal nutrient restriction during mid-gestation on the performance of the dam, feeding performance, carcass composition, and genetic expression in the offspring. Cows (n = 46) were randomly assigned to one of two treatments based on global nutrient intake: 1) fed to achieve and/or maintain a BCS of 5 or 6 (Positive Energy Status [PES]; n = 24) or 2) fed to lose one BCS during 99 days of mid-gestation (Negative Energy Status [NES]; n = 22). Cows in NES had greater decreases (P < 0.05) in body fat stores and muscle mass, elevated NEFA concentrations, and displayed decreased (P < 0.05) DMI, HP, and GHG emissions.
