Many studies have been conducted on Notophthalmus viridescens viridescens but none have occurred in West Virginia. There are variations in the life history of newts depending on location. In this study, I studied a population of newts in Wayne County, West Virginia from February 1996 to June 1997. I examined population structure, seasonal activity, and reproductive biology.

The population structure can be divided into four classes: egg, larvae, eft, and adult. Eggs at Shoals were deposited in mid-May and larvae were present from mid-June to early September. Larvae began to transform at an approximate total length of 32.8 mm. Red …

Prior to 1988, two subspecies of the mosquitofish Gambusia affinis (G. a. affinis and G. a. holbrooki) were recognized along the Gulf and Atlantic Coasts of the United States. Based on electrophoretic analysis, subsequent investigators recognized G. holbrooki and G. affinis as separate species. Populations in drainages west of Mobile Bay were considered G. affinis, and those east of this divide being G. holbrooki. The only mosquitofish population in West Virginia is found in the Meadow River wetlands (1392 ha). This wetland complex is located at the western end of Greenbrier County in the Allegheny Mountain Province. …

The grass pickerel, Esox americanus vermiculatus, is listed as Undetermined on the Vertebrate Species List of Concern in West Virginia. The central mudminnow, Umbra limi, is a disjunct population in Green Bottom Swamp. The lentic, vegetated areas that are required for spawning for the grass pickerel and the central mudminnow have been reduced by residential, agricultural, and industrial developments. Green Bottom Swamp, a naturally occurring wetland of 58 ha, and a nearby mitigated wetland of 29 ha, provides spawning habitat for the grass pickerel and the mudminnow. Various fungal products have immunomodulating activity and some have been studied regarding prevention of transplantation rejection. Prior to this investigation, the mycotoxin, gliotoxin (GT), has never been investigated as an immunotherapeutic drug for autoimmune disease. GT is a fungal secondary metabolite and a member of the epipolythiodioxopiperazine (ETP) family which has been shown to inhibit phagocytosis, induction of cytolytic T cells and the proliferation of T cells following mitogen stimulation. GT also induces in vitro apoptosis in certain immune cell types. More importantly, GT exhibits selective activity towards cells of hemopoietic origin.

We evaluated the rate of fatty acid and triglyceride synthesis and storage content in red skeletal muscles, liver, inguinal fat, and retroperitoneal fat of the LA/N-fa^k (“corpulent”) and Zucker fa obese rat strains. Green Bottom Wildlife Management Area (GBWMA), Cabell County, West Virginia, is a wetlands system adjacent to the Ohio River composed of wetlands varying in age from 4 to 60 years. This system is annually flooded by the Ohio River, yet insect communities remain unique to each vegetation type or microhabitat. Nine vegetative communities from two wetlands were sampled and compared using chi-square analysis to determine if the insect communities were unique. A total of 34,975 specimens, representing 23 families, were collected over ten sample intervals. A study was conducted on Desmognathus quadramaculatus in the northern periphery of its range with some aspects of its life history, surface density, and seasonal foraging habits. The results in this study are compared to studies done on D. quadramaculatus in the southern portion of its range. Female D. quadramaculatus have been found tending egg clutches from June to September in the southern portion of its range and data collected in this study shows that egg deposition also occurs during this period in the northern population. Studies were conducted on the comparative microhabitat requirements and feeling habits of the ravine salamander, Plethodon richmondi and the redback salamander, P. cinereus in northern West Virginia. Plethodon richmondi had a mean SVL of 49.7 ± 7.1 mm and were significantly larger than P. cinereus which had a mean SVL of 37.2 ± 8.0 mm. Both species had biennial egg laying patterns and deposited eggs in late spring or early summer. Juvenile P. richmondi were not surface active except for the early spring and neonates did not become surface active until the ensuing spring; conversely, P. cinereus juveniles exhibited more …

The (3)H-tetracycline bone labeling procedure was employed to monitor bone resorption from urinary loss curves in male and female rats of various ages. In addition, whole body dry skeletal mass and the loss of (3)H-tetracycline from individual bones was determined. It was found that the dry skeletal mass/body mass ratio of 24-week-old females was 30-40% greater than that for males. The urinary loss of (3)H-tetracycline was described by a double exponential equation indicating the presence of two distinct and independent exchangeable bone compartments. This differentiation is accompanied by an increase in protein kinase Ca (PKCα) protein level and selective enrichment in nuclear-associated PKCα. PKC is thought to regulate gene expression through the TPA response element (TRE). This element is specifically recognized by the AP-1 transcription factor composed of jun and fos family members. The pigment cell-specific expression of tyrosinase and TRP1 has been shown to be important for the production of melanin in pigmented cells. Using a pigmented cell line, B16 mouse melanoma, we obtained evidence that PKC plays a major role in regulating melanogenesis. Chronic treatment with phorbol dibutyrate (PDBu) leads to downregulation of PKC activity and protein levels. This is accompanied by a loss of pigmentation which is correlated with a 50% reduction and a complete loss of TRP1 and tyrosinase respectively. Similar results were obtained with Northern and Western blotting indicating that PKC may regulate the steady state levels of …

The blacknose dace, Rhinichthys atratulus, is a cyprinid minnow common to shallow streams throughout North America. It has been shown to be sensitive to anthropogenic stress and a study of the genetic nature of R. atratulus populations may yield information about their habitats. In West Virginia, two subspecies of R. atratulus are present, R. a. atratulus and R. a. obtusus, which are separated by the Allegheny divide. Postnatal retinal development was studied in a marsupial opossum, Monodelphis domestica using light microscopy and 3H-thymidine autoradiography. For the light microscopic study, opossum neonates at 1, 3, 7, 14, 21, 25 and 28 days of age were euthanized, fixed and processed into paraffin sections for hematoxylin and eosin staining. The distinct ganglion cell layer, first observed on postnatal day (P) 7, was separated from the outer neuroblasts by the inner plexiform layer. A survey of the soricid (shrew) fauna of southern West Virginia was conducted between May 16, 1994 and May 27, 1995. Twenty-five sites were established in Mercer and Summers counties within seven habitat types. Pitfall traps were used and through a limitation of fluid depth within the trap the selectivity for soricids was increased to over 83% of the total capture. Geographic and nongeographic variation was assessed on 12 cranial and 4 external characters in approximately 400 specimens of Microtus (M. ochrogaster, M. chrotorrhinus, M. pinetorum, M. pennsylvanicus) in West Virginia. Univariate and multivariate statistical analyses were used to examine age-related variation, sexual dimorphism, elevation and geographic variation and litter size for each species of Microtus in the state. Cranial characters were more beneficial than external in detecting variation. Utilization of wood duck (Aix sponsa, L.) nest boxes was evaluated at the Green Bottom Wildlife Management Area, a 1,110-acre mid-Ohio Valley floodplain. Primary focus was directed upon nest box utilization by the wood duck but use by other wildlife was observed and recorded. Utilization of the nest boxes by the wood duck was analyzed with respect to plant community type, orientation and juxtaposition to other wood duck nest boxes. Desmognathus ochrophaeus, the mountain dusky salamander, is a medium-sized Desmognathine, which inhabits moist woodlands. It is a slender species with a well rounded, slim and tapering tail (Bishop 1967), and 14 costal grooves (Green and Pauley 1987). The average total length is between 70 mm and 100 mm with the tail accounting for approximately half this length (Conant and Collins, 1991). An ecological study of the wood turtle, Clemmys insculpta (LeConte), was conducted in West Virginia from May 1991 to March 1993. Population structure at the primary study area (WV #1) was estimated to be 46% juvenile (H-8 yrs. old), 26% female and 28% male. Total population estimate for WV #1 was 337 wood turtles, with an aquatic density of 1,980 turtles per Hectare of river and a terrestrial density of 22.5 turtles per ha of terrestrial habitat. The bowfin, Amia calva Linnaeus, is the only extant species of the family Amiidae. The Green Bottom Wildlife Management Area (GBWMA) is the home of the only known reproducing bowfin population in the state of West Virginia. The GBWMA (38°35' 35" N, 82°14' 55" W) is located along the Ohio River 26 km northeast of Huntington, West Virginia. A study was conducted to determine various aspects of the ecology of Plethodon kentucki in West Virginia. Results of studies on range and distribution revealed that P. kentucki is limited to southwestern counties in West Virginia. A population of P. kentucki at Beech Fork State Park was extremely seasonal with regard to activity. Seasonal activity was not significantly correlated with air temperature, soil temperature, air relative humidity, or soil pH. Seasonal activity was significantly correlated with soil moisture. Much has been written about the ecology of the green salamander, Aneides aeneus (Cope and Packard). The life history and ecology of southern populations of this species have been studied intensively by Gordon (1952), Woods (1968), and Snyder (1971). Gordon collected data in Highlands, North Carolina on distribution, habitat selection, population dynamics, and the ability of aeneus to withstand water loss as compared to Plethodon jordani melaventris. Plethodon cinereus is found throughout the state except for part of the Allegheny Plateau Province where P. richmondi occurs. A survey of the adult Leptoceridae of West Virginia revealed 27 species, 25 of which were state records. The most abundant genus in the state was Ceraclea which had ten different species. Triaenodes had six different species, while Oecetis had five. The most common species in the state included C. cancellatus, C. maculate, O. avara, and O. inconspicua. Species that were abundant only in the larger rivers of the western portion of the state included C. flava, C. maculata, Nectopsyche pavida, and T. ignita. A study of the reproductive and early developmental stages was the primary objective. A description of the morphology, a shell aberration, a study of its ability to estivate, a correlation between sex types in lentic and lotic environments and explanations for its expansion to West Virginia are included as well. Effects of pituitary stalk-transection on plasma concentrations of luteinizing hormone (LH), follicle stimulating hormone (FSH) prolactin (PRL) and progesterone were investigated during the estrous cycle of ewes. Pituitary stalk (SS) or sham (SH) transection was performed on day 1 (estrus = day 0) of the estrous cycle. A Teflon or Silastic barrier was placed between the cut ends of the stalk to prevent reorganization of the portal vasculature. Aspidogaster conchicola and Cotylaspis insignis were only two aspidogastrid trematodes recovered in a year long study involving 500 bivalves (22 species) collected from a thirteen county area in western West Virginia. Also collected were three species of aquatic mites identified as: Union­ icola fossulata, Unionicola formosa, and Najadicola ingens. High elevation, low elevation and cave populations of Plethodon wehrlei (Fowler and Dunn) in West Virginia were compared to determine the influence of habitat variation on morphology and physiology. Discriminant analysis of 14 external morphological characters and 11 skeletal characters revealed a similar morphology for the three populations. Comparisons of critical thermal maxima showed a positive correlation to habitat temperature. Dehydration studies revealed that the high elevation population was more resistant and tolerant to water loss than the cave and low elevation populations.

A survey of the adult Hydropsychiclae revealed 39 species, 14 of which were state records. The adult hydropsychids were collected from various locations throughout West Virginia. The most common species Hydropsychidae were Hydropsyche morosa, H. sparna, Cheumatopsyche analis, C. campyla, and Diplectrona modesta. Most of the species did not appear to be limited to any one particular drainage basin. Several species exhibited a preference for stream order (headwaters, middlereaches, large rivers). Aphropsyche monticola, Parapsyche apicalis, Cheumatopsvche wrighti, C, gyra, and Hydropsyche alhedra preferred the fast-flowing headwater streams in the higher elevations of the state. A survey of the Unionacea of Twelvepole Creek, West Virginia was conducted coupled with a qualitative study of commensal algae found on selected mussel species. The naiads and the algae were found to have diverse populations. Naiads collected during the survey were represented by 23 species, 16 genera and 2 families. Corbicula fluminea was found to be confined largely to the main branch of Twelvepole, with no individuals being collected from the East or West Forks and only limited collections near the mouth of Beechfork Creek.
