Dollar sunfish (Lepomis marginatus) at the Savannah River Site were sampled from two radiocesium contaminated ponds with a history of thermal elevation and from reference ponds with no history of radioactive or thermal contamination. Fluorescent sequencing of a portion of the mitochondrial control region showed no genetic variation within or between populations. Centrarchidae microsatellite loci Lma 120 (6 alleles, N=124 from 5 populations), Lma 20 (10 alleles, N=37 from 3 populations) and Rb 7 (14 alleles. N=17 from 2 populations) were variable for dollar sunfish. An additional 9 microsatellite primer pairs were designed for dollar sunfish. Testing of the designed …

Elephants from the tropical forests of Africa are morphologically distinct from savannah or bush elephants. Dart-biopsy samples from 195 free-ranging African elephants in 21 populations were examined for DNA sequence variation in four nuclear genes (1732 base pairs). Phylogenetic distinctions between African forest elephant and savannah elephant populations corresponded to 58% of the difference in the same genes between elephant genera Loxodonta (African) and Elephas (Asian). Large genetic distance, multiple genetically fixed nucleotide site differences, morphological and habitat distinctions, and extremely limited hybridization of gene flow between forest and savannah elephants support the recognition and conservation management of two African …

A Cytochrome-B Perspective On Passerina Bunting Relationships, John Klicka, Adam J. Fry, Robert M. Zink, Christopher W. Thompson

We sequenced the complete mitochondrial cytochrome-b gene (1,143 nucleotides) for representatives of each species in the cardinalid genera Passerina (6 species), Guiraca (1 species), and Cyanocompsa (3 species), and used a variety of phylogenetic methods to address relationships within and among genera. We determined that Passerina, as presently recognized, is paraphyletic. Lazuli Bunting (P. amoena) is sister to the much larger Blue Grosbeak (Guiraca caerulea). Indigo Bunting (P. cyanea) and Lazuli Bunting are not sister taxa as generally thought. In all weighted parsimony trees and for the gamma-corrected HKY tree, Indigo Bunting is …

The Taxonomic Rank Of Spizella Taverneri: A Response To Mayr And Johnson, John Klicka, Robert M. Zink, Jon C. Barlow, W. Bruce Mcgillivray, Terry J. Doyle

Mayr and Johnson suggest that Spizella taverneri should be a subspecies of the biological species S. breweri, because it is possibly not reproductively isolated. We originally concluded that evidence from mitochondrial DNA sequences, habitat preferences, timing of breeding, vocalizations, and morphology supported the recognition of S. taverneri as a phylogenetic and biological species. Nothing in the commentary by Mayr and Johnson causes us to change that conclusion. We believe that it is probable that these two allopatric taxa are isolated. Contrary to Mayr and Johnson, we believe that more information is given by ranking S. taverneri as a species, because …

Canine parvovirus (CPV) enters and infects cells by a dynamin-dependent, clathrin-mediated endocytic pathway, and viral capsids colocalize with transferrin in perinuclear vesicles of cells shortly after entry (J. S. L. Parker and C. R. Parrish, J. Virol. 74:1919–1930, 2000). Here we report that CPV and feline panleukopenia virus (FPV), a closely related parvovirus, bind to the human and feline transferrin receptors (TfRs) and use these receptors to enter and infect cells. Capsids did not detectably bind or enter quail QT35 cells or a Chinese hamster ovary (CHO) cell-derived cell line that lacks any TfR (TRVb cells). However, capsids bound and …

Analysis of the genetic variation of an endangered population is an important component for the success of conservation. Animals from two local Romanian pig breeds, the Mangalitsa and Bazna, were analyzed for variation at a number of genetic loci using PCR-based DNA tests. Polymorphism was assessed at loci which 1) are known to cause phenotypic variation, 2) are potentially involved in trait differences or 3) are putative candidate genes. The traits considered are disease resistance, growth, coat color, meat quality and prolificacy. During a visit to Isla Guadalupe from 31 May to 3 June 1996, we documented three species new to the island, the Barn Owl, Swainson's Thrush, and Hooded Oriole, and established first breeding records for the European Starling and Western Meadowlark. Red-shafted Flickers are now breeding on the island, representing a recent recolonization from the mainland following the extinction of the endemic population. We investigated the validity of Colaptes auratus rufipileus and concluded that it doesn’t meet the standard for phylogenetic species but differs from C. a. collaris at the 75% level usually associated with subspecific rank. Ordered gene maps of mammalian species are becoming increasingly valued in assigning gene variants to function in human and animal models, as well as recapitulating the natural history of genome organization. To extend this power to the domestic cat, a radiation hybrid (RH) map of the cat was constructed integrating 424 Type I-coding genes with 176 microsatellite markers, providing coverage over all 20 feline chromosomes. Alignment of parallel RH maps of human and cat reveal 100 conserved segments ordered (CSOs) between the species, nearly three times the number observed with reciprocal chromosome painting analyses. Historically, a paucity of comparative morphological characters has led to much debate regarding relationships within and among the major lineages of New World nine-primaried oscines. More recently, DNA-DNA hybridization studies have provided novel and testable hypotheses of relationships, although no consensus has been reached. For 40 songbird taxa, we obtained 1,929 base pairs (bp) of DNA sequence from the mitochondrial cytochrome-b (894 bp) and NADH dehydrogenase subunit 2 (1,035 bp) genes. Phylogenetic analyses confirm the monophyly of this assemblage as traditionally defined. Source of Primers. PCR primers were designed from the published sequence for porcine vitamin D-25-hydroxylase (CYP25) cDNA (GenBank accession no. Y16417). The position of the forward and reverse primers corresponds to 506 to 530 bp and 830 to 854 bp, respectively, of the cDNA. The primer pair amplified a 348-bp fragment of the cDNA and an 862-bp fragment from the pig genomic DNA. The 862-bp fragment (Gen- Bank accession no. The objective of this research was to determine whether plasma concentration of FSH was genetically correlated with ovulation rate and thus was a useful trait for indirect selection. Blood samples were collected from 619 animals from five lines of pigs. Line I was selected for increased index of ovulation rate and embryonal survival, and Line C was its randomly selected control. Pigs sampled from Lines I and C were from generations 12 and 13. Pigs from three additional lines that were derived from eighth-generation pigs of Lines I and C also were used. The January tetra (Hemigrammus hyanuary Durbin) exhibits two eye color phenotypes. These have a silver iris, which is characteristic of the species, and a green color variant. Segregation patterns observed in the progenies from 12 different crosses support an hypothesis for the monogenic inheritance of iris coloration in this species.

A comparative gene map of the horse genome composed of 127 loci was assembled based on the new assignment of 68 equine type I loci and on data published previously. PCR primers based on consensus gene sequences conserved across mammalian species were used to amplify markers for assigning 68 equine type I loci to 27 horse synteny groups established previously with a horse-mouse somatic cell hybrid panel (SCHP, UC Davis). This increased the number of coding genes mapped to the horse genome by over 2-fold and allowed refinements of the comparative mapping data available for this species. In conjunction with …

We examined patterns of variation in skeletal morphometrics (29 characters), allozymes (34 loci), mitochondrial DNA (mtDNA) restriction sites (n = 74) and fragments (n = 395), and mtDNA sequences (1,739 bp from cytochrome b, ND2, ND6, and the control region) among all species of Toxostoma. The phenetic pattern of variation in skeletal morphometrics generally matched traditional taxonomic groupings (based on plumage patterns) with the exceptions of T. redivivum, which because of its large size clusters outside of its proper evolutionary group (lecontei), and T. occelatum, which did not cluster with T. curvirostre. The Timberline Sparrow (Spizella taverneri), although originally described as a species, is currently classified as a subspecies of the more widespread Brewer's Sparrow (S. breweri). We investigated the taxonomic status and recent evolutionary history of these species by comparison of both morphological and molecular characters. Morphometric comparisons using 6 external and 18 skeletal measurements show that S. taverneri specimens from two widely separated populations (Yukon and southwestern Alberta, Canada) are indistinguishable with respect to size yet are significantly larger (by 3%) than representatives of several breweri populations. The aim of this study was to estimate genetic and phenotypic (coroariances between objective measures and carcass visual scores, as a test of the potential value afvisual scores in selection programmes to improve carcass composition in crossbred lambs. In each of 1986, 1987 and 1988, 22 Suffolk rams were chosen with either high or low scores on an index designed to increase lean growth rate. The purpose of this experiment was to determine if the estrogen receptor gene (ESR) could be used as a genetic marker for litter size in sheep. The estrogen receptor gene was chosen because it has been successfully used as a genetic marker for litter size in swine. In this study, DNA dilutions from two experimental flocks, totaling approximately 200 animals, were used. Amplification of exon 1/intron 1 of the ESR gene was performed using PCR. We report a phylogenetic analysis of primate malaria parasites based on the gene encoding the cytochrome b protein from the mitochondrial genome. We have studied 17 species of Plasmodium, including 14 parasitic in primates. In our analysis, four species were used for rooting the Plasmodium phylogenetic tree: two from closely related genera (Hepatocystis sp. and Haemoproteus columbae) and two other Apicomplexa (Toxoplasma gondii and Theileria parva). We found that primate malaria parasites form a monophyletic group, with the only exception being the Plasmodium falciparumPlasmodium reichenowi lineage. The exact nature of the mechanisms and the regulation of vascular smooth muscle contraction is not well understood. To better understand these processes, we examined two systems involved in smooth muscle contraction, the cytoskeleton and the protein kinases. In order to study the role of the cytoskeleton in smooth muscle contraction, we examined the contractile and mechanical effects of cytoskeleton disruption. We found that the relationship between passive tension applied to aortic rings and the resulting increase in tissue length was nearly linear over the range of 1 g to 15 g. In this study, isoelectric focusing is used to separate proteins of Rhinichthys cataractae and R. atratulus and conventional staining is used to identify alleles of six genes. The objective of this work is to determine whether these methods can effectively assay isozymes that may be used to establish the relationship between anthropogenic stress and genetic diversity. Alleles for LDH-A, LDH-B, EST-A, EST-B, EST-C, and EST-D loci were identified for two species of fish, R. atratulus and R. cataractae. An experiment was conducted to determine the effects of dietary protein intake after a period of weight loss on the wool components of staple strength for sheep with a history of low or high staple strength (18.0 vs 34 Nlktex). After being fed to lose 15% of their liveweight over 10 weeks, sheep within each staple strength group were assigned in equal numbers to either a low or high protein diet designed to re-gain initialliveweight in 8 weeks. The use of molecular techniques for the assessment of familial relationships among social species of mammals has become relatively commonplace. However, some species represent poor candidates for such studies due to naturally low levels of genetic diversity, leading to unacceptably large standard errors associated with estimates of relatedness. Here, we report on a preliminary study of genetic diversity within two populations of a social species of ground squirrel, Gunnison's prairie dog (Cynomys gunnisoni) using DNA fingerprinting. We observed low levels of diversity in the form of large mean coefficients of genetic similarity among individuals occupying the same population. …
