Genetic Analysis Of Methionine Restriction-Associated Mitochondrial Unfolded Protein Response Signaling., Josh D. Dodge Jan 2025

Genetic Analysis Of Methionine Restriction-Associated Mitochondrial Unfolded Protein Response Signaling., Josh D. Dodge

Biology Dissertations - Archive

The lifespan of an organism is dictated by various genetic and environmental factors. One such determinant is the health of mitochondria which are organelles that mediate various critical cell functions including a significant role in generating the vast supply of cellular energy. Indeed, the gradual decline of mitochondrial function is believed to be a hallmark and possible cause of aging. Paradoxically, mild mitochondrial dysfunction early in life extends lifespan in various model organisms. Cells use diverse approaches to promote mitochondrial homeostasis, including the transcriptional reprogramming of cells by mitochondrial unfolded protein response (UPRmt) to mitigate mitochondrial stress. Consistently, UPRmt activation …

Patterns Of Kinship And Sociality In North American River Otters, Whitney E. Vickers Jan 2025

Patterns Of Kinship And Sociality In North American River Otters, Whitney E. Vickers

Cal Poly Humboldt theses and projects

Previous studies of river otters (Lontra canadensis) in coastal Humboldt County, California have documented varying social group compositions over the last four decades. It has been assumed that otters travel between coastal wetland sites, but the extent of movement has been unknown. Socially, river otters have been observed alone, in small family groups that consisted of an otter mom and pup(s), in larger helper groups that consisted of an otter mom, pup(s), and at least one additional adult or juvenile (helpers), and in all-adult social groups. Until now it was unknown whether the composition of social groups was …

‘Who’S Your Daddy?’: Ddradseq Parentage In Broodstock-Offspring Pairs Of Diadema Antillarum In A Population Restoration Initiative, Bianca R. Rosendahl

Undergraduate Scholarship and Creative Works

In the early 1980s Diadema antillarum, or the long-spined urchin, was hit by a catastrophic mass mortality event across the western Atlantic and Gulf of Mexico, inclusive of the Florida Keys. The cause of this die-off event was likely triggered by a pathogen or environmental stressor that decimated native populations by up to 98%. The species has not yet fully recovered and is struggling with recruitment success which is why it may benefit from an ex-situ recovery program. In this study, I used single nucleotide polymorphisms (SNPs) to identify genetic variation and analyze parentage in a captive population of …

Karyotyping As A Method To Verify Phylogenetic Relationships Among The Enneacanthus Species, Ophelia A. Anderson Jan 2025

Karyotyping As A Method To Verify Phylogenetic Relationships Among The Enneacanthus Species, Ophelia A. Anderson

Theses and Dissertations

The Enneacanthus genus consists of three species of freshwater fish endemic to eastern North America: E. obesus (Banded Sunfish), E. gloriosus (Blue Spotted Sunfish), and E. chaetodon (Black Banded Sunfish). Previous studies using molecular data suggest that E. obesus and E. gloriosus are more closely related, with E. chaetodon as the most distantly related species in the genus. This study investigates the phylogenetic relationship among the Enneacanthus species, using karyotype characteristics - chromosome number, morphology and fundamental number - as comparative metrics. Chromosome spreads were obtained from scale and gill epithelium samples collected between 1993 and 1994. Prepared slides …

An Edna Assessment Of The Diamond Darter (Crystallaria Cincotta) In Its Current And Historic Distribution Range, Cameron Moore Layne

Graduate Theses, Dissertations, and Problem Reports (ETD)

Environmental DNA (eDNA) can serve as a useful monitoring tool for cryptic and imperiled freshwater stream fishes. eDNA can fill knowledge gaps regarding species’ distribution, site abundance, and habitat usage. The last known stronghold of the Diamond Darter (Crystallaria cincotta) in the Elk River of West Virginia provides an excellent test case to examine the utility of this non-invasive method of surveying rare species. Diamond Darters are one of the most newly recognized freshwater fishes in North America and are among the most imperiled U.S. fishes, having been federally listed as endangered in 2013. The cryptic behavior and limitations of …

Relationship Between Host Genomics And Microbiomics In Beef Cattle, Andrew D. Lakamp Dec 2024

Relationship Between Host Genomics And Microbiomics In Beef Cattle, Andrew D. Lakamp

Department of Animal Science: Dissertations, Theses, and Student Research

As sequencing technology becomes more affordable and throughput increases, microbiome information is becoming more readily available. For beef cattle selection, microbial information has a variety of uses including being a target for genetic prediction or used as a means to explicitly describe additional phenotypic variability in other traits.

Infectious bovine keratoconjunctivitis (IBK), commonly known as pinkeye, is a disease that infects the ocular surface and surrounding tissue which is an animal health and producer economic concern. Vaccinations have shown to have variable effectiveness, while limited genetics studies have suggested that direct genetic selection for resistance would be slow. Therefore, an …

Porcine Peritoneal Macrophages Are Susceptible To Porcine Reproductive And Respiratory Syndrome Virus Infection, Kassandra Durazo-Martinez, Jayeshbhai Chaudhari, Sushmita Kumari, Hiep Vu Nov 2024

Porcine Peritoneal Macrophages Are Susceptible To Porcine Reproductive And Respiratory Syndrome Virus Infection, Kassandra Durazo-Martinez, Jayeshbhai Chaudhari, Sushmita Kumari, Hiep Vu

Department of Animal Science: Faculty Publications

Previous studies have suggested that porcine peritoneal macrophages (PPMs) are resistant to PRRSV infection, whereas porcine alveolar macrophages (PAMs) are highly susceptible. This contrast is intriguing, as both cell types belong to the same monocyte/macrophage family. The current study aimed to investigate the host factors contributing to the differing susceptibility of PPMs and PAMs to PRRSV infection. We found that PPMs exhibit a higher frequency of CD14+ cells compared to PAMs, suggesting a more immature macrophage phenotype in PPMs. Importantly, PPMs expressed both CD163 and CD169, the key receptors for PRRSV entry, although the frequency and intensity of CD163 and …

Protracted Maternal Malnutrition Induces Aberrant Changes In Maternal Uterine Artery Hemodynamics And The Metabolic Profiles Of The Dam And Neonate, Jennifer F. Thorson, Ligia D. Prezotto Nov 2024

Protracted Maternal Malnutrition Induces Aberrant Changes In Maternal Uterine Artery Hemodynamics And The Metabolic Profiles Of The Dam And Neonate, Jennifer F. Thorson, Ligia D. Prezotto

Department of Animal Science: Faculty Publications

Malnutritional stress during gestation is a well-established driver of metabolic disfunction in offspring. Extended exposure to malnutrition requires metabolic plasticity as the animal shifts toward a catabolic state. In this paper we demonstrate the influence of malnutrition throughout gestation on uterine artery hemodynamics and the metabolism of the dam and neonate. We hypothesized that gestational malnutrition reduces blood flow of the maternal uterine artery and regulates the metabolic profile of the dam and offspring. Further, the combination of these factors consequently influences the concentration of metabolites in the cerebrospinal fluid of the neonate at birth. To test our hypotheses, pregnant …

Adding Gastrointestinal Parasite Resistance To The Breeding Objective In Hair Sheep: Initial Step, Robert M. Forbes, Thomas W. Murphy, Joan M. Burke, David R. Notter, Matthew L. Spangler, Michael D. Macneil, Ronald M. Lewis

Department of Animal Science: Faculty Publications

The U.S. Maternal Hair Index was designed by the National Sheep Improvement Program (NSIP) to increase total weight of lamb weaned per ewe lambing (TW). Producers are interested in adding gastrointestinal nematode (GIN) resistance to this breeding objective since parasitism causes substantial economic losses. The NSIP provides producers with estimated breeding values (EBV) for postweaning fecal egg count (PFEC), an indicator of GIN resistance. Our objective was to gauge the effects of including PFEC as another selection criterion and goal trait. Selection index theory was used to construct 11 indexes. …

Department of Animal Science: Faculty Publications

Whole genome sequences (WGS) of 185 North American Thoroughbred horses were compared to quantify the number and frequency of variants, diversity of mitotypes, and autosomal runs of homozygosity (ROH). Of the samples, 82 horses were born between 1965 and 1986 (Group 1); the remaining 103, selected to maximize pedigree diversity, were born between 2000 and 2020 (Group 2). Over 14.3 million autosomal variants were identified with 4.5–5.0 million found per horse. Mitochondrial sequences associated the North American Thoroughbreds with 9 of 17 clades previously identified among diverse breeds. Individual coefficients of inbreeding, estimated from ROH, averaged 0.266 (Group 1) and …

Enhanced Bmp Signaling Via Alk2 In Osteoclasts Decreases Bone Density In Mice, Yolanda V. Gutierrez, Hiroyuki Yamaguchi, Yuji Mishina, Yoshihiro Komatsu

Research Colloquium

Bone remodeling is a complex biological process that has been extensively studied. Bone Morphogenetic Proteins (BMPs) are recognized as one of the critical growth factors that coordinate bone remodeling. Previous studies have demonstrated that BMP signaling in osteoclasts has a positive effect on osteoclast function. However, little is known about how each BMP type I receptors control osteoclastogenesis. To investigate this question, we utilized the Cre-LoxP system to specifically activate BMP signaling through ALK2 in mice. We utilized Cathepsin K (Ctsk)-Cre driver to activate BMP signaling in osteoclasts in mice. Compared with aged- and gender-matched controls, gain-of-function of BMP mutant …

Technical Note: Assessing Gps Sensor Accuracy Using Real-Time Kinematic Device For Livestock Tracking, Biquan Zhao, Rebecca Mcdermott, Galen E. Erickson, Yijie Xiong

Department of Animal Science: Faculty Publications

The technical aspects of global positioning system (GPS) sensors have been improved substantially over the years, making them valuable and popular tools for livestock tracking. Using GPS sensors allows producers and researchers to locate grazing livestock, monitor their behavior and distribution, and gather referencing information about the health status of the animals and pastures. However, interpreting the behavior of grazing livestock, such as cattle, from GPS data can be difficult due to positioning inaccuracies. Without knowing the positioning accuracy range of GPS sensors, achieving high-level confidence in determining grazing densities and identifying abnormal livestock movement patterns is challenging. …

Investigation Of P27 Tumor Suppressor Gene (Cdkn1b) Polymorphisms In Dogs With Malignant Mammary Tumors, Gizem Kırmızıoğlu, Iraz Akış

The Thai Journal of Veterinary Medicine

Mammary tumors are the most common neoplasm in dogs, with a high mortality rate. The development of canine mammary tumors (CMT) is multifactorial, but different incidence rates between breeds suggest the effects of genetic risk factors. Many CMT-associated candidate genes were reported, mainly involved in cell cycle control. This study aims to investigate p27 tumor suppressor gene polymorphisms in dogs with mammary tumors and analyze the association between variations and CMTs. For this purpose, case and control groups were formed from 22 dogs diagnosed with malignant mammary tumors and 10 dogs with healthy mammary glands. The whole canine p27 gene …

Knockdown Of Gonadotropin-Releasing Hormone Ii Receptor Impairs Ovulation Rate, Corpus Luteum Development, And Progesterone Production In Gilts, Amy T. Desaulniers, Rebecca Cederberg, Clay A. Lents, Brett White Aug 2024

Knockdown Of Gonadotropin-Releasing Hormone Ii Receptor Impairs Ovulation Rate, Corpus Luteum Development, And Progesterone Production In Gilts, Amy T. Desaulniers, Rebecca Cederberg, Clay A. Lents, Brett White

Department of Animal Science: Faculty Publications

Reproduction is classically controlled by gonadotropin-releasing hormone (GnRH-I) and its receptor (GnRHR-I) within the brain. In pigs, a second form (GnRH-II) and its specific receptor (GnRHR-II) are also produced, with greater abundance in peripheral vs. central reproductive tissues. The binding of GnRH-II to GnRHR-II has been implicated in the autocrine/paracrine regulation of gonadal steroidogenesis rather than gonadotropin secretion. Blood samples were collected from transgenic gilts, with the ubiquitous knockdown of GnRHR-II (GnRHR-II KD; n = 8) and littermate controls (n = 7) at the onset of estrus (follicular) and 10 days later (luteal); serum concentrations of 16 steroid hormones …

Experiment 1 utilized 320 yearling steers (initial BW = 413 kg; SD = 25 kg) fed in 32 pens (10 steers/pen) and assigned to one of four treatments (n = 8 pens/treatment). The four treatments consisted of a blended DRC:HMC corn control diet (CON), de-oiled modified distillers grains plus solubles included at 40% of diet DM …

The Role Of Elevation In The Migration, Physiology, And Genomic Diversification Of Birds, Jessie L. Williamson Aug 2024

The Role Of Elevation In The Migration, Physiology, And Genomic Diversification Of Birds, Jessie L. Williamson

Biology ETDs

Elevational gradients cause profound eco-climatic variation across short distances. Partial pressure of oxygen (PO2) is a severe challenge that organisms face at high elevations; due partly to specialization on PO2, most Neotropical bird species have narrow elevational ranges. However, a small fraction are elevational generalists that span sea level to high elevations, and a subset undertake extreme migratory journeys from low to high elevations biannually. My dissertation combines diverse approaches across levels of biological organization to understand how elevation impacts the ecology, evolution, physiology, and migration of montane birds. I examined ‘extreme’ elevational …

Investigating Associations Between Activity And Performance Traits Across Three Breeds Of Swine Using NuTrack, Dalton R. Obermier Aug 2024

Investigating Associations Between Activity And Performance Traits Across Three Breeds Of Swine Using NuTrack, Dalton R. Obermier

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Feed costs are the largest contributor towards the variable costs of pork production. To decrease feed cost while not sacrificing output requires an improvement in feed efficiency (FE). A potential approach to expand available phenotypes associated with FE is with activity tracking, yet the relationship between activity and performance is not well defined in swine. Therefore, the objective of this study was to quantify associations between activity and feed intake traits. Average daily feed intake (ADFI) was collected on 1,766 purebred boars of three genetic lines (Duroc, Landrace, and Yorkshire) using FIRE (Osborne Industries, Inc., Osborne, Kansas) at a testing …

Assessment Of Genetic Diversity And Population Structure Of U.S. Polypay Sheep From Breed Origins To Future Genomic Selection, Carrie S. Wilson, Jessica Lynn Petersen, Luiz F. Brito, Brad A. Freking, Sara M. Nilson, J. Bret Taylor, Thomas W. Murphy, Ronald M. Lewis

Department of Animal Science: Faculty Publications

Knowledge of past and present genetic diversity within a breed is critical for the design and optimization of breeding programs as well as the development of strategies for the conservation of genetic resources. The Polypay sheep breed was developed at the U.S. Sheep Experiment Station (USSES) in 1968 with the goal of improving productivity in Western U.S. range flocks. It has since flourished in the more intensively managed production systems throughout the U.S. The genetic diversity of the breed has yet to be documented. Therefore, the primary objective of this study was to perform a comprehensive evaluation of the genetic …

Conservation Genomic Investigation Of A Threatened Trout Species Endemic To The Gila River, David Thomas Camak Jul 2024

Conservation Genomic Investigation Of A Threatened Trout Species Endemic To The Gila River, David Thomas Camak

Biology ETDs

Climate change is a looming threat to the persistence of species, especially aquatic, imperiled species. Species native to desert streams are at a particular disadvantage, as many species already face drought, floods, wildfires, and threats from nonnative species, all of which are thought to be exacerbated by climate change. Species such as the threatened Gila Trout (Oncorhynchus gilae) already have small ranges and persist in fragmented and isolated habitats that limits gene flow between populations. Therefore, it is important to understand the current genomic status of such populations to be able to predict the future effects of climate …

Hybridization Between The Rare Gray-Headed Chickadee And The Abundant Boreal Chickadee In The Midst Of Shifting Climate, Matthew R. Armstrong Jul 2024

Hybridization Between The Rare Gray-Headed Chickadee And The Abundant Boreal Chickadee In The Midst Of Shifting Climate, Matthew R. Armstrong

School of Natural Resources: Dissertations, Theses, and Student Research

As species respond to changing climate, distributions and abundances may shift and alter species interactions. Hybridization, a relatively widespread phenomenon becoming more common with climate change, can have beneficial and detrimental effects on population growth rates and genetic integrity. Beneficial effects due to the introduction of advantageous alleles and increased genetic diversity may result from hybridization. Species may also accrue fitness costs associated with changing climates if mismatches occur between environmental variables and phenotypes. The gray-headed chickadee, Poecile cinctus lathami, is an extremely rare songbird that has experienced marked declines in recent decades within its restricted distribution in Alaska …
