Iron deficiency chlorosis (IDC) is a significant yield-limiting problem in several major soybean [Glycine max (L.) Merr.] production regions in the United States. Soybean plants display a variety of symptoms that range from a slight yellowing of the leaf to interveinal chlorosis, to stunted growth that reduces yield. The objective of this analysis was to employ single nucleotide polymorphism (SNP)-based genome-wide association mapping to uncover genomic regions associated with IDC tolerance. The micronutrients iron (Fe), zinc (Zn), and copper (Cu) are essential for plants and the humans and animals that consume plants. Increasing the micronutrient density of staple crops, or biofortification, will greatly improve human nutrition on a global scale. This review discusses the processes and genes needed to translocate micronutrients through the plant to the developing seeds, and potential strategies for developing biofortified crops.

The use of velocity traverses to measure in-situ air flow rate of ventilation fans can be subject to significant errors. The Fan Assessment Numeration System (FANS) was developed by the USD-ARS Southern Poultry Research Laboratory and refined at the University of Kentucky to measure air flow of fans in-situ. The procedures for using the FANS unit to test fans in-situ are not completely standardized. Processes that regulate site of fat deposition in beef cattle are poorly understood. For the producer to procure the greatest profit, it is ideal to maximize intramuscular fat. Furthermore, to understand the physiological mechanisms affecting fat depots, it is necessary to evaluate hormones involved in growth regulation. Using a 2 x 2 factorial design of treatments, four experiments were conducted to examine two adipogenic compounds, chlortetracycline and dexamethasone. Synovex-S® and Revalor-S® were used to investigate potential interactions between growth implants and adipogenic compound. Growth performance, carcass quality, organ and fat mass and plasma hormone concentrations were measured in these studies. …

Sonchus yellow net virus (SYNV), Potato yellow dwarf virus (PYDV) and Lettuce Necrotic yellows virus (LNYV) are members of the Rhabdoviridae family that infect plants. SYNV and PYDV are Nucleorhabdoviruses that replicate in the nuclei of infected cells and LNYV is a Cytorhabdovirus that replicates in the cytoplasm. LNYV and SYNV share a similar genome organization with a gene order of Nucleoprotein (N), Phosphoprotein (P), putative movement protein (Mv), Matrix protein (M), Glycoprotein (G) and Polymerase protein (L). PYDV contains an additional predicted gene between N and P, denoted as X, that has an unknown function. In order to gain …

Tall fescue, a cool-season grass native to Europe, central Asia, and northern Africa, has been widely distributed throughout the U.S. for use as turf and forage. Following its widespread planting, its ability to associate with a toxic fungal endophyte, Neotyphodium coenophialum, was discovered. Research has linked this fescue-endophyte association with increased biotic and abiotic stress resistance in endophyte-infected (E+) versus endophyte-free (E-) plants, and these differences may affect the ability of land managers to eradicate tall fescue and restore native grasslands. Switchgrass (Panicum virgatum), a native warm-season grass, has been investigated as a renewable energy crop that may provide viable wildlife habitat. This study investigated small mammal populations in switchgrass, hay, and corn to assess the relative habitat quality. Four, three-night trapping sessions were conducted at four locations in Kentucky using Sherman livetraps. Trapping occurred in spring (before first hay harvest), summer, fall (before switchgrass and corn harvest), and winter (post-harvest). Relative abundance of small mammals, calculated using a capture per unit effort index (per 100 trapnights), and mean taxonomic richness were used to compare habitats. Switchgrass had a …

A computational method for estimating off-target application areas based on the machine-controlled section width and the field shape was developed and implemented in software with a graphical user interface written in the MatLab environment. The program, which is called the Field Coverage Analysis Tool (FieldCAT), includes three modules: data import, data preparation, and coverage analysis. Nine field boundaries were evaluated to test the software using controlled section widths from 0.5 to 27 m and various swath orientations. Optimisation of the optical signal from the laser multi-spot beam generator employed in a photonic based real-time plant discrimination sensor for use in selective herbicide spraying systems is presented. The plant detection sensor uses a three-wavelength laser diode module that sequentially emits identically-polarized laser light beams through a common aperture, along one optical path. Each laser beam enters a multi-spot beam generator which produces 15 parallel laser beams over a 240mm span. The intensity of the reflected light from each spot is detected by a high-speed line scan image sensor. Plant discrimination is based on calculating the slope of the …

The goal of this dissertation is to address the role of government payments in the allocation of off-farm labor through three essays. Government payments have been a part of agriculture since 1933 and at no time has the government stated a policy objective of decreasing the agricultural labor force. Using time series data and new econometric techniques, the first essay finds agricultural policy may have an unintended impact on labor migration. Specifically, we find that government payments increased labor migration from the farm. From 1939 to 2007, increased direct government payments resulted in greater migration of labor from agriculture. The …

The abandonment of fixed exchange rate systems has caused exchange rate movements to become a major concern for traders, policy makers and researchers. During the previous four decades of floating exchange rates, numerous studies have been conducted to determine whether exchange rate volatility affected international trade flows. Researchers have not yet reached a general consensus as to the magnitude and direction of the impact of exchange rate volatility on trade flows. The objective of this study was to determine if experimental infection of neonatal calves with Mycobacterium avium subsp. paratuberculosis would invoke changes in the percentages of total B cells in the peripheral blood mononuclear cell population and of subpopulations of B cells as determined by CD5, CD25, and CD45RO markers during a 12-month period. Experimental infection groups included control (noninfected), oral (infected with M. avium subsp. paratuberculosis strain K-10), oral/DXM (pretreatment with dexamethasone before oral inoculation), i.p. In the 40th Annual Phi Delta Kappa/Gallup Poll of the Public’s Attitudes toward the Public Schools, Americans rated discipline as the second largest problem facing public education (Bushaw & Gallup, 2008). This poses a substantial problem for administrators as they strive to employ school reform policies, address public demands and meet the needs of contemporary students. A review of literature revealed a large body of research which examines disciplinary practice; however, it also showcased a literary gap regarding administrative disciplinary philosophies. Due to a close genetic relatedness, there is no known antibody that detects Mycobacterium avium subspecies paratuberculosis (MAP), which causes Johne’s disease in cattle and sheep, and does not cross-react with other M. avium subspecies. In the present study, a monoclonal antibody (MAb; 17A12) was identified from mice immunized with a cell membrane fraction of MAP strain K-10. This antibody is 100% specific as it detected a 25-kDa protein in all 29 MAP whole cell lysates, but did not bind to any of the 29 non- paratuberculosis strains tested in immunoblot assays. Three independent studies were conducted to quantify ultimate biochemical oxygen demand (UBOD) and the corresponding decomposition rate constant for production pond (average 21.5 ha each) waters and effluents on six semi-intensively managed marine shrimp (Litopenaeus vannamei) farms in Honduras. Samples were collected during the rainy season in studies 1–3 and during the dry season only in Study 1. Pond water samples were collected during the culture period and/or at harvest. The dry season 30-d biochemical oxygen demand (BOD30) of pond waters across all farms was 26% lower and UBOD was 54% lower, but the decomposition rate constant was …

The issue of food security is complex. By analyzing the relationship between international, national and local society, one can better contextualize food security issues. Using an embedded research design (with a qualitative leaning) supported by quantitative data, research is conducted via surveys, interviews and focus groups. This research design was chosen to offset limited sample sizes with quantitative data to strengthen findings. Research findings were cross analyzed to identify three emergent themes. Kentucky reported a solid 13.1% growth in certified organic land from 1997 to 2002. The relative lack of research on disease management practices in Kentucky consistent with organic regulations is an issue that needs to be addressed to provide more reliable information to local farmers. Epichloë species and their anamorphs, Neotyphodium species, are fungal endophytes that inhabit cool-season grasses and often produce bioprotective alkaloids. These alkaloids include lolines, which are insecticidal and insect feeding deterrents. Lolines are exo-1-aminopyrrolizidines with an oxygen bridge between carbons 2 and 7, and are usually methylated and formylated or acetylated on the 1-amine. In previously published studies lolines were shown to be derived from the amino acids L-proline and L-homoserine. In addition the gene cluster involved in loline-alkaloid biosynthesis has also been characterized. While forage plays an important role in equine nutrition, little research has been conducted evaluating fiber utilization by young horses. Therefore, studies were conducted to compare in vivo digestibility and digesta passage in weanlings and mature horses (Exp 1) and yearlings and mature horses (Exp 2). All horses were fed forage-based diets at the same rate (on a metabolic BW basis; Exp 1: 67% alfalfa cubes, 33% concentrate; Exp 2: 75% timothy cubes, 25% concentrate). The Southern California area is experiencing increasing water shortages resulting in the need for plant material that has a high ornamental value as well as a low water requirement. The objectives of our project were to find, plant, and observe succulent plant material in the Southern California area to determine which succulents have a low water requirement, are low maintenance, disease and pest resistant, while adding beauty to the landscape. Trap grease offers a promising and less expensive alternative to food grade vegetable oils for biodiesel production, but its commercial marketability could be affected by its intense and offensive odor. Identification, quantitation and eventual elimination of the odorant compounds are vital challenges that need to be addressed before commercialization can be achieved. A rapid method using HS-SPME and GC/MS was developed in this study to identify and quantify the major volatile odorant compounds present in trap grease. The main odorant compounds were identified as short chain free fatty acids (SC-FFA) from C4 - C10.

The influence of asparaginase on acrylamide formation, as well as colour development, in short dough biscuits was studied. In particular, asparaginase concentration, incubation time and temperature were changed according to an experimental design. As acrylamide formation was found to vary significantly between biscuits obtained by using the same ingredients and process, a mixed effect model was used to model variation of acrylamide concentration. By contrast a fixed effect model was used for colour polynomial analysis. Within the range of study, the overall results allowed the best conditions for minimising acrylamide formation to be found.
