Mutational Analysis Of The Major Soybean Uref Paralogue Involved In Urease Activation, Joe C. Polacco, D. L. Hyten, Mônica Medeiros-Silva, David A. Sleper, Kristin D. Bilyeu
Department of Agronomy and Horticulture: Faculty Publications
The soybean genome duplicated ~14 and 45 million years ago and has many paralogous genes, including those in urease activation (emplacement of Ni and CO2 in the active site). Activation requires the UreD and UreF proteins, each encoded by two paralogues. UreG, a third essential activation protein, is encoded by the single-copy Eu3, and eu3 mutants lack activity of both urease isozymes. eu2 has the same urease-negative phenotype, consistent with Eu2 being a single-copy gene, possibly encoding a Ni carrier. Unexpectedly, two eu2 alleles co-segregated with missense mutations in the chromosome 2 UreF paralogue (Ch02UreF), suggesting …

Soybean (Glycine max) is a self-pollinating species that has relatively low nucleotide polymorphism rates compared with other crop species. Despite the low rate of nucleotide polymorphisms, a wide range of heritable phenotypic variation exists. There is even evidence for heritable phenotypic variation among individuals within some cultivars. Williams 82, the soybean cultivar used to produce the reference genome sequence, was derived from backcrossing a Phytophthora root rot resistance locus from the donor parent Kingwa into the recurrent parent Williams. To explore the genetic basis of intracultivar variation, we investigated the nucleotide, structural, and gene content variation of different …

Iron deficiency chlorosis (IDC) is a significant yield-limiting problem in several major soybean [Glycine max (L.) Merr.] production regions in the United States. Soybean plants display a variety of symptoms that range from a slight yellowing of the leaf to interveinal chlorosis, to stunted growth that reduces yield. The objective of this analysis was to employ single nucleotide polymorphism (SNP)-based genome-wide association mapping to uncover genomic regions associated with IDC tolerance. Two populations [2005 (n = 143) and 2006 (n = 141)] were evaluated in replicated, multilocation IDC trials. After controlling for population structure and individual relatedness, and selecting …

Restoration Of Tall Fescue Pastures To Native Warm Season Grasslands: Does A Fungal Endophyte Symbiosis Play A Role In Restoration Success?, Sarah Lynn Hall
University of Kentucky Doctoral Dissertations
Tall fescue, a cool-season grass native to Europe, central Asia, and northern Africa, has been widely distributed throughout the U.S. for use as turf and forage. Following its widespread planting, its ability to associate with a toxic fungal endophyte, Neotyphodium coenophialum, was discovered. Research has linked this fescue-endophyte association with increased biotic and abiotic stress resistance in endophyte-infected (E+) versus endophyte-free (E-) plants, and these differences may affect the ability of land managers to eradicate tall fescue and restore native grasslands. I conducted three studies to examine whether E+ tall fescue plants respond differently to management than E- plants, …
Intermediate Steps Of Loline Alkaloid Biosynthesis, Jerome Ralph Faulkner
Intermediate Steps Of Loline Alkaloid Biosynthesis, Jerome Ralph Faulkner
University of Kentucky Doctoral Dissertations
Epichloë species and their anamorphs, Neotyphodium species, are fungal endophytes that inhabit cool-season grasses and often produce bioprotective alkaloids. These alkaloids include lolines, which are insecticidal and insect feeding deterrents. Lolines are exo-1-aminopyrrolizidines with an oxygen bridge between carbons 2 and 7, and are usually methylated and formylated or acetylated on the 1-amine. In previously published studies lolines were shown to be derived from the amino acids L-proline and L-homoserine. In addition the gene cluster involved in loline-alkaloid biosynthesis has also been characterized. In this dissertation a survey of plant-endophyte symbioses revealed a phenotype with only N-acetylnorloline. This …

Books & book chapters
The department was not always such a significant organisation; it came from very small beginnings. However, it has been a major component of the engine which has produced an agricultural industry in a Mediterranean climate zone which is superior to all others operating in this zone. This book is a summary of its work. There are many more fascinating chapters to be recorded of work on specific issues of development, investigation or conservation. I hope they will be written by someone one day. With this in mind the department has decided to put the book on a web page where …

Biosecurity published reports
Biosecurity is the protection of the economy, environment and human health from the negative impacts of pests, weeds and diseases, including the management of invasive species if they become established (definition adopted by AusBIOSEC).
This requires steps to prevent the entry of harmful organisms, as well as the timely and efficient management of them if they do enter.
Biosecurity threats range from catastrophic diseases such as Foot and Mouth Disease, which causes rapid and widespread animal deaths, to more subtle and slower spreading problems such as invasive weeds, mussels that foul vessel hulls and fungus-induced dieback disease of trees. These …

Soil Texture Fractions And Fractal Dimension Of Particle Size Distribution As Predictors Of Interrill Erodibility, Mohammad Reza Neyshabouri, Abbas Ahmadi, Hassan Rouhipour, Hossein Asadi, Mehdi Irannajad
Turkish Journal of Agriculture and Forestry
Choosing a particular textural fraction as an erodibility predictor is often confusing because various fractions of soil particles have been introduced as erodibility index by many researchers. Recently, advances in fractal theory have introduced a scaling parameter for characterizing soil fragments. The objectives of this study were (i) to test the applicability of fractal dimension of particle size distribution (PSD) for estimation of interrill erodibility and (ii) to study the relationship between interrill erodibility and soil texture components. Samples from 36 soil series with contrasting characters were collected from northwest Iran. The sand fractions were obtained by sieving, while silt …

Some Characteristics Of The Stagnation Stage In The Development Of Silver Fir (Abies Alba Mill.) Trees In Selection Forests In Serbia, Damjan Pantic, Stanisa Bankovic, Milan Medarevic, Snezana Obradovic
Turkish Journal of Agriculture and Forestry
A period of stagnation in silver fir development has been recorded in all felled trees in selection forests of Serbia (233 trees on Mt. Goc and 451 trees on Mt. Tara). In the selection forests on Mt. Goc, the stagnation stage ranges between 40 and 330 years, but on Mt. Tara, it ranges from 15 to 185 years. It was concluded that the duration was caused not by the ecological and productivity potential (site class) of the soil for fir development, but primarily by the growth space, resulting from the application of single-tree selection or group selection systems. It was …

Entomopathogenic nematodes (EPNs) in families Heterorhabditidae and Steinernematidae have considerable potential as biological control agents against soil-inhabiting insect pests. In the present study, the potential of Steinernema feltiae as a biological control agent against cabbage maggot (CM) Delia radicum (Diptera: Anthomyiidae), persistence of the nematode in areas infested by CM larvae, and foraging behavior of the nematode toward CM and/or oilseed rape roots were examined in the laboratory. The results of the laboratory experiment showed that more than 80% control was achieved against the last instars of D. radicum. Steinernema feltiae persisted in the soil in high numbers for 12 …

The following study was conducted in order to compare the modified single-seed descent method of selection used in soybean breeding at the Institute of Field and Vegetable Crops in Novi Sad, Serbia, with the standard bulk method and the pedigree method. The source materials for the development of hybrid populations were 15 genotypes, and a total of 10 cross combinations were made. Comparison of the lines developed by the different breeding methods was done in the F6 generation. Genetic gain from selection was taken as the main indicator of efficiency of the tested methods. The modified single-seed descent method produced …
