Rice blast caused by Magnaporthe oryzae poses a major threat to world food supply. Conditions conducive to the disease include insufficient light. Our previous studies found that light-dependent phosphorylation of light-harvesting complex II protein (LHCB5) mediates rice immunity; however, the underlying mechanisms, in particular, the upstream regulators of LHCB5 phosphorylation, remain unknown. Here, we show that OsLIKE1 functions as a lectin receptor-like kinase (LecRLK) to play a critical role in rice resistance against M. oryzae. OsLIKE1 interacts with and phosphorylates LHCB5. In addition, we find two phosphorylation sites in the kinase domain of OsLIKE1 important for its auto-phosphorylation and phosphorylation …

Neurological disorders are linked to mitochondrial dysfunction and calcium overload. Mitochondrial calcium uptake is mediated by the mitochondrial calcium uniporter (mtCU), regulated by MICU1, which can be either homodimerized or heterodimerized with MICU2 or MICU3. Though MICU2 is scarce in the adult brain, MICU2 loss in patients leads to a neurodevelopmental disorder. We hypothesized that MICU2 is required for developmental calcium signaling and neuronal migration. MICU2 is present in the developing mouse brain but disappears by maturation, contrasting with other mtCU subunits that increase. Potentially inappropriate prescribing (PIP) is an avoidable factor leading to adverse drug events in older adults, particularly in the emergency department (ED). This study aimed to train and validate a machine learning (ML) model to predict the probability of PIP in elderly patients in the ED. This retrospective analysis encompassed patients aged 65 years and above who presented to the Al-Qatif Central Hospital. PIPs were identified using a validated screening tool designed for older persons to ensure appropriate treatment. Background
Alcohol-induced liver injury occurs in the pericentral region of the liver and can induce mitochondrial remodeling and exacerbate Alzheimer's Disease (AD) progression. Subpopulations of mitochondria: general mitochondria (GM), peridroplet mitochondria (PDM) and endoplasmic reticulum(ER)-bound mitochondria (ERM) maintain cellular homeostasis via energy synthesis, lipid homeostasis, and regulation of intracellular calcium. Hepatic amyloid precursor protein (APP) is a source of peripheral amyloid beta (aB) and affects AD pathology in the brain. Understanding the localization and expression of hepatic APP in mitochondrial subpopulations are critical to understanding aB metabolism and may play a role in identifying a potential mechanism for metabolic dysfunction. …

The loss function is a mathematical representation of the costs experienced by a forecaster when observed outcomes differ from what was predicted. Prior studies suggest that USDA forecasts are not optimal based on an assumed mean-zero quadratic loss function. This study proposes an alternative view of forecast evaluation, which assumes all USDA forecasts are produced to minimize the forecasters’ costs, and searches for the dimensions of the loss function under which optimality holds. We illustrate the degree to which USDA loss functions vary across a series of WASDE price forecasts. A better understanding of USDA forecasters’ costs will benefit forecasters …

Perspectives beyond the CD4 T cell interferon (IFN)-γ paradigm are needed to understand immunity in tuberculosis (TB). Growing data in patients across a spectrum of TB highlight that changes in antibody Fc domain glycosylation and Fc effector functions correlate with disease and impact Mycobacterium tuberculosis (Mtb) infection. How antigen-specific antibodies within polyclonal responses affect bacteria is less clear. This study examines antibodies targeting ESAT-6 and CFP-10, Mtb virulence proteins essential for pathogenesis. Our Goal is to identify how colorectal cancer (CRC) arises in the single-layered cell epithelium (simple columnar epithelium) that lines the luminal surface of the large intestine. Background: We recently reported that the dynamic organization of cells in colonic epithelium is encoded by five biological rules and conjectured that colon tumorigenesis involves an autocatalytic tissue renewal reaction. The Profession of Microbiology (POM) embodies the bulk of the American Society for Microbiology (ASM) members and represents the career preparation arm of the ASM for academia, industry, and clinical lab professions. Perinatal T cells have distinctive phenotypes and functions that may be due in part to age-associated features of stromal cells in the perinatal thymus. We identify age-associated changes in mouse thymic epithelial cells, mesenchyme, endothelium, and hematopoietic antigen-presenting cells from birth to one month of age using single-cell transcriptional profiling, flow cytometry, and imaging. Coordinated cellular and molecular changes occur at 7-14 days of age, designated "transitional ages," as thymus growth switches to homeostasis. E2F target gene expression declines, and the expression of type I interferon response genes increases across diverse cell types at transitional ages. Alterations in thymic stromal …

Mantle cell lymphoma (MCL) is an incurable subtype of B-cell non-Hodgkin lymphoma. Despite multiple approved Bruton tyrosine kinase inhibitors (BTKis), resistance to BTKi continues to pose a major clinical challenge. The transcription factor sex determining region Y-box 11 (SOX11) is expressed in most patients with MCL and is associated with poor outcomes. We have previously demonstrated SOX11-dependent B-cell receptor (BCR) signaling in transgenic models of MCL. Here, we report that SOX11 drives BCR signaling via the transcriptional activation of the PAX5/CD19 axis. Lead is a persistent environmental toxin with pronounced neurotoxic effects, particularly on the central nervous system. This study evaluated the impact of lead acetate on the cerebellar cortex of adult Wistar rats and assessed the neuroprotective effects of aqueous Bryophyllum pinnatum extract. Twenty adult Wistar rats (180–220 g) were randomly assigned to four groups (n = 5): Control (distilled water), T1 (0.075 mg/kg lead acetate), T2 (100 mg/kg B. pinnatum), and T3 (lead + B. pinnatum). Qat (Catha edulis) chewing is a common practice in Yemen, associated with oral mucosal irritation. The objective of the current study is to compare cytological findings of buccal mucosa between Qat chewers and non-chewers. A comparative cross-sectional study was designed. Buccal smears were collected from chewers (n = 100) and non-chewers (n = 100), fixed, and stained with Harris hematoxylin staining. Smears were assessed for inflammatory changes and cytological atypia by two blinded examiners; disagreements were resolved by consensus. Green tea extract (GTE) is widely recognised for its bioactive compounds, which exhibit potential health benefits, including modulation of gut microbiota. However, the concentration-dependent effects of GTE on specific probiotic strains remain underexplored. The aim of this study was to systematically evaluate the concentration-dependent effects of green tea extract (GTE) on Lacticaseibacillus rhamnosus (L. rhamnosus). The promoting and inhibiting effects of GTE in the range of 0–50 mg/mL were analysed in vitro by OD₆₀₀ growth curve, pH change and colony forming unit (CFU) count. Injury recovery in sport involves not only physical rehabilitation but also significant psychological challenges, particularly for female athletes in amateur and elite environments. This study aimed to explore the perceived social support received by injured female Gaelic Athletic Association (GAA) athletes and the effect it had on their mental health during injury. A qualitative research design was employed to gain an in-depth understanding of the experiences of injured female athletes. Consumer uptake of a sustainable diet is critical for addressing environmental challenges and promoting public health. Understanding why consumers resist adopting sustainable eating habits can inform targeted interventions and policies. This pilot study examines whether a newly developed scale can effectively detect consumer resistance to a sustainable diet. Background:
Heavy metals, like lead, arsenic, and cadmium, are linked to increased inflammation even in early life; however, anti-inflammatory diets may offset these effects.
Methods:
We evaluated associations between heavy metals and inflammatory biomarkers, and potential effect modification by diet in 399 adolescents aged 10 to 18 at baseline who attended two study visits 2 years apart (773 observations). At baseline, blood lead, urinary arsenic, and cadmium concentrations were measured, and the Children’s Dietary Inflammatory Index (C-DII) was calculated. Fasting serum interleukin (IL)-4, IL-10, IL-1β, IL-6, IL-8, high-sensitivity C-reactive protein, and tumor necrosis factor-alpha were measured for both visits. Generalized …

In 2024, Utah State University conducted a study of Utah's farmers, ranchers, and food makers related to value-added food production and processing. This fact sheet provides an overview of the survey results related to respondents, their operations, products, and marketing outlets. It also discusses respondents’ familiarity with key regulatory and resource organizations, the types of products and business resources they find difficult to obtain or access, and the primary obstacles to their processing operations. Actinomycetota bacteria have specialized in the biosynthesis of antibacterial natural products (NPs), and extract and fraction libraries made from those strains remain a promising source of NP drug leads. Herein, we present a high-throughput screen (HTS), based on engineered Escherichia coli strains expressing the human (Trm5) or bacterial (TrmD) m1G37 tRNA methyltransferase, to discover NPs as novel anti-Gram-negative antibiotic leads. Background: Epithelial cell adhesion molecule (EpCAM) is a pleotropic transmembrane glycoprotein comprising an extracellular domain (EpEX), a single transmembrane domain, and an intracellular domain (EpICD). For several types of cancer, high EpCAM expression is associated with tumor progression, metastasis, immune evasion, and overall poor prognosis. A rare but aggressive form of thyroid cancer, anaplastic thyroid cancer (ATC), has a mean survival time of only 3–6 months after diagnosis.

Background/Objectives: Immune checkpoint blockade (ICB) therapies have significantly improved outcomes in metastatic melanoma. However, immune-related adverse events (irAEs) and persistent chronic toxicities (CTs) among this emerging survivor population likely influence different facets of quality of life. This study characterized CT, patient-reported outcomes (PROs), diet, physical activity and gut microbiome features in a cohort of long-term survivors with a history of ICB-treated metastatic melanoma.

We employ density functional theory calculations to investigate the energetics and diffusion mechanisms of alkali metal ions (Na+, K+, Rb+, and Cs+) within the analcime framework. While Na+preferentially adsorbs at the center of the 8-member ring, DFT calculations show that larger ions, such as Cs+, occupy the center of the cage due to steric constraints. The computed exchange energies indicate that exchange of Na+by Cs+is thermodynamically favorable. Multiplex immunofluorescence and immunohistochemistry (mIF/IHC) are increasingly employed antibody-based technologies that use tissue sparingly and facilitate the detection of co-localized or neighboring biomarkers. Specifically, these platforms enable spatial analyses of the tumor microenvironment as well as extended applications, for example, describing normal tissue anatomy, autoimmunity, infectious diseases, etc. mIF/IHC has greatly enhanced biomarker discovery efforts, and a growing number of studies suggest superiority to traditional IHC. Standardization of staining approaches, reporting of image analysis strategies and resultant data is critical for facilitating cross-study comparisons, validation, deployment, and generalization of findings. This paper presents a comprehensive study of a multi-gigahertz chaotic generator of light pulses based on solid-state laser sources, including fiber lasers, governed by carefully designed positive and negative feedback loops. It harnesses the inherent nonlinear dynamics within a solid-state laser controlled by a combination of two inertial feedback loops, enabling the realization of complex chaotic behavior, including the logistic map scenario, under moderate amplification conditions. The laser system dynamics are rigorously investigated through theoretical modeling, employing a nonlinear map approach, and high-resolution picosecond simulations. Non-small cell lung carcinoma (NSCLC) is the most periodic type of lung cancer and the second most diagnosed cancer globally. Syzygium cumini is plant that extensively used in cuisine and traditional medicine. However, its potential for NSCLC treatment has not yet been elucidated. This study determined the potential of S. cumini as anti-NSCLC using in silico approaches. The in silico study was applied to perform active compound analysis, selection of target candidates, network pharmacology, functional annotation, molecular docking, and molecular dynamics simulation, respectively.
