Occurrence Of Botryosphaeriaceae Species Associated With Grapevine Dieback In Algeria, Faiza Ammad, Messaoud Benchabane, Mohamed Toumi, Nesrine Belkacem, Amel Guesmi, Cherif Ameur, Pascal Lecomte, Othmane Merah

Turkish Journal of Agriculture and Forestry

Grapevine dieback is an increasing problem in grape vineyards in Algeria. We conducted field surveys each spring between 2006 and 2012 to determine the fungal pathogens associated with grapevine decline in various grape-growing regions. We isolated and identified 3 species of Botryosphaeriaceae (Botryosphaeria dothidea, Diplodia seriata, and Neofusicoccum parvum) from infected wood using a combination of morphological and cultural characters, as well as phylogenetic analyses of ribosomal DNA internal transcribed spacer (ITS) regions and fruiting body. We identified 20 isolates of Diplodia seriata, 13 isolates of Botryosphaeria dothidea, and 7 isolates of Neofusicoccum parvum. Inoculation of grapevine plantlets with the …

Application Of A Multicriteria Decision-Making Approach For Rice Land Suitability Analysis In Turkey, İsmai̇l Sezer, Orhan Dengi̇z

Turkish Journal of Agriculture and Forestry

The main objective of this study was to implement a spatial model with a multicriteria assessment approach for rice (Oryza sativa L.) land suitability by using geographic information system (GIS) techniques. The land mapping units resulting from the overlay of the selected theme layers had unique land quality information on which land suitability was based. The selected theme layers of rice include topographic factor (slope), soil physical factors (soil depth, soil texture, drainage, stoniness, hydraulic conductivity), and soil chemical factors (pH, electrical conductivity, CaCO_3, soil fertility). These theme layers were collected from existing information. Spatial information of soil physical and …

Optimization Of Orius Majusculus Release: Photoperiodic Sensitivity At Different Temperatures And Storage Of Diapausing Adults, Şeri̇fe Ünal Bahşi̇, İrfan Tunç

Turkish Journal of Agriculture and Forestry

The photoperiodic sensitivity and storage of Orius majusculus (Reuter) from Antalya was investigated to obtain data for the optimization of its release. In the current study, it was not possible to induce a high (up to 100%) incidence of diapause unless all the nymphal instars and adults were successively exposed (i.e. full exposure) to a short day length (11.5 h) at constant temperatures of 18 and 26 °C, and fluctuating temperatures of 10/26 °C. Hence an intermittent exposure to long day lengths, even for short periods of time (only during instars IV–V or adult stage), would avoid a high incidence …

Rabies in bats is considered enzootic throughout the New World, but few comparative data are available for most countries in the region. As part of a larger pathogen detection program, enhanced bat rabies surveillance was conducted in Guatemala, between 2009 and 2011. A total of 672 bats of 31 species were sampled and tested for rabies. The prevalence of rabies virus (RABV) detection among all collected bats was low (0.3%). Viral antigens were detected and infectious virus was isolated from the brains of two common vampire bats (Desmodus rotundus). RABV was also isolated from oral swabs, lungs and …

Evaluating The Effect Of Short-Term Capture And Handling On Fecal Glucocorticoid Metabolite Levels In Mourning Doves, D. S. Jachowski, T. W. Wong, B. E. Washburn, J. H. Schulz, J. J. Millspaugh Jan 2014

Evaluating The Effect Of Short-Term Capture And Handling On Fecal Glucocorticoid Metabolite Levels In Mourning Doves, D. S. Jachowski, T. W. Wong, B. E. Washburn, J. H. Schulz, J. J. Millspaugh

United States Department of Agriculture Wildlife Services: Staff Publications

The measurement of stress hormones (i.e., glucocorticoids) has greatly advanced animal conservation. Fecal glucocorticoid metabolite assays are valuable because they are noninvasive, but their ability to detect responses to short-term (<30 >min) stressors in a way similar to blood serum assays is comparatively less well understood. We evaluated whether fecal glucocorticoid metabolites (FGMs) increased in captive wild mourning doves (Zenaida macroura) exposed to either a brief (<2 >min) capture, handling and release (CHR) or capture stress protocol (CSP; i.e., capture, hold for 30 min, release) treatment. Previous studies have shown that mourning doves exhibit elevated FGMs within 2-3 …

The Socioecology Of Monk Parakeets: Insights Into Parrot Social Complexity, Elizabeth A. Hobson, Michael L. Avery, Timonthy F. Wright Jan 2014

The Socioecology Of Monk Parakeets: Insights Into Parrot Social Complexity, Elizabeth A. Hobson, Michael L. Avery, Timonthy F. Wright

United States Department of Agriculture Wildlife Services: Staff Publications

In many species, individuals benefit from social associations, but they must balance these benefits with the costs of competition for resources. Understanding how these competing factors generate diversity in social systems is a major goal of behavioral ecology, but one that has been hampered by a lack of basic data quantifying many aspects of social structure and associations. Although parrots are generally assumed to have complex social groups, few studies have quantitatively examined these assumptions about parrot social structure. We critically assessed 4 assumptions about parrot socioecology using data from captive and wild groups of Monk Parakeets (Myiopsitta monachus …

Trichinella spp. and Toxoplasma gondii are important zoonotic parasites that infect warmblooded animals and humans worldwide. Among domesticated food animals, pigs are themain host for Trichinella spiralis. Pigs, chickens, sheep, and goats are known to be infectedwith T. gondii at varying rates, depending on husbandry. Infections in wildlife with theseparasites are generally higher than in domesticated species. Feral swine act as reservoirsof infection in the sylvatic ecosystem for Trichinella spp. and T. gondii, acting as sourcesof infection for peridomestic carnivores whose home ranges overlap with domestic pigs.Feral swine can have direct contact with non-biosecure domestic pigs, presenting oppor-tunity …

Coqui Frog Populations Are Negatively Affected By Canopy Opening But Not Detritus Deposition Following An Experimental Hurricane In A Tropical Rainforest, Paul D. Klawinski, Ben Dalton, Aaron B. Shiels

United States Department of Agriculture Wildlife Services: Staff Publications

Hurricanes, cyclones, and typhoons are common disturbances in many island and coastal forests. There is a lack of understanding of the importance to forest biota of the two major physical aspects that occur simultaneously during a hurricane: canopy disturbance and detritus (debris) deposition onto the ground. Using a replicated factorial design, our study involved experimentally determining the independent and interactive effects of canopy opening and debris additions to the forest floor on densities of coqui frogs (Eleutherodactylus coqui). Coquies are the dominant amphibian, and second most common vertebrate species, in the Luquillo Experimental Forest (LEF), a montane, tropical …

Background: Wild raccoons have been shown to be naturally exposed to avian influenza viruses (AIV). However, the mechanisms associated with these natural exposures are not well-understood.

Methodology/Principal Findings: We experimentally tested three alternative routes (water, eggs, and scavenged waterfowl carcasses) of AIV transmission that may explain how raccoons in the wild are exposed to AIV. Raccoons were exposed to 1) water and 2) eggs spiked with an AIV (H4N6), as well as 3) mallard carcasses experimentally inoculated with the same virus. Three of four raccoons exposed to the high dose water treatment yielded apparent nasal shedding of >102.0 PCR …

Eqtl Networks Reveal Complex Genetic Architecture In The Immature Soybean Seed, Yung-Tsi Bolon, D. Hyten, James H. Orf, Carroll P. Vance, Gary J. Muehlbauer

Department of Agronomy and Horticulture: Faculty Publications

The complex network of regulatory factors and interactions involved in transcriptional regulation within the seed is not well understood. To evaluate gene expression regulation in the immature seed, we utilized a genetical genomics approach on a soybean [Glycine max (L.) Merr.] recombinant inbred line (RIL) population and produced a genome-wide expression quantitative trait loci (eQTL) dataset. The validity of the dataset was confirmed by mapping the eQTL hotspot for flavonoid biosynthesis-related genes to a region containing repeats of chalcone synthase (CHS) genes known to correspond to the soybean inhibitor locus that regulates seed color. We then identified eQTL for …

Glyphosate-Resistant Giant Ragweed (Ambrosia Trifida) Control In Glufosinate-Resistant Soybean, Simranpreet Kaur, Lowell D. Sandell, John L. Lindquist, Amit J. Jhala

Department of Agronomy and Horticulture: Faculty Publications

Glyphosate-resistant giant ragweed is one of the most competitive weeds of agronomic crops in the United States. Early emergence and rapid growth rate makes giant ragweed a competitive weed early in the season and reduces crop yields. Therefore, early spring control of giant ragweed using a preplant herbicide is critical. Glufosinate is an alternative POST herbicide for weed control in glufosinate-resistant soybean. Field experiments were conducted at David City, NE, in 2012 and 2013 to evaluate the efficacy of preplant herbicides followed by glufosinate applied alone or in tank mixes for control of glyphosate-resistant giant ragweed in glufosinate-resistant soybean. Preplant …

Shattercane X Als-Tolerant Sorghum F1 Hybrid And Shattercane Interference In Als-Tolerant Sorghum, Rodrigo Werle, Jared J. Schmidt, John Laborde, Angela Tran, Cody F. Creech, John L. Lindquist

Department of Agronomy and Horticulture: Faculty Publications

ALS-tolerant grain sorghum cultivars are expected to be available for farmers within the next few years. Knowing that: (i) crosses between sorghum and shattercane are likely to occur resulting in crop-to-weed gene flow; (ii) ALS-susceptible shattercane X ALS-tolerant grain sorghum F1 hybrids (hybrids) were ultimately resistant to ALS-herbicides under field conditions; and (iii) hybrid fitness is equal to, or greater than, the wild parent, we conducted a greenhouse study to compare the competitive effect of shattercane and hybrid on sorghum, and whether or not herbicide application would influence the competitive ability of the hybrid …

Department of Agronomy and Horticulture: Faculty Publications

Phytophthora root rot (PRR) is a devastating disease in soybean [Glycine max (L.) Merr.] production. Michigan elite soybean E00003 is resistant to Phytophthora sojae and has been used as a resistance source in breeding. Genetic control of PRR resistance in this source is unknown. To facilitate marker-assisted selection (MAS), the PRR resistance loci in E00003 and their map locations need to be determined. In this study, a genetic mapping approach was used to identify major PRR -resistant loci in E00003. The mapping population consists of 240 F4–derived lines developed by crossing E00003 with the P. sojae susceptible …

Common bean (Phaseolus vulgaris L.) is the most important grain legume for human consumption and has a role in sustainable agriculture owing to its ability to fix atmospheric nitrogen. We assembled 473 Mb of the 587-Mb genome and genetically anchored 98% of this sequence in 11 chromosome-scale pseudomolecules. We compared the genome for the common bean against the soybean genome to find changes in soybean resulting from polyploidy. Using resequencing of 60 wild individuals and 100 landraces from the genetically differentiated Mesoamerican and Andean gene pools, we confirmed 2 independent domestications from genetic pools that diverged before human colonization. …

The Interaction Of Climate Change, Land Cover, And Political Representation In The Usa, Brady W. Allred, Dirac L. Twidwell, Samuel D. Fuhlendorf

Department of Agronomy and Horticulture: Faculty Publications

The difficulties in tackling climate change are inherently complex and primarily centered on political and social values. This is evident in the United States where political divisions and polarizations are fundamental barriers to advancing national policies, which in turn hinder international agreements, mitigation, and adaptation. Within the United States, the vast majority of agricultural and natural resource lands are projected to incur significant climate departures and are represented by the Republican Party. The resources and economic sectors that will be directly affected by climate change are represented by national leadership that is unlikely to accept policies to prevent or adapt …

Porocercospora Seminalis Gen. Et Comb. Nov., The Causal Organism Of Buffalograss False Smut, Bimal S. Amaradasa, Hugo Madrid, Johannes Z. Groenewald, Pedro W. Crous, Keenan L. Amundsen

Department of Agronomy and Horticulture: Faculty Publications

False smut caused by Cercospora seminalis is an important disease of buffalograss (Buchloe dactyloides) affecting seed production. The pathogen prevents normal caryopsis development and causes considerable yield loss and reduced seed germination. The current taxonomic placement of the false-smut causal pathogen in the genus Cercospora is incorrect based on its morphological characteristics and DNA phylogeny. In the present study the phylogenetic position of C. seminalis is clarified based on DNA sequence analysis of three loci namely the internal transcribed spacer (ITS) region, partial nuclear ribosomal large subunit (LSU) and partial sequences of the RNA polymerase II second largest …

Drought conditions in 2012 were some of the most severe in recent history. The purpose of this study is to examine the impact of drought on quality, quantity, and theoretical ethanol yield (TEY) of three bioenergy feedstocks, corn stover, mixed grasses from Conservation Reserve Program lands, and Miscanthus × giganteus. To assess drought effects on these feedstocks, samples from 2010 (minimal to no drought) and 2012 (severe drought) were compared from multiple locations in the US. In all feedstocks, drought significantly increased extractives and reduced structural sugars and lignin; subsequently, TEYs were reduced 10–15%. Biomass yields were significantly reduced …

Triacylglycerol (TAG) metabolism is a key aspect of intracellular lipid homeostasis in yeast and mammals, but its role in vegetative tissues of plants remains poorly defined. We previously reported that PHOSPHOLIPID:DIACYLGLYCEROL ACYLTRANSFERASE1 (PDAT1) is crucial for diverting fatty acids (FAs) from membrane lipid synthesis to TAG and thereby protecting against FA-induced cell death in leaves. Here, we show that overexpression of PDAT1 enhances the turnover of FAs in leaf lipids. Using the trigalactosyldiacylglycerol1-1 (tgd1-1) mutant, which displays substantially enhanced PDAT1- mediated TAG synthesis, we demonstrate that disruption of SUGAR-DEPENDENT1 (SDP1) TAG lipase or PEROXISOMAL TRANSPORTER1 (PXA1) severely decreases …

An Analysis Of A University Reclassification Effect On Applications Following A Move To A New Intercollegiate Athletic Association, Dylan P. Williams

LSU Doctoral Dissertations

Scholars have argued that athletics are utilized by universities to advertise their school to all individuals (Collins, 2012; Dwyer, Eddy, Havard, & Braa, 2010; Toma & Cross, 1998; Washington & Ventresca, 2004; Weaver, 2010). Expectedly, university officials are willing to contribute resources in order to develop an effective athletics program to establish an institution’s legitimacy among other universities (Collins, 2012; Toma & Cross, 1998; Washington & Ventresca, 2004). One tactic employed by schools focuses on the process of athletic association reclassification into the National Collegiate Athletic Association (NCAA) from other member associations (e.g., National Association for Intercollegiate Athletics). Officials consider …

Numerical Study Of Lipid Translocation Driven By Nanoporation Due To Multiple High-Intensity, Ultrashort Electrical Pulses, Viswanadham Sridhara, Ravindra P. Joshi

Electrical & Computer Engineering Faculty Publications

The dynamical translocation of lipids from one leaflet to another due to membrane permeabilization driven by nanosecond, high-intensity (>100 kV/cm) electrical pulses has been probed. Our simulations show that lipid molecules can translocate by diffusion through water-filled nanopores which form following high voltage application. Our focus is on multiple pulsing, and such simulations are relevant to gauge the time duration over which nanopores might remain open, and facilitate continued lipid translocations and membrane transport. Our results are indicative of a N1/2 scaling with pulse number for the pore radius. These results bode well for the use of pulse …

Evaluations Of A Mechanistic Hypothesis For The Influence Of Extracellular Ions On Electroporation Due To High-Intensity, Nanosecond Pulsing, V. Sridhara, R. P. Joshi

Electrical & Computer Engineering Faculty Publications

The effect of ions present in the extracellular medium on electroporation by high-intensity, short-duration pulsing is studied through molecular dynamic simulations. Our simulation results indicate that mobile ions in the medium might play a role in creating stronger local electric fields across membranes that then reinforce and strengthen electroporation. Much faster pore formation is predicted in higher conductivity media. However, the impact of extracellular conductivity on cellular inflows, which depend on transport processes such as electrophoresis, could be different as discussed here. Our simulation results also show that interactions between cations (Na+ in this case) and the carbonyl oxygen of …

Identification Of Differentially Expressed Genes Between Sorghum Genotypes With Contrasting Nitrogen Stress Tolerance By Genome-Wide Transcriptional Profiling, Malleswari Gelli, Yongchao Duo, Anji Reddy Konda, Chi Zhang, David R. Holding, Ismail M. Dweikat

Department of Agronomy and Horticulture: Faculty Publications

Background: Sorghum is an important cereal crop, which requires large quantities of nitrogen fertilizer for achieving commercial yields. Identification of the genes responsible for low-N tolerance in sorghum will facilitate understanding of the molecular mechanisms of low-N tolerance, and also facilitate the genetic improvement of sorghum through marker-assisted selection or gene transformation. In this study we compared the transcriptomes of root tissues from seven sorghum genotypes having differential response to low-N stress.

Results: Illumina RNA-sequencing detected several common differentially expressed genes (DEGs) between four low-N tolerant sorghum genotypes (San Chi San, China17, KS78 and high-NUE bulk) and three …

Background: Association analysis is an alternative to conventional family-based methods to detect the location of gene(s) or quantitative trait loci (QTL) and provides relatively high resolution in terms of defining the genome position of a gene or QTL. Seed protein and oil concentration are quantitative traits which are determined by the interaction among many genes with small to moderate genetic effects and their interaction with the environment. In this study, a genome-wide association study (GWAS) was performed to identify quantitative trait loci (QTL) controlling seed protein and oil concentration in 298 soybean germplasm accessions exhibiting a wide range of …
