Ferns are the only major lineage of vascular plants not represented by a sequenced nuclear genome. This lack of genome sequence information significantly impedes our ability to understand and reconstruct genome evolution not only in ferns, but across all land plants. Azolla and Ceratopteris are ideal and complementary candidates to be the first ferns to have their nuclear genomes sequenced. They differ dramatically in genome size, life history, and habit, and thus represent the immense diversity of extant ferns. Together, this pair of genomes will facilitate myriad large-scale comparative analyses across ferns and all land plants. Here we review the …

Activity Of 3\Beta-Hydroxysteroid Dehydrogenase Associated With Progesterone Production In Bovine Granulosa Cells Cultured Under Different Concentrations Of Serum, Insulin-Like Growth Factor I, And Gonadotropin, Özkan Şi̇mşek, Monika Mihm

Turkish Journal of Veterinary & Animal Sciences

Three-\beta-hydroxysteroid dehydrogenase (3\beta-HSD) is the enzyme responsible for progesterone production. This study aimed to determine whether 3\beta-HSD activity can be shown to reflect progesterone production by granulosa cells cultured under different serum conditions and follicle stimulating hormone (FSH), luteinising hormone (LH), and insulin-like growth factor I (IGF-I) concentrations. Large bovine follicles were dissected from abattoir ovaries to recover granulosa cells. Cells were washed, stained for viability, and plated for 48 h in basic medium with or without 5% foetal calf serum (FCS). Subsequently, cells were exposed to FSH (1 ng/mL), LH (10 or 100 ng/mL), or FSH (1 ng/mL) + …

Inhibition Of Phosphodiesterase10a Attenuates Morphine-Induced Conditioned Place Preference, Ying Mu, Zhaoxiang Ren, Jia Jia, Bo Gao, Longtai Zheng, Eitan Friedman

Publications and Research

Background: Phosphodiesterase (PDE) 10A is selectively expressed in medium spiny neurons of the striatum. Nucleus accumbens (NAc) is a key region that mediates drug reward and addiction-related behaviors. To investigate the potential role of PDE10A in the reinforcement properties of morphine, we tested the effect of MP-10, a selective inhibitor of PDE10A, on acquisition, expression, and extinction of morphine-induced conditioned place preference (CPP).

Results: The results show that 2.5 mg/kg MP-10, administered subcutaneously, significantly inhibited the acquisition of morphine-induced CPP. The same dose of MP-10 alone did not result in the CPP. Moreover, MP-10 did not alter the expression of …

A Small Subset Of Fruitless Subesophageal Neurons Modulate Early Courtship In Drosophila, David Tran, Geoffrey Meissner, Rachael French, Bruce Baker Jan 2014

A Small Subset Of Fruitless Subesophageal Neurons Modulate Early Courtship In Drosophila, David Tran, Geoffrey Meissner, Rachael French, Bruce Baker

Faculty Publications, Biological Sciences

We show that a small subset of two to six subesophageal neurons, expressing the male products of the male courtship master regulator gene products fruitlessMale (fruM), are required in the early stages of the Drosophila melanogaster male courtship behavioral program. Loss of fruM expression or inhibition of synaptic transmission in these fruM(+) neurons results in delayed courtship initiation and a failure to progress to copulation primarily under visually-deficient conditions. We identify a fruM-dependent sexually dimorphic arborization in the tritocerebrum made by two of these neurons. Furthermore, these SOG neurons extend descending projections to the thorax and abdominal ganglia. These anatomical …

As the main witnesses of the ecological and economic impacts of invasions on ecosystems around the world, ecologists seek to provide the relevant science that informs managers about the potential for invasion of specific organisms in their region(s) of interest. Yet, the assorted literature that could inform such forecasts is rarely integrated to do so, and further, the diverse nature of the data available complicates synthesis and quantitative prediction. Here we present a set of analytical tools for synthesizing different levels of distributional and/or demographic data to produce meaningful assessments of invasion potential that can guide management at multiple phases …

Survival And Recovery Following Wildfire In The Southern Range Of The Coast Redwood Forest, Rachel Lazzeri-Aerts, Will Russell

Faculty Publications, Environmental Studies

Fire plays a central role in determining structure, composition, and recruitment in many forest types. In coast redwood forests, the role of fire is not well understood and scant literature exists on post-fire response, particularly in the southern part of the range. In order to better understand patterns of survival and recruitment following fire for coast redwood (Sequoia sempervirens [lamb. ex D. Don] Endl.) and associated tree species, three sites in the Santa Cruz Mountains, California, USA, were sampled following wildfire. Randomly selected 10 m diameter plots were used to collect data on survivorship and post fire regeneration in order …

Walking The Edge With Controversial Use Of Preimplantation Genetic Diagnosis (Pgd): Opinions And Attitudes Of Genetic Counselors, Kristen Everton

Theses and Dissertations

Purpose: This study explored opinions and attitudes of genetic counselors regarding three controversial applications of preimplantation genetic diagnosis (PGD): PGD for early-onset Alzheimer, use of embryos that are BRCA positive after PGD revealed no disease-free embryos to be available, and PGD to select against a variant of unknown significance (VUS) for Marfan syndrome. Methods: Genetic counselors were contacted through the National Society of Genetic Counselors (NSGC) electronic mailing list. Inclusion criteria required that a participant was currently practicing as a genetic counselor, was a member of the NSGC, and has counseled patients about PGD. Twenty-nine participants volunteered to participate and …

Identification Of Clinical Markers That Predict The Outcomes Of Methicillin-Resistant Staphylococcus Aureus Infections And Identification Of Synergistic Antibiotic Combinations For The Treatment Of These Infections, Virginia Long Jan 2014

Identification Of Clinical Markers That Predict The Outcomes Of Methicillin-Resistant Staphylococcus Aureus Infections And Identification Of Synergistic Antibiotic Combinations For The Treatment Of These Infections, Virginia Long

Master's Theses

Methicillin-resistant Staphylococcus aureus(MRSA) is a prominent nosocomial infection. Vancomycin is the mainstay of treatment, but it has a high rate of failure. Combination therapies can be used, but synergy is often not tested in clinical laboratories. Physicians are at a disadvantage because they cannot predict MRSA outcomes. The first aim of the study was to identify clinical and demographic markers that could predict infection recurrence or prolonged bacteremia. This was accomplished using retrospective chart review and statistical analysis. The second aim of the study was to identify which combinations, of those commonly used for MRSA treatment at Loyola, demonstrate the …

Catalytic Mechanism And Maturation Of The Metalloenzyme Nitrile Hydratase, Natalie I. Gumataotao Jan 2014

Catalytic Mechanism And Maturation Of The Metalloenzyme Nitrile Hydratase, Natalie I. Gumataotao

Dissertations

Nitrile hydratases are metalloenzymes that catalyze the hydration of nitriles to their corresponding amides in a specific manner at ambient pressures and temperatures at neutral pH. Traditional industrial methods require high temperature and pressure, extreme pH, and heavy metals. NHases are used as biocatalysts in the large scale industrial production of amide precursors to textiles, animal feedstock, and polymers. Notably, NHase is used in the production of ~100,000 tons of acrylamide annually by the Mitsubishi Corporation.

Despite being used extensively in industry, questions remain about NHase. The catalytic mechanism is not defined. Understanding the way in which the nitrile is …

Host Proteins Interact With The Hiv-1 Core To Facilitate And Restrict, Zana Lukic Jan 2014

Host Proteins Interact With The Hiv-1 Core To Facilitate And Restrict, Zana Lukic

Dissertations

Host cell proteins, termed restriction factors, which inhibit viral replication at various stages of the viral life cycle, determine the species-specific tropism of numerous retroviruses. Many members of the TRIM family of proteins act as viral restriction factors. One well-characterized example is the ability of TRIM5á from rhesus macaques (rhTRIM5á) to inhibit human immunodeficiency virus type-1 (HIV-1) soon after viral entry but prior to reverse transcription (RT). It is well established that the restriction requires an interaction between the viral capsid lattice and the B30.2/SPRY domain of TRIM5á. Following the binding of the viral core, TRIM5á mediates an event or …

Investigating The Role Of The Pgf2 Alpha/Calcineurin-Signaling Pathway In The Regulation Of Adipogenesis, Damodaran Annamalai Jan 2014

Investigating The Role Of The Pgf2 Alpha/Calcineurin-Signaling Pathway In The Regulation Of Adipogenesis, Damodaran Annamalai

Dissertations

Prostaglandin F2α (PGF2α) is a potent physiological inhibitor of adipocyte differentiation. In previous studies, we demonstrated that PGF2α inhibits adipogenesis via activation of the calcium-regulated protein phosphatase, calcineurin. In this current study, we have now extended these findings to identify the IL-11 cytokine and the Nurr1 orphan nuclear hormone receptor as functionally important downstream transcriptional targets of the PGF2α/calcineurin-pathway involved in the inhibition of adipocyte differentiation. In the case of IL-11, we show that this cytokine acts in an autocrine fashion to inhibit adipogenesis via the essential actions of the gp130 cytokine co-receptor signaling subunit. Further, by using a well-characterized …

Adenovirus Vectors As Potent Adjuvants In Vaccine Development, Kathleen Ann Mcguire Jan 2014

Adenovirus Vectors As Potent Adjuvants In Vaccine Development, Kathleen Ann Mcguire

Dissertations

Due to their ability to activate the immune system, replication-defective Adenoviruses (Ad) are potential vaccine vectors for several pathogens. The proinflammatory response to Ad contributes to the response to vaccine antigens. We found that reactive oxygen species (ROS) are an important signal in the proinflammatory response to Ad. We identified that serotype 5 adenovirus (Ad5) elicits ROS by inducing mitochondrial membrane damage, a process that is dependent on endosomal membrane rupture and Cathepsin release. This mitochondrial dysfunction contributes to NLRP3 inflammasome- and NFκB-dependent innate immune activation. The ROS-dependent inflammatory response likely contributes to the adaptive immune response by supporting DC …

The Molecular Components Of Estrogen Receptor Beta (Erβ) Signaling In Neuronal Sytems, Natasha Mott Jan 2014

The Molecular Components Of Estrogen Receptor Beta (Erβ) Signaling In Neuronal Sytems, Natasha Mott

Dissertations

With increasing life expectancy, women are now living upwards of 50 years without circulating estrogens, therefore, it is essential to investigate how the brain is changed by estrogen deprivation and also how aging influences these changes. The Women's Health Initiative (WHI) study spurred rigorous debate regarding estrogen therapy for postmenopausal women due to dichotomous effects of estrogens in menopausal and post-menopausal women. Meta-analyses of the WHI study revealed that after circulating estrogens are depleted for many years re-exposure may cause aberrant, negative health effects, indicating that there is an age-related `switch' in estrogen signaling around menopause. These age-related effects of …

Unveiling The Impact Of Human Influence On Species Distributions In Vietnam: A Case Study Using Babblers (Aves: Timaliidae), Laurel R. Yohe, Johnathan Flanders, Hoang Minh Duc, Long Vu, Sushma Reddy Jan 2014

Unveiling The Impact Of Human Influence On Species Distributions In Vietnam: A Case Study Using Babblers (Aves: Timaliidae), Laurel R. Yohe, Johnathan Flanders, Hoang Minh Duc, Long Vu, Sushma Reddy

Biology: Faculty Publications and Other Works

As developing countries give priority to economic growth, the effects of development threaten natural habitats and species distributions. Over the course of two decades, Vietnam has rapidly developed, especially in the expansion of agricultural production. However, no study has quantitatively measured the effects of recent human impact on the effects of past species distributions in Vietnam. We use locality data collected from multiple natural history collections, including several in Vietnam, to infer past species distributions. We assess habitat availability of five common babbler species (Aves: Timaliidae) using distribution models with data prior to rapid development that followed political reform. Overlaying …

The Effect Of Dietary Fat Intake On Hepatic Gene Expression In Lg/J And Sm/J Mice, Charlyn G. Partridge, Gloria L. Fawcett, Bing Wang, Clay F. Semenkovich, James M. Cheverud Jan 2014

The Effect Of Dietary Fat Intake On Hepatic Gene Expression In Lg/J And Sm/J Mice, Charlyn G. Partridge, Gloria L. Fawcett, Bing Wang, Clay F. Semenkovich, James M. Cheverud

Biology: Faculty Publications and Other Works

Background

The liver plays a major role in regulating metabolic homeostasis and is vital for nutrient metabolism. Identifying the genetic factors regulating these processes could lead to a greater understanding of how liver function responds to a high-fat diet and how that response may influence susceptibilities to obesity and metabolic syndrome. In this study we examine differences in hepatic gene expression between the LG/J and SM/J inbred mouse strains and how gene expression in these strains is affected by high-fat diet. LG/J and SM/J are known to differ in their responses to a high-fat diet for a variety of obesity- …

A Framework For A Marine Biodiversity Observing Network Within Changing Continental Shelf Seascapes, Frank Muller-Karger, Maria Kavanaugh, Enrique Montes, William Balch, Mya Breitbart

Marine Science Faculty Publications

Continental shelves and the waters overlying them support numerous industries as diverse as tourism and recreation, energy extraction, fisheries, transportation, and applications of marine bio-molecules (e.g., agribusiness, food processing, pharmaceuticals). Although these shelf ecosystems exhibit impacts of climate change and increased human use of resources (Halpern et al., 2012; IPCC, 2013, 2014; Melillo et al., 2014), there are currently no standardized metrics for assessing changes in ecological function in the coastal ocean. Here, we argue that it is possible to monitor vital signs of ecosystem function by focusing on the lowest levels of the ocean food web. Establishment of biodiversity, …

How Do Patients With Advanced Cancer Cope With An Uncertain Disease Trajectory? Implications For Grief Counselling, Elizabeth A. Lobb Jan 2014

How Do Patients With Advanced Cancer Cope With An Uncertain Disease Trajectory? Implications For Grief Counselling, Elizabeth A. Lobb

Health Sciences Papers and Journal Articles

A growing number of cancer patients are living longer with incurable disease. This paper describes strategies that patients use to cope with the uncertain trajectory of their disease. Twenty-seven patients with a prognosis of 12 months were recruited from the oncology and palliative care service at three metropolitan Sydney hospitals. A semi-structured face-to-face interview was conducted, which was audiotaped and transcribed verbatim. The patients coped with the uncertain trajectory of their disease through avoidance, maintaining a normal life, comparing themselves favourably with others in a similar situation and remaining positive. Participants indicated that they did not wish referral for psychological …

An Eof-Based Algorithm To Estimate Chlorophyll A Concentrations In Taihu Lake From Modis Land-Band Measurements: Implications For Near Real-Time Applications And Forecasting Models, Lin Qi, Chuanmin Hu, Hongtao Duan, Brian B. Barnes, Ronghua Ma

Marine Science Faculty Publications

For near real-time water applications, the Moderate Resolution Imaging Spectroradiometers (MODIS) on Terra and Aqua are currently the only satellite instruments that can provide well-calibrated top-of-atmosphere (TOA) radiance data over the global aquatic environments. However, TOA radiance data in the MODIS ocean bands over turbid atmosphere in east China often saturate, leaving only four land bands to use. In this study, an approach based on Empirical Orthogonal Function (EOF) analysis has been developed and validated to estimate chlorophyll a concentrations (Chla, μg/L) in surface waters of Taihu Lake, the third largest freshwater lake in China. The EOF approach analyzed the …

Elucidating The Transcriptional Regulation Of Col2a1a In Zebrafish, Sonja Dabizljevic Jan 2014

Elucidating The Transcriptional Regulation Of Col2a1a In Zebrafish, Sonja Dabizljevic

Master's Theses

Cartilage is an important tissue in vertebrates beginning in the early embryo where it lays down the scaffolding for the skeleton, and continuing through adulthood where it makes up joints and intervertebral disks. The major component of cartilage is a filamentous protein known as Collagen type II, alpha 1 (Col2a1). Mutations in col2a1 in humans can lead to multiple congenital disorders and the early onset of joint and retinal deterioration. Our laboratory is interested in understanding the conserved transcriptional regulation of this critical structural gene utilizing the zebrafish, Danio rerio. Previously, we identified a 310 bp regulatory region (R2) 1.7 …

Life Histories, Diets, And Secondary Production Of Odonata Along A Temperature Gradient On The Copper River Delta, Alaska, Nicole Emilie Furlan Jan 2014

Life Histories, Diets, And Secondary Production Of Odonata Along A Temperature Gradient On The Copper River Delta, Alaska, Nicole Emilie Furlan

Master's Theses

Dragonflies (Odonata: Epiprocta) and damselflies (Odonata: Zygoptera) are a conspicuous aspect of the biota in ponds on southcentral Alaska's Copper River Delta (CRD). Odonate densities, secondary production, and diets were assessed in sixteen ponds classified by delta region (east vs. west) and landscape type (outwash plain (OP) vs uplifted marsh (UM)).

Enallagma boreale (Coenagrionidae) comprised 48.5% of collected odonates. Leucorrhinia hudsonica (Libellulidae) and Aeshna juncea (Aeshnidae) comprised 36.6% and 10.4% of collected odonates, respectively. L. hudsonica densities and secondary production were significantly higher (p<0.001) in west UM ponds than in other pond types. Ostracods (Ostracoda) and water boatmen (Corixidae) dominated west OP A. juncea diets. Midge larvae (Chironomidae) dominated A. juncea diets in remaining pond types, occurring in 68% of foreguts. 27% of A. juncea foreguts demonstrated intraguild predation, and 6% of foreguts demonstrated cannibalism. Foreguts containing threespine stickleback (Gasterosteus aculeatus) revealed A. juncea's apex predator role in CRD ponds.

Reconsidering The Model Of Trim5Α Assembly: The Role Of The Linker2 (L2) Region In Trim5Α Assembly, Laura Johnsen

Master's Theses

The TRIM5α protein from rhesus macaques (TRIM5αrh) exhibits a remarkable ability to potently inhibit infection by Human Immunodeficiency Virus Type-1 (HIV-1). Extensive studies have shown that TRIM5α is capable of self-associating at many levels, eventually leading to the formation of a hexameric assembly that can superimpose on the hexameric lattice of the HIV-1 capsid. The mechanism underlying the self-association of TRIM5α and the molecular determinants of self-association remain to be completely understood. In this study, we show that the Linker 2 (L2) region of TRIM5rh is important for dimerization and higher order self-association, both of which are independent processes. Additionally, …

Characterizing The Mechanisms By Which Community Associated Methicillin-Resistant Staphylococcus Aureus Influences Keratinocyte Innate Immune Responses During Recurrent Infection, Ashley Lynn Larm Jan 2014

Characterizing The Mechanisms By Which Community Associated Methicillin-Resistant Staphylococcus Aureus Influences Keratinocyte Innate Immune Responses During Recurrent Infection, Ashley Lynn Larm

Master's Theses

Community associated–methicillin resistant Staphylococcus aureus (CA–MRSA) infection has become a major health concern. In human epidermal keratinocytes, S. aureus is mainly recognized through toll–like receptor 2 (TLR2) and its co–receptor, CD14. We hypothesize that CA–MRSA isolates cause recurrent infections by interrupting TLR2–mediated inflammation in keratinocytes. Recurrent CA–MRSA bacterial culture supernatant exposure to keratinocytes in vitro resulted in significant decreases in pro and anti–inflammatory cytokine and HMGB1 secretion from keratinocytes as assessed by ELISAs. Recurrent CA–MRSA live infection did not result in significant changes in cytokine or HMGB1 secretion, surface receptor expression, or NFκB activation post infection as assessed by ELISA …

History Of Freshwater Invasions In Illinois: Learning From The Past To Inform The Future, Abigail Jacobs Jan 2014

History Of Freshwater Invasions In Illinois: Learning From The Past To Inform The Future, Abigail Jacobs

Master's Theses

Rates of introduction and spread of non-native species continue to increase worldwide, with freshwater ecosystems highly impacted. Many non-native species crossed between the Great Lakes and Mississippi watersheds by traveling through waterways in Illinois. I assembled a comprehensive database of occurrences of aquatic non-native species (n=99) in Illinois inland waters. The arrival of non-native species accelerated since 1873. The Great Lakes Basin was the main source of established species into Illinois. From 1990-2012 the dominant vector was unintentional release of established species non-native to North America. Sixty of the 99 introduced species established. Eighteen established species had high or very …
