Model Studies Towards The Total Synthesis Of Lyconadin A Via An Acyl Radical Cascade Reaction, Koudi Zhu Jun 2006

Model Studies Towards The Total Synthesis Of Lyconadin A Via An Acyl Radical Cascade Reaction, Koudi Zhu

Theses and Dissertations

Lyconadin A is an alkaloid possessing a unique structure and antitumor activity. The total synthesis of Lyconadin A was proposed via an acyl radical cascade reaction. To investigate the possibility and stereoselectivity of the cascade cyclization, phenyl selenoester 16 was chosen as a model substrate to study the 7-exo-5-exo radical cyclization. A synthetic route to phenyl selenoester 16 was developed. The 7-exo-5-exo radical cyclization was found to occur with a high yield and excellent stereoselectivty. Attempts were also tried to synthesize another radical precursor 14 albeit with less success. A synthetic pathway to the synthesis of 14 as well as …

"Preface" To Selenium: Its Molecular Biology And Role In Human Health, Second Edition, Dolph L. Hatfield, Marla J. Berry, Vadim N. Gladyshev

Vadim Gladyshev Publications

The purpose of the new edition book is to inform the reader of many new discoveries and to examine our present knowledge of the molecular biology of selenium, its incorporation into proteins as selenocysteine and the role that this element and selenium-containing proteins (selenoproteins) play in health. The book's emphasis is on our understanding of selenium metabolism in mammals and the role of this element in human health. The book begins with a brief history of selenium and how its face has changed through the years fiom one of a toxin and possible carcinogen to one of an essential micronutrient …

Vadim Gladyshev Publications

Mouse models have been generated to assess the roles of selenoproteins involved with housekeeping tasks and/or stress-related phenomena in development and health. Each mouse model has taken advantage of the fact that the synthesis of all selenoproteins is dependent on the expression of two selenocysteine (Sec) tRNA[Ser]Sec isoforms that differ fiom each other by a single methyl group on the ribosyl moiety at position 34. The endogenous (Sec) tRNA[Ser]Sec population was selectively altered by generating mouse models involving 1) transgenic animals carrying mutant or wild type (Sec) tRNA[Ser]Sec transgenes, 2) conditional knockout animals carrying a floxed (Sec) tRNA[Ser]Sec gene that …

Selenoproteins And Selenoproteomes, Vadim N. Gladyshev Jun 2006

Selenoproteins And Selenoproteomes, Vadim N. Gladyshev

Vadim Gladyshev Publications

In the past several years, progress in genome sequencing and development of specialized bioinformatics tools allowed efficient identification of selenocysteine-containing proteins encoded in completely sequenced genomes. Information is currently available on selenoproteomes from a variety of organisms, including humans, which contain 25 known selenoprotein genes. This review provides basic information about mammalian selenoproteins and other known selenoprotein families. Analysis of full sets of selenoproteins in organisms provides exciting avenues for examining selenoprotein evolution and dependence of organisms on the trace element selenium and allows linking selenoproteins with specific biological and biomedical effects of dietary selenium.

Selenium And Methionine Sulfoxide Reduction, Hwa-Young Kim, Vadim N. Gladyshev Jun 2006

Selenium And Methionine Sulfoxide Reduction, Hwa-Young Kim, Vadim N. Gladyshev

Vadim Gladyshev Publications

Methionine residues in proteins can be readily oxidized to a diastereomeric mixture of methionine sulfoxides by reactive oxygen species. In most organisms, methionine sulfoxides are reversibly and stereospecifically reduced back to methionine by two distinct classes of repair enzymes, methionine-S-sulfoxide reductase (MsrA) and methionine-R-sulfoxide reductase (MsrB). Methionine sulfoxide reduction is thought to be an essential pathway that protects cells from oxidative stress and regulates protein function. This pathway is also implicated in delaying the aging process in organisms from yeast to mammals. The first selenoprotein identified using bioinformatics methods, SelR (also known as SelX or MsrB1), was recently found to …

Evolution Of Selenocysteine Decoding And The Key Role Of Selenophosphate Synthetase In The Pathway Of Selenium Utilization, Gustavo Salinas, Héctor Romero, Xue-Ming Xu, Bradley A. Carlson, Dolph L. Hatfield, Vadim N. Gladyshev Jun 2006

Gladyshev

Vadim Gladyshev Publications

The complete sequencing of genomes and the development of in silico methods for identification of genes encoding selenocysteine (Sec)-containing proteins have greatly contributed to shape our view on the evolution of selenium utilization in nature. Current evidence is consistent with the idea that Sec decoding is a late addition to the genetic code and it evolved once, before the separation of archaeal, bacterial and eukaryal domains. Many organisms have lost the Sec decoding trait, but recent evidence has shown that the loss is not irreversible. The distribution of organisms that use UGA as a Sec codon suggests that Sec decoding …

Selenoproteins In Parasites, Gustavo Salinas, Alexei V. Lobanov, Vadim N. Gladyshev Jun 2006

Selenoproteins In Parasites, Gustavo Salinas, Alexei V. Lobanov, Vadim N. Gladyshev

Vadim Gladyshev Publications

Parasites, which cause an enormous burden in the population of the third world, are a diverse group of organisms, many of which are sensitive to oxidative stress imposed by their hosts. In recent years, several selenoprotein families, some with antioxidant properties, have been described and characterized in metazoan parasites. Glutathione peroxidase and thioredoxin glutathione reductase (TGR) appear to be essential selenoproteins in flatworms (phylum Platyhelminthes). TGR is the single enzyme that provides reducing equivalents to both thioredoxin and glutathione pathways, in contrast to hosts, which evolve parallel pathways. In roundworms (phylum Nematoda), selenoproteins have recently been described, revealing …

The 15-Kda Selenoprotein (Sep15): Functional Analysis And Role In Cancer, Vyacheslav M. Labunskyy, Vadim N. Gladyshev, Dolph L. Hatfield

Vadim Gladyshev Publications

The 15-kDa selenoprotein (Sep15) was identified several years ago as a protein of unknown function. In recent years, several lines of evidence implicated Sepl5 in the effect of dietary selenium in cancer prevention. These lines of evidence include: 1) protein expression patterns in normal and malignant cells; 2) identification of polymorphic sites that regulate Sep15 levels and differentially respond to selenium supplementation; 3) location of the Sep15 gene in the human genome; and 4) correlation between Sep15 haplotype and susceptibility to cancer. Functional analyses revealed a specific interaction between Sep15 and a protein folding sensor in the endoplasmic reticulum of …

Vadim Gladyshev Publications

Selenocysteine (Sec) tRNA occupies a prominent position in the expression of selenoproteins as it is essential for their synthesis and it provides the means by which selenium is co-translationally inserted into protein as the amino acid, Sec. Thus, Sec tRNA is regarded as the principle constituent in selenoprotein synthesis. Many features unique to this tRNA have been characterized over the years in mammals and other eukaryotes. In the last five years, the major advances have been in an elucidation of the different roles that the two major Sec tRNA isoforms play in selenoprotein biosynthesis and in Sec biosynthesis. One isoform …

Perspective Greenhouse Gas Mitigation By Covers On Livestock Slurry Tanks And Lagoons?, Soren O. Petersen, Daniel N. Miller

Department of Animal Science: Faculty Publications

Liquid manure (slurry) storage facilities are important point sources of atmospheric pollution. Being point sources, containment of gaseous emissions via improved storage conditions may be possible, and permeable surface covers (natural crusts and artificial covers) are increasingly recognized for their capacity to reduce various gaseous emissions. Microbial transformations in permeable surface covers include bacterial-methane oxidation, but this interface between nitrogen-rich slurry and the atmosphere is also an environment with intense nitrogen turnover that can lead to nitrous oxide emissions. Both methane and nitrous oxide are greenhouse gases, and strategies to reduce environmental impact of slurry stores must consider the total …

Anadromous Rainbow Smelt And Tomcod In Connecticut: Assessment Of Populations, Conservation Status, And Need For Restoration Plan, Heather A. Fried, Eric T. Schultz Jun 2006

Anadromous Rainbow Smelt And Tomcod In Connecticut: Assessment Of Populations, Conservation Status, And Need For Restoration Plan, Heather A. Fried, Eric T. Schultz

EEB Articles

(beginning of rainbow smelt executive summary)

Evidence indicates that anadromous rainbow smelt (Osmerus mordax) populations in Connecticut and elsewhere in the northeast United States have severely declined. Several sampling programs have documented declines in Connecticut’s smelt populations over the last three decades (Marcy 1976a, Marcy 1976b, Millstone Environmental Laboratory 2005). Similar declines have also been documented in the Hudson River (ASA Analysis & Communication 2005) and in Massachusetts (personal communication, Brad Chase, MA Division of Marine Fisheries 2004). Recreational and commercial fisheries in the region for this species have virtually ceased (Blake and Smith 1984). The Connecticut Fish Advisory Committee …

A Short Tutorial On Reconstructability Analysis, Martin Zwick Jun 2006

A Short Tutorial On Reconstructability Analysis, Martin Zwick

Complex Systems Faculty Publications and Presentations

This talk will present the basic ideas of Reconstructability Analysis. RA is a general modeling methodology developed in the systems community since the early 60s (pioneered by Ross Ashby) for exploratory (data mining, machine learning) and confirmatory modeling. RA is based in graph theory, set theory, information theory, and statistics. It is applicable to qualitative (nominal) and, via binning, to quantitative (continuous) data, with both statistical & non-statistical applications. It overlaps and augments more widely known methods, e.g., log-linear modeling and Bayesian networks.

Keywords: Reconstructability Analysis, Log-Linear Modeling, categorical data, multivariate statistics, exploratory modeling, confirmatory modeling

Multivariate Analysis And Visualization Of Splicing Correlations In Single-Gene Transcriptomes, Mark C. Emerick, Giovanni Parmigiani, William S. Agnew Jun 2006

Multivariate Analysis And Visualization Of Splicing Correlations In Single-Gene Transcriptomes, Mark C. Emerick, Giovanni Parmigiani, William S. Agnew

Johns Hopkins University, Dept. of Biostatistics Working Papers

Through ‘combinatorial splicing’, RNA metabolism may create enormous structural diversity in the proteome. Functional interactions among multiple alternative domains can have a disproportionate impact on the phenotype, requiring integrated RNA-level regulation of molecular composition. Splicing correlations within molecules expressed from a single gene, where these effects would be greatest, provide valuable clues to functional relationships and targets for splicing regulation. We present tools to visualize complex splicing patterns in full-length cDNA libraries. Developmental changes in pair-wise correlations are presented vectorially in ‘clock plots’ and linkage grids. Higher-order correlations are assessed via a loglinear model and Monte Carlo analysis with an …

Red Rock Desert Learning Center: Evaluation Of Curriculum: Final Report, Lori Olafson, Gregory Schraw

Reports (RRLC)

The UNLV Center for Evaluation and Assessment developed an assessment instrument to evaluate curriculum for the proposed Red Rock Desert Learning Center science school’s fifth grade program. A 25-item Likert-type scale was created and administered before and after 68 pre-service and in-service teachers participated in each of five curriculum pilot sessions. The survey consisted of questions related to knowledge, pedagogy, and attitudes. Two open-ended questions were included on the post-survey.

Findings revealed significant gains in knowledge, pedagogy, and attitudes in each of five events from pre- to post-test. Gains over time were greatest for knowledge. Some events were rated more …

Nested Deletion Analysis Of Wheat Streak Mosaic Virus Hc-Pro: Mapping Of Domains Affecting Polyprotein Processing And Eriophyid Mite Transmission, Drake C. Stenger, Gary L. Hein, Roy C. French Jun 2006

Nested Deletion Analysis Of Wheat Streak Mosaic Virus Hc-Pro: Mapping Of Domains Affecting Polyprotein Processing And Eriophyid Mite Transmission, Drake C. Stenger, Gary L. Hein, Roy C. French

Department of Plant Pathology: Faculty Publications

A series of in-frame and nested deletion mutations which progressively removed 5′-proximal sequences of the Wheat streak mosaic virus (WSMV) HC-Pro coding region (1152 nucleotides) was constructed and evaluated for pathogenicity to wheat. WSMV HC-Pro mutants with 5′- proximal deletions of 12 to 720 nucleotides systemically infected wheat. Boundary sequences flanking the deletions were stable and unaltered by passage through plants for all deletion mutants except HCD12 (lacking HC-Pro codons 3–6) that exhibited strong bias for G to A substitution at nucleotide 1190 in HC-Pro codon 2 (aspartic acid to asparagine). HC-Pro mutants with 5′-proximal deletions of up to 720 …

The Effect Of Male Competition On Female Choice In Brandt’S Vole, Lasiopodomys Brandti, Jianjun Zhang, Dazhao Shi, Lixing Sun

All Faculty Scholarship for the College of the Sciences

The effect of male competition on female choice in the Brandt’s vole (Lasiopodomys brandti) was studied in the laboratory. Using a two-way choice test, we showed that when males did not compete, estrous females preferred and mated with the dominant males, but non-estrous females preferred subordinate males. However, when males competed, all dominant males gained the priority to mate, and the amicable behaviour between males, regardless of their dominant status, and between non-estrous females increased, compared with the behaviour between males and the estrous females. These results showed that interactions among Brandt’s voles were not only influenced by …
