Total and intra-mitochondrial gonadal cholesterol concentrations are decreased in fish exposed to the phytoestrogen beta-sitosterol (beta-sit). The present study examined the potential for beta-sit to disrupt de novo cholesterol synthesis in the gonads of goldfish exposed to 200 microgram/g beta-sit and 10 microgram/g 17beta-estradiol (E2; estrogenic control) by intra-peritoneal Silastic® implants for 21 days. The de novo cholesterol synthetic capacity was estimated by incubating gonadal tissue with 14C-acetate for a period of 18 hours, followed by chloroform/methanol lipid extraction and thin layer chromatography (TLC) lipid separation. Lipid classes were confirmed using infrared spectroscopy. Plasma testosterone (T) and total cholesterol concentration …

Effects Of Cheatgrass Control On Wyoming Big Sagebrush In Southeastern Utah, Daniel Blaine Eddington
Effects Of Cheatgrass Control On Wyoming Big Sagebrush In Southeastern Utah, Daniel Blaine Eddington
Theses and Dissertations
Critical mule deer (Odocoileus hemionus) winter ranges in southeastern Utah dominated by Wyoming big sagebrush (Artemisia tridentata var. wyomingensis [Beetle and A. Young] Welsh) have developed dense cheatgrass (Bromus tectorum L.) understories. These communities are currently characterized by predominately mature to decadent stands of sagebrush with few perennial grasses and forbs. Sagebrush seedlings and perennial grasses compete for limited resources with annual grasses and forbs. To determine the affects of cheatgrass control on sagebrush growth and reproductive characteristics, imazapic (PLATEAU®, AC 263,222) herbicide was sprayed at 438.5 ml/ha with water and methylated seed oil during active fall growth of cheatgrass …

Effect Of Prebiotic Oligosaccharides On Enteropathogenic Escherichia Coli Adherence, Kari Shoaf
Effect Of Prebiotic Oligosaccharides On Enteropathogenic Escherichia Coli Adherence, Kari Shoaf
Department of Food Science and Technology: Dissertations, Theses, and Student Research
Antiadhesion therapy has been found to be a very promising means to prevent and treat infections in the respiratory, urinary, and gastrointestinal tract. Despite the considerable research interest in antiadhesives, relatively little is known about the ability of commercial dietary oligosaccharides, such as prebiotics, to inhibit pathogen adherence. The goal of this research was to determine if commercially available prebiotic oligosaccharides could function as antiadhesive agents against enteropathogenic E. coli (EPEC).
To investigate the ability of prebiotics to inhibit EPEC adherence, inhibition assays were performed by comparing adherence rates of EPEC on HEp-2 and Caco-2 cells in the presence of …

MicroRNAs (miRNAs) are capable of controlling gene expression by targeting complimentary sequences in many mRNAs. Thus, a small number of miRNAs are capable of regulating expression of many different genes. miRNAs have been found in all animals from Drosophila to human and they are highly conserved. This work was undertaken in order to identify such RNAs in the newt eye.

The sex pheromone of the scarab beetle, Phyllophaga anxia, is a blend of the methyl esters of two amino acids, L-valine and L-isoleucine. A field trapping study was conducted, deploying different blends of the two compounds at 59 locations in the United States and Canada. More than 57,000 males of 61 Phyllophaga species (Coleoptera: Scarabaeidae: Melolonthinae) were captured and identified. Three major findings included: (1) widespread use of the two compounds [of the 147 Phyllophaga (sensu stricto) species found in the United States and Canada, males of nearly 40% were captured]; (2) in most species intraspecific male …
Capillary Electrophoresis Of Proteins With Selective On-Line Affinity Monoliths, Jenny Marcela Armenta Blanco
Capillary Electrophoresis Of Proteins With Selective On-Line Affinity Monoliths, Jenny Marcela Armenta Blanco
Theses and Dissertations
The analysis of proteins in biological fluids by capillary electrophoresis (CE) is of interest in clinical chemistry. However, due to low analyte concentrations and poor concentration limits of detection (CLOD), protein analysis by this technique is frequently challenging. Coupling preconcentration techniques with CE greatly improves the CLOD. An on-line preconcentration-CE method that can selectively preconcentrate any protein for which an antibody is available would be very useful for the analysis of low abundance proteins and would establish CE as a major tool in biomarker discovery. To accomplish this, an on-line protein G monolithic preconcentrator CE system for enrichment and separation …
Comparative Rates Of Ndna Degradation Using Rat Models In Southeastern Tennessee, Matthew F. Pullen
Comparative Rates Of Ndna Degradation Using Rat Models In Southeastern Tennessee, Matthew F. Pullen
Honors Theses
The advent of DNA identification methods has revolutionized forensic science. It is now possible to identify criminals from trace evidence, eliminate false leads, and discover the identity of a deceased individual long after their death. As DNA plays such important roles in criminal investigatious, determining the quality of DNA due to its exposure time enviroment is imperative. This experiment seeks to help narrow down the time frame that useful nuclear DNA (nDNA) can be extracted from a decaying body. The rate of DNA degradation was measured in Sprague-Dawlwy rat models placed in varied environmnts of Southeastern Tennessee. The environments included …

We have generated monthly resolved, stable isotope (δ18O and δ13C) and Sr/Ca time series from a massive Porites coral from Rabaul (4°S, 152°E): a site located in the warmest sector of the Western Pacific Warm Pool (WPWP). The coral δ18O and Sr/Ca time series are well correlated to each other and positive excursions in both records coincide with times of ENSO warm phase events. These time series contain abundant interannual variability that exhibits the well‐recognized pattern of low amplitude ENSO variation between ∼1920–1960 and high amplitude ENSO variation between 1880–1920 and 1960–1997. Vulnerability Of Pathogenic Biofilms To Micavibrio Aeruginosavorus, Daniel Kadouri, Nel C. Venzon, George A. O'Toole
Dartmouth Scholarship
The host specificity of the gram-negative exoparasitic predatory bacterium Micavibrio aeruginosavorus was examined. M. aeruginosavorus preyed on Pseudomonas aeruginosa, as previously reported, as well as Burkholderia cepacia, Klebsiella pneumoniae, and numerous clinical isolates of these species. In a static assay, a reduction in biofilm biomass was observed as early as 3 hours after exposure to M. aeruginosavorus, and an ∼100-fold reduction in biofilm cell viability was detected following a a 24-h exposure to the predator. We observed that an initial titer of Micavibrio as low as 10 PFU/well or a time of exposure to the predator as short as 30 …

Kinesiology and Public Health
We explored the relationships among girls’ weight status, dairy servings, and total energy intake. The hypothesis that consuming dairy could reduce risk for overweight was evaluated by comparing energy intake and weight status of girls who met or consumed less than the recommended three servings of dairy per day. Participants included 172 11-year-old non-Hispanic white girls, assessed cross-sectionally. Intakes of dairy, calcium, and energy were measured using three 24-hour recalls. Body mass index and body fat measures from dual-energy x-ray absorptiometry were obtained. Because preliminary analyses suggested systematic underreporting of energy intake, the relationships among dairy servings and measures of …

Penalized Likelihood And Bayesian Methods For Sparse Contingency Tables: An Analysis Of Alternative Splicing In Full-Length Cdna Libraries, Corinne Dahinden, Giovanni Parmigiani, Mark C. Emerick, Peter Buhlmann
Johns Hopkins University, Dept. of Biostatistics Working Papers
We develop methods to perform model selection and parameter estimation in loglinear models for the analysis of sparse contingency tables to study the interaction of two or more factors. Typically, datasets arising from so-called full-length cDNA libraries, in the context of alternatively spliced genes, lead to such sparse contingency tables. Maximum Likelihood estimation of log-linear model coefficients fails to work because of zero cell entries. Therefore new methods are required to estimate the coefficients and to perform model selection. Our suggestions include computationally efficient penalization (Lasso-type) approaches as well as Bayesian methods using MCMC. We compare these procedures in a …
Multiple Testing With An Empirical Alternative Hypothesis, James E. Signorovitch
Multiple Testing With An Empirical Alternative Hypothesis, James E. Signorovitch
Harvard University Biostatistics Working Paper Series
An optimal multiple testing procedure is identified for linear hypotheses under the general linear model, maximizing the expected number of false null hypotheses rejected at any significance level. The optimal procedure depends on the unknown data-generating distribution, but can be consistently estimated. Drawing information together across many hypotheses, the estimated optimal procedure provides an empirical alternative hypothesis by adapting to underlying patterns of departure from the null. Proposed multiple testing procedures based on the empirical alternative are evaluated through simulations and an application to gene expression microarray data. Compared to a standard multiple testing procedure, it is not unusual for …

Independent data from the Gulf of Mexico are used to develop and test the hypothesis that the same sequence of physical and ecological events each year allows the toxic dinoflagellate Karenia brevis to become dominant. A phosphorus-rich nutrient supply initiates phytoplankton succession, once deposition events of Saharan iron-rich dust allow Trichodesmium blooms to utilize ubiquitous dissolved nitrogen gas within otherwise nitrogen-poor sea water. They and the co-occurring K. brevis are positioned within the bottom Ekman layers, as a consequence of their similar diel vertical migration patterns on the middle shelf. Upon onshore upwelling of these near-bottom seed populations to CDOM-rich …
The Circuitous Path To The Comparison Of Simulated Values From Crop Models With Field Observations, Albert Weiss, Wally Wilhelm
The Circuitous Path To The Comparison Of Simulated Values From Crop Models With Field Observations, Albert Weiss, Wally Wilhelm
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
The Journal of Agricultural Science, Cambridge has been a fixture in dissemination of crop simulation models and the concepts and data upon which they are built since the inception of computers and computer modeling in the mid-20th century. To quantify the performance of a crop simulation model, model outputs are compared with observed values using statistical measures of bias, i.e. the difference between simulated and observed values. While applying these statistical measures is unambiguous for the experienced user, the same cannot always be said of determining the observed or simulated values. For example, differences in accessing crop development can be …

The retinal pigment epithelium (RPE) is a monolayer of cells located directly behind the photoreceptor cells in the retina. These cells are involved in a variety of functions that support the visual process in the eye, namely 1) they form a blood-retina barrier which separates the neural retina from the choroid's blood supply, 2) the apical processes of RPE cells diurnally phagocytose the outer segments of photoreceptor cells, and 3) they participate in the renewal of the photopigment 11-cis retinal. Age-related macular degneration (AMD) is the leading cause of blindness in people over the age of 50 years in North …

Ontologies enable us to elevate syntactic and structural processing in an information system/Web to an information system/Web powered with semantic processing. Experience has shown that monolithic and tightly coupled approaches seldom succeed, and majority of information systems and applications will need to deal with plurality of ontologies in a loosely coupled environment (i.e., independently evolving ontologies and inter-ontology relationships, existence of different contexts for different users/applications etc.) Development of such loosely-coupled multi-ontology environments entails development of techniques for ontology mapping/alignment, multi-ontology query processing, and much more.

The healthcare industry is rapidly advancing towards the widespread use of electronic medical records systems to manage the increasingly large amount of patient data and reduce medical errors. In addition to patient data there is a large amount of data describing procedures, treatments, diagnoses, drugs, insurance plans, coverage, formularies and the relationships between these data sets. While practices have benefited from the use of EMRs, infusing these essential programs with rich domain knowledge and rules can greatly enhance their performance and ability to support clinical decisions. Active Semantic Electronic Medical Record (ASEMR) application discussed here uses Semantic Web technologies to …

An Overview Of The Classification And Evolution Of The Major Scarab Beetle Clades (Coleoptera: Scarabaeoidea) Based On Preliminary Molecular Analyses, Andrew B. T. Smith, David C. Hawks, John M. Heraty
University of Nebraska State Museum: Entomology Papers
We present a preliminary overview of our molecular phylogenetics research on the superfamily Scarabaeoidea. The molecular data consists of 28S ribosomal DNA sequences (mainly D2 and D3 expansion regions) for over 600 taxa and 18S ribosomal DNA sequences (mainly E17 to E35 expansion regions) for over 150 representative taxa within the lineages sampled. Based on our preliminary molecular phylogenetic results, Scarabaeoidea includes three major groups: 1) Geotrupidae, Passalidae, and Pleocomidae; 2) Lucanidae, Diphyllostomatidae, Trogidae, and Glaresidae; and 3) Hybosoridae, Ochodaeidae, Glaphyridae, and Scarabaeidae. The broad evolutionary patterns within the Scarabaeoidea are discussed with respect to phylogeny and evolution.

A Review Of The Family-Group Names For The Superfamily Scarabaeoidea (Coleoptera) With Corrections To Nomenclature And A Current Classification, Andrew B. T. Smith
University of Nebraska State Museum: Entomology Papers
For the first time, all family-group names in the superfamily Scarabaeoidea (Coleoptera) are evaluated using the International Code of Zoological Nomenclature to determine their availability and validity. A total of 383 family-group names were found to be available, and all are reviewed to scrutinize the correct spelling, author, date, nomenclatural availability and validity, and current classification status. Numerous corrections are given to various errors that are commonly perpetuated in the literature. A reversal of precedence is used to preserve the prevailing usage of the following family-group names: Eupariini Schmidt, 1910 (over Ataeniini Harold, 1868); Pachydemini Burmeister, 1855 (over Elaphocerini Blanchard, …
Characterization Of Cucurbituril Complex Ions In The Gas Phase Using Electrospray Ionization Fourier Transform Ion Cyclotron Resonance Mass Spectrometry, Haizhen Zhang
Theses and Dissertations
Host-guest interactions have been well studied in the new century to obtain fundamental insights into supramolecular chemistry. Most of the pioneering works have been done using techniques such as NMR, X-ray crystallography, IR spectroscopy and so on. However, none of these techniques is universal for the investigation of all types of supramolecules, and usually they have one or more limiting factors such as relatively large sample consumption, matrix effects from solvents, etc. Electrospray mass spectrometry has been widely used to investigate host-guest interactions in the gas phase. A particular advantage of gas phase host-guest research is that the experimental results …

The degree of similitude or closeness between two different isolates or strains of PRRSV is very important for deciding which strains should be used for immunization, by either vaccinating with commercial vaccines or attempting to stabilize a herd with planned wt PRRSV infection. As previously known for certain important viral diseases, such as in Foot and Mouth Disease, the reciprocal (cross-) neutralization titers between two strains may be of utmost importance to establish the degree of similarity or difference between these. There is currently a void of methods that would allow distinguishing or grouping strains of PRRSV in a manner …

Why Technology Provides Compelling Reasons To Apply A Daubert Analysis To The Legal Standard Of Care In Medical Malpractice Cases, Nichole Hines
Duke Law & Technology Review
Traditionally, courts have applied a "customary practice" standard in determining the legal standard of care in medical malpractice cases. Recently, a few courts have abandoned this dated standard and instead applied a Daubert analysis to the standard of care, which focuses on medical evidence that is scientifically based . In light of these recent holdings, this iBrief argues that with the increasing amounts of technologies improving evidence-based medicine, the customary practice standard is no longer a useful or appropriate test for determining the standard of care in medical malpractice cases. By applying a Daubert analysis to an expert’s testimony on …
