Meiotic Cohesion Requires Accumulation Of Ord On Chromosomes Before Condensation, Eric M. Balicky, Matthew W. Endres, Cary Lai, Sharon E. Bickel
Dartmouth Scholarship
Cohesion between sister chromatids is a prerequisite for accurate chromosome segregation during mitosis and meiosis. To allow chromosome condensation during prophase, the connections that hold sister chromatids together must be maintained but still permit extensive chromatin compaction. In Drosophila, null mutations in the orientation disruptor (ord) gene lead to meiotic nondisjunction in males and females because cohesion is absent by the time that sister kinetochores make stable microtubule attachments. We provide evidence that ORD is concentrated within the extrachromosomal domains of the nuclei ofDrosophila primary spermatocytes during early G2, but accumulates on the meiotic chromosomes by …

Biological Sciences
Temporal and spatial variability of physical, biological, and optical properties on scales of minutes to months and meters to ~50 km are examined using an extensive data set collected on the New York Bight continental shelf during the Hyperspectral Coastal Ocean Dynamics Experiment. Measurements from a midshelf mooring and bottom tripod (~25 km offshore, 24 m water depth) and two nearshore profiling nodes (~5 km offshore, 15 m water depth) are utilized to quantify and correlate midshelf and nearshore variability. Towed shipboard undulating profilers and a high-frequency radar (CODAR) array provide complementary spatial data. We show that phytoplankton and dissolved …

Maternal Behavior In Mice Selected For Large Litter Size, Chung-Feng Chiang, Rodger . K. Johnson, Merlyn K. Nielsen
Department of Animal Science: Faculty Publications
A total of 96 lactating mice and their pups from each of three replications of lines selected 21 generations for increased litter size (LS) followed by 23–25 generations of random selection and contemporary unselected control lines (LC) were characterized for maternal behavior. Sixteen dams and their pups from each replication of LS and LC lines were sampled. Litter sizes at birth for LS versus LC in the first, second, and third replicates were 17.3 versus 13.2, 15.9 versus 13.1, and 12.3 versus 10.6 pups, respectively, with LS dams averaging 2.87 ± .70 pups more. One-half of the litters were standardized …

Wolbachia Infection In Cotesia Sesamiae (Hymenoptera: Braconidae) Causes Cytoplasmic Incompatibility: Implications For Biological Control, Moses B. Mochiah, Adele J. Ngi-Song, William A. Overholt, Richard Stouthamer

Cotesia sesamiae (Hymenoptera: Braconidae) is an indigenous, gregarious, larval endoparasitoid that attacks mid- to late-instar of the stem borer larvae. Although the parasitoid is distributed widely throughout Africa, not all local populations appear to be equally effective in controlling stem borers. Consequently, there is an interest in releasing the more effective strains in areas that already have very low populations of C. sesamiae. Some C. sesamiae populations are infected with bacterial symbionts in the genus Wolbachia, which may induce cytoplasmic incompatibility. Using an antibiotic treatment, we have established that the Wolbachia infection causes cytoplasmic incompatibility of the female mortality type …

The concentration of photosynthetic pigments (i.e., chlorophylls a and b, and carotenoids) and chlorophyll degradation enzyme (i.e., chlorophyllase, oxidative bleaching, and Mg-dechelatase) activities on aphid-damaged and non-damaged regions of the infested leaves were determined with two infestation periods (6 and 12 days). Russian wheat aphid [Diuraphis noxia (Mordvilko) (Hemiptera: Aphididae)] feeding caused significant losses of chlorophylls a and b and carotenoids in the damaged regions. However, bird cherry-oat aphid [Rhopalosiphum padi (L.) (Hemiptera: Aphididae)] feeding did not, except a significantly lower level of carotenoids was observed in the damaged regions from the short-infestation (6-day) samples. Interestingly, the non-damaged …

Monte Carlo Simulation On The Indirect Exchange Interactions Of Co-Doped Zno Film, Abraham F. Jalbout, Hanning Chen, Scott L. Whittenburg
Chemistry and Biochemistry Faculty Publications
Monte Carlo simulations using a three-dimensional lattice model studied the Ruderman–Kittel–Kasuya–Yosida (RKKY) indirect exchange interaction of doped magnetic Co ions in ZnOfilms. The results of the calculations show that the RKKY interaction in Co-doped ZnO is long ranged and its magnitude is proportional to (inverse of the distance from a central ion). The sign oscillates with a frequency that depends on the concentration of the carrier. The long-distance sum of the RKKY indirect exchange energies is positive indicating that these materials are ferromagnetic, in direct correlation with previously reported results.

John Owens: Speeches and Appearances
I’d like to start tonight with the words of the late anthropologist Margaret Mead, who once said: “Never doubt that a small group of thoughtful citizens can change the world. Indeed, it is the only thing that ever has.”
As the LEAD22 class I have no doubt that you are a group of thoughtful Nebraska citizens, and we look forward with enthusiastic anticipation to your important participation in various agricultural and other organizations when you complete this wonderful program. We in the Institute of Agriculture and Natural Resources look forward to all you can and will do in …

John Owens: Speeches and Appearances
Thank you so much for inviting my wife Virginia and me to be with you today. It delights me to talk about land-grant universities in general, and about the land-grant university mission we take so very seriously in the Institute of Agriculture and Natural Resources in particular, because I am so proud of the way our faculty and staff continually strive to contribute to and improve the economic and societal well-being of rural Nebraska, as well as all of Nebraska.

Early studies showed that in addition to GTP, the pyrimidine nucleotides UTP and CTP support activation of the adenylyl cyclase (AC)-stimulating G(s) protein. The aim of this study was to elucidate the mechanism by which UTP and CTP support G(s) activation. As models, we used S49 wild-type lymphoma cells, representing a physiologically relevant system in which the beta(2)-adrenoreceptor (beta(2)AR) couples to G(s), and Sf9 insect cell membranes expressing beta(2)AR-Galpha(s) fusion proteins. Fusion proteins provide a higher sensitivity for the analysis of beta(2)AR-G(s) coupling than native systems. Nucleoside 5'-triphosphates (NTPs) supported agonist-stimulated AC activity in the two systems and basal AC …

A Novel High-Cell-Density Protein Expression System Based On Ralstonia Eutropha, Sriram Srinivasan, Gavin C. Barnard, Tillman U. Gerngross
Dartmouth Scholarship
We describe the development of a novel protein expression system based on the industrial fermentation organism Ralstonia eutropha (formerly known as Alcaligenes eutrophus) NCIMB 40124. This new system over- comes some of the shortcomings of traditional Escherichia coli-based protein expression systems, particularly the propensity of such systems to form inclusion bodies during high-level expression. Using a proteomics approach, we identified promoters that can be induced by simple process parameters or medium compositions in high-density cell culture or shake flasks, respectively. By combining newly developed molecular biological tools with a high-cell-density fermentation process, we were able to produce high levels ( …

Dispersal is a phenomenon of central importance in ecology and evolution. Yet many of its fundamental aspects remain poorly understood or barely investigated. This excellent, broad-ranging volume is a collection of 26 short reviews derived from a Centre National de la Recherche (CNRS)-National Science Foundation (NSF) sponsored workshop held in 1999. As stated by the editors, this book is mainly comprised of “reviews and more theoretical approaches, with a limited number of empirical examples” (p. xx) on dispersal.

We have investigated the intracellular traffic of PrPc, a glycosylphosphatidylinositol (GPI)-anchored protein implicated in spongiform encephalopathies. A fluorescent functional green fluorescent protein (GFP)-tagged version of PrPc is found at the cell surface and in intracellular compartments in SN56 cells. Confocal microscopy and organelle-specific markers suggest that the protein is found in both the Golgi and the recycling endosomal compartment. Perturbation of endocytosis with a dynamin I-K44A dominant-negative mutant altered the steady-state distribution of the GFP-PrPc, leading to the accumulation of fluorescence in unfissioned endocytic intermediates. These pre-endocytic intermediates did not seem to accumulate GFP-GPI, a minimum GPI-anchored protein, suggesting that …
