Diseased Thyrocytes Have Aberrant Communication And Tnf Receptor Secretion, Mary Labue Dec 1996

Diseased Thyrocytes Have Aberrant Communication And Tnf Receptor Secretion, Mary Labue

Loma Linda University Electronic Theses, Dissertations & Projects

Autoimmune diseases are a world-wide health concern and are an intense area of investigation. However, one area of research has been largely ignored. This involves the contribution of the target tissue to the disease process. We hypothesized that cell-cell communication is disrupted by lymphocytic infiltration and advancing fibrosis and this disruption results in altered target cell responses to immune components produced at the site of inflammation, including Tumor Necrosis Factor-alpha (TNF- α).

We have used thyroiditis as our model of autoimmunity and have documented the development of spontaneous thyroiditis in MRL-lpr/lpr mice. We have found aberrant connexin expression in …

Expression And Regulation Properties Of Pseudomonas L-Arabinose Utilization Plasmids, Mary E. Barecki Dec 1996

Expression And Regulation Properties Of Pseudomonas L-Arabinose Utilization Plasmids, Mary E. Barecki

Masters Theses

The L-arabinose utilization pathway in Pseudomonas fluorescens is a unique metabolic pathway. In Pseudomonas, L-arabinose is converted to the end product α-ketogluterate and does not involve phosphorylated intermediates. This differs from the well studied L-arabinose pathway found in Escherichia coli which contains phosphorylated intermediates.

The enzymes of the L-arabinose pathway have been shown to be induced in the presence of L-arabinose in P. saccharophila. The current study determined regulatory properties of previously cloned chromosomal fragments of P. fluorescens which confer an L-arabinose positive phenotype to the surrogate host, Pseudomonas putida. Cells of P. putida containing plasmids pPZ221, …

The Effects Of Benzo(A)Pyrene And Dimethylbenzanthracene On Cultured Porcine Aortic Endothelial Function, Michelle Lynn Kagey Dec 1996

The Effects Of Benzo(A)Pyrene And Dimethylbenzanthracene On Cultured Porcine Aortic Endothelial Function, Michelle Lynn Kagey

Masters Theses

Polycyclic aromatic hydrocarbons are environmental contaminants which have been shown to initiate and promote atherosclerosis and carcinogenesis in various animal species. These findings have been based primarily on the increase in incidence and size of smooth muscle cell tumors in these animals were exposed to benzo(a)pyrene (BaP) or dimethylbenzanthracene (DMBA). The endothelium has also been identified as an important mediator in vascular disease and atherogenesis, although the endothelial response to these PAHs has not been investigated. Thus, the focus of this study was on characterization of the endothelial response to BaP and DMBA. Concentration/response relationships for these chemicals were examined, …

About The Cellular Tropism Of The Gammaherpesvirus Bovine Herpesvirus Type 4, Fernando A. Osorio, J. A. Galeota-Wheeler, D. E. Reed, O. Lopez

Nebraska Center for Virology: Faculty Publications

We read with interest the recent report by Egyed et al. (4) about the distribution in tissue, in the natural host, of bovine herpesvirus type 4 (BHV-4), a member of the gammaherpesvirus group. Using PCR, those authors were able to trace BHV-4 (Movar strain) in several tissues of experimentally infected cattle (mostly spleen, lung, trachea, and nasal epithelium tissues) at different times, ranging from 3 to 48 days postinfection. The authors also report consistent detection of BHV-4 DNA in what they consider to be a pure population of lymphocytes obtained from peripheral blood of the BHV-4- infected cattle, and then …

"Notes On Bird Sightings In Nebraska," From Nebraska Bird Review (December 1996) Dec 1996

"Notes On Bird Sightings In Nebraska," From Nebraska Bird Review (December 1996)

Nebraska Bird Review

House Finches. This species has been appearing at my backyard feeders, 10 to 30 feet from our window, for several years, increasing in number each year. They came nearly every day in 1996 except during a period that I associate with post-breeding molt. Numbers varied from one to a maximum of 17 at one time, with 11 counted for the 1996 Christmas Bird Count. We commonly use 7-8 power binoculars to look for a rare Purple Finch. House Finches are noticeably smaller than House Sparrows and slightly larger than American Goldfinches, and feed with both species.

Males have varied in …

Phylogenetic Analysis Of Opisthadena (Digenea: Hemiuridae), Virginia León-Règagnon, Gerardo Pérez-Ponce De León, Daniel R. Brooks

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Systematic study of the closely related hemiurid genera Mitrostoma, Neopisthadena and Opisthadena revealed that Opisthadena cortesi is a junior synonym of Opisthadena dimidia and Opisthadena fujianensis and Opisthadena marina are species inquirendae. Phylogenetic analysis of the 7 recognized species, based on 21 transformation series derived from comparative morphologic characters, produced a single tree with a consistency index of 0.735. The five species of the genus Opisthadena constitute a monophyletic clade and Neopisthadena habei is their sister group. Mitrostoma nototheniae is the sister group of the other 6 members of the clade. Geographic distribution suggests an ancient circum-Pacific distribution of …

Gross And Histological Features Of A Myofascial Trigger Point In The Upper Trapezius, Kathryn E. Levee Dec 1996

Gross And Histological Features Of A Myofascial Trigger Point In The Upper Trapezius, Kathryn E. Levee

Electronic Theses and Dissertations

The purpose of this study was to precisely locate, in living humans, a myofascial trigger point associated with the upper portion of the trapezius muscle (TrP1) that refers pain to the head and neck and to determine if this point is associated with anatomical structures. This study is descriptive and utilizes data from measurements of the location of TrP1 in relation to anatomical landmarks, of pressure sensitivity overlying the trigger point and electromyography recordings in localizing the trigger point. Information obtained from living humans was used to determine anatomical correlation to structures in cadavers. Results indicated there is little variability …

Expression Of Α4Β1 Integrin On Tissue Eosinophils In The August Rat, John C. Phipps Dec 1996

Expression Of Α4Β1 Integrin On Tissue Eosinophils In The August Rat, John C. Phipps

Masters Theses

The integrin α4β1 is expressed on a wide variety of peripheral leukocytes, and functions in recruitment of these cells to sites of inflammaiton. We infected august (AUG) rats with the helminth parasite Nippostrongylus brasiliensis and later used leukocytes from the peritoneum of the sensitized animals and antibodies against α4β1 integrin to localize that adhesion molecule on the cell surfaces after antigen challenge. We found that after antigen challenge, eosinophils, monocytes, and small lymphocytes, but not mast cells, expressed α4β1 integrin if the animals had been previously infected, but cells from uninfected animals did not show expression of this molecule. Our …

Numa Assembles Into An Extensive Filamentous Structure When Expressed In The Cell Cytoplasm, Alejandro Saredi, Louisa Howard, Duane A. Compton

Dartmouth Scholarship

NuMA is a 236 kDa protein that participates in the organization of the mitotic spindle despite its strict localization in the nucleus during interphase. To test how cells progress through mitosis when NuMA is localized in the cytoplasm instead of the nucleus, we have deleted the nuclear localization sequence of NuMA using site-directed mutagenesis and transiently expressed this mutant protein (NuMA-DeltaNLS) in BHK-21 cells. During interphase, NuMA-DeltaNLS accumulates in the cytoplasm as a large mass approximately the same size as the cell nucleus. When cells enter mitosis, NuMA-DeltaNLS associates normally with the mitotic spindle without causing any apparent deleterious effects …

Great Basin Naturalist

The gastrointestinal tracts, lungs, and urinary bladders from 77 Bufo microscaphus, 61 Bufo woodhousii, and 8 of their hybrids were examined for helminths. One species of trematode (Glypthelmins quieta), 1 species of cestode (Distoichometra bufonis), and 5 species of nematodes (Aplectana incerta, A. itzocanensis, Rhabdias americanus, Physaloptera sp., and Physocephalus sp.) were found. The greatest prevalence (41%) and mean intensity (231.7) were recorded for Aplectana incerta in Bufo woodhousii. It appears hybrids harbor fewer parasites than either parent species.

A Scanning Electron Microscopy Study Of Taxus Leaves As Related To Taxonomy, G.A. Strobel, W. M. Hess Nov 1996

A Scanning Electron Microscopy Study Of Taxus Leaves As Related To Taxonomy, G.A. Strobel, W. M. Hess

Scanning Microscopy

Scanning electron microscopy, when applied to the surfaces of the needles of Taxus spp. (yew) revealed features that appear useful in the taxonomy of this tree species which yields the important anticancer drug, taxol. For instance, all of the four North American species have 3-5 rows of stomata on one-half of the abaxial leaf surface, whereas all of the others, including those from Europe and Asia, have 7-10 rows of stomata. The appearance of individual or fused papilliform epidermal cells and their arrangement on the leaf surface also is a feature that varies between species. Patterns of wax formation appeared …

Structures Of The Extracellular Domain Of The Type I Tumor Necrosis Factor Receptor, James H. Naismith, Tracey Q. Devine, Tadahiko Kohno, Stephen R. Sprang

Biological Sciences Faculty Publications

Background: Tumor necrosis factor (TNF) is a powerful cytokine that is involved in immune and pro-inflammatory responses. Two TNF receptors that belong to the cysteine-rich low affinity nerve growth factor receptor family (TNF-R1 and TNF-R2) are the sole mediators of TNF signalling. Signalling is thought to occur when a trimer of TNF binds to the extracellular domains of two or three receptor molecules, which permits aggregation and activation of the cytoplasmic domains. The complex is then internalized within an endocytic vesicle, whereupon it dissociates at low pH. Structure of the soluble extracellular domain of the receptor (sTNF-R1) both in the …

Structure Of The Gdp-Pi Complex Of Gly203→Ala G(Iα1): A Mimic Of The Ternary Product Complex Of Galpha-Catalyzed Gtp Hydrolysis, Albert M. Berghuis, Ethan Lee, André S. Raw, Alfred G. Gilman, Stephen R. Sprang

Biological Sciences Faculty Publications

Background: G proteins play a vital role in transmembrane signalling events. In their inactive form G proteins exist as heterotrimers consisting of an α subunit, complexed with GDP and a dimer of βγ subunits. Upon stimulation by receptors, G protein α subunits exchange GDP for GTP and dissociate from βγ. Thus activated, α subunits stimulate or inhibit downstream effectors. The duration of the activated state corresponds to the single turnover rate of GTP hydrolysis, which is typically in the range of seconds. In G(iα1), the Gly203→Ala mutation reduces the affinity of the substrate for Mg2+, inhibits a key …

The Chromatin Structure Of Well-Spread Demembranated Human Sperm Nuclei Revealed By Atomic Force Microscopy, M. J. Allen, E. M. Bradbury, R. Balhorn Nov 1996

The Chromatin Structure Of Well-Spread Demembranated Human Sperm Nuclei Revealed By Atomic Force Microscopy, M. J. Allen, E. M. Bradbury, R. Balhorn

Scanning Microscopy

The fundamental structure formed when genomic DNA is packaged by protamine in the human sperm nucleus still remains essentially unresolved. It is known that the binding of protamine, a small arginine-rich protein, to DNA generates a large dense, hydrophobic complex making the sperm chromatin structure difficult to study microscopically. To visualize the internal nuclear structures, isolated human sperm nuclei were swollen extensively in saline buffer using only a reducing agent. The nuclei were swollen during deposition onto coverglass and then imaged in the atomic force microscope (AFM). The two main results obtained from imaging individual well-spread nuclei indicate that native …

Protein-Protein And Protein-Dna Interactions At The Bacteriophage T4 Dna Replication Fork. Characterization Of A Fluorescently Labeled Dna Polymerase Sliding Clamp, Daniel J. Sexton, Theodore E. Carver, Anthony J. Berdis, Stephen J. Benkovic Nov 1996

Protein-Protein And Protein-Dna Interactions At The Bacteriophage T4 Dna Replication Fork. Characterization Of A Fluorescently Labeled Dna Polymerase Sliding Clamp, Daniel J. Sexton, Theodore E. Carver, Anthony J. Berdis, Stephen J. Benkovic

Chemistry Faculty Publications

The T4 DNA polymerase holoenzyme is composed of the polymerase enzyme complexed to the sliding clamp (the 45 protein), which is loaded onto DNA by an ATP-dependent clamp loader (the 44/62 complex). This paper describes a new method to directly investigate the mechanism of holoenzyme assembly using a fluorescently labeled cysteine mutant of the 45 protein. This protein possessed unaltered function yet produced substantial changes in probe fluorescence intensity upon interacting with other components of the holoenzyme. These fluorescence changes provide insight into the role of ATP hydrolysis in holoenzyme assembly. Using either ATP or the non-hydrolyzable ATP analog, adenosine …

We report the use of a model system that examines the dynamics of biological energy availability in organic matter in a sphagnum peat potting mix critical to sustenance of microorganism-mediated biological control of pythium root rot, a soilborne plant disease caused by Pythium ultimum. The concentration of readily degradable carbohydrate in the peat, mostly present as cellulose, was characterized by cross-polarized magic-angle spinning 13C nuclear magnetic resonance spectroscopy. A decrease in the carbohydrate concentration in the mix was observed during the initial 10 weeks after potting as the rate of hydrolysis of fluorescein diacetate declined below a critical threshold level …
