The Use Of Lactic Acid Bacteria To Control The Growth Of Foodborne Pathogens On Fresh-Cut Fruits And Sprout Vegetables, Franca Gabriela Rossi Jun 2016

The Use Of Lactic Acid Bacteria To Control The Growth Of Foodborne Pathogens On Fresh-Cut Fruits And Sprout Vegetables, Franca Gabriela Rossi

Master's Theses

Growing consumer awareness of the health benefits associated with fruits and vegetables and demand for easy to prepare products has prompted the development of a wide variety of minimally processed fruits and vegetables. Minimally processed fruits and vegetables are often peeled, cut, or diced which compromise the produces’ natural protective barriers, exposing a nutrient rich medium and providing an ideal environment for the growth of microorganisms, including foodborne pathogens. The germination conditions of sprout vegetables consisting of relatively high temperatures and humidity, low light and abundance of nutrients are also conducive to the proliferation of foodborne pathogens. Recent outbreaks and …

Using Hadoop To Identify False Positives In Bacterial Strain Typing From Dna Fingerprints, Colin C. Adams Jun 2016

Using Hadoop To Identify False Positives In Bacterial Strain Typing From Dna Fingerprints, Colin C. Adams

Computer Science and Software Engineering

Pyroprinting is a novel technique used by the Department of Biological Sciences to obtain “fingerprints” from the DNA of E. coli isolates in order to categorize them into strains. To determine the number of false positives that occur in the pyroprinting process, isolates with the same pyroprints needed to be sequenced to see if their underlying alleles match. If they do match, this shows they are indeed the same strain and are a true positive. If the alleles don’t match, they are different strains and are a false positive. To do this 100 isolates with nucleotide identifiers were sequenced. Over …

Physical Testing Of Potential Football Helmet Design Enhancements, Michael Jeremy Schuster Jun 2016

Physical Testing Of Potential Football Helmet Design Enhancements, Michael Jeremy Schuster

Master's Theses

Football is a much loved sport in the United States. Unfortunately, it is also hard on the players and puts them at very high risk of concussion. To combat this an inventor in Santa Barbara brought a new design to Cal Poly to be tested.

The design was tested in small scale first in order to make some preliminary conclusions about the design. In order to fully test the helmet design; however, full scale testing was required. In order to carry out this testing a drop tower was built based on National Operating Committee on Standards for Athletic Equipment, NOCSAE, …

OBJECTIVE. The impact of exclusively breastfeeding (EBF) on child body mass index (BMI) from birth through 60 months of age was investigated.

METHODS. 60,190 mothers and children attending Women, Infant and Children (WIC) clinics in the Santa Barbara and San Luis Obispo counties of California provided data on breastfeeding duration, maternal pre-pregnancy BMI, and child weight-for-length (WL) was measured at 3 month intervals until 5 years of age. Missing time points were interpolated in determining child BMI/WL z-score trajectories plotted on the Center of Disease Control (CDC) infant and child growth curves. Multivariate analysis of variance was used to contrast …

Evaluating The Myth Of Allelopathy In California Blue Gum Plantations, Kristen Marie Nelson Jun 2016

Evaluating The Myth Of Allelopathy In California Blue Gum Plantations, Kristen Marie Nelson

Master's Theses

It is widely accepted that allelopathy is not only significant, but more or less singular, in the inhibition of understory vegetation in California Eucalyptus globulus (blue gum) plantations. However, there is no published documentation of allelopathy by blue gums against California native species. Here, we present evidence that germination and early seedling growth of five California native species are not inhibited by chemical extracts of blue gum foliage, either at naturally-occurring or artificially concentrated levels. In the greenhouse, seeds were germinated in field-collected soil from mature blue gum plantations and the adjacent native, coastal scrub communities. In petri plates, seeds …

Stress Ecology Of The Pacific Rattlesnakes (Crotalus Oreganus And Crotalus Helleri), Natalie Claunch

Master's Theses

Stress is a physiological state induced by disturbance or adverse environmental conditions and is modulated by the glucocorticoid hormone corticosterone (CORT) in reptiles. Stressors can have various impacts on vertebrate trait expression and may affect survival or reproduction. Little is known about the effects of chronically elevated CORT in free-ranging reptiles, or the effect of disturbance stress on venom composition in captive snakes.

In chapter 1, we investigated the effects of researcher induced disturbance on CORT levels and venom composition in a group of captive Northern Pacific rattlesnakes (Crotalus oreganus). Venom protein concentration and plasma CORT levels were …

Evaluation Of The Estrogenic And Osmoregulatory Impacts Of Exposure To 4-Nonylphenol Pollution In The Estuarine Arrow Goby, Clevelandia Ios, Kaitlin Marie Johnson Jun 2016

Evaluation Of The Estrogenic And Osmoregulatory Impacts Of Exposure To 4-Nonylphenol Pollution In The Estuarine Arrow Goby, Clevelandia Ios, Kaitlin Marie Johnson

Master's Theses

Recent evidence indicates that some of California’s coastal estuaries are contaminated with the chemical 4-nonylphenol (4-NP). Tissue burdens of 4-NP detected in the intertidal arrow goby (Clevelandia ios) in California are among the highest recorded worldwide, however, it remains unknown whether these fish are impacted by this 4-NP contamination. 4-NP is an established endocrine disrupting compound with estrogenic properties that can alter reproductive function. Furthermore, evidence that estrogens can modulate iono- and osmo-regulatory function in fishes implies that estuarine fishes exposed to 4-NP may also experience an impaired ability to maintain hydromineral balance. In Chapter 1 of this …

Genes Encoding Flower- And Root-Specific Functions Are More Resistant To Fractionation Than Globally Expressed Genes In Brassica Rapa, Naiyerah F. Kolkailah Jun 2016

Genes Encoding Flower- And Root-Specific Functions Are More Resistant To Fractionation Than Globally Expressed Genes In Brassica Rapa, Naiyerah F. Kolkailah

Master's Theses

Like many angiosperms, Brassica rapa underwent several rounds of whole genome duplication during its evolutionary history. Brassica rapa is particularly valuable for studying genome evolution because it also experienced whole genome triplication shortly after it diverged from the common ancestor it shares with Arabidopsis thaliana about 17-20 million years ago. While many B. rapa genes appear resistant to paralog retention, close to 50% of B. rapa genes have retained multiple, paralogous loci for millions of years and appear to be multi-copy tolerant. Based on previous studies, gene function may contribute to the selective pressure driving certain genes back to singleton …

The Effects Of Quorum Sensing And Temperature On The Soluble Proteome Of Vibrio Salmonicida, Christopher L. Massey Jun 2016

The Effects Of Quorum Sensing And Temperature On The Soluble Proteome Of Vibrio Salmonicida, Christopher L. Massey

Master's Theses

Vibrio salmonicida causes cold-water vibriosis in salmon populations around the world and causes financial damage to fisheries designed to farm these salmon. Very little is known about the physiology of how V. salmonicida causes disease and measures to contain vibriosis are restricted to either vaccinating individual fish against disease or administering antibiotics when an outbreak is detected. These procedures are costly and increase the risk for selection of antibiotic-resistant V. salmonicida strains. A recent reoccurrence of outbreaks in Norwegian fisheries provided incentive to better understand the virulence mechanisms of V. salmonicida. In this thesis, a proteomic approach was used to …

Rearing Temperature Affects The Expression Of Proteins In The Adhesive Of The Striped Acorn Barnacle, Balanus Amphitrite, Melissa J. Daugherty Jun 2016

Rearing Temperature Affects The Expression Of Proteins In The Adhesive Of The Striped Acorn Barnacle, Balanus Amphitrite, Melissa J. Daugherty

Master's Theses

Barnacles are dominant hard–fouling organisms in marine waters. They attach to substrates by secreting a complex proteinaceous adhesive. Understanding the chemical composition of this multi–protein underwater adhesive and how it is affected by environmental variables, such as oceanic temperatures, is critical for developing nontoxic solutions to control biofouling. Previous experiments in our lab revealed an inverse relationship between critical removal stress (CRS) and temperatures at which barnacles were reared. Further investigations showed that this correlation is not attributed to differences in physical properties such as barnacle size or short–term changes in the viscosity of adhesive. Therefore, the observed effects may …

Comparison Of Foaming Properties Between Chelated Reconstituted Smp And Caseinates, Boya Liu Jun 2016

Comparison Of Foaming Properties Between Chelated Reconstituted Smp And Caseinates, Boya Liu

Master's Theses

Caseinate powders have been well accepted because of their foaming properties. In this study, 10% solution of reconstituted skim milk powder (SMP) chelated with sodium hexametaphosphate (SHMP) and trisodium citrate (TSC) at 1 mEq, 50 mEq, and 100 mEq were prepared to conduct a comparison with sodium caseinate, potassium caseinate, and calcium caseinate solutions. Foamability, foam stability as well as the preferential locations of αs-casein, β-casein and !-casein in their foams were analyzed. It was hypothesized that the foamability, foam stability and the preferential locations of these three caseins in the milk foams are different from treatment to treatment. Milk …

Limestone is a distinctive substrate that has significant effects on soils and plants. The present study characterizes the diversity of vascular plants, bryophytes, and lichens at the Simonton Corner Quarry Preserve, an abandoned limestone quarry in Rockport, Maine, USA, which was in operation in the late 1800s. We document vascular plant diversity and associated edaphic features (i.e., soil pH and elemental chemistry) using 30 535 meter plots spread throughout the site. For vascular plants, 114 species in 96 genera and 50 families were observed; few of these species are known to prefer calcareous environments, and 38% are nonnative. Conversely, the …

Separation And Characterization Of Reconstituted Skim Milk Powder Treated With Mineral Chelators, Amy Kringle May 2016

Separation And Characterization Of Reconstituted Skim Milk Powder Treated With Mineral Chelators, Amy Kringle

Master's Theses

The proteins found in milk are largely important in the functionality of many dairy products and dairy processes. The casein micelle system in milk is a complex and highly studied system. The micelle is thought to be a sponge like structure containing four caseins, αs1, αs2, β, and κ casein, and bound together with colloidal calcium phosphate. When a chelating agent such as a citrate, phosphate, or polyphosphate are added to milk systems, the CCP is bound to the chelator and removed from the micelle. It has been shown through past research that the use of …

A Comparative Study Of The Flora And Soils Of Great Duck And Little Duck Islands, Maine, Usa, Luka Negoita, Matthew Dickinson, Glen H. Mittlehauser, N. Rajakaruna

Biological Sciences

Strong environmental gradients and varied land-use practices have generated a mosaic of habitats harboring distinct plant communities on islands on the coast of Maine. Botanical studies of Maine’s islands, however, are generally limited in number and scope. Baseline studies of Maine’s islands are necessary for assessing vegetation dynamics and changes in habitat conditions in relation to environmental impacts imposed by climate change, rising sea levels, invasive species, pests and pathogens, introduced herbivores, and human disturbance. We conducted a survey of the vascular plants and soils of forest, field, and ocean-side communities of Great Duck and Little Duck Islands, ME. These …

The human immunodeficiency virus (HIV) has infected millions of people since the start of the epidemic in the early 1980s. HIV is the etiologic agent of the acquired immunodeficiency syndrome (AIDS), which has taken millions of lives. It is extremely difficult for particular populations to receive medication to treat HIV infection and prevent the progression of HIV to AIDS. Multiple studies have examined the effect of vitamin A on HIV infection, due to its immune stimulating effects. It is unclear whether an association exists between vitamin A and different aspects of HIV. This review examined the effect of vitamin A …

The Effect Of Xenon Pulsed-Light Technology On Biofilm Adhered To Stainless Steel Surfaces, Stephanie Jacquez Mar 2016

The Effect Of Xenon Pulsed-Light Technology On Biofilm Adhered To Stainless Steel Surfaces, Stephanie Jacquez

Master's Theses

In food processing, inadequate surface sanitation procedures lead to the formation of biofilms in which bacteria attach and aggregate in a hydrated polymeric matrix of their own synthesis. Formation of these sessile communities and their inherent resistance to existing sanitation procedures and agents are at the root of the risk of bacterial infections for consumers. Due to this existing problem, an effective method for reducing biofilm formation in dairy processing equipment is necessary for dairy products processing. Ultraviolet Pulsed light Technology has shown a positive effect in eliminating microorganism populations on food products. The objective of this work is to …

Partial Calcium Depletion During Membrane Filtration Affects Gelation Of Reconstituted Milk Protein Concentrates, H. Eshpari, R. Jiménez-Flores, P. S. Tong, M. Corredig

Dairy Science

Milk protein concentrate powders (MPC) with improved rehydration properties are often manufactured using processing steps, such as acidification and high-pressure processing, and with addition of other ingredients, such as sodium chloride, during their production. These steps are known to increase the amount of serum caseins or modify the mineral equilibrium, hence improving solubility of the retentates. The processing functionality of the micelles may be affected. The aim of this study was to investigate the effects of partial acidification by adding glucono-δ-lactone (GDL) to skim milk during membrane filtration on the structural changes of the casein micelles by observing their chymosin-induced …

Resource Partitioning Along Multiple Niche Axes Drives Functional Diversity In Parrotfishes On Caribbean Coral Reefs, Thomas C. Adam, Megan Kelley, Benjamin I. Ruttenberg, Deron E. Burkepile Dec 2015

Resource Partitioning Along Multiple Niche Axes Drives Functional Diversity In Parrotfishes On Caribbean Coral Reefs, Thomas C. Adam, Megan Kelley, Benjamin I. Ruttenberg, Deron E. Burkepile

Biological Sciences

The recent loss of key consumers to exploitation and habitat degradation has significantly altered community dynamics and ecosystem function across many ecosystems worldwide. Predicting the impacts of consumer losses requires knowing the level of functional diversity that exists within a consumer assemblage. In this study, we document functional diversity among nine species of parrotfishes on Caribbean coral reefs. Parrotfishes are key herbivores that facilitate the maintenance and recovery of coral-dominated reefs by controlling algae and provisioning space for the recruitment of corals. We observed large functional differences among two genera of parrotfishes that were driven by differences in diet. Fishes …
