The Expression Pattern Of The Tonoplast Intrinsic Protein Γ-Tip In Arabidopsis Thaliana Is Correlated With Cell Enlargement, Edward Himelblau, Herman Hofte, Dolors Ludevid, Maarten J. Chrispeels

Biological Sciences

The vacuolar membrane (tonoplast) contains an abundant intrinsic protein with six membrane-spanning domains that is encoded by a small gene family. Different isoforms of tonoplast intrinsic protein (TIP) are expressed in different tissues or as a result of specific signals. Using promoter-β-glucuronidase (GUS) fusions and in situ hybridization, we have examined the expression of γ-TIP in Arabidopsis thaliana. GUS staining of plants transformed with promoter-GUS fusions showed that γ-TIP gene expression is high in recently formed tissues of young roots. In the shoot, γ-TIP gene expression was highest in the vascular bundles of stems and petioles, as well as …

Nest Usurpation, Supersedure And Colony Failure Contribute To Africanization Of Commercially Managed European Honey Bees In Venezuela, Robert G. Danka, Richard L. Hellmich, Thomas E. Rinderer Dec 1992

Nest Usurpation, Supersedure And Colony Failure Contribute To Africanization Of Commercially Managed European Honey Bees In Venezuela, Robert G. Danka, Richard L. Hellmich, Thomas E. Rinderer

Richard L Hellmich

The fates of 58 colonies in 4 apiaries managed commercially for honey production in northwest Venezuela were determined during 15 months. All colonies had been previously requeened with marked European queens. Heritage of new queens (European daughter replacements versus Africanized usurpers) was traced by mitochondrial DNA and morphological characteristics of worker progeny. Three usurpations (5% of colony fates, at a rate of 4.7% of colonies per year) occurred. These colonies showed congruent African-derived mitochondrial DNA and Africanized morphology. Supersedures accounted for much of the introgression of African genes into the test apiaries. Twenty-eight colonies (48%) superseded their queens once, three …

Isolation And Characterization Of The Tricarboxylate Transporter From Pea Mitochondria, Cecilia A. Mcintosh, David J. Oliver Dec 1992

Isolation And Characterization Of The Tricarboxylate Transporter From Pea Mitochondria, Cecilia A. Mcintosh, David J. Oliver

David J. Oliver

The tricarboxylate transporter was solubilized from pea (Pisum sativum) mitochondria with Triton X-114, partially purified over a hydroxylapatite column, and reconstituted in phospholipid vesicles. The proteoliposomes exchanged external [14C]citrate for internal citrate or malate but not for preloaded D,L-isocitrate. Similarly, although external malate, succinate, and citrate competed with [14C]citrate in the exchange reaction, D,L-isocitrate and phosphoenolpyruvate did not. This tricarboxylate transporter differed from the equivalent activity from animal tissues in that it did not transport isocitrate and phosphoenolpyruvate. In addition, tricarboxylate transport in isolated plant mitochondria, as well as that measured with the partially purified and reconstituted transporter, was less …

Dynamics Of The Arctic Fox Population On St. Lawrence Island, Bering Sea, Francis H. Fay, Robert L. Rausch

Harold W. Manter Laboratory of Parasitology: Faculty and Staff Publications

English abstract: We hypothesized that the arctic fox, Alopex lagopus (Linnaeus), population on St. Lawrence Island was cyclic and that its fluctuations in size. structure, and productivity were correlated with the relative size of the population of northern voles, Microtus oeconomus Pallas, the primary prey. Based on a nine-year study, we determined that the variations in size of the fox and vole populations were similar, but they both were of low amplitude and not closely correlated. The high pregnancy rate (mean, 86%/yr) and numbers of young conceived (mean, 11.5/pregnancy) did not vary significantly among years, probably because of the consistently …

Biological Sciences Faculty Publications

Soils with a history of 2,4-dichlorophenoxyacetic acid (2,4-D) treatment at field application rates and control soils with no prior exposure to 2,4-D were amended with 2,4-D in the laboratory. Before and during these treatments, the populations of 2,4-D-degrading bacteria were monitored by most-probable-number (MPN) enumeration and hybridization analyses, using probes for the tfd genes of plasmid pJP4, which encode enzymes for 2,4-D degradation. Data obtained by these alternate methods were compared. Several months after the most recent field application of 2,4-D (approximately 1 ppm), soils with a 42-year history of 2,4-D treatment did not have significantly higher numbers of 2,4-D-degrading …

Processing And Localization Of Dengue Virus Type 2 Polyprotein Precursor Ns3-Ns4a-Ns4b-Ns5, Luwen Zhang, P. Maruthi Mohan, R. Padmanabhan Dec 1992

Processing And Localization Of Dengue Virus Type 2 Polyprotein Precursor Ns3-Ns4a-Ns4b-Ns5, Luwen Zhang, P. Maruthi Mohan, R. Padmanabhan

Nebraska Center for Virology: Faculty Publications

Processing of dengue virus type 2 polyprotein precursor NS3-NS4A-NS4B-NS5 could be mediated by the catalytically active NS3 protease domain and NS2B in trans at the dibasic sites NS3-NS4A and NS4B-NS5. Subcellular localization of the unprocessed precursor NS3-NS4A-NS4B-NS5 showed that it was confined to a distinct subcellular organelle in the cytoplasm, which was distinct from the distribution of the mature NS5.

Hanson, Peter L. Nara, Radha K. Maheshwari, Girmel S. Sidhu, John G. Bernbaum, David Hoekzema, Monte S. Meltzer, Howard Gendelman

Nebraska Center for Virology: Faculty Publications

Levels of human immunodeficiency virus (HIV) DNA, RNA, or p24 antigen and reverse transcriptase activity in T-cell cultures treated with 500 IU of recombinant alpha interferon (rIFNα) per ml were comparable to those in control cultures. Radioimmunoprecipitation analysis of proteins in lysates of IFN-treated T cells documented a marked accumulation of HlV proteins. Localization of gp120 by immunofluorescence showed a diffuse pattern in IFN-treated cells quite distinct from the ring pattern in untreated control cells. That large quantities of a120 in aberrant cell compartments might affect HlV morphogenesis was confirmed in infectivity studies: virions from IFN-treated cells were 100- to …

Effects Of Dietary Calcium And Fasting On Osteoclast Recession And Recruitment In Calcium Deficient Rats, Kenneth R. Wright Dec 1992

Effects Of Dietary Calcium And Fasting On Osteoclast Recession And Recruitment In Calcium Deficient Rats, Kenneth R. Wright

Loma Linda University Electronic Theses, Dissertations & Projects

The osteoclast, a multinucleated giant cell, has as its role the resorption of bone matrix with consequent release of calcium. The osteoclast is thought to derive from a pluripotent stem cell which also gives rise to monocytes and macrophages. The development, activation, and inhibition of osteoclasts are influenced by a number of systemic and local factors. The release of these factors is influenced by changes in the dietary status of the animal. Calcium deficiency is known to stimulate the recruitment and activity of large numbers of osteoclasts. These osteoclasts can subsequently be inhibited by increasing dietary calcium intake. Disappearance of …

The Entner-Doudoroff Pathway In Escherichia Coli Is Induced For Oxidative Glucose Metabolism Via Pyrroloquinoline Quinone-Dependent Glucose Dehydrogenase, Ronda Fliege, Suxiang Tong, Annemarie Shibata, Kenneth W. Nickerson, Tyrrell Conway Dec 1992

The Entner-Doudoroff Pathway In Escherichia Coli Is Induced For Oxidative Glucose Metabolism Via Pyrroloquinoline Quinone-Dependent Glucose Dehydrogenase, Ronda Fliege, Suxiang Tong, Annemarie Shibata, Kenneth W. Nickerson, Tyrrell Conway

Papers in Microbiology

The Entner-Doudoroff pathway was shown to be induced for oxidative glucose metabolism when Escherichia coli was provided with the periplasmic glucose dehydrogenase cofactor pyrroloquinoline quinone (PQQ). Induction of the Entner-Doudoroff pathway by glucose plus PQQ was established both genetically and biochemically and was shown to occur in glucose transport mutants, as well as in wild-type E. coli. These data complete the body of evidence that proves the existence of a pathway for oxidative glucose metabolism in E. coli. PQQ-dependent oxidative glucose metabolism provides a metabolic branch point in the periplasm; the choices are either oxidation to gluconate followed …

The Effect Of Trimethyltin On The Cholinergic System Of The Rat Hippocampus, Richard L. Cannon Dec 1992

The Effect Of Trimethyltin On The Cholinergic System Of The Rat Hippocampus, Richard L. Cannon

Electronic Theses and Dissertations

Trimethyltin (TMT) is a neurotoxin occurring in the environment. Exposure to (TMT) is known to destroy specific neuronal components of the hippocampus in the rat and to cause clinical symptoms in exposed humans, including mnemonic deficits, that indicate hippocampal involvement. In addition to hippocampal cell loss TMT causes significant increases in cholinergic markers such as acetylcholinesterase (AChE) stain density and choline acetyltransferase (ChAT) activity in the hippocampus of rats. However, despite these observations the effect of TMT on hippocampal cholinergic system has not been investigated in detail. The purpose of the present study was to elucidate more fully the consequences …

Neurophysiologic Bases For Abnormal Muscle Coactivation Patterns In Hemiparetic Stroke, Julius P.A. Dewald Dec 1992

Neurophysiologic Bases For Abnormal Muscle Coactivation Patterns In Hemiparetic Stroke, Julius P.A. Dewald

Loma Linda University Electronic Theses, Dissertations & Projects

To study disturbed muscle activation patterns in hemiparetic stroke, the EMG activity in spastic-paretic and contralateral elbow muscles of 10 stroke subjects were compared during graded directionally varying voluntary isometric contractions. We observed significant shifts in the peak EMG angle and increases in the angular range of EMG activity in the impaired upper limb. Furthermore, we observed stereotypic coactivation of many muscles acting at the same joint or at different joints across all subjects.

The effect of elbow angle changes on muscle coactivation patterns was studied by examining isometric torque-angle relations under passive and active conditions in 8 stroke subjects. …
