Purification And Applications Of A Bacteriophage-Derived Enzyme Chapk Against Problematic Staphylococci., Niamh Marie O'Shea

The bacteriophage K-derived peptidase CHAPk (Cysteine/Histidine-dependant amidohydrolase peptidase) can be produced from E. coli XL 1-Blue. Standard operating procedures for the production, purification and quality assessment of the peptidase were developed and set out in detail in this thesis. CHAPk exhibits strong antistaphylococcal properties and the purified enzyme was evaluated for its potential to eliminate S. aureus from the bovine udder and from human skin. As a teat dip CHAPk reduced the levels of S. aureus by three to five log cycles on both animal and human surfaces. CHAPk activity in milk was evaluated using challenge assays against the bovine …

Staphylococci are a diverse group of bacteria that cause diseases ranging from minor skin infections to life-threatening bacteremia. Methicillin-resistant S. aureus (MRSA) is the most common multidrug-resistant nosocomial pathogen in Europe estimated to cause 170,000 infections resulting in more than 5,000 deaths and additional healthcare costs of approximately €380 million annually (Kock et al., 201 1). Whilst rarely life-threatening, coagulase-negative staphylococci (CoNS), in particular S. epidermidis are responsible for the majority of indwelling medical device failures, causing intrinsically resistant biofilm infections, estimated to cost $2 billion per year in additional healthcare costs in the U.S. alone (Otto, 2009).

The genus Cronobacter accommodates 16 biogroups of the emerging opportunistic pathogen known formerly as Enterobacter sakazakii. Cronobacter are occasional contaminants of milk powder and, consequently, powdered infant formula (PIT). Since it is a pathogen that is transmitted in infant formula it represents a significant health risk to neonates. These bacteria are opportunistic pathogens (can only cause disease when the host's resistance is low) and are linked with life-threatening infections in neonates. Some of the clinical symptoms of Cronohacter infection include necrotizing enterocolitis, bacteremia, and meningitis, with case fatality rates of 50-80% being reported (Healy et al. 2010). Genomics and bioinformatics are topics of increasing interest in undergraduate biological science curricula. Many existing exercises focus on gene annotation and analysis of a single genome. In this paper, we present two educational modules designed to enable students to learn and apply fundamental concepts in comparative genomics using examples related to bacterial pathogenesis. Students first examine alignments of genomes of Escherichia coli O157:H7 strains isolated from three food-poisoning outbreaks using the multiple-genome alignment tool Mauve. Students investigate conservation of virulence factors using the Mauve viewer and by browsing annotations available at the A Systematic Annotation Package for Community Analysis of …

Recent molecular phylogenies suggest the surprising reacquisition of posthatching metamorphosis within an otherwise directdeveloping clade of lungless salamanders (family Plethodontidae). Metamorphosis was long regarded as plesiomorphic for plethodontids, yet the genus Desmognathus, which primarily includes metamorphosing species, is now nested within a much larger clade of direct-developing species. The extent to which the putative reacquisition of metamorphosis in Desmognathus represents a true evolutionary reversal is contingent upon the extent to which both larva-specific features and metamorphosis were actually lost during the evolution of direct development. Background: The bacterium Pelobacter carbinolicus is able to grow by fermentation, syntrophic hydrogen/formate transfer, or electron transfer to sulfur from short-chain alcohols, hydrogen or formate; it does not oxidize acetate and is not known to ferment any sugars or grow autotrophically. The genome of P. carbinolicus was sequenced in order to understand its metabolic capabilities and physiological features in comparison with its relatives, acetate-oxidizing Geobacter species.
Results: Pathways were predicted for catabolism of known substrates: 2,3-butanediol, acetoin, glycerol, 1,2-ethanediol, ethanolamine, choline and ethanol. Background: A new strain of Geobacter sulfurreducens, strain KN400, produces more electrical current in microbial fuel cells and reduces insoluble Fe(III) oxides much faster than the wildtype strain, PCA. The genome of KN400 was compared to wildtype with the goal of discovering how the network for extracellular electron transfer has changed and how these two strains evolved.
Results: Both genomes were re-annotated, resulting in 14 fewer genes (net) in the PCA genome; 28 fewer (net) in the KN400 genome; and ca. 400 gene start and stop sites moved. 96% of genes in KN400 had clear orthologs with conserved synteny in …

Mamejvo (Enicostemma axillare Raynal) is a perennial herb with diversified health benefits. It was found to be affected by a witches’ broom disease under the field conditions at Anand, Gujarat, India. Affected plants were 53.33% reduced in height due to shortened internodes, leading to typical witches’ broom symptoms. Flower size was reduced significantly in the affected plants compared with the healthy ones and their petal color gradually turned green (virescence). Affected plants produced 2.5 times more dry herbage than the normal plants but with less active ingredient (swertiamarin). Symptomatic plants died early while root suckers from such plants failed to …
