Viruses-like particles (VLPs) are frequently being used as platforms for polyvalent display of foreign epitopes of interest on their capsid surface to improve their presentation enhancing the antigenicity and host immune response. In the present study, we used the VLPs of Tomato bushy stunt virus (TBSV), an icosahedral plant virus, as a platform to display 180 copies of 16 amino acid epitopes of ricin toxin fused to the C-terminal end of a modified TBSV capsid protein (NΔ52). Live-fire military training can deposit millimeter-sized particles of high explosives (HE) on surface soils when rounds do not explode as intended. Rainfall-driven dissolution of the particles then begins a process whereby aqueous HE solutions can enter the soil and groundwater as contaminants. We dripped water onto individual particles of TNT, Tritonal, Comp B and Octol to simulate how surface-deposited HE particles might dissolve under the action of rainfall and to use the data to verify a model that predicts HE dissolution as a function of particle size, particle composition and rainfall rate. Complementary and alternative medicine (CAM) therapies present a growing information management challenge for physicians because nearly 40% of their patients may be using and another 50% may be considering use of CAM as part of their health care regimen. The National Health Statistics Reports for 2007 described the most commonly used nonvitamin, nonmineral therapy as natural products (eg, herbals at 17.7%). More than5% of children under the age of 18 years used CAM for allergic conditions including asthma. The 2006/07 and 2007/08 US-ITASE traverses from Taylor Dome to South Pole in East Antarctica provided opportunities to survey the subglacial and englacial environments using 3MHz and 200MHz radar. We present first results of these new ground-based radar data. A prominent basal deformation layer indicates different ice-flow regimes for the northern and southern halves of the Byrd Glacier drainage. Buried dune stratigraphy that appears to be related to the megadunes towards the west occurs at depths of up to 1500 m. At least two new water-filled subglacial lakes were discovered, while two recently drained lakes identified from repeat ICESat surface …

We examine the photophysics of a colloidal suspension of C60 particles in a micellar solution of Triton X-100 and water, prepared via a new synthesis which allows high-concentration suspensions. The particle sizes are characterized by transmission electron microscopy and dynamic light scattering and found to be somewhat polydisperse in the range of 10-100 nm. The suspension is characterized optically by UV-vis spectroscopy, femtosecond transient absorption spectroscopy, laser flash photolysis, and z-scan. The ground-state absorbance spectrum shows a broad absorbance feature centered near 450 nm which is indicative of colloidal C60. The transient absorption dynamics, presented for the …

We describe and validate a model that retrieves fractional snow-covered area and the grain size and albedo of that snow from surface reflectance data (product MOD09GA) acquired by NASA's Moderate Resolution Imaging Spectroradiometer (MODIS). The model analyzes the MODIS visible, near infrared, and shortwave infrared bands with multiple endmember spectral mixtures from a library of snow, vegetation, rock, and soil. We derive snow spectral endmembers of varying grain size from a radiative transfer model specific to a scene's illumination geometry; spectra for vegetation, rock, and soil were collected in the field and laboratory. Piercing of various parts of the body has gained tremendous popularity among adolescent and adult populations in recent years. There are no exact statistical reports on the number of body piercings being performed in the United States. One study among 481 college students in New York revealed that 42% of men and 60% of women had body piercing. In the past, the majority of complications of ear piercing were associated with the lobule. Torn ear lobes, localized infection, hypertrophic scars, allergic contact dermatitis, and keloid formations were among the most common, with no significant morbidity. Mercury is deposited to the Polar Regions during springtime atmospheric mercury depletion events (AMDEs) but the relationship between snow and ice crystal formation and mercury deposition is not well understood. The objective of this investigation was to determine if mercury concentrations were related to the type and formation of snow and ice crystals. On the basis of almost three hundred analyses of samples collected in the Alaskan Arctic, we suggest that kinetic crystals growing from the vapor phase, including surface hoar, frost flowers, and diamond dust, yield mercury concentrations that are typically 2-10 times higher than that reported for snow …

Only a limited number of complete mitochondrial genome sequences belonging to Native American haplogroups were available until recently, which left America as the continent with the least amount of information about sequence variation of entire mitochondrial DNAs. In this study, a comprehensive overview of all available complete mitochondrial DNA (mtDNA) genomes of the four pan-American haplogroups A2, B2, C1, and D1 is provided by revising the information scattered throughout GenBank and the literature, and adding 14 novel mtDNA sequences. During live fire training exercises, large amounts of explosives are consumed. Low order detonations of high explosive payloads result in the patchy dispersal of particles of high explosive formulations over large areas of firing range soils. Dissolution of explosives from explosive formulation particles into soil pore water is a controlling factor for transport, fate, and effects of explosive compounds. We developed an empirical method to evaluate soils based on functionally defined effective dissolution rates. The superoxide radical anion (SO) is a critical biomarker for monitoring cellular stress responses. Electrochemical SO biosensors are frequently constructed through the covalent immobilization of cytochrome c (Cyt c) onto self-assembled monolayers (SAMs); however, a detailed comparison of these systems as well as configuration influence on SO detection is needed to enable robust applications. Background: Immunization with RTS,S/AS02 consistently protects some vaccinees against malaria infection in experimental challenges and in field trials. A brief immunization schedule against falciparum malaria would be compatible with the Expanded Programme on Immunization, or in combination with other prevention measures, interrupt epidemic malaria or protect individuals upon sudden travel to an endemic area.

To assess the association between influenza immunization and subsequent diagnosis of group A streptococcus (GAS)-illness in Army recruits during influenza seasons 2002–2006. A case–control study was employed with cases as trainees with outpatient GAS diagnosis (ICD-9-CM codes: 034.0, 035, 038.0, 041.01, 320.2, 390–392, 482.31) during the influenza season, and controls as trainees with no outpatient GAS diagnosis during the influenza season. Primary exposure was influenza immunization during 1st September to 30th April of each season. Estimated protective effects of influenza immunization against GAS-illness ranged from 50% to 77%. Lipid biomarker analysis has proven valuable in testing the hypothesis that attributes of the extant microbiota can directly reflect the occurrence of contaminant biodegradation. Two past research efforts have demonstrated this utility and are described here.
A 4.5 m vertical core was obtained from a diesel fuel oil contamination plume. Core material was assayed for total petroleum hydrocarbons (TPH) and bacterial membrane phospholipids (PLFA) via a single solvent extraction. Microbial viable biomass and the relative abundance of Gram-negative bacterial PLFA biomarkers were found to be significantly correlated with TPH concentration. Zinc (Zn) is an essential nutrient that affects immune function, especially within the digestive system, although the underlying mechanisms are not well understood. This study examined the effects of short-term moderate Zn restriction on intestinal health and immune function in lipopolysaccharide (LPS)-challenged mice through plasma cytokine profiling and histological evaluation of intestinal tissue sections. Adult male mice were fed with a Zn-adequate (40 ppm) or a Zn-marginal (4 ppm) diet for 4 weeks, and then a bacterial challenge was simulated by intraperitoneal injection of LPS (10 μg/g body weight [BW]) or saline (control). Perchlorate is an oxidizer that has been routinely used in solid rocket motors by the Department of Defense and National Aeronautics and Space Administration. Royal Demolition Explosive (RDX) is a major component of military high explosives and is used in a wide variety of munitions. Perchlorate bearing wastewater typically results from production of solid rocket motors, while RDX is transferred to Army industrial wastewaters during load, assemble and pack operations for new munitions, and hot water or steam washout for disposal and deactivation of old munitions (commonly referred to as demilitarization, or simply demil).

Organic mulch is a complex organic material that is typically populated with its own consortium of microorganisms. The organisms in mulch breakdown complex organics to soluble carbon, which can then be used by these and other microorganisms as an electron donor for treating RDX and HMX via reductive pathways. A bench-scale treatability study with organic mulch was conducted for the treatment of RDX- and HMX-contaminated groundwater obtained from a plume at the Pueblo Chemical Depot (PCD) in Pueblo, Colorado. Thirty weak base 1,2,4-dispiro trioxolanes (secondary ozonides) were synthesized. Amino amide trioxolanes had the best combination of antimalarial and biopharmaceutical properties. Guanidine, aminoxy, and amino acid trioxolanes had poor antimalarial activity. Lipophilic trioxolanes were less stable metabolically than their more polar counterparts.

Amodiaquine (AQ) and tebuquine are 4-aminoquinoline antimalarials with Mannich base side chain and are highly effective against chloroquine (CQ)-resistant strains of Plasmodium falciparum. Clinical use of AQ has been severely restricted due to hepatoxicity and agranulocytosis side effects associated with its long term use. Lysosomal accumulation and bioactivation to generate reactive quinoneimine metabolite are implicated to be the cause of the observed AQ toxicities. Thermally responsive bulk polymer films utilizing reversible Diels-Alder chemistry have been developed. Gold nanoparticles (AuNPs) were passivated with thiol-terminated poly(styrene)-b-poly(ethylene glycol) (PS-b-PEG) copolymer ligand, where the PS and PEG blocks are joined via a Diels-Alder (DA) linkage. The ligand-functionalized nanoparticles were dispersed within a microphase-separated PS-b-poly(methyl methacrylate) (PS-b-PMMA) block copolymer. Nanoparticle location was dictated by the compatibility of the external shell with the block copolymer matrix. As cast, the PEG shell compatibilized the nanoparticles with the PMMA domains. Ricin is a potent toxin associated with bioterrorism for which no vaccine or specific countermeasures are currently available. A stable, non-toxic and immunogenic recombinant ricin A-chain vaccine (RTA 1-33/44-198) has been developed by protein engineering. We identified optimal formulation conditions for this vaccine under which it remained stable and potent in storage for up to 18 months, and resisted multiple rounds of freeze–thawing without stabilizing co-solvents. Reformulation from phosphate buffer to succinate buffer increased adherence of the protein to aluminum hydroxide adjuvant from 15 to 91%, with a concomitant increase of nearly threefold in effective antigenicity in a mouse model. …

We previously identified structurally diverse small molecule (non-peptidic) inhibitors (SMNPIs) of the botulinum neurotoxin serotype A (BoNT/A) light chain (LC). Of these, several (including antimalarial drugs) contained a 4-amino-7-chloroquinoline (ACQ) substructure and a separate positive ionizable amine component. The same antimalarials have also been found to interfere with BoNT/A translocation into neurons, via pH elevation of the toxin-mediated endosome. Thus, this structural class of small molecules may serve as dual-function BoNT/A inhibitors. We report the first safety and immunogenicity trial of the Plasmodium falciparum vaccine candidate FMP2.1/AS02A, a recombinant E. coli-expressed protein based upon the apical membrane antigen-1 (AMA-1) of the 3D7 clone formulated with the AS02A adjuvant. We conducted an open-label, staggered-start, dose-escalating Phase I trial in 23 malaria-naıve volunteers who received 8, 20 or 40 _g of FMP2.1 in a fixed volume of 0.5mL of AS02A on a 0, 1, and 2 month schedule. Nineteen of 23 volunteers received all three scheduled immunizations. The most frequent solicited local and systemic adverse events associated with immunization were injection site pain (68%) …

The reactivation of nerve agent-inhibited acetylcholinesterase (AChE) by oxime is the most important step in the treatment of nerve agent poisoning. Since the evaluation of nerve agent antidotes cannot be conducted in humans, results from animal experiments are extrapolated to humans. Guinea pig is one of the animal models that is frequently used for conducting nerve agent antidote evaluations. Our investigations in 2007 focused on continuing collection and analyses of tree rings from living trees and building a long ring-width series from interstadial wood. These interstadial forests were extensive and their preservation, although discontinuous and fragmented, is remarkable. In situ stumps, still rooted in growth position, and logs held within sediments remain from forests that existed at various times between glaciations over approximately the last 10,000 years.
We have analyzed previous and new collections of ring-width tree-ring series, including a multiple species collection from near Excursion Ridge in the southeast portion of the Park. These tree-ring series have a …
