The resistive pulse technique characterizes single analytes by detecting fluctuations in the ion current as they pass through a narrow orifice connecting two electrolyte-filled chambers, each containing a single electrode. The amplitude, shape, frequency, and duration of these fluctuations primarily provide information about the analyte’s size, shape, concentration, and net surface charge, respectively. One configuration for resistive pulse measurements is of an electrolyte-filled nanopipette immersed in a bath, where the nanopipette tip acts as the nanoscale orifice. Graphene has generated substantial interest as a filler due to its exceptional strength, flexibility, and conductivity but faces obstacles in supply and implementation. A renewable, plant-based graphene nanoparticle (pGNP) presents a more accessible and sustainable filler with the same properties as mineral graphenes. In this study, the mechanisms of graphene reinforcement in carbon fiber reinforced plastic (CFRP) were examined, along with the resulting improvements to mechanical strength, resistance to crack propagation, electrical and thermal conductivity at elevated temperatures. pGNP, produced from renewable biomass, was shown to have a graphitic structure with flakes 3-10 layers thick and a median lateral size …

Shale oil reservoirs are prolific on the short term due to hydraulic fracturing and horizontal drilling but experience significant production decline, leading to poor ultimate recovery and leaving billions of barrels of oil buried in the ground. In this study, a systematic multi-scale investigation of an enhanced oil recovery (EOR) process using relatively inexpensive silicon dioxide nanoparticles and carbon dioxide for shale oil reservoirs was conducted. From pharmaceuticals to biofuels, fermentation bioprocessing has gained traction in many industries reaching a global market value of almost $60 billion in 2018. However, applications of bioprocessing have been limited in scope due to high capital costs, long development times, and difficulties maintaining productivity during scale-up. Furthering development and scale-up of fermentation bioprocesses require careful monitoring and control through analytical chemistry. Currently, there are no small molecule quantification techniques that are sufficiently accurate, inexpensive, and high-throughput. This investigation explores viability of a novel quantification assay for amino acids in fermentation broth leveraging molecularly imprinted polymers (MIPs) and gold nanoparticles (AuNPs).

Herein, we developed a novel artificial tongue using machine learning and 12 nanoassemblies (2D-NAs) to identify and analyzed different kinds of milk beverages for quality control. This biomimetic sensor array was trained to “taste” different milk types as an “artificial tongue” which is the first time we demonstrated that this sensor array can be implemented to complex systems. Two-dimensional nanoparticles (2D-nps) and nine fluorescently labeled single stranded oligonucleotides (ssDNA) with different length and nucleobases were assembled to create 12 2D-NAs. In this dissertation, four nanoparticle reaction schemes were developed as substrates for halochromic dyes or nucleic acids. The reaction schemes include the use of two substrates: silica nanoparticles and halloysite nanotubes. The protocols can incorporate silica (SiO2) nanoparticles and halloysite aluminosilicate (AlO2SiO2) nanotubes due to the presence of silane groups on the surface of either substrate. High clinical resistance to molecularly targeted therapeutics has pushed interest again towards inhibiting universal biochemical hallmarks of cancer. Recent evidence suggests that malignant tumors acidify the local extracellular environment to activate proteases for degrading the tumor matrix, which facilitates metastasis, and explains why more aggressive tumors are more acidic. Novel methods are needed to traverse the blood-brain barrier (BBB) and deliver drugs to specific targets in the brain. To this end, MS2 bacteriophage was explored as a multifunctional transport and targeting vector. The MS2 capsid exterior was modified with two different targeting moieties for delivery across the BBB and targeting specific regions of interest in the brain. Successful modification of MS2 capsids with a brain targeting peptide and NMDAR2D-targeting antibody was confirmed by immunoblotting and fluorescence detection. Previous experimental studies show that nanoparticle-stabilized supercritical CO2 foams (or, NP CO2 foams) can be applied as an alternative to surfactant foams, in order to reduce CO2 mobility in gas injection enhanced oil recovery (EOR). Magnetic nanoparticles (MNPs) are used in a variety of applications, including as agents for magnetic resonance imaging, generation of local hyperthermia, and as platforms for drug delivery. Iron-based MNPs are often coated with a shell, such as silica or gold, to increase biocompatibility for drug delivery applications. The purpose of this work was to recombinantly produce a 45- amino acid long metal binding peptide that is useful for nanoparticle synthesis. Using splicing by overlap extension PCR, a synthetic gene containing the fusion of the metal binding peptide with Green Fluorescent Protein (GFPUV) was constructed. The cell interior is a crowded chemical space, which limits the diffusion of molecules and organelles within the cytoplasm, affecting the rates of chemical reactions. We provide insight into the relationship between non-specific intracellular diffusion and cytoskeletal integrity. Quantum dots entered the cell through microinjection and their spatial coordinates were captured by tracking their fluorescence signature as they diffused within the cell cytoplasm. Particle tracking revealed significant enhancement in the mobility of biocompatible quantum dots within fibrosarcoma cells versus their healthy counterparts, fibroblasts, as well as in actin destabilized fibroblasts versus untreated fibroblasts. In this dissertation, a detailed investigation on the influence of various macrocyclic resorcinarene surfactants in determining the morphology, stabilization and self-assembly of mono- and bi- metallic nanoparticles was undertaken. Chapter 2 describes the influence of resorcinarene surfactants functionalized with amine- and thiol- headgroups in determining the morphology of monometallic Pt nanoparticles synthesized via the Brust-Schiffrin reaction. We found that while resorcinarene benzylthiol can lead to the formation of highly branched Pt nanostructures, resorcinarene amine can lead to the formation of anisotropic crystalline Pt nanoparticles. This dissertation presents an investigation into the effect of nanoparticle fillers and a polydopamine adhesive primer on the tribological performance of thin PTFE films. The principal objective of this investigation was to reduce wear in PTFE films, an issue which precludes the use of PTFE films in tribological applications requiring high durability. The friction and wear of the composite films were evaluated using a ball-on-flat configuration in linear reciprocating motion. It was found that the use of a polydopamine adhesive primer reduces the wear of PTFE films more than 600 times. X-ray photoelectron spectroscopy (XPS) results show that a tenacious …

The CoFe2O4/GNS was synthesized by a simple one-pot solvothermal method. The results obtained by X-ray diffraction (XRD) and transmission electron microscopy (TEM) showed that the CoFe2O4 particles with sizes of 15 nm dispersed on the surface of graphene uniformly. Electrochemical tests indicated that the CoFe2O4/GNS nanocomposite exhibited excellent cyclic stability with almost 95.8% capacity retention after 100 cycles at the current density of 500 mA·g-1 and delivered a high reversible capacity of 482 mAh·g-1 at the current density of 2 A·g-1.

This thesis describes the surface modification of barium strontium titanate nanoparticles for use in polymer/ceramic composite thin film capacitors with resultant improved dielectric and film-making properties. Phosphonic acid-type ligands proved to be most effective for surface conjugation to the surface of the barium strontium titanate nanoparticles. Amine-terminated ligands proved to be effective at removing surface adsorbed water before being almost entirely removed during the sample washing stage. Carboxylic acid terminated ligands proved to adhere less well to the nanoparticle than the phosphonic acid, but resulted in thin films with a higher dielectric constant, which was more stable in the measured …

Engineered nanomaterials (ENMs) are currently used in hundreds of commercial products and industrial processes, with more applications being investigated. Nanomaterials have unique properties that differ from bulk materials. While these properties may enable technological advancements, the potential risks of ENMs to people and the environment are not yet fully understood. Certain low solubility nanoparticles are more toxic than their bulk material, such that existing occupational exposure limits may not be sufficiently protective for workers. Risk assessments are currently challenging due to gaps in data on the numerous emerging materials and applications as well as method uncertainties and limitations.

The main focus of this research was to evaluate the ability of a novel multifunctional nanoparticle to mediate drug delivery and enable a non-invasive approach to measure drug release kinetics in situ for the treatment of cancer. These goals were approached by developing a nanoparticle consisting of an inorganic core (i.e. gadolinium sulfoxide doped with europium ions or carbon nanotubes). This was coated with an external amphiphilic polymer shell comprised of a biodegradable polyester (i.e. poly(lactide) or poly(glycolide)), and poly(ethylene glycol) block copolymer. Project Summary Technical Abstract This Small Business Innovation Research Phase I project, presented by Talamo Inc., is to test an ovarian cancer therapeutic delivery system, comprised of a nanoscale biocompatible vesicle that carries a bioactive small interfering RNA molecule (siRNA), in vivio in mice. The siRNA has shown to reduce the expression of an enzyme (Sod2) that is highly expressed in multiple ovarian cancer types. The reduced expression of Sod2 will allow the tumor to become increasingly susceptible to chemotherapy agents, while simultaneously diminishing tumor progression. Modification of nanostructures via laser processing is of great interest for a wide range of applications such as aerospace and the storage of nuclear waste. The primary goal of this dissertation is to improve the understanding of nanostructures through two primary routes: the modification of aerogels and pulsed laser ablation in ethanol. A new class of materials, patterned aerogels, was fabricated by photopolymerizing selected regions of homogeneous aerogel monoliths using visible light. The characterization and fabrication of functionally graded, cellular and compositionally anisotropic aerogels and ceramics is discussed.
