We develop six-dimensional single-molecule orientation-localization microscopy (SMOLM) to measure the 3D positions and 3D orientations simultaneously of single fluorophores. We show how careful optimization of phase and polarization modulation components can encode phase, polarization, and angular spectrum information from each fluorescence photon into a microscope’s dipole-spread function. We used the transient binding and blinking of Nile red (NR) to characterize the helical structure of fibrils formed by designed amphipathic peptides, KFE8L and KFE8D, and the pathological amyloid-beta peptide Aβ42. We also deployed merocyanine 540 to uncover the interfacial architectures of biomolecular condensates.

Conventional magnetophoresis techniques for manipulating biocarriers and cells predominantly rely on large-scale electromagnetic systems, which is a major obstacle to the development of portable and miniaturized cell-on-chip platforms. Herein, a novel magnetic engineering approach by tailoring a nanoscale notch on a disk micromagnet using two-step optical and thermal lithography is developed. Versatile manipulations are demonstrated, such as separation and trapping, of carriers and cells by mediating changes in the magnetic domain structure and discontinuous movement of magnetic energy wells around the circumferential edge of the micromagnet caused by a locally fabricated nano-notch in a low magnetic field system. Since the early 2000s heavy-metal-free quantum dots (QDs) such as CuInS2/ZnS have attempted to replace CdSe, their heavy-metal-containing counterparts. CuInS2/ZnS is synthesized in a two-step process that involves the fabrication of CuInS2 (CIS) nanocrystals (NCs) followed by the addition of zinc precursors. Instead of the usual core/shell architecture often exhibited by binary QDs, coating CIS QDs results in alloyed and/or partially alloyed cation-exchange (CATEX) QDs. The effect that zinc has on the properties of CIS NCs was studied by incorporating zinc during the first step of the synthesis. Considerable research work is going on worldwide among the therapeutic scientific communities to use magnetic nanoparticles (NPs) as a controlled drug delivery system. Challenges to use those NPs for such in-vivo applications successfully are mainly biocompatibility and the functionality of those NPs. Addressing the needs, in this project, we synthesized carbon-coated cobalt ferrite (CFO) core-shell nanoparticles (CSNPS) by ablating a solution of toluene and CFO NPs with varying number of pulsed laser shots. Defect engineering is a strategy that has been widely used to design active semiconductor photocatalysts. However, understanding the role of defects, such as oxygen vacancies, in controlling photocatalytic activity remains a challenge. Here, we report the use of chemically triggered fluorogenic probes to study the spatial distribution of active regions in individual tungsten oxide nanowires using super-resolution fluorescence microscopy. The nanowires show significant heterogeneity along their lengths for the photocatalytic generation of hydroxyl radicals. Through quantitative, coordinate-based colocalization of multiple probe molecules activated by the same nanowires, we demonstrate that the nanoscale regions most active for the photocatalytic generation of …

Herein, we developed a novel artificial tongue using machine learning and 12 nanoassemblies (2D-NAs) to identify and analyzed different kinds of milk beverages for quality control. This biomimetic sensor array was trained to “taste” different milk types as an “artificial tongue” which is the first time we demonstrated that this sensor array can be implemented to complex systems. Two-dimensional nanoparticles (2D-nps) and nine fluorescently labeled single stranded oligonucleotides (ssDNA) with different length and nucleobases were assembled to create 12 2D-NAs. Biologically, deoxyribonucleic acid (DNA) molecules have been used for information storage for more than 3 billion years. Today, modern synthesis tools have made it possible to use synthetic DNA molecules as a material for engineering nanoscale structures. These self-assembling structures are capable of both resolutions as fine as 4 angstroms and executing programed dynamic behavior. Numerous approaches for creating structures from DNA have been proposed and validated, however it remains commonplace for engineered systems to exhibit unexpected behaviors such as low formation yields, poor performance, or total failure. A confocal micro-particle image velocimetry (C-μPIV) technique along with associated post-processing algorithms is detailed for obtaining three dimensional distributions of nano-particle velocity and concentrations at select locations of the 2.5D (pseudo 3D) Poly(methyl methacrylate) (PMMA) and ceramic micro-model. The designed and fabricated 2.5D micro-model incorporates microchannel networks with 3D wall structures with one at observation wall which resembles fourteen morphological and flow parameters to those of fully 3D actual reservoir rock (Boise Sandstone) at resolutions of 5 and 10 μm in depth and 5 and 25 μm on plane. As drinking water sources become increasingly impaired with nutrients and wastewater treatment plant (WWTP) effluent, formation of disinfection byproducts (DBPs) – such as trihalomethanes (THMs), dihaloacetonitriles (DHANs), and N-nitrosamines – during water treatment may also increase. N-nitrosamines may comprise the bulk of the chronic toxicity in treated drinking waters despite forming at low ng/L levels. This research seeks to elucidate physicochemical properties of carbon nanotubes (CNTs) for removal of DBP precursors, with an emphasis on total N-nitrosamines (TONO).

As drinking water sources become increasingly impaired, enhanced removal of natural organic matter (NOM) may be required to curb formation of disinfection byproducts (DBPs) upon chlor(am)ination. While carbon nanotubes (CNTs) can adsorb NOM, their properties for DBP precursor adsorption have not been elucidated. Nine types of CNTs were assessed for trihalomethane (THM), dihaloacetonitrile (DHAN), and total N-nitrosamine (TONO) precursor adsorption. Batch isotherm experiments were completed with lake water and, to simulate an impaired condition, effluent from a wastewater treatment plant (WWTP). A new fluorescence enhancement technical platform based on nanostructured aluminum oxide (NAO) has been discovered. Experiments reveal that this new platform can enhance the fluorescence signals up to two or three orders of magnitude compared to a traditional glass substrate platform. In order to realize a large-scale arrayed fluorescence biosensing platform for high throughput applications, a rapid and cost-effective process has been developed to fabricate the micropatterned NAO of a variety of shapes. Ligand cross-linking is known to improve the colloidal stability of nanoparticles, particularly in aqueous solutions. However, most cross-linking is performed chemically, in which it is difficult to limit interparticle cross-linking, unless performed at low concentrations. Photochemical cross-linking is a promising approach but usually requires ultraviolet (UV) light to initiate. Using such high-energy photons can be harmful to systems in which the ligand–nanoparticle bond is fairly weak, as is the case for the commonly used semiconductor quantum dots (QDs). Phase contrast microscopy (PCM) is a widely used analytical method for airborne asbestos, but it is unable to distinguish asbestos from non-asbestos fibers and requires time-consuming and laborious manual counting of fibers. Previously, we developed a high-throughput microscopy (HTM) method that could greatly reduce human intervention and analysis time through automated image acquisition and counting of fibers. In this study, we designed a dual-mode HTM (DM-HTM) device for the combined reflection and fluorescence imaging of asbestos, and automated a series of built-in image processing commands of ImageJ software to test its capabilities. A microfluidic reactor for synthesizing cadmium selenide (CdSe) quantum dots (QDs) was synthesized out of a silicon wafer and Pyrex glass. Microfabrication techniques were used to etch channels into the silicon wafer. Holes were wet-drilled into the Pyrex glass using a diamond-tip drill bit. The Pyrex wafer was anodically bonded to the etched silicon wafer to enclose the microfluidic reactor. Conditions for anodic bonding were created by exposing the stacked substrates to 300V at ~350oC under 5.46N of force. The majority of pharmacologically active chemotherapeutics are poorly water soluble. Solubilization enhancement by the utilization of organic solvents often leads to adverse side effects. Nanoparticle-based cancer therapy, which is passively targeted to the tumor tissue via the enhanced permeation and retention effect, has been vastly developed in recent years. Nanocrystals, which exist as crystalline and carry nearly 100% drug loading, has been explored for delivering antineoplastic agents. Quantum dots are semiconductive particles whose properties are highly influenced by the presence of at least one electron. Cadmium selenide quantum dots were synthesized via colloidal synthesis. Contrary to previous preparations, more focus was placed on the temperature rather than the duration of time at which they form. A series of colored solutions were obtained because the excited quantum dots of various sizes emitted specific wavelengths of light. The emission spectra showed that the temperature-dependent quantum dots were successfully synthesized. Stimulated emission depletion (STED) microscopy is one of the breakthrough technologies that belong to far-field optical microscopy and can achieve nanoscale spatial resolution. We demonstrate a far-field optical nanoscopy based on continuous wave lasers with different wavelengths, i.e., violet and green lasers for excitation and STED, respectively. Fluorescent dyes Coumarin 102 and Atto 390 are used for validating the depletion efficiency. Fluorescent nanoparticles are selected for characterizing the spatial resolution of the STED system. Purple membrane (PM, λ" role="presentation">λmax" role="presentation">max 570 nm) of H. halobium on treatment with sulphuric acid changes its colour to blue (λ" role="presentation">λmax" role="presentation">max 608 nm). The purple chromophore can be regenerated from the blue chromophore by exogeneous addition of anions such as CI−" role="presentation">− and HPO42−" role="presentation">2−4. Chloride ion is found to be more effective than the dibasic phosphate ion in regenerating the purple chromophore. Nevertheless, one thing common to the anion regeneration is that both CI−" role="presentation">− and HPO42−" role="presentation">2−4 show marked pH effect. At pH 1.0 the efficiency of …
