Drug theophylline is a bronchodilator used to treat asthma and chronic obstructive pulmonary disease (COPD), but precise monitoring is required to avoid side effects. Therefore, this study aimed to develop a screen-printed electrode (SPE) modified with a composite of green-synthesized silver nanoparticles and boron-doped diamond nanoparticles using tea polyphenols (Ag-BDDTP/SPE) for detecting theophylline. Formation of AgNPs was confirmed by UV-Vis spectroscopy, which showed a localized surface plasmon resonance peak at ~450 nm. Transmission electron microscopy revealed polydisperse nanoparticles with an average diameter of 39.94 nm. Ocular melanoma is commonly treated using plaque brachytherapy; however, radiation-induced damage to healthy ocular tissues frequently results in partial or complete vision loss. This work investigates the development of an injectable, magnetically responsive silicone magnetic fluid designed to localize adjacent to the tumor and attenuate low-energy gamma radiation during treatment.
The material system consists of iron oxide nanoparticles surface-functionalized with a siloxane polymer, in which the nanoparticles provide magnetic responsiveness and radiation attenuation, while the polymer coating ensures colloidal stability, injectability, and biocompatibility. Pancreatic ductal adenocarcinoma is a highly lethal malignancy characterized by poor prognosis and resistance to conventional therapies. Novel treatment strategies are needed to overcome this resistance. Non-thermal physical modalities, including nanosecond pulsed electric field (nsPEF) and cold atmospheric pressure plasma discharge (CAP), have emerged as potential anticancer approaches; however, the mechanisms underlying their combined effects remain unclear.

We present an overview of the composition, function, energetics, and dynamics of extracellular matrix and its relation to nanoscale structures and phenomena. These concepts are then related to the development of synthetic and biosynthetic materials that aim to mimic the extracellular matrix for regenerative medicine and tissue engineering technologies. Notable successes and advancements towards the goal of biomimicry are outlined, while remaining challenges and knowledge gaps towards that goal are highlighted. Finally, we frame the remaining challenges in the field in terms of nanoscience-related research opportunities that if solved, might prove to be transformative steps forward in the discipline.

This study evaluated the remineralization potential of a ceramic–polyelectrolyte system, based on a novel combination of hydroxyapatite nanoparticles (HAp NPs) and a hyaluronic acid (HY) matrix, for dental enamel, a tissue that remains challenging to repair in clinical dentistry. Nano-Hydroxyapatite (nanoHAp) powder was synthesized and characterized using X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), field-emission scanning electron microscopy (FE-SEM), dynamic light scattering (DLS), and zeta potential analysis to confirm their crystalline structure, functional groups, morphology, particle size distribution, and colloidal stability. Continuous glucose monitoring is critical for effective diabetes management; however, conventional benchtop potentiostats are bulky, costly, and unsuitable for decentralized point-of-care (PoC) applications. To address these limitations, this work presents a miniaturized, low-cost electrochemical sensing platform integrating a non-enzymatic glucose sensor with a portable potentiostat. The sensing electrode is based on laser-induced graphene modified with zinc oxide and platinum nanostructures via electrodeposition to enable sensitive glucose detection under physiological conditions. A custom-designed portable potentiostat was developed to control electrode potentials and perform electrochemical measurements, and its performance was experimentally validated against a commercial Metrohm system. The endometrium is a vital mucosal tissue which undergoes cyclical regeneration, differentiation, and remodeling upon hormonal, cellular, and molecular signaling networks. Dysregulation of these processes can trigger a range of pathological conditions including chronic inflammatory disorders, hyperplastic lesions, malignancies, and infertility, necessitating the need for effective therapeutic interventions. This thesis investigates the mechanisms of correlated neuronal activity in the globus pallidus externus (GPe), a key structure in the basal ganglia. Neuronal activity in the GPe is reportedly decorrelated in healthy subjects but can become highly correlated in Parkinson’s disease. However, high-density neuronal recordings in the GPe of healthy rats reveal a surprising degree of correlated activity. This raises the question of whether intrinsic properties or network connectivity alone can account for such patterns. This work presents the development of innovative electrochemical sensors for selective and sensitive dopamine (DA) detection using flexible laser-induced graphene (LIG) electrodes modified with advanced nanocomposites. An LIG electrode synthesized from pyralux film was functionalized with Nb4C3Tx MXene and silver nanoparticles (AgNPs), significantly enhancing its electrochemical performance. The modified electrode exhibited a peak anodic current increase from 150 μA to 330.4 μA and demonstrated a wide linear detection range (100 nM to 10 μM) with a low detection limit of 1 nM and high sensitivity (160.96 μA/nM cm -2). Nanosecond pulsed electric field (nsPEF) employs nanosecond-duration, high voltage pulses to induce oxidative stress, leading to temporary or permanent damage to cells or tissue (also known as reversible and irreversible electroporation), and has been considered a promising approach for cancer therapy. The cosmetic industry has developed and commercialized numerous products using new technologies, making them increasingly appealing to the public. The use of metallic nanoparticles (MtNPs) as key ingredients in cosmetics has become more widespread due to their demonstrated benefits. Improper healing of bone fractures and limited regeneration of bone following bone tumor resection remain a prevalent complication worldwide. Long-term effects of this include joint instability, limited movement, and arthritis. Bone infections (osteomyelitis), reactive oxygen species (ROS) generation via excessive inflammation, and underlying conditions such as osteoporosis can contribute to suboptimal bone tissue regeneration. ROS accumulation stimulates osteoblast apoptosis, while promoting osteoclast differentiation. These processes inhibit mineralization and osteogenesis. This study successfully implemented microcrystal electron diffraction (microED) and X-ray powder diffraction (XRPD) for the crystal structure determination of a new phase, TAF-CNU-1, Ni(C₈H₄O₄)·3H₂O, solved by microED from single microcrystals in the powder and refined at the kinematic and dynamic electron diffraction theory levels. This nickel metal–organic framework (MOF), together with its cobalt and manganese analogues with formula M (C₈H₄O₄)·2H₂O with M = Mn II or CoII, were synthesized in aqueous media as one-pot preparations from the corresponding hydrated metal chlorides and sodium terephthalate, as a promising `green' synthetic route to moisture-stable MOFs. Protein-based self-assembling biomaterials, known as Thermo-Responsive Assembled Proteins (TRAP), present meaningful potential as drug delivery carriers. These materials enable the controlled, slow release of poorly soluble chemotherapeutic agents, such as doxorubicin (Dox), while potentially reducing systemic off-target effects. In collaboration with multiple NYU labs, this study evaluated the efficacy of two TRAP variants—TRAP and F-TRAP—as drug delivery carriers in a xenograft mouse model of glioblastoma multiforme (GBM).

Orthovoltage X-ray beams are attractive for surface and intra-operative radiotherapy. Yet, their clinical utility is constrained by rapid attenuation and the limited dose that can be delivered without harming healthy tissue. High-Z hafnium oxide (HfO2) nanoparticles can boost local energy deposition through the photoelectric effect, while catalytic surface defects accelerate water radiolysis. Harnessing both mechanisms in a single nano-heterostructure could enable potent radiosensitization at diagnostic-range energies.

Nanoparticles (NPs), specifically polymer-modified NPs, have illustrated unique therapeutic advantages compared to bulk materials. Cellax NPs have provided a promising approach to cancer therapy by improving drug delivery, targeting the tumor microenvironment, and potentially overcoming drug resistance, all while reducing overall toxicity compared to traditional taxane treatment. This dissertation investigates the development and optimization of pathology-responsive polymersomes for simultaneous enhanced magnetic resonance imaging (MRI) and enzyme replacement therapy (ERT) in the treatment of GM1 gangliosidosis (GM1). While therapeutic strategies exist for similar, non-neuropathic lysosomal storage disorders, implementation towards GM1 faces significant challenges due to the blood-brain barrier.

The clinical translation of nanoparticle (NP) therapeutics for brain injury has been slow despite preclinical promise. The causes of disconnect between preclinical and clinical outcomes is multifactorial, but insufficient delivery is noteworthy. Preclinical delivery characterization often lacks, obscuring mechanisms of failure, slowing development. Multimodal imaging is a promising strategy to improve delivery characterization, combining the strengths of various modalities. Traumatic Brain Injury (TBI) causes a cascade of detrimental events within neurons, contributing to a host of neurological diseases relating to dementia. In this study we have performed a meta-analysis comparing differentially expressed genes (DEG) in specific cell types such as neurons and microglia revealing inflammation as a common driver of neurological deficiencies post TBI. Inflammation and hypoxia are well characterized features of TBI; however, the molecular mechanisms causing tau hyperphosphorylation by tauopathy associated genes (TAG) post-TBI are currently poorly understood. This dissertation endeavors to advance the frontiers of analytical chemistry by addressing critical challenges across three distinct chapters. In the first chapter, the focus is on the evaluation of digestion methods for the analysis of trace metals in mammalian tissues and the Standard Reference Material NIST 1577c. The primary goal is to optimize methodologies, laying the groundwork for reliable trace metal analysis and furthering our understanding of their roles in biological systems.

Early cancer detection is paramount for effective treatment and potential cures. This research explores the application of perovskite materials, specifically Sr2+-doped Lanthanum Cobaltite (La1-xSrxCoO3) nanomaterials, in cancer detection, with a focus on rats as an experimental model. The ferroelectric nature of these materials, synthesized through a combination of sol-gel and molten-salt processes, was examined at varying Sr2+ doping levels (1-20 wt%). Rigorous characterization, employing X-ray diffraction and scanning electron microscopy, confirmed the uniform morphology of nano cubes, laying the foundation for subsequent investigations. Precision medicine enables healthcare providers to tailor medical care to an individual's distinct genetic and environmental makeup. We contrast the switching of photoluminescence (PL) of PbS quantum dots (QDs) cross-linked with photochromic diarylethene molecules with different end groups, 4,4′-(1-cyclopentene-1,2-diyl)bis[5-methyl-2-thiophenecarboxylic acid] (1C) and 4,4′-(1-cyclopentene-1,2-diyl)bis[5-methyl-2-thiophenethiocarboxylic acid] (2T). Our results show that the QDs cross-linked with the carboxylic acid end group molecules (1C) exhibit a greater amount of switching in photoluminescence intensity compared to QDs cross-linked with the thiocarboxylic acid end group (2T). We also demonstrate that regardless of the molecule used, greater switching amounts are observed for smaller quantum dots. Varying these parameters allows for the fabrication of photoswitches with tunable PL change. We relate these observations to the …

Conventional magnetophoresis techniques for manipulating biocarriers and cells predominantly rely on large-scale electromagnetic systems, which is a major obstacle to the development of portable and miniaturized cell-on-chip platforms. Herein, a novel magnetic engineering approach by tailoring a nanoscale notch on a disk micromagnet using two-step optical and thermal lithography is developed. Versatile manipulations are demonstrated, such as separation and trapping, of carriers and cells by mediating changes in the magnetic domain structure and discontinuous movement of magnetic energy wells around the circumferential edge of the micromagnet caused by a locally fabricated nano-notch in a low magnetic field system.
