Nucleosome core particles (NCP) are the building blocks that form a highly organized and compact chromatin structure. Nucleosomes package DNA in the nucleus of eukaryotic cells. The NCP consists of about 147 base pairs of DNA wrapped around the histone octamer, with 1.65 superhelical turns in a left-handed manner. The histone octamer is composed of two copies of H3, H4, H2A, and H2B. Together with histone H1 and linker DNA, they further assemble into a higher-order chromatin structure. The nucleosome complex is stabilized by electrostatic interactions between positively charged histone residues and the negatively charged DNA backbone. To effectively access …

The hypersonic regime poses numerous challenges that researchers face in the development of hypersonic flight vehicles. Due to their excellent thermomechanical properties, ultra-high-temperature ceramics (UHTCs) have risen as a promising solution to act as a protective barrier between the harsh environment and surface materials of these flight bodies. The mechanical operation of a portable hypersonic simulation device was developed in-house and tested at Argonne National Laboratories (ANL) to gather in-situ material response of prospective UHTC samples when exposed to a hypersonic regime. Atomization of non-Newtonian, high-viscosity fluids is a challenging task with many applications. One method of atomizing these types of flows is the Wave Augmented Varicose Explosions (WAVE) atomizer. Using this framework, the study establishes an analysis methodology, investigates the relationship between 2D axisymmetric (2DA) and 3D models, and provides analysis of geometric, non-geometric, and non-dimensional parameter influences on the atomizer characterization in 2DA models. The study found that 2DA models can provide viable predictions of certain 3D model characteristics, established several mathematical models for parameter relationships among 2DA models, and identified certain metrics as inadequate predictors of atomizer characteristics in …

This research investigates aerodynamic drag reduction on a 25° Slanted Ahmed Body (SAB) by integrating porous media model rods through combined experimental and computational methods at a Reynolds number of 1.16×10⁴. Two porous media configurations: short rods (6.75% of the model height) and long rods (20.0% of the model height), both featuring cylindrical rods with 80% porosity, were systematically compared against a baseline SAB. In-depth analyses were performed to investigate the wake flow topology, recirculation region characteristics, pressure coefficient distribution, Reynolds stress distributions and drag coefficient. The acoustic guitar is a stringed instrument, often made of wood, that transduces vibrational energy of steel strings into coupled vibrations of the wood and acoustic pressure waves in the air. Variations in wood selection and instrument geometry have been shown to affect the timbre of the acoustic guitar. Computational methods were utilized to investigate the impact of material properties on acoustic performance. Mechanical Exfoliation of Graphene is an often-overlooked portion of the fabrication of quantum devices, and to create more devices quickly, optimizing this process to generate better flakes is critical. In addition, it would be valuable to simulate test pulls quickly, to gain insight on flake quality of various materials and exfoliation conditions. Physical pulls of graphene at various temperatures, pull forces, and pull repetitions were analyzed and compared to the results of ANSYS simulations, solved for similar results. Combining vibrating mesh nebulizers with additional new technologies leads to substantial improvements in pharmaceutical aerosol delivery to the lungs across therapeutic administration methods. In this dissertation, streamlined components, aerosol administration synchronization, and/or Excipient Enhanced Growth (EEG) technologies were utilized to develop and test several novel devices and aerosol delivery systems. The powder-bed fusion (PBF) process is a subdivision of Additive Manufacturing (AM) technology where a heat source at a controlled speed selectively fuses regions of a powder-bed material to form three-dimensional (3-D) parts. Two of the most effective PBF processes are selective laser melting (SLM) and electron beam additive manufacturing (EBAM), which can fabricate full-density metallic parts in a layer-by-layer fashion. Modeling works which simulate the proton-exchange membrane fuel cell with the computational fluid dynamics approach involve the simultaneous solution of multiple, interconnected physics equations for fluid flows, heat transport, electrochemical reactions, and both protonic and electronic conduction. Modeling efforts vary by how they treat the physics within and adjacent to the membrane-electrode assembly (MEA). Certain approaches treat the MEA not as part of the computational domain, but rather an interface connecting the anode and cathode computational domains. Buildings of high importance that are critical to community function, such as hospitals, often utilize supplemental damping devices like supplemental viscous fluid dampers or base isolators to reduce the overall demands on the structural system. An anguilliform swimming robot replicating an idealized motion is a complex marine vehicle necessitating both a theoretical and experimental analysis to completely understand its propulsion characteristics. The ideal anguilliform motion within is theorized to produce ``wakeless'' swimming (Vorus, 2011), a reactive swimming technique that produces thrust by accelerations of the added mass in the vicinity of the body. Pacific Northwest National Laboratory (PNNL) is currently developing an ultra-low background liquid scintillator counter (ULB LSC) in the shallow underground laboratory. At a depth of 35-meters water-equivalent, the underground laboratory has a multi-layered shielding to keep out cosmic-ray induced background. The ULB LSC, which is located in a clean room facility, is a multi-layered design made up of various materials, including plastic scintillator veto panels, borated polyethylene, lead and copper. The objective of the project was to develop the processes and procedures to create an optimized physical and visual bond between layers to minimize vibratory dissipation, thus maximizing sustain.
