Contemporary research suggests that shear force is a major contributor to discomfort and pressure injury. However, the variation of shear forces during human-exoskeleton interaction dynamics is a highly unexplored field. The high cost of commercial triaxial force sensors may be a major factor in the notable lack of research in this field. Therefore, in this paper, we present a low-cost, 3D-printed triaxial force sensor designed specifically to measure the triaxial interaction forces between an exoskeleton and its user. An accurate representation of cellular mechanobiology necessitates the inclusion of subcellular elements characterized by minute masses and dimensions. These minute objects yield multiscale dynamic models with disproportionate terms, which require inordinate amounts of computational time to simulate. The computational requirements limit the time span of the simulation to time histories shorter than one second, even when employing supercomputers. This work presents a high-speed approach to simulating the mechanobiology of stem cells. This thesis presents the development, calibration, and initial implementation of transcranial focused ultrasound (tFUS) for neuromodulation in a human study. The primary objectives were to design a system capable of delivering precise and safe ultrasound stimulation to targeted brain regions and to conduct preliminary trials to assess its effects on brain activity. A custom calibration device was developed to ensure accurate measurement of acoustic pressure, addressing challenges such as voltage alterations caused by changes in system impedance. The author of this thesis conceived and investigated an unexpected electrical signal that may not be noticed during regular electroglottography (EGG) when electrodes are conventionally placed on the anterior surface of the throat. It appears that there is a measurable electrical signal from the EGG equipment when electrodes are placed over the frontal cheek (belly of zygomatic muscle), and the tongue is elevated to contact the hard palate, or the tongue is lowered to break the contact. Therefore, this phenomenon is designated as unconventional electroglottography (UEGG, Dr. Steven Cloud, private communication, July 17, 2022).

This project seeks to develop an updated version of a microwave imaging device for use in conjunction with breast MRI, improving upon existing technology and developing novel concepts for the device. It posits three primary redesign targets for updating the previous system: resizing the system height, making the device more iteration- friendly, and improving the overall manufacturability of the device by replacing custom components with commercially available alternatives. Porous scaffolds mechanically stimulated with ultrasound are used for the treatment of bone conditions such as osteoporosis or traumatic bone injury. However, one current issue in these treatment techniques is designing a scaffold structure with a high structural integrity that also does not impede tissue growth. A larger ultrasound wave transmission would also allow more of a scaffold to receive mechanical stimulation from ultrasound. Microscale propulsion impacts a diverse array of fields, with simplistic microrobots allowing for novel innovations in microscale surgery and drug delivery. Propulsion at the microscale is constrained by physics, with time-reversal and geometric symmetries limiting available propulsion mechanisms. However, certain fluid environments and surface coatings allow for the propulsion of microparticles through externally applied magnetic fields. Presented here is a detailed analysis of microparticles propelling using spontaneous symmetry breaking, flagella surface coatings, and multi-modal actuation mechanisms. In this research, two actuation systems were introduced, inertial and magnetic actuation. In the inertial actuation, the robot used the transfer of momentum to navigate, and this momentum could be generated by spinning masses and wheels. Recent studies in our System Laboratory proved that a wide range of inertially actuated locomotion systems could be generated. This can be achieved by using a family tree approach, starting from a very simple system, and progressively evolving it to more complex ones. Photoacoustic (PA) tomography (PAT) is a novel imaging modality that combines the fine lateral resolution from optical imaging and the deep penetration from ultrasonic imaging, and provides rich optical-absorption–based images. PAT has been widely used in extracting structural and functional information from both ex vivo tissue samples to in vivo animals and humans with different length scales by imaging various endogenous and exogenous contrasts at the ultraviolet to infrared spectrum. The C-Arm is a large “C” shaped and manually maneuvered arm that provides surgeons and X-ray technicians the ability to take quick quality X-rays during surgery. In impact mechanics, the collision between two or more bodies is a common, yet a very challenging problem. Producing analytical solutions that can predict the post-collision motion of the colliding bodies require consistent modeling of the dynamics of the colliding bodies. This dissertation presents a new method for solving the two and multibody impact problems that can be used to predict the post-collision motion of the colliding bodies. In E. coli, fatty acid synthesis is catalyzed by the enzyme acetyl-CoA carboxylase (ACC), which converts acetyl-CoA into malonyl-CoA. Malonyl-CoA is a major building block for numerous of bioproducts. Multiple parameters regulate the homeostatic cellular concentration of malonyl-CoA, keeping it at a very low level. Understanding how these parameters affect the bacterial production of malonyl-CoA is fundamental to maximizing it and its bioproducts. To this end, competing pathways consuming malonyl-CoA can be eliminated, and optimal nutritional and environmental conditions can be provided to the fermentation broth. This paper presents a systematic method to analyse the stability of systems with single delay in which the coefficient polynomials of the characteristic equation depend on the delay. Such systems often arise in, for example, life science and engineering systems. A method to analyze such systems was presented by Beretta and Kuang in a 2002 paper, but with some very restrictive assumptions. This work extends their results to the general case with the exception of some degenerate cases. It is found that a much richer behavior is possible when the restrictive assumptions are removed. In this work, the approach to the manipulation of alginate artificial cell soft-microrobots, both individually and in swarms is shown. Fabrication of these artificial cells were completed through centrifugation, producing large volumes of artificial cells, encapsulated with superparamagnetic iron oxide nanoparticles; these artificial cells can be then externally stimulated by an applied magnetic field. The construction of a Permeant Magnet Stage (PMS) was produced to manipulate the artificial cells individually and in swarms. The stage functionalizes the permanent magnet in the 2D xy-plane. This work is to develop an experiment-validated numerical model to elucidate the wave transmission mechanisms through a surrogate head under blast loading. Repeated shock tube tests were conducted on a surrogate head, i.e., water-filled polycarbonate shell. Surface strain on the skull simulant and pressure inside the brain simulant were recorded at multiple locations. A numerical model was developed to capture the shock wave propagation within the shock tube and the fluid-structure interaction between the shock wave and the surrogate head. In this paper, simulation of the brain based on an artificial spiking neuron model is used to create a self-learning algorithm. The spiking neuron simulation is used to demonstrate a neuromodulation program in which the reward seeking properties of dopamine, the risk-adverse effects of serotonin, and the attention-focusing effects of the cholinergic and noradrenergic systems are applied to a mobile robotic platform as it moves autonomously throughout an environment. External stimuli is recorded by the program as spiking “events” that result in corresponding amounts of dopamine and serotonin influenced spiking patterns. Magnetic resonance elastography (MRE) is a potentially transformative imaging modality allowing local and non-invasive measurement of biological tissue mechanical properties. It uses a specific phase contrast MR pulse sequence to measure induced vibratory motion in soft material, from which material properties can be estimated. Compared to other imaging techniques, MRE is able to detect tissue pathology at early stages by quantifying the changes in tissue stiffness associated with diseases. This dissertation presents innovate methodologies for locating, extracting, and separating multiple incoherent sound sources in three-dimensional (3D) space; and applications of the time reversal (TR) algorithm to pinpoint the hyper active neural activities inside the brain auditory structure that are correlated to the tinnitus pathology. Specifically, an acoustic modeling based method is developed for locating arbitrary and incoherent sound sources in 3D space in real time by using a minimal number of microphones, and the Point Source Separation (PSS) method is developed for extracting target signals from directly measured mixed signals.
