Design And Characterization Of A Pulsed Electric Field Chamber For Microalgae Lysis, Connor Strutin

Microalgae are a promising renewable source of biofuel due to their high lipid content and rapid growth rates; however, algae cells must be electroporated (lysed) to release these lipids. The system developed in this project applies short (40 μs–10 ms), high-intensity (up to 33 kV/cm) pulsed electric fields (PEF) across cell membranes to enable electroporation.
Our lab-scale PEF chamber generates uniform, high-intensity electric fields using parallel-plate electrodes with sub-millimeter spacing. The system is designed to efficiently lyse microalgae cells for lipid extraction and biofuel production.

This project developed and evaluated an optical sensing system for detecting changes associated with glucose concentration. The system combined a laser-diode, cuvette sample holder, photodiode, resistive-feedback transimpedance amplifier, high-resolution analog-to-digital converter, and microcontroller. Parametric testing evaluated the effects of input current and feedback resistance on transimpedance gain, output range, and linearity. Firmware was developed to configure the ADC, average repeated conversions, monitor measurement variation, convert raw digital counts into voltage using a source-meter calibration equation, and compare sample measurements with a water reference. A cuvette enclosure maintained alignment between the laser-diode, sample, and photodiode while reducing external optical interference. Testing …

The project presents the design, fabrication, and implementation of a PCB test bench that interfaces with a custom VLSI chip designed to emulate the neural activity of a Central Pattern Generator (CPG). The test bench provides a precise and reliable platform for supporting signal operations and validating neuromorphic functionality. The design integrates both analog and digital circuitry to provide stable biasing, signal routing, and measurement access for chip characterization. The incorporation of op-amp buffers and adjustable potentiometers enables precise voltage tuning, resulting in controlled neuron-like behavior within the custom IC. Arteriovenous (AV) grafts are commonly used to provide vascular access for hemodialysis in patients with end-stage renal disease. Despite their widespread use, AV grafts are prone to complications such as stenosis and thrombosis. Early detection of these conditions remains challenging with current monitoring methods too costly or insufficient. This work presents the design, fabrication, and validation of an LC pressure sensor embedded within a model AV graft to enable real-time monitoring. With the continued growth of the biopharmaceutical industry, the demand for scalable, robust, and resource-efficient platforms for large-scale mammalian cell culture is amplified. Recent developments in microfluidic technology, such as precise control of the microenvironment, showed the potential to improve the performance of cell culture systems. However, constrained by scalability and operational efficiency, applying such approaches to large-scale cell culture and biopharmaceutical production presents challenges. Transient plasma ignition (TPI) utilizes non-equilibrium plasmas, produced by nanosecond high-voltage pulses, to improve lean-fuel combustion performance and reduce emission. It is known that the relatively high reduced electric field (E/N) in TPI plays an important role in generating energetic electrons and facilitating energy-efficient radical productions, resulting in reliable ignition for lean combustion. Automated nail disease diagnostics provide a non-invasive pathway for identifying underlying systemic health conditions; however, conventional centralized deep learning approaches often raise concerns related to privacy, fairness, and interpretability. Although the original NeuroNail-SNN framework demonstrated an energy-efficient and edge-ready diagnostic solution, its broader clinical adoption remained limited by unresolved trust, transparency, and ethical considerations. In this experiment, the goal was to design, build, and test an Electroencephalogram (EEG) circuit that would then be paired with a touchscreen display as a portable unit for use in Schizophrenic research at East Tennessee State University at the Quillen College of Medicine. Cell separation techniques are widely used in many biomedical and clinical applications for the development of screening, diagnosis and therapeutic tests. Current 3D microfluidics-based cell separation methods have limited applications in part due to low throughput and technical complexity. To address these critical needs, we have developed a 2D microfluidics surface which is the miniaturized version of a 3D microfluids cell separation device. The fabrication of microfluidic devices often requires specialized methods. The development of these methods requires careful characterization and understanding of the processes involved. Using primarily PolyJet 3D printing technology, microfluidics offers a wide scope of applications such as microfluidic benthic microbial fuel cells (MBMFCs) and electrostatic artificial muscles. MBMFCs benefit from the confinement of the microbes resulting in close proximity between the electrode and the organisms. Using a modular design called the Sponge, assembly and upscaling is possible. This work describes the design of an EEG-based neurofeedback system which provides users with real-time feedback on their level of focus or relaxation. By analyzing the spectral content of the brain activity measured via scalp electrodes, focus and relaxation levels can be quantified. Based on these measurements, live feedback in the form of moving bar graphs is provided to users, allowing them to gain awareness of their mental-state and more efficiently learn how to consciously relax or focus. Traditional electrodes used for electrophysiology recording, characterized by their hard, dry, and inanimate nature, are fundamentally mismatched with the soft, moist, and bioactive characteristics of biological tissues, leading to suboptimal skin-electrode interfaces. Hydrogel materials, mirroring the high water content and biocompatibility of biological tissues, emerge as promising candidates for epidermal electronic materials due to their adjustable physicochemical properties. Recent advancements in bioimpedance technology have demonstrated significant promise in the application of cardiac health monitoring. This research explores the design and development of a forearm-based wearable bioimpedance device for non-invasive measurement of heart rate and respiratory rate at an accuracy level comparable to medical-grade monitors. In this work, we developed and investigated the cancer cell killing efficacy of different instances using real-time high-sensitivity measurements facilitated by Guided-Mode Resonance (GMR) sensors. The GMR biosensor technology detects shifts in the spectral peak caused by changes in the refractive index of the surrounding medium, offering a label-free method for monitoring cellular events.

Breast Cancer Related Lymphedema (BCRL) is a common co-morbidity in cancer survivors following neoadjuvant therapies such as chemotherapy, radiation, and/or surgery. It is brought about by the disruption in the lymphatic system (think lymph node biopsy) that leads to a buildup of lymphatic fluid in the arm. This study focuses on the development of an adaptive ride-on toy specifically designed for a 2.5-year-old child with spastic diplegic cerebral palsy. As a feasibility study, the primary objective of this innovative device is to enhance motor function, foster autonomy, and improve the overall quality of life for the child. The purpose of this study was to investigate and compare the corrosion mechanisms between wrought and additive-manufactured (3D-printed) copper and stainless steel. The experimental procedure consisted of measuring the open circuit potential, electrochemical impedance spectroscopy, linear sweep voltammetry, Tafel analysis, surface topology, and scanning electron microscopy for each metal within salt water, tap water, sulfuric acid, and synthetic body fluid (excluding copper in synthetic body fluid).
Overall, printed stainless steel was more corrosion-resistant than wrought stainless steel in tap water and synthetic body fluid based on OCP, LSV, and surface topology results. Additionally, printed copper was more corrosion-resistant than wrought …
