Physical therapy requires patients to perform repeated actions to achieve meaningful results in rehabilitation. This thesis explores production methods and various sensor systems by utilizing rapid prototyping, inertial measurement units (IMUs), and capacitive sensor arrays (CSAs). CSAs can be made from a wide ar- ray of materials and techniques including 3d printing and laser ablation–to rapidly create CSAs that can be custom fit to enable proximity, force, and touch detection. Breast-conserving therapy (lumpectomy) is one of the most performed breast cancer surgeries in the United States. The best outcome of lumpectomy surgery is achieved when the surgical margins are free of cancer. When remnants of cancer are detected at the surgical margins after the initial operation, a second operation will be required to remove the cancer. Unfortunately, a significant number of patients undergo breast conserving surgery (BCS) at local hospitals that do not have access to immediate on-site pathology leading to high rates of re-excision or reoperation (greater than 30%). The severe side effects of acute dehydration and blood loss may be prevented if assessed and treated quickly. As amplifier technology has improved, small peripheral venous pressure (PVP) signals collected using a transducer on an intravenous catheter can be analyzed to monitor patient volume status. This thesis details the development of an embedded system prototype that quickly and accurately assesses volume status using PVP analysis. The volume prediction algorithm classified 10 seconds of PVP data as “dehydrated” or “resuscitated”. Nicotinamide adenine dinucleotide (NAD) is a cofactor that serves to shuttle electrons during metabolic processes such as glycolysis, the tricarboxylic acid cycle, and oxidative phosphorylation (OXPHOS) [1-5]. The reduced form of NAD, NADH, is autofluorescent, making it a measurable characteristic in the investigation of metabolic changes. The fluorescence lifetime of NADH is dependent on the state of protein binding, specifically free or bound NADH, allowing for detection of metabolic shifts between glycolysis (free NADH) and OXPHOS (bound NADH) [6-16]. The goal of this research is to characterize the electromagnetic properties of biological and non-biological materials at terahertz (THz), millimeter-wave, and microwave frequency bands. The biological specimens are measured using the THz imaging and spectroscopy system, whereas the non-biological materials are measured using the microwave and millimeter-wave free-space system. Peripheral venous pressure (PVP) can be used to measure blood volume status with a minimally invasive procedure. The pediatric cohort undergoing surgery for pyloric stenosis was studied to determine how arterial circulation and patient factors linearly impact PVP. The relationship between PVP and these confounding factors can provide valuable information for future PVP researchers.
To investigate the linear relationship between PVP and electrocardiogram (ECG) the waveforms were transformed into the frequency domain. Electrodeposited conductive copolymer films with predictable relative properties (quantities of functional groups for further modification and capacitance) are of interest in sensors, organic electronic materials and energy applications. Potentiodynamic copolymerization of films in aqueous solutions of two different thiophene derivatives, (2,3-dihydrothieno[3,4-b]dioxin-2-yl) methanol (1) and 4-((2,3-dihydrothieno[3,4-b][1,4]dioxin-2-yl)-methoxy)-4-oxobutanoic acid (2), containing 0.02 M total monomer (0, 25, 34, 50, 66, 75, 100 mol% 2), 0.05 M sodium dodecyl sulfate, and 0.1 M LiClO4, on gold microelectrodes in an array was investigated. Microdialysis (µD) sampling is a diffusion-limited sampling method that has been widely used in different biomedical fields for greater than 35 years. Device calibration for in vivo studies is difficult for current non-steady state analytes of interest correlated with both inflammatory response and microbial signaling molecules (QS); which exist in low ng/mL to pg/mL with molecular weights over a wide range of 170 Da to 70 kDa. This thesis proposes the use of a mesh morphing algorithm for the quantitative evaluation of terahertz (THz) images. This work differs from traditional evaluation methods based on qualitative evaluation because it provides a fair and quantitative measurement of the THz imaging system's performance. The objective of the algorithm is to match the alignment, shape, and resolution of the THz and reference pathology images. Therefore, the proposed morphing method provides a pathology reference for a pixel-by-pixel evaluation of the region classification in the THz image. 3D printing was used to fabricate porous artificial knee meniscus material from biocompatible polymeric blends of polycarbonate-urethane (PCU) and ultra-high-molecular-weight polyethylene (UHMWPE) to enable “weep” lubrication that mimics the native meniscus. 3D printed and molded pure PCU, as well as molded PCU and UHMWPE, were used for comparison. Preliminary printing was done to evaluate the impact of process parameters on the results. This research investigated the use of carbon nanotubes (CNTs) as a treatment to increase the permeability of a bacterial cell wall. Recombinant Escherichia coli BL21 (DE3) containing a plasmid that expressed Green Fluorescent Protein (GFP) and -lactamase were exposed to CNTs under various levels of agitation for different times. Fluorescence assay for GFP, optical absorbance for -lactamase activity, and Transmission Electron Microscopy (TEM) were used to determine the amount of released protein, and visually examine the permeability enhancement of the cells, respectively. Previous studies, both theoretical and experimental, of network level dynamics in the cerebral cortex show evidence for a statistical phenomenon called criticality; a phenomenon originally studied in the context of phase transitions in physical systems and that is associated with favorable information processing in the context of the brain. Electrospinning is an inexpensive technique that is used to produce nanofibers for a variety of applications. In electrospinning, a polymer solution is dispensed from a hypodermic-like syringe where an intense electric field attracts the solution to a collector while drawing the polymer into a very thin fiber. The diameter of the fiber can be controlled by tuning the process parameters such as the applied electric field, solution flow rate, distance between syringe tip and collector, and the collector geometry. Bone at the nanoscale consists of type I collagen and hydroxyapatite (HAP). Type I collagen and HAP [Ca10(PO4)6(OH)2] are responsible for most of the structural integrity of bone. Collagen fibrils contain HAP platelets of varying size dispersed between the collagen. We investigate heterotrimeric collagen interaction with HAP using Steering Molecular Dynamics to obtain the force-displacement relation as the collagen is undergoing shearing and peeling on the surface of HAP. A transparent nanofluidic system with embedded sensing electrodes was designed and fabricated by integrating Atomic Force Microscopy (AFM) nanolithography, Focused Ion Beam (FIB) milling and metal deposition, and standard microfabrication processing. The fabrication process started with the evaporation of chrome/gold (Cr/Au) onto a Pyrex 7740 wafer followed by photolithography and wet etching of the microchannels. In this dissertation, the relationship between the geometry of ion-beam sculpted solid-state nanopores and their ability to analyze single DNA molecules using resistive pulse sensing is investigated. To accomplish this, the three dimensional shape of the nanopore is determined using energy filtered and tomographic transmission electron microscopy. It is shown that this information enables the prediction of the ionic current passing through a voltage biased nanopore and improves the prediction of the magnitude of current drop signals when the nanopore interacts with single DNA molecules. Solid State nanopores that are fabricated by the ion beam sculpting process and electron beam drilling have shown great promise as a sensing device for DNA and protein molecules. Even though biological pores such as the alpha-Haemolysin have been in use for quite some time, the use of solid state Nanopores in single biomolecule detection has been on the rise since the mid 1990s.
