This project calibrated two software programs used for estimating the traffic impacts of work zones. The WZ Spreadsheet and VISSIM programs were recommended in a previous study by the authors. The two programs were calibrated using field data from two work zones in Missouri. Both work zones involved a single lane closure on a three-lane section of roadway. The I-44 work zone was a long-term work zone, while the I-70 work zone was temporary, lasting only a few hours. This study investigated the effect of an alternative merge sign configuration within a freeway work zone. In this alternative configuration, the graphical lane closed sign from the MUTCD was compared with a MERGE/arrow sign on one side and a RIGHT LANE CLOSED sign on the other side. The study measured driver behavior characteristics including speeds and open lane occupancies. The measurements were taken at two identical work zones on I-70 in Missouri, one with the new test sign and the other with the standard MUTCD sign. The objective of this study was to develop evaluation bases for the repair, rehabilitation, and replacement of existing railway bridges. Considering the importance of bridges to the railway network, the research involved the review and analysis of major factors that influence structural performance, e.g., degree of corrosion, loss of material (section loss, holes), fatigue problems, and loose connections. In particular, the research focused on connections, and their importance relative to the evaluation of bridge performance. The new Highway Safety Manual (HSM) contains predictive models that need to be calibrated to local conditions. This calibration process requires detailed data types, such as crash frequencies, traffic volumes, geometrics, and land-use. The HSM does not document in detail techniques for gathering such data, since data systems vary significantly across states. The calibration process also requires certain decisions, such as the correct sampling approach, determination of the minimum segment length, the treatment of left-turn phasing, and the inclusion or exclusion of speed-change lane crashes. Quantifying actual evapotranspiration (ETa) of riparian zones is important for more robust water balance analyses that will enable better planning, managing, and allocating of water resources as well as developing strategies to protect delicate riparian ecosystem functions. To increase RAP materials by up to 75% by binder replacement, a fractionation method was applied to the RAP stockpile by discarding RAP materials passing No. 16 sieve. This fractionation method was effective in improving volumetric properties of the HMA mixture with high RAP contents. The mix designs were then performed for both HMA and WMA with varying fractionated RAP amounts accounting for a replacement of 20, 30, 40, 50 and 75% of the mixture’s optimum asphalt binder content. The Missouri HAL manual is used to identify, analyze, and correct high crash locations, and has not been updated since 1999. This new edition brings the manual up to date, while incorporating the methodology of the national Highway Safety Manual (HSM). This 4th edition represents a complete re-working of all existing chapters of the manual. The changes are both stylistic and substantive. A contemporary book-style stylesheet was used to improve the appearance of figures, tables, headings, and labels. Livestock manure treatment and storage structures are potential environmental sources of antimicrobials and antimicrobial resistance genes (ARGs). In this study, the occurrence of antimicrobials and ARGs was investigated in the water and the sludge compartments of beef cattle storage ponds and swine lagoons. Analysis was focused on two families of antimicrobials (sulfonamide and tetracycline) and the corresponding ARGs (sul1, sul2, tetO, tetQ and tetX). Results showed that the pseudo-partitioning coefficients of tetracyclines were higher than those of sulfonamides, suggesting different distributions of these two classes of antimicrobials between water and sludge. Extreme wet and dry years (± 1 standard deviation, respectively), as well as the top 95 percentile (P95) of daily precipitation events, derived from tropical cyclone (TC) and nontropical cyclone (NTC) rainfall, were analyzed in coastal river basins in Southern Oaxaca, Mexico (Río Verde, Río Tehuantepec, and the Southern Coast). The study is based on daily precipitation records from 47 quality-controlled stations for the 1961 to 1990 period and TC data for the Eastern Tropical Pacific (EPAC). Research shows that a high percentage of crashes that take place on high-speed rural expressways occur at intersections with minor roads. One low-cost alternative design for improving the safety of at-grade intersections on such expressways is the J-turn. In the last few years, the Missouri Department of Transportation has converted some two-way stop controlled (TWSC) intersections into J-turns. This study evaluated the effectiveness of the J-turn intersection design in Missouri utilizing field studies, a public survey, crash analysis, and traffic conflict analysis. The objective of this paper is to investigate the performance of adhesive joints of carbon/epoxy wind turbine blade subjected to combined bending and tension loadings through finite element method. The influence of adhesive material properties and geometrical details including fillet and imperfections was examined in terms of interlaminar stresses in the adhesive layer. The variation of stress intensity with change in adhesive shear modulus has also been investigated, while contour integral method was used for evaluating the stress intensity factors (SIF) at the imperfection tip. Beef feedlot manure distributed to row crop production areas is a potential surface water contaminant source of the steroid hormones commonly used in beef cattle production. This article reports on research conducted at the University of Nebraska Haskell Agricultural Laboratory near Concord, Nebraska, in July 2009. Manure, collected from beef feedlot pens, was stockpiled for ten months prior to application to a row crop field. Previous research identified that the detection frequency of steroid hormones in beef manure varies greatly. This study was conducted to measure the effects of slurry application method, swine growth stage, and flow rate on runoff nutrient transport. Swine slurry was obtained from production units containing grower pigs, finisher pigs, or sows and gilts. The swine slurry was applied using broadcast, disk, or injection methods at a rate required to meet annual nitrogen requirements for corn. Three 30 min simulated rainfall events, separated by 24 h intervals, were applied to the experimental plots at an intensity of 70 mm h-1. Hourly evapotranspiration (ET) crop coefficients (Kc) are needed to optimize the effectiveness and efficiency of high-frequency micro- and sprinkler irrigation practices involving the application of water multiple times a day. However, not much is known about the daily and seasonal patterns and magnitudes in hourly Kc values for soybean. Few studies have been conducted to investigate the potential contamination of groundwater from manureborne steroid hormones. The objective of this study was to monitor leaching of steroid hormones at the bottom of 2.4 m deep percolation lysimeters installed in plots treated with beef cattle manure. Soil samples were also collected from surrounding field plots. Treatments consisted of application of manure from two treatment methods (stockpiling and composting) or no manure application. Laboratory analyses of manure, soil, and leachate samples used liquid chromatography and tandem mass spectrometry to identify 17 steroid hormones and metabolites. Subsurface drip irrigation (SDI) was used to irrigate corn and dry beans in a crop rotation study for four years from 2005 to 2008 to evaluate the accumulative effect of irrigation amount on yield and irrigation water use efficiency (IWUE). This study was conducted on a very fine sandy loam soil using a short season corn hybrid typical for the area. The senior capstone experience within the Department of Biological Systems Engineering at the University of Nebraska-Lincoln is a two-semester, two-course sequence intended to give senior students realistic design experience, working with real projects, real clients, faculty consultants, and teammates to produce a deliverable that meets the client’s needs. Students within this course sequence come from two different degree programs (agricultural engineering and biological systems engineering) and within each degree program from a variety of “emphasis areas”, e.g. biomedical engineering, environmental engineering, machine design. Background—DNA delivery systems, which transport exogenous DNA to cells, have applications that include gene therapy, tissue engineering and medical devices. Although the cationic nonviral DNA carrier polyethyleneimine (PEI) has been widely studied, the molecular factors and pathways underlying PEI-mediated DNA transfer remain largely unknown, preventing the design of more efficient delivery systems.
Methods—HEK 293 T cells were treated with polyplexes formed with PEI and pEGFPLuc encoding for green fluorescent protein (GFP). Transfected cells expressing GFP were flowseparated from treated, untransfected cells. Studies in regions of extensive irrigation practices have revealed a significant influence of evaporative cooling on regional temperatures as a result of surface energy redistribution during evaporation. In the U.S. High Plains, maximum temperatures during the last quarter of the 20th century have been decreasing. We investigated the trends in evapotranspiration (ET or latent heat) fluxes originating from increasing irrigation practices in the High Plains region from 1981 to 2008. Platoons are created by signalized intersections which control travel patterns of vehicles. It is common for platoons to disperse during normal traffic operations. While platoon dispersion has been studied extensively under homogenous and lane disciplined traffic conditions, this characteristic under heterogeneous conditions has not been addressed adequately. This study will be one of the first steps in this direction and will lead to better understanding and simulation of platoon dispersion under Indian conditions.

Due to the use of antimicrobials in livestock production, residual antimicrobials and antimicrobial resistance genes (ARGs) could enter the environment following the land application of animal wastes and could further contaminate surface and groundwater. The objective of this study was to determine the effect of various manure land application methods on the fate and transport of antimicrobials and ARGs in soil and runoff following land application of swine manure slurry.
