The assessment and characterization of atmospheric aerosols and their optical properties are of great significance for several applications such as air pollution studies, atmospheric visibility, remote sensing of the atmosphere, and impacts on climate change. Particle mass and number concentration measurements during different seasonal periods were conducted to study the effects caused by both privately own vehicles (POV) and commercial operated vehicles at the International Bridge of the Americas (BOTA), El Paso, Texas. This thesis is a presentation of particulate matter data collected from February 2007 to June 2008. Personal environment monitors (PEM), which measured 7-day PM2.5 concentrations, had been deployed at 14 homes during the 2007 year and deployed in 16 different homes during the 2008 sampling year. Within each sampling year, sampling occurred for two one-week (7-day) periods during each season. In addition to PEM sampling, a tapered element oscillating microbalance (TEOM) continuous sampler had been deployed in both indoors and outdoors at one pre-selected household per week. Traffic-related air pollution can be a major public health concern in any urban area. This problem is compounded in the Paso del Norte region that has experienced rapid economic growth, and a substantial number of people living in close vicinity of major roadways. The desert surroundings, arid weather, frequent temperature inversions, heavy border traffic at the international ports of entry between El Paso and Ciudad Juárez, and poorly maintained vehicle fleet further exacerbates this problem. A growing body of air quality and epidemiologic research has discerned the linkage between traffic emissions and respiratory disorders.

This Dissertation characterizes the first asphaltene DSSC (dye-sensitized solar cell), in which light to electricity conversion efficiency has improved from 0 % to 1.8 %. Four natural organic asphaltene factions, namely, un-fractionated asphaltene, Fr. 1 Asphaltene, Fr.2 asphaltene, Fr.3 and Fr.4 were extracted and tested for various solar cell parameters of TiO2 based dye-sensitized solar cell DSSC. DSSCs were fabricated using the four individual asphaltene dyes. The photovoltaic performance of the cells was examined by determining the various parameters such as open-circuit voltage, short-circuit current, fill factor, and series resistance. Recent decades have brought an increasing concern of potential adverse human and ecological health effects resulting from Endocrine Disruptor Compounds (EDCs). In particular from new emerging compounds such as natural estrogens (e.g., 17©¬-estradiol, estrone), synthetic estrogens (e.g., 17Â¥á-ethynylestradiol), bisphenol A (BPA), nonylphenol ethoxylates (NPEOS), and nonylphenol (NP). Apparent soil electrical conductivity (ECa) is typically mapped to define soil spatial variability within an agricultural field. Knowledge of the vertical variability of ECa is desired to define the site‐specific behavior of the soil profile. A pneumatic angular scanning system (PASS) was developed to sense horizontal and vertical changes of ECa on‐the‐go with an electromagnetic induction (EMI) instrument using an angular scanning method. This sensor system consists of a sled with a rotating mechanism, an EMI sensor, an inclinometer, and a pneumatic actuator. An earlier version of a storm runoff simulator to test conservation buffers reproduced target hydrographs and sedigraphs using uniform, fine sand; however, it was unable to uniformly mix and deliver native sediment. The objectives of this work reported were to create a method to process native agricultural sediment, mix a uniform sediment slurry at a target concentration, and create a control system that will deliver the slurry in varying flow rates corresponding to a target sedigraph. Holocarboxylase synthetase (HCS) is a chromatin protein that is essential for mediating the covalent binding of biotin to histones. Biotinylation of histones plays crucial roles in the repression of genes and repeats in the human genome. We tested the feasibility of DNA adenine methyltransferase identification (DamID) technology to map HCS binding sites in human mammary cell lines. Full-length HCS was fused to Dam for subsequent transfection into breast cancer (MCF-7) and normal breast (MCF-10A) cells. Flocculation is a common and inexpensive method for harvesting algae from solution. After nitrogen starvation, it was shown that 83 + 3% of the wall-deficient cells of the cw 15 mutant of Chlamydomonas reinhardtii flocculated from 12 mL samples within 15 min after the addition of 15 mM calcium chloride at pH 8.4. Only 24 2% of the wildtype strain flocculated under these conditions, thus demonstrating how a simple mutation might facilitate process design. The data suggested that algae grown in waters with similar calcium concentrations (e.g. certain wastewaters) might be harvested through simple pH adjustment. It was also discovered …

Estimation of actual evapotranspiration (ET), especially its partitioning into plant transpiration (T) and soil evaporation (E), in agricultural fields is important for effective soil water management and conservation and for understanding the interactions between ET, T, and E with the management practices. This Extension Circular describes various irrigation efficiency, crop water use efficiency, and irrigation uniformity evaluation terms that are relevant to irrigation systems and management practices currently used in Nebraska, in other states, and around the world. The definitions and equations described can be used by crop consultants, irrigation district personnel, and university, state, and federal agency personnel to evaluate how efficiently irrigation water is applied and/or used by the crop, and can help to promote better or improved use of water resources in agriculture.

The use of precision agriculture technologies such as automatic boom section control allows producers to reduce off-target application when applying herbicides. While automatic boom section control provides benefits, pressure differences across the spray boom resulting from boom section actuation may lead to off-rate application errors. Off-rate errors may also result from spray rate controller compensation for ground speed changes or velocity variation across the spray boom during turning movements. Imaging is essential in accurately detecting, staging, and treating primary liver cancer (hepatocellular carcinoma [HCC]), one of the most prevalent and lethal malignancies. We developed a novel multifunctional nanoparticle (NP) specifically targeting glypican-3 (GPC3), a proteoglycan implicated in promotion of cell growth that is overexpressed in most HCCs. Quantitative real-time polymerase chain reaction was performed to confirm the differential GPC3 expression in two human HCC cells, Hep G2 (high) and HLF (negligible). These cells were treated with biotin-conjugated GPC3 monoclonal antibody (aGPC3) and subsequently targeted using superparamagnetic iron oxide NPs conjugated to streptavidin and Alexa Fluor 647. Identifying molecular mechanisms or therapeutic targets is typically based on large-scale cellular analysis that measures the abundance of mRNA or protein; however, abundance does not necessarily correlate with activity. We report a method for direct large-scale quantification of active pathways that employs a cellular array with parallel gene delivery of constructs that report pathway activity. The reporter constructs encode luciferase, whose expression is influenced by binding of transcription factors (TFs), which are the downstream targets of signaling pathways. Luciferase levels are quantified by bioluminescence imaging (BLI), which allows for rapid, noninvasive measurements. Activity profiles by BLI of 32 TFs were …

Inefficient gene delivery is a critical factor limiting the use of nonviral methods in therapeutic applications including gene therapy and tissue engineering. There have been few efforts to understand or engineer the molecular signaling pathways that dictate the efficacy of gene transfer. Microarray analysis was used to determine endogenous gene expression profiles modulated during nonviral gene transfer. Nonviral DNA lipoplexes were delivered to HEK 293T cells. Flow cytometry was used to isolate a population of transfected cells. Expression patterns were compared between transfected and nontransfected samples, which revealed three genes that were significantly upregulated in transfected cells, including RAP1A, …

We present measurements which show that the tidal Ems River in Germ any is extremely muddy over a 30 km + turbid zone, with fluid mud o f 1-2 m thickness covering the bed with suspended sediment concentrations (SSC) o f greater than 50 kg.m-3. Moreover, we show that these elevated SSC contain large quantities of organic material which deplete dissolved oxygen (DO) and produce summertime hypoxic zones. Using mathematical modeling, we develop simplified representations o f the estuary physics that reproduce the tidally-averaged circulation, SSC distribution, and oxygen depletion. Buildings are a significant energy consumer and are responsible for an increasingly large percent of worldwide greenhouse gas emissions, currently between 30 and 40 percent. Energy efficiency presents unique opportunities for building owners to reduce their environmental footprint and add value through cost savings, tax deductions, and increased market value. An analysis of 183 samples of efficiency measures in seven technology categories found that 74% of efficiency investments had a positive net present value. Building automation system and chiller plant improvements had the highest mean energy and carbon dioxide savings per square foot. Additionally, building automation systems had, on average …

A system dynamics model was developed to estimate the carbon dioxide (CO2) emissions associated with conveyance of water from the water source to the distribution laterals of the Las Vegas Valley. In addition, the impact of several water management policies, including water conservation, reuse, and population growth rate change was evaluated. The results show that, at present, nearly 0.53 million metric tons of CO2 emissions per year are released due to energy use for water conveyance in distribution laterals of the Valley from Lake Mead, located 32.2 km (20 miles) southeast of the Las Vegas at an …

In the rural areas of Madagascar only 29% of the population has access to clean water and 10% has access to improved sanitation. It has been estimated that environmental risk factors, such as inadequate access to clean drinking water and proper sanitation, are responsible for 94% of the diarrheal disease burden. This study was focused on testing a point-of-use technology called the Tulip filter, which is a siphon ceramic candle filter impregnated with silver. The purpose is to assess its feasibility for implementation in rural regions of Madagascar through a laboratory study performed at the University of South Florida.

With increasing water reuse applications and upcoming stringent regulations for treated wastewater effluent discharge, wastewater plants need to consider alternative technologies beyond conventional treatment processes. The new regulations, Numeric Nutrient Criteria (NNC), may regulate discharge nitrogen and phosphorus concentrations to as low as 0.5 mg/L as N and 10 μg/L as P respectively. To meet these target requirements, system retrofitting to incorporate chemical or advanced nutrient removal systems possibly with membrane technology will most likely be required. The high incidence of upper respiratory diseases, contamination of waterways due to pathogens and nutrients from human and animal wastes, unsustainable deforestation, gender disparities in burden of disease due to unequal exposure to indoor air pollutants, and carbon black emissions from the burning of solid fuels are interrelated problems in many developing countries. Biofouling, or the formation of biofilm on membrane surfaces, can decrease the performance (decreased flux and/or increased operating pressure) of a reverse osmosis (RO) membrane system in a water treatment plant. However, biofilms have been used in water treatment systems to remove organic carbon from water via biofilters and successfully reduce biofilm growth downstream. This research investigates the possibility that the heterotrophic biofilm present on membrane surfaces removes nutrients from the treatment water, thereby making it nutrient deprived as it travels along the treatment train. Natural contamination of groundwater by arsenic (As) has become a critical public health threat in many parts of the world. The well-known regions associated with As contamination of groundwater are Bangladesh and West Bengal, India where approximately 100 million people are exposed to high levels of arsenic by drinking arsenic-contaminated groundwater and about 35 million are already affected. Long-term drinking of arsenic-contaminated water leads to arsenicosis, which is characterized by cancers of the skin, organ disease and certain other types of cancer. The United Nations Millennium Development goals aim to make improvements in access to potable water. In the Bocas del Toro region of Panama, rainwater harvesting systems are making that goal more attainable. Rainwater harvesting, especially in rural, difficult access areas, may be a viable water source improvement that will allow a greater population to access improved water. This study uses the Carnegie Mellon University Economic Input-output Life Cycle Assessment tool to assess the embodied energy of plastic and ferrocement rainwater harvesting systems in rural Panama. Soil-aquifer treatment (SAT) has been proposed as a method for reusing treated municipal wastewater. SAT is characterized by alternating cycles of aerobic and anaerobic conditions in the subsurface, in response to alternating cycles of flooding and drainage of a surface impoundment. It is not yet known how these alternating redox conditions affect the removal of potentially harmful endocrine-disrupting compounds (EDCs) from treated effluent.

This research estimates nitrogen removal from stormwater runoff using a denitrifying bioretention system using the USEPA Storm Water Management Model Version 5 (SWMM-5). SWMM-5 has been used to help planners make better decisions since its development in 1971. A conventional bioretention system is a type of Low Impact Development (LID) technology, which designed without a media layer specifically for achieving nitrogen removal. More recently studies have showed that high TN removal efficiencies are possible when incorporating a denitrification media layer. These systems are known as denitrifying bioretention systems, or alternative bioretention systems. LID projects are currently being designed and developed …
