TEMPO modified nanocellulose membranes are utilized across various laboratory and industrial applications from protein purification to advancements in kidney dialysis. This study investigates the effect of varying coagulation temperatures on the morphology and transport properties of these membranes to enhance their performance for targeted filtration. It was hypothesized that higher temperatures would accelerate the solvent-nonsolvent exchange during phase inversion, resulting in larger pore structures. Conversely, lower temperatures were expected to slow solvent diffusion, yielding smaller, more uniform pores. The membranes[JH1] were synthesized using 2,2,6,6-tetramethylpiperidinyl-1-oxyl (TEMPO) to improve selectivity and antifouling characteristics. Chronic kidney disease occurs when the kidneys lose the ability to effectively filter the blood, which causes waste build up. End-stage kidney disease (ESKD) is the most severe case of kidney disease and requires either dialysis or a kidney transplant. Hemodialysis, the most common form of dialysis, is an extremely time-intensive and physically burdensome process that is currently limited by membrane performance. This research evaluates the effectiveness of polymeric inhibitors at suppressing scaling during direct contact membrane distillation (DCMD). Anionic polyacrylic acid (PAA), neutral polyethylene glycol (PEG), and cationic polyethyleneimine (PEI), were investigated. Experiments were conducted using both hypersaline single-salt and multi-component mixed-salt feeds to determine how the presence of different ions influences performance. Results from single-salt experiments showed that PAA and PEG effectively maintained permeate flux through chelation and steric hindrance, respectively, while PEI caused rapid membrane failure due to interfacial aggregation and pore wetting. In complex mixed-salt solutions, PEG was the additive, maintaining a steady flux with only a 7.33% …

In pursuit of sustainability, we explored replacing conventional dissolved air floatation (DAF) in poultry processing wastewater (PPW) treatment with a precisely tuned 0.02 µm stainless-steel ultrafiltration (SSUF) membrane. SSUF is a robust, homogenously porous membrane with strong chemical resistance, ease of cleaning, and exceptional resistance to organic fouling. Unlike polymeric membranes, it can be regenerated multiple times, making it a cost-effective choice due to its compatibility with harsh chemical cleaning. The PPW used for the study was untreated wastewater from all processing units and post-initial screening. Our study revealed the SSUF membrane’s exceptional efficiency at eliminating contaminants. Biocompatibility and the ability to mediate the appropriate flux of ions, urea, and uremic toxins between blood and dialysate components are key parameters for membranes used in dialysis. Oxone-mediated TEMPO-oxidized cellulose nanomaterials have been demonstrated to be excellent additives in the production and tunability of ultrafiltration and dialysis membranes. In the present study, nanocellulose ionic liquid membranes (NC-ILMs) were tested in vitro and ex vivo. An increase in flux of up to two orders of magnitude was observed with increased rejection (about 99.6%) of key proteins compared to that of polysulfone (PSf) and other commercial membranes. Produced water (PW) generated from hydraulic fracturing operations was treated using an integrated electrocoagulation, ultrafiltration, membrane distillation, and crystallization process (EC UF MDC). The aim was to determine the viability of this integrated process for maximizing water recovery. The results obtained here indicate that optimizing the various unit operations could lead to increased recovery of PW. Membrane fouling limits all membrane separation processes. A pretreatment step to suppress fouling is essential. Here, removal of total suspended solids (TSS) and total organic carbon (TOC) was achieved by electrocoagulation (EC) followed by ultrafiltration (UF). Society relies on plastic products, whether they are single use or durable. A downside of plastic is that the most common type is a product of oil and oil is not only a limited resource but also a climate-damaging resource. Polylactic acid (PLA) is a bio-based, biodegradable plastic. However, the process of converting biomass to polylactic acid polymer has the largest environmental impact of the PLA production process, so alternative methods of conversion are needed (Moretti et al., 2021). Chapter 1 of this dissertation introduces these challenges and covers previous research that has been done in those areas. Additionally, the objectives of this dissertation are stated. In chapter 2, a study was done in which polyethersulfone ultrafiltration membranes were functionalized with copper oxide nanoparticles and zwitterions for the purpose of reducing fouling on the membrane surface. Virus filters are an essential component of the overall virus clearance strategy. Microcystin-LR (MC-LR) is a toxin produced by cyanobacteria that can bloom in freshwater supplies. This study describes a new strategy for remediation of MC-LR that combines linearization of the toxin using microcystinase A, MlrA, enzyme with rejection of linearized byproducts using membrane filtration. The MlrA enzyme was expressed in Escherichia coli (E. coli) and purified via a His-tag with 95% purity. Additionally, composite membranes made of 95% polysulfone and 5% sulfonated polyether ether ketone (SPEEK) were fabricated and used to filter a solution containing cyclic and linearized MC-LR. Micro- and ultra-filtration membranes are commonly used in biopharmaceutical applications such as filtering viruses and separating proteins from a carrier solution. A groundwater treatment facility was designed for Lawton, Oklahoma to address potential water scarcity due to drought conditions in southwest Oklahoma. The facility will produce five million gallons per day (MGD) of treated water. The plant will have the capacity of treating 4,085 gallons per minute of influent water at an 86% recovery. The water to be treated will come from the Arbuckle-Timbered Hills Aquifer. Studies have been conducted around the aquifer to identify the best well site locations.

Membrane distillation is an emerging technology for treating hydraulic fracturing flowback and produced waters. Suppression of membrane fouling by inorganic and polar and non-polar organic compounds is a challenge. Here polyhydroxyethyl methacrylate, polyacrylic acid, polvinylallyl imidazolium bromide and polyvinylhexyl imidazolium bromide chains have been grafted from the membrane surface. Fouling is initially due to adsorption of organic compounds followed by scale formation. When challenged with produced water, membranes modified with polvinylallyl imidazolium bromide chains provided the greatest resistance to fouling. For EC pretreated produced water and synthetic produced water that contained mainly inorganic species, the flux decline was much less.

Virus filtration is an integral part of the downstream purification of mammalian cell culture-derived biotherapeutics to assure the viral safety of the products. Virus filtration membranes remove viruses based on a size-exclusion mechanism. Commercial parvovirus filers possess unique membrane structure and are designed to remove smaller non-enveloped parvoviruses with size 18-26 nm. Electrospun membranes are an attractive alternative to flat sheet membranes as absorbent with numerous advantages like high porosity, large specific surface area and ease of functionalization. This doctoral dissertation focuses on fabricating novel polymeric membrane adsorbents for protein separations and ammonium ion removal. Three distinctly different preparation methods including UV-initiated polymerization, atom transfer radical polymerization, and mixed-matrix formation, have been employed to fabricate the electrospun membranes. Polyaniline coated cellulose membranes show impressive conductive properties that may be used to innovate traditional charged separation techniques, such as electrodeionization. However, these membranes are not sold to consumers, so they cannot be easily integrated into such systems. In this work, we have made and characterized a pair of immunobiosensors for detecting the green fluorescent protein (GFP) in an aqueous matrix. An anti-GFP antibody-based biosensor was assembled to detect GFP, while a novel peptoid (N-substituted oligomers of glycine designated as IOS-1) biosensor was also assembled for GFP detection. A quartz crystal microbalance (QCM) gold sensor was used as the supporting substrate for self-assembly of the immunobiosensors. Gravimetric measurements of the QCM gold sensor during immunobiosensor construction and operation were available in real-time using a QCM instrument. Noble metal nanoparticles and two-dimensional (2D) transition metal dichalcogenide (TMD) crystals offer unique optical and electronic properties that include strong exciton binding, spin-orbital coupling, and localized surface plasmon resonance. Controlling these properties at high spatiotemporal resolution can support emerging optoelectronic coupling and enhanced optical features. Highly conserved throughout evolution, lipins are dual functioning proteins found from yeast to humans. Functioning in the cytoplasm as phosphatidate phosphatase enzymes (PAP), lipins produce diacylglycerol that serves as a precursor for neutral fats and membrane phospholipids. Alternatively, nuclear lipins are responsible for the regulation of metabolic genes. Interestingly, both the mammalian lipin 1 paralog and the single Drosophila Lipin ortholog are highly phosphorylated proteins. Target of rapamycin (TOR) has previously been identified as one of the kinases that controls the subcellular localization of both lipin 1 and Drosophila Lipin.
