Ethanol stress assays are commonly used to evaluate microbial tolerance, metabolic adaptation, and fermentation performance. However, manual liquid handling introduces variability across replicate wells and small-volume pipetting steps, limiting reproducibility and throughput. This study developed an automated OT-2 robotic workflow to generate replicated ethanol concentration gradients for high-throughput inhibition assays in engineered thermophilic biofuel strains. Kinetic plate-reader measurements were used to quantify ethanol-dependent growth responses under anaerobic fermentation conditions. The reasearch question is: How do engineered thermophilic biofuel strains differ in ethanol-dependent growth inhibition under anaerobic fermentation conditions, and can automated robotic assays improve the reproducibility of these measurements? Heavy soils, with low rates of soil-water movement, such as those found in the South-West Irrigation Area (SWIA), require closely spaced (2-6 m apart) subsoil drainage systems to provide sufficient water movement to control the effects of salt-waterlogging on pastures. Australia has undertaken a strategic review of current and historical deep drainage projects. A 'rapid appraisal' methodology was utilised, based principally on existing hydrological investigations and interpretation and anecdotal evidence provided by landholders to clarify the role of drainage in managing water in dry land rural landscapes.
The objectives of this discussion paper are to:
1. Review the current status of groundwater drainage practice

The need to fence for efficient livestock control was recognised early in the history of the pastoral industry in Western Australia. Fence design, materials and layout have all undergone many changes since the 1880s. Modern fences have 4 or 5 strands of high tensile light gauge wires with post spacings up to 50m, and strains up to 4000m. These modern fences contain less material and are faster to erect than traditional fences, yet they provide an equally good, if not superior, barrier. The happy coincidence of strength and cost effectiveness will be apparent through this manual.
