Cancer is a heterogeneous disease caused by unchecked cell proliferation. The disease contributed to 9.7 million deaths worldwide in 2022, with hematological malignancies (leukemias, lymphomas, and myelomas) accounting for over 1.4 million deaths in the United States from 1999 to 2020. This thesis focuses on the treatment of these cancers, with an emphasis on non-Hodgkin lymphomas (NHLs). R-CHOP (Rituximab-Cyclophosphamide, Doxorubicin hydrochloride, Oncovin/Vincristine, and Prednisone) is the standard of care first-line treatment for diffuse large B-cell lymphoma (DLBCL), the most common subtype of NHL, and encompasses a mixture of chemotherapy, immunotherapy, and corticosteroids. Inherited metabolic diseases (IMDs) affect approximately 1 in 800 individuals. The only curative treatment for IMDs is liver transplantation. However, transplantation carries significant risks, including organ rejection and a lifelong need for immunosuppression. Therefore, gene editing has emerged as a promising alternative to liver transplantation for treating IMDs.

Familial hypercholesterolemia (FH) is and IMD characterized by elevated LDL-C levels resulting in premature cardiovascular disease. CRISPR-Cas9-mediated gene editing to disrupt the gene encoding angiopoietin-like 3 (ANGPTL3) in the liver represents a novel therapeutic approach for FH. Intracellular reactive oxygen species (ROS) generation is widely suggested as a trigger for biological consequences of electric field exposures, such as those in electroporation applications. ROS are linked with membrane barrier function degradation, genetic damage, and complex events like immunological cell death. Dihydroethidium (DHE) is commonly used to monitor ROS in cells. DHE is linked to intracellular ROS by a primary oxidation product, Ethidium (Eth+), that shows increased fluorescence upon binding to polynucleotides. We observed changes in DHE-derived fluorescence in Chinese hamster ovary (CHO) cells post 300-ns electric pulse exposures, comparing them to tert-butyl-hydroperoxide (t-BHP) induced oxidative stress. …

Reversing the pulse polarity, i.e., changing the electric field direction by 180°, inhibits electroporation and electrostimulation by nanosecond electric pulses (nsEPs). This feature, known as “bipolar cancellation,” enables selective remote targeting with nsEPs and reduces the neuromuscular side effects of ablation therapies. We analyzed the biophysical mechanisms and measured how cancellation weakens and is replaced by facilitation when nsEPs are applied from different directions at angles from 0 to 180°. Monolayers of endothelial cells were electroporated by a train of five pulses (600 ns) or five paired pulses (600 + 600 ns) applied at 1 Hz or 833 kHz. Reversing …

Inherited metabolic diseases (IMDs) of the liver have a collective occurrence of about 1 in 800 births. Current therapies for IMDs of the liver are limited, and the only curative option for patients is orthotopic liver transplantation. Due to shortages of organ donors, immunosuppression following the transplantation, and mortality risks associated with the procedure, alternative treatment options are necessary for curing IMDs of the liver. A large-scale in vitro 3D tumor model was generated to evaluate gene delivery procedures in vivo. This 3D tumor model consists of a "tissue-like" spheroid that provides a micro-environment supportive of melanoma proliferation, allowing cells to behave similarly to cells in vivo. This functional spheroid measures approximately 1 cm in diameter and can be used to effectively evaluate plasmid transfection when testing various electroporation (EP) electrode applicators. In this study, we identified EP conditions that efficiently transfect green fluorescent protein (GFP) and interleukin 15 (IL-15) plasmids into tumor cells residing in the 3D construct. Statistical computations are an important tool for the analysis of stochastic phenomena and processes that are characterized by variability. Biological systems (e.g., cells, tissues etc.) are perfect examples wherein response to a given external stimulus can be varied and needs to be adequately considered. The Monte Carlo method of analysis has now been recognized as the most effective way of treating stochastic variability.

Nanosecond pulsed electric fields (nsPEFs) are hypothesized to affect intracellular structures in living cells providing a new means to modulate cell signal transduction mechanisms. The effects of nsPEFs on the release of internal calcium and activation of calcium influx in HL-60 cells were investigated by using real time fluorescent microscopy with Fluo-3 and fluorometry with Fura-2. nsPEFs induced an increase in intracellular calcium levels that was seen in all cells. With pulses of 60 ns duration and electric fields between 4 and 15 kV/cm, intracellular calcium increased 200-700 nM, respectively, above basal levels (similar to100 nM), while the uptake of …

The purpose of the present research work is to analyze the flow of ions through pores formed in the plasma membrane of biological cells during electroporation. In this regard, simulations have been conducted to track the movement of ions in 3-D space based on a Brownian dynamics model, which is mostly deterministic but has some stochastic properties to account for ionic scattering. Pulsed electric fields (PEF's) above a certain voltage threshold cause electroporation for microsecond pulses and intracellular effects for submicrosecond pulses. Models describing these effects often depend on the electrical properties of the cell, which are altered by the PEF. Time domain dielectric spectroscopy was used to provide data for these models and to measure changes in the conductivity of HL-60 cell suspensions due to single 50 μs and 10 ns PEF's of the same energy. For 1.1 kV/cm, 50 μs pulses, the conductivity rose within a minute after the pulse and dropped dramatically approximately forty minutes after the pulse. For …
