In this work, the potential for PEGylated gold nanoparticles for use as a platform for stem cell delivery in treatment of myocardial infarction (MI) is preliminarily investigated. Cardiovascular disease is currently the leading cause of death worldwide, with majority of mortality resulting from coronary artery disease and associated MI. The ensuing ischemic conditions resulting from MI cause substantial heart muscle tissue loss and scarring in the heart. Adverse tissue remodeling creates a significant loss in heart function that can result in the formation of cardiac hypertrophy, ventricular dilation and arrythmias. Natural and synthetic polymers and biopolymers have been studied for a variety of applications in food emulsion, biopharmaceutical purification, tissue engineering, and biosensor. The structure and property of polymers and biopolymers are critically important to determine their functions. Molecular dynamics (MD) simulations have a unique advantage to explore the structure and property of polymers and biopolymers from the molecular level. In the dissertation, MD simulations were conducted to study the mechanisms of various biological and chemical processes controlled by polymers and biopolymers based on real-world experimental results.

Stroke is the leading cause of disability and a primary cause of mortality, resulting in an estimated $33 billion dollars spent on healthcare, rehabilitation and lost productivity in the United States each year. The most prevalent cause of stroke incidents are acute ischemic events, manifested as blood clots in the vasculature supplying the brain. Current gold standard treatments have improved since 2006 with the introduction of mechanical stent retrievers; however, several issues with the current treatments to acute ischemic stroke remain. Oxidative metabolism is required to produce adequate energy to sustain human life. A primary example of deteriorating oxidative capacity is seen in the cardiac musculature during chronic heart failure. This suggests that by improving oxidative potential, chronic heart disease could be mitigated and one approach to accomplish this may be through targeting the mt-mRNA translation system. The goal of this work is to investigate terahertz technology for assessing the surgical margins of breast tumors through electromagnetic modeling and terahertz experiments. The measurements were conducted using a pulsed terahertz system that provides time and frequency domain signals. Three types of breast tissues were investigated in this work. The first was formalin-fixed, paraffin-embedded tissues from human infiltrating ductal and lobular carcinomas. The second was human tumors excised within 24-hours of lumpectomy or mastectomy surgeries. The third was xenograft and transgenic mice breast cancer tumors grown in a controlled laboratory environment to achieve more data for statistical analysis.

The objective of this project was to develop the chicken as a convenient animal model to examine tissue targeting of intravenously (i.v.)-injected iron oxide (IO) nanoparticles. Volumetric muscle loss (VML) is an injury to skeletal muscle characterized by a loss of more than 20% of a muscles volume. The combination of the bulk loss of tissue, transection and separation of myofibers proximal and distal to the injury, loss of innervation and blood supply, and the depletion of muscle progenitor cells results in permanent fibrosis and functional deficits due to loss of contractile tissue. The accumulation of amyloid-beta (Aβ) oligomers is believed to be the driving force behind Alzheimer’s disease (AD) pathogenesis. Due to the metastable nature of Aβ oligomers, the knowledge of Aβ aggregation and accumulation is not well understood. Here, we use capillary electrophoresis (CE) and photo-induced crosslinking of unmodified proteins (PICUP) to learn about the aggregation of Aβ.

The performance of extracellular matrix (ECM) biological scaffolds for the treatment of volumetric muscle loss (VML) has shown promising results with regenerating native muscle in animal models and human patients. However, the limitations of these scaffolds include non-specific characteristics based on the original parameters of the muscle that is lost and this is why our group chose to characterize human skeletal ECM. Approximately 40,000 women will die from breast cancer in the United States in 2014. About 90% of these deaths will be due to metastases, rather than the primary tumor and majority of metastases are due to the recurrence and progression of non-metastatic disease. Current adjuvant treatments, such as chemotherapy and radiation, have severe side effects and may result in overtreatment and drug resistance.

Incomplete recovery during microdialysis sampling is hindering important research in neurology, proteomics, and immunology. Although the current generalized solution, decreasing volumetric flow rates (Q), has been and will remain to be a useful strategy it has reached it's a physical limitation due to evaporation at the collection site. A proof-of-concept microfluidic device combined with heparin-immobilized magnetic beads was created to concentrate cytokine proteins collected from microdialysis samples. Cytokines are known to be related to several diseases such as cancer, and Parkinson's diseases, so to be able to develop more effective diseases treatments their interactions have to be well understood. Amine-functionalized polystyrene and carboxyl-functionalized magnetic microspheres of ~6.0 ìm in diameter were used to immobilize heparin.
