Mesenchymal stem cells (MSCs) are a population of stromal cells found traditionally in the bone marrow and adipose tissues. They can also be found in other tissues including fallopian tube, core blood, peripheral blood, fetal liver, and lungs. Mesenchymal stem cells have profound effects in regenerative engineering, tissue repair and drug discovery owing to the excellent properties such as proliferation, self-renewal, and multipotency generating multiple cell types including adipocytes, osteocytes, cardiomyocytes (CMs), pericytes (PCs), and chondrocytes. The purpose of this project was to characterize the potential of various strains of purple non-sulfur bacteria for the production of fixed nitrogen fertilizers for the manned Mars missions. Six strains of Rhodopseudomonas palustris (CGA009, CGA010, TIE-1, NifA*, and PB23) were all investigated. Through initial growth trial experiments, R. palustris NifA* and PB23 were selected for their engineered nitrogen fixation and rapid growth respectively. Growth curves and ammonium concentrations were collected over time in pilot scale batch photobioreactors (200 mL). Biomass production was then scaled up to benchtop photobioreactors (1.5 L) and then 14 L total volume for both NifA* …

The traditional method for the carbonation of beer includes bubbling CO2 through a pressurized brite tank until the desired level of carbonation concentration is reached. The gas either dissolves in the liquid volume or passes through the beer into the gas headspace above the bulk volume of beer. The gas that passes through the liquid can strip the beer of flavors, and this undissolved gas is vented to the atmosphere. Astronauts traveling or researching in space are subject to a harsh environment that includes high levels of ionizing radiation and microgravity. Ionizing radiation is dangerous to overall health because it causes double-stranded DNA breaks, increases the production of free radicals or reactive oxygen species, and leads to an enhanced risk of cardiovascular disease and cancer over a person’s lifetime. Central Valley Water Reclamation Facility (CVWRF) in Salt Lake City is the largest municipal wastewater treatment plant in Utah and must meet new and rigorous nutrient effluent standards–over 95% reduction in phosphorus output by 2025. Filtrate from CVWRF anaerobic digesters contains high levels of nitrogen, phosphorus, and magnesium. Supersaturation of these constituents leads to nuisance struvite precipitation that clogs belts, pumps, and pipes downstream of anaerobic digesters. Struvite is a mineral precipitate composed of equimolar magnesium, ammonium, and phosphate. Controlled precipitation of struvite helps prevent clogging and scaling, removes phosphate from wastewater, and generates a marketable fertilizer product.

Choroidal neovascularization (CNV) is a major cause of blindness in patients with age-related macular degeneration. CNV is characterized by new blood vessel growth and subretinal fluid accumulation, which results in mechanical pressure on retinal pigment epithelial (RPE) cells. The overexpression of RPE-derived angiogenic factors plays an important role in inducing CNV. In this work, we investigated the effect of mechanical stress on the expression of angiogenic factors in porcine RPE cells and determined the impact of conditioned medium on in-vitro angiogenesis.

The economics of algae biofuels and bioproducts would be improved by increased biomass productivity. Two studies on this potential are described in this thesis – one on a locally isolated filamentous yellow-green alga and the other on a planktonic strain genetically improved via selective enrichment.

Polycultures have been viewed as productive, stable, and, in some cases, harvestable by natural bioflocculation. Local native strains might have higher productivity than culture collection strains because they are already adapted to local outdoor conditions. Nanofertilizers are increasingly explored for sustainable micronutrient delivery in agriculture. Pre-treating seeds with nanofertilizers prior to planting (i.e., seed priming) reduces concerns about nanoparticle (NP) fertilizer non-target dispersion; however, priming formulations and concentrations must be carefully selected to avoid germination inhibition and toxicity. Here we investigate changes in corn seed germination and seedling development after seed priming with ZnO NPs, ZnO bulk and ZnCl2. Through 2016 and 2017, a team led by Canadian virologist David Evans, and funded by an American pharmaceutical company, attempted to synthesize the previously extinct horsepox virus. After just six months and an expenditure of $100,000, the research team was able to successfully construct the virus “using only commercially available information, technology and tools.” In January of 2018, the team went on to publish their information in an American-based journal, PLOS ONE.

Acute respiratory distress syndrome (ARDS) is a life-threatening condition that causes diffuse alveolar damage and a loss of the extracellular matrix (ECM). This leads to pulmonary edema and lung function deterioration. Our lab has created decellularized porcine lung, electrosprayed ECM nanoparticles that have been previously shown to have pro-regenerative capabilities in vitro.

In this study, the ECM nanoparticle effects on young murine lungs were tested in vivo. Imaging of BF distributions helps characterize many diseases associated with tissue hypoxia/ischemia. The purpose of this study was to develop and validate a novel, noninvasive, noncontact, high-density camera-based speckle contrast diffuse correlation tomography (scDCT) device for use in both animal and human studies. Recent advances in three-dimensional (3D) cell culture systems have provided key insights into the understanding of biochemical and physiological states of native tissue. A significant progress in the field of mechanobiology involves measuring cellular traction forces in a more native 3D environment. Parkinsonian rigidity is caused by the inability of the muscles to relax and extend properly, due to reduced dopamine levels and often begins on one side of the body before spreading contralaterally. The current standard for determining joint rigidity in a clinical setting is a test completed by the clinician based on the feel of the relaxed wrist and elbow joints as they are passively flexed and extended and a series of ordinal rating scales, the Movement Disorder Society’s – Unified Parkinson’s Disease Rating Scale (MDS-UPDRS), Hoehn and Yahr Scale (H&Y), and Parkinson’s Disease Questionnaire-39 (PDQ-39). Patients with severe lung pathologies, such as Acute Respiratory Distress Syndrome (ARDS), often require mechanical ventilation as a clinical intervention; however, this procedure frequently exacerbates the original pulmonary issue and produces an exaggerated inflammatory response that potentially leads to sepsis, multisystem organ failure, and mortality. This acute lung injury (ALI) condition has been termed Ventilator-Induced Lung Injury (VILI). Alveolar overdistension, cyclic atelectasis, and biotrauma are the primary injury mechanisms in VILI that lead to the loss of alveolar barrier integrity and pulmonary inflammation.
