Salmonella Typhimurium is a foodborne pathogen associated with raw and undercooked eggs, poultry, beef, fruits, and vegetables. In the United States, Salmonella is responsible for approximately 1.2 million illnesses, 23,000 hospitalizations, and 450 deaths annually. For many years, conventional detection methods such as culture-dependent and PCR-based methods have been the “golden standards” for the detection of this pathogen due to their high sensitivity and reliability. Virus filtration is an integral part of the downstream purification of mammalian cell culture-derived biotherapeutics to assure the viral safety of the products. Virus filtration membranes remove viruses based on a size-exclusion mechanism. Commercial parvovirus filers possess unique membrane structure and are designed to remove smaller non-enveloped parvoviruses with size 18-26 nm. However, some filters face issues, such as pre-mature fouling, the decline of filtrate flux, and reduction in virus retention. Climate change affects water resources differently across geospatial regions in the United States (U.S). There is a concern of how water availability will be affected by changes in long-term temperature and precipitation patterns, specifically in major production regions for eight fruit and vegetable crops. The effects on surface water available for irrigation use and supply stress in five regions containing 31 Agricultural Statistics Districts (ASDs) were assessed. For this Honors Thesis project, I designed and fabricated a ventilation and recirculation airway sub-system prototype for a hydroponic demonstration unit to be used as an educational demonstration for prospective students, illustrating the capabilities of biological engineers. This thesis focuses on the design of an autonomous air system that toggles between ventilation and recirculation modes based upon sensor output. In the literature review I discuss how ventilation can be used to achieve a targeted climate, the benefits of hands-on projects for education, and the optimum parameters for growing herbs in a hydroponic system.

The traditional method for the carbonation of beer includes bubbling CO2 through a pressurized brite tank until the desired level of carbonation concentration is reached. The gas either dissolves in the liquid volume or passes through the beer into the gas headspace above the bulk volume of beer. The gas that passes through the liquid can strip the beer of flavors, and this undissolved gas is vented to the atmosphere. Necrotic skin flaps are difficult to predict and treat due to the lack of quantitative biomarkers. Label-free multiphoton microscopy is well suited for non-invasively monitoring skin metabolism through NAD(P)H and other intrinsic fluorophores, and offers immediate future directions for assessing necrosis in the clinic. The objective of this study was to assess whether phasor FLIM could be used to evaluate skin flap status and treatment efficacy in ex vivo skin sections. Phasor maps revealed differences in growth factor treatment and region, but changes in skin flap autofluorescence at 755nm excitation and 460nm emission were not just related to NAD(P)H. A …

Biofiltration without pre-ozonation has the capability to remove natural organic matter (NOM) fractions that serve as precursors of disinfection byproducts (DBPs), which include the four regulated trihalomethanes (THMs) and dichloroacetonitrile (DCAN). Rapid small-scale column tests (RSSCTs) and Pilot Plant filters operated at empty-bed contact times (EBCTs) of 4, 8, and 16 minutes were used to evaluate the performance of nutrient-amended (free ammonia and phosphorus) biofiltration for THM and DCAN precursor removal, as measured using formation potential (FP) tests. This study investigated the effect of nutrient addition on algal growth in three United States Forest Service lakes for fishery management in Arkansas. In fishery managed lakes, fertilization works by manipulating algae growth, a basal food resource in lakes, to promote the growth of the fish population. For the nutrient addition experiments, water was collected from each lake in cubitainers and spiked with nutrients; the treatments included the control, nitrogen (+N), phosphorus (+P), and nitrogen and phosphorus (+N +P). The objective of this project was to examine the effect of fungal treatment and liquid hot water pretreatment of switchgrass combine in view of increasing glucose release. The fungal treatment consisted of incubating Pleurotus ostreatus in square switchgrass bales, at 50% moisture content for 25 days, 54 days, and 82 days. The digestibility of the switchgrass biomass was subsequently evaluated using Accelerase 1500 enzyme. Lignin is an important barrier to enzymatic hydrolysis, and it was stipulated that incubation with P. ostreatus would disrupt plant cell walls, resulting in enhanced saccharification. Sweetgum bark extract has been known to show biological activities such as antimicrobial and antioxidant capabilities. The storage capacity of the extract, however, was unknown and previously thought to diminish over time. However, upon experimentation, the freeze-dried sweetgum bark extract showed no signs that storage time or storage temperature had any significant effect on the biological activities. There was no significant difference across storage temperature over time in the experiment (ANOVA RM, P0.05). In an effort to expand the pool of bacterium useful for biotechnology applications, Pseudomonas fluorescens, a common gram negative microbe, was examined for its ability to function in a recombinant setting. P. fluorescens is ubiquitous in nature and was initially identified as a soil bacterium found in dirt and is typically associated with plant material. This study investigated the effects of source water quality in Beaver Lake on the amount of chlorine (Cl) needed to develop decision support system to help guide chlorination practices in pre-treatment of source water. To produce fermentable sugars from lignocellulosic biomass feedstock, severe pretreatment conditions are needed (either high acid concentration, temperature, or retention times). High severities can produce toxic byproducts which inhibit enzymatic hydrolysis or fermentation. In order to reduce pretreatment severities (and thus increase enzyme and fermentation efficiency), the white-rot fungus Pleurotus ostreastus was seeded into square and round bales of Kanlow switchgrass (Panicum virgastum L.) and left in the field over a period of 9 month. The effect of compounds produced during biomass pretreatment on cellulolytic enzyme was investigated. Liquid prehydrolyzates were prepared by pretreating switchgrass using 24 combinations of temperature, time, and sulfuric acid concentration based on a full factorial design. Temperature was varied from 140°C to 180°C; time ranged from 10 to 40 min; and the sulfuric acid concentrations were 0.5% or 1% (v/v). Identified products in the prehydrolyzates included xylose, glucose, hydroxymethylfurfural (HMF), furfural, acetic acid, formic acid, and phenolic compounds at concentration ranging from 0 to 21.4 g/L.
