Detection And Interpretation Of Fluorescence Signals Generated By Excitable Cells And Tissues, Anthony J. Costantino Jan 2017

Detection And Interpretation Of Fluorescence Signals Generated By Excitable Cells And Tissues, Anthony J. Costantino

Graduate Dissertations and Theses

Part 1: High-Sensitivity Amplifiers for Detecting Fluorescence
Monitoring electrical activity and Cai 2+ transients in biological tissues and individual cells increasingly utilizes optical sensors based on voltage-dependent and Cai 2+-dependent fluorescent dyes. However, achieving satisfactory signal-to-noise ratios (SNR) often requires increased illumination intensities and/or dye concentrations, which results in photo-toxicity, photo-bleaching and other adverse effects limiting the utility of optical recordings. The most challenging are the recordings from individual cardiac myocytes and neurons. Here we demonstrate that by optimizing a conventional transimpedance topology one can achieve a 10-20 fold increase of sensitivity with photodiode-based recording systems (dependent on …

With the increasing prevalence of diabetes in the United States and worldwide, blood glucose monitoring must be accurate and reliable. Current enzymatic sensors have numerous disadvantages that make them unreliable and unfavorable among patients. Recent research in glucose affinity sensors correct some of the problems that enzymatic sensors experience. Dextran and concanavalin A are two of the more common components used in glucose affinity sensors. When these sensors were first explored, a model was derived to predict the response time of a glucose affinity sensor using concanavalin A and dextran. However, the model assumed the system was linear and fell …

Synthesis, Characterization, And Activity Of Co/Fe Alumina/Silica Supported Ft Catalysts And The Study Of Promoter Effect Of Ruthenium, Sunday Azubike Esumike Jan 2017

Synthesis, Characterization, And Activity Of Co/Fe Alumina/Silica Supported Ft Catalysts And The Study Of Promoter Effect Of Ruthenium, Sunday Azubike Esumike

Doctoral Dissertations

The alumina and hybrid alumina-silica FT catalyst were prepared by one-step solgel/oil-drop methods using metal-nitrate-solutions (method-I), and nanoparticle-metaloxides (method-2). The nanoparticle-metal-oxides did not participate in solubility equilibria in contrast to metal nitrate in method-1 causing no metal ion seepage; therefore, method-2 yields higher XRF metal loading efficiency than method-1. The thermal analysis confirmed that the metal loading by method-1 and method-2 involved two different pathways. Method-1 involves solubility equilibria in the conversion of metal-nitrate to metal- hydroxide and finally to metal-oxide, while in method-2 nanoparticle-metal-oxide remained intact during sol-gel-oil-drop and calcination steps.

The alumina supported catalysts were dominated by γ-alumina …

The demand for energy is continually increasing overtime and the key to meeting future demand in a sustainable way is with energy storage. Li-ion batteries employing layered transition metal oxide cathodes are one of the most technologically important energy storage technologies. However, current Li-ion batteries are unable to access their full theoretical capacity and suffer from performance limiting degradation over time partially originating from the cathode and partially from the interface with the electrolyte. Understanding the fundamental limitations of layered transition metal oxide cathodes requires a complete understanding of the surface and bulk of the materials in their most delithiated …

Interplanetary Mission Design With Applications To Guidance And Optimal Control Of Aero-Assisted Trajectories, Peter J. Edelman Dec 2016

Interplanetary Mission Design With Applications To Guidance And Optimal Control Of Aero-Assisted Trajectories, Peter J. Edelman

Open Access Dissertations

A method for finding optimal aerogravity-assist tours of the solar system is developed using indirect methods. Two cost functionals are used in the optimization; finding the minimum required maximum lift-to-drag ratio, with and without a convective heating-rate path constraint, and the path which provides the minimum total stagnation point convective heat load. It is found that using present or near-future thermal protection system materials will suffice for certain aerogravity assist trajectories at Mars. Minimum heat load optimal trajectories are found for aerocapture maneuvers at Uranus and Neptune. With a large radius, and short rotational periods, atmospheric rotation must be taken …

Statistical Modeling And Simulation Of Variability And Reliability Of Cmos Technology, Khaled Hassan Md Dec 2016

Statistical Modeling And Simulation Of Variability And Reliability Of Cmos Technology, Khaled Hassan Md

Open Access Dissertations

The introduction of High-κ Metal Gate transistors led to higher integration density, low leakage current, and faster switching speed. However, this transition in technology roadmap brought about new failure mechanisms such as Positive Bias Temperature Instability and Stress Induced Leakage Current. In addition, the relentless downscaling of devices to keep up with Moore's law has significantly increased the time-zero variability caused by Random Dopant Fluctuation, Mean Gate Length Deviation, and Line Edge Roughness. Because of their possible correlation with time dependent aging effects, the quantification of reliability has become more complex than ever. To that effect, we propose a framework …

Computational Science and Engineering (CSE) has been denominated as the third pillar of science and as a set of important skills to solve the problems of a global society. Along with the theoretical and the experimental approaches, computation offers a third alternative to solve complex problems that require processing large amounts of data, or representing complex phenomena that are not easy to experiment with. Despite the relevance of CSE, current professionals and scientists are not well prepared to take advantage of this set of tools and methods. Computation is usually taught in an isolated way from engineering disciplines, and therefore, …

Passive Thermal Management Using Phase Change Materials, Yash Yogesh Ganatra Dec 2016

Passive Thermal Management Using Phase Change Materials, Yash Yogesh Ganatra

Open Access Theses

The trend of enhanced functionality and reducing thickness of mobile devices has led to a rapid increase in power density and a potential thermal bottleneck since thermal limits of components remain unchanged. Active cooling mechanisms are not feasible due to size, weight and cost constraints. This work explores the feasibility of a passive cooling system based on Phase Change Materials (PCMs) for thermal management of mobile devices. PCMs stabilize temperatures due to the latent heat of phase change thus increasing the operating time of the device before threshold temperatures are exceeded. The primary contribution of this work is the identification …

A Sharp Interface Isogeometric Strategy For Moving Boundary Problems, Tao Song Dec 2016

A Sharp Interface Isogeometric Strategy For Moving Boundary Problems, Tao Song

Open Access Dissertations

The proposed methodology is first utilized to model stationary and propagating cracks. The crack face is enriched with the Heaviside function which captures the displacement discontinuity. Meanwhile, the crack tips are enriched with asymptotic displacement functions to reproduce the tip singularity. The enriching degrees of freedom associated with the crack tips are chosen as stress intensity factors (SIFs) such that these quantities can be directly extracted from the solution without a-posteriori integral calculation.

As a second application, the Stefan problem is modeled with a hybrid function/derivative enriched interface. Since the interface geometry is explicitly defined, normals and curvatures can be …

Implementing A Wireless Monitoring Solution For A Biomechanical Telemetry System, Hyung Suk Kim

Open Access Theses

Sport-related Traumatic Brain Injury (TBI) is one of the major concerns for collision based sports athletes and their families. The ability to measure impacts on the head is imperative to monitor brain injury and prevent serious TBI. Recent research by the Purdue Neurotrauma Group (PNG) indicates that accumulation of subconcussive impacts may cause permanent neurological damage. Currently available commercial impact monitoring systems are designed as an event-based models which collect impact data above pre-determined acceleration thresholds. To track long-term effects of sub-concussive impacts, event-based modeling is inadequate. The PNG developed a biomechanical sports telemetry system that is capable of storing …

Optical Sub-Diffraction Limited Focusing For Confined Heating And Lithography, Luis M. Traverso

Open Access Dissertations

Electronics and nanotechnology is constantly demanding a decrease in size of fabricated nanoscale features. This decrease in size has become much more difficult recently due to the limited resolution of optical systems that are fundamental to many nanofabrication methods. A lot of effort has been made to fabricate devices smaller than the diffraction limit of light. Creating devices that are capable of confining fields by means of interference patterns of propagating wave modes and surface plasmon, has proven successful to confine light into smaller spot sizes.

Zone plate diffraction lenses generate spots with dimensions very close to the diffraction limit. …
