Compartmentalization Of Dna-Based Molecular Computing Elements Using Lipid Bilayers, Aurora Fabry-Wood

This dissertation will present a progression from the detection of double-stranded DNA using a combination of toehold-mediated strand displacement and DNAzyme reactions in dilute saline solutions, to the generation of separate compartments to allow standardization of DNA computing elements, by protecting from complementary strands. In well-mixed solutions complementary regions cause spurious interactions. Importantly, these compartments also provide protection from nucleases. Along the way we will also explore the use of silica microsphere supported lipid bilayers to run compartmentalized DNA reactions on a fluid surface and the design of a molecule capable of DNA-based transmembrane signal transduction.

Spatial irrigation management has been steadily advancing over the last several years. A current issue with managing irrigation spatially on sub-field scale is the inability to readily collect the spatial field data necessary to properly manage irrigation. Multispectral and thermal infrared imagery used in informing irrigation management decisions was previously collected by satellite and manned aircraft remote sensing platforms. These remote sensing platforms pose issues concerning economic feasibility, revisit intervals, and weather factors that inhibit the collection of data. Two novel, sustainable UV-LED advanced oxidation processes (AOPs) were evaluated for their ability to improve inactivation efficacy of P22 bacteriophage and E. coli bacteria. The first objective of this research was to determine the treatment efficacy of P22 bacteriophage and E. coli bacteria using UV-LED at wavelengths of 255, 265, and 285 nm. The UV-LED system demonstrated high inactivation potential for both contaminants at all wavelengths, suggesting it could be a potential alternative to typical low-pressure and medium-pressure mercury-based UV systems. The purpose of this study was to validate a 3D musculoskeletal model in OpenSim and assess OpenSim’s ability to determine muscle-length variation during ambulation. An 18 camera motion capture system was used to analyze 20 healthy individuals between the ages of 18 and 35. Following data collection, the data was processed through OpenSim and Visual3D. The kinematic output processed through the OpenSim model was then compared to the kinematic output of the validated Visual3D model to validate the OpenSim model. Detection and quantification of benzene, toluene, ethylbenzene, and xylenes (BTEX) in liquid phase is of interest because of the significant public health hazards posed by these compounds. BTEX are volatile organic compounds (VOCs) commonly used as additives in gasoline. Shear horizontal surface acoustic wave (SH-SAW) devices have shown promise in liquid phase sensing applications and is used to address this problem. SH-SAW devices can be made chemically sensitive by depositing a polymer coating on the device surface. In this thesis, a novel method for tracker fusion is proposed and evaluated for vision-based tracking. This work combines three distinct popular techniques into a recursive Bayesian estimation algorithm. First, semi supervised learning approaches are used to partition data and to train a deep neural network that is capable of capturing normal visual tracking operation and is able to detect anomalous data. We compare various methods by examining their respective receiver operating conditions (ROC) curves, which represent the trade off between specificity and sensitivity for various detection threshold levels. Prior work at Marquette University developed the Marquette Prosthesis, an active transtibial prosthesis that utilized a torsional spring and a four-bar mechanism. The controls for the Marquette Prosthesis implemented a finite state control algorithm to determine the state of gait of the amputee along with two lower level controllers, a PI moment controller to control the moment during stance and a PID position controller to control the position during stance. The Marquette Prosthesis was successful in mimicking the gait profile presented by Winter. This work provides a framework for Design Day analysis. First, we estimate the temperature conditions which are expected to be colder than all but one day in N years. This temperature is known as the Design Day condition. Then, we forecast an upper bound on natural gas demand when temperature is at the Design Day condition. Natural gas distribution companies (LDCs) need to meet demand during extreme cold days. However, they tend to lack the accuracy and precision achieved by fixed manipulators, especially in scenarios where both the manipulator and the autonomous vehicle move simultaneously. The maintenance of infrastructure is critical to the well-being of society. This work focuses on a novel method for inspecting the exterior of dams using unmanned aerial vehicles (UAVs) in an automated fashion. The UAVs are equipped with optical sensors capturing still images. The resulting images are used to generate three-dimensional (3D) models using Structure from Motion (SfM) computer software. The SfM models are then used to inspect the exterior of the dam. One area of exploration is the development of fuel cells, which includes biofuel cells that can extract energy from carbohydrates obtained from biomass. This is due to their low inertia and inherent ability to more safely interact with the world without damaging themselves or the people around them. This work enables accelerated fluid recovery in oil and gas reservoirs by automatically controlling fluid height and bottomhole pressure in wells. Several literature studies show significant increase in recovered oil by determining a target bottomhole pressure but rarely consider how to control to that value. This work enables those benefits by maintaining bottomhole pressure or fluid height. Moving Horizon Estimation (MHE) determines uncertain well parameters using only common surface measurements. A Model Predictive Controller (MPC) adjusts the stroking speed of a sucker rod pump to maintain fluid height. Molybdenum-Oxide/Carbon (MoO2/C) composite nanofibers were synthesized using Forcespinning method and subsequent heat treatment, for use as anode materials for lithium-ion batteries. Due to the layered structure of MoO2, the composite nanofibers are very promising for use as anode material for LIBs. The MoO2/C composite fiber anodes exhibit good electrochemical performance and cycling stability.

In this study, heparin-loaded poly-ε-caprolactone (PCL) fibrous mats were prepared and characterized based on their physical, cytotoxic, thermal, and biological properties. The main objective of the work described here was to test the hypothesis that incorporation of heparin into a PCL carrier could serve as bio-compatible material capable of inhibiting Human Papillomavirus (HPV) infection. The idea of firmly anchoring heparin to capture soluble virus, vs. a slow heparin release to inhibit a virus in solution was tested. In this paper, parametric excitation of a repulsive force electrostatic resonator is studied. A theoretical model is developed and validated by experimental data. A correspondence of the model to Mathieu's Equation is made to prove the existence and location of parametric resonance. The repulsive force creates a combined response that shows parametric and subharmonic resonance when driven at twice its natural frequency. The resonator can achieve large amplitudes of almost 24 μm and can remain dynamically stable while tapping on the electrode. Because the pull-in instability is eliminated, the beam bounces off after impact instead of sticking to the electrode. …

Effects Of Nb And Ta Additions On The Strength And Coarsening Resistance Of Precipitation-Strengthened Al-Zr-Sc-Er-Si Alloys, Dinc Erdeniz, Anthony De Luca, David N. Seidman, David C. Dunand

A dilute Al-0.07Zr-0.02Sc-0.005Er-0.06Si (at.%) alloy was microalloyed with 0.08 at.% Nb or Ta. Atom-probe tomography reveals that, upon aging, Nb and Ta partition to the coherent L12-Al3(Zr,Sc,Er) nanoprecipitates (with average concentrations of 0.2 and 0.08 at.%, respectively), with both segregating at the matrix/nanoprecipitate heterophase interface. This is consistent with the Nb- and Ta-modified alloys exhibiting, as compared to the unmodified alloy: (i) higher peak microhardness, from a higher nanoprecipitate volume fraction and/or latticeparameter mismatch; and (ii) improved aging resistance, from slower nanoprecipitate coarsening due to the small diffusivities of niobium and tantalum in aluminum. This study presents an approach to co-optimization of transactive flexibility, energy, and optimal injection-range of Variable Energy Resource (VER). Flexibility receives immense attention, as it is the essential resource to accommodate VERs in modern power systems. With a novel concept of transactive flexibility, the proposed approach proactively positions the flexible resources and optimizes the demand of flexibility. A surrogate affine approximation (SAA) method is proposed to solve the problem with variable infinite-constraint range in polynomial time. Temperature control in friction stir welding (FSW) is of interest because of the potential to improve the mechanical and microstructure characteristics of a weld. Two types of active temperature control have been previously implemented for steady-state friction stir welding conditions: PID Feedback Control and Model Predictive Control. In this dissertation, the rate of the BFD during body armor impact is characterized with fiber Bragg gratings for the first time ever. The depth rate is characterized using a single fiber optic sensor, while the entire shape rate can be characterized using multiple fiber optic sensors. This is done with a final depth accuracy of less than 10% and a timing accuracy of 15% for BFDs as deep as 50 mm and impact event of less than 1 millisecond.
