Masters Theses and Doctoral Dissertations

In terms of mesh resolution requirements, higher-order finite element discretization methods offer a more economic means of obtaining accurate simulations and/or to resolve physics at scales not possible with lower-order schemes. For simulations that may have large relative motion between multiple bodies, overset grid methods have demonstrated distinct advantages over mesh movement strategies. Combining these approaches offers the ability to accurately resolve the flow phenomena and interaction that may occur during unsteady moving boundary simulations. Additionally, overset grid techniques when utilized within a finite element setting mitigate many of the difficulties encountered in finite volume implementations. This research presents the …

Temporally And Spatially Resolved Quantification Of Hemodynamic Forces And Endothelial Mechanics, Lori M. Lambert May 2016

Temporally And Spatially Resolved Quantification Of Hemodynamic Forces And Endothelial Mechanics, Lori M. Lambert

Department of Mechanical and Materials Engineering: Dissertations, Theses, and Student Research

The endothelium is a thin layer of endothelial cells that line the interior surface of an artery. Due to their direct contact with blood flow, endothelial cells experience varying hemodynamic forces and respond to these forces by altering their morphology. When plaque and other substances accumulate in the walls of arteries, i.e., atherosclerosis, endothelial cells have abnormal responses to blood flow. Studying atherosclerosis progression is, therefore, a two-fold investigation into 1) the hemodynamic forces that cause endothelial responses, and 2) the biological and mechanical responses of endothelial cells. The ultimate goal of this study was to develop an experimental …

Development And Characterization Of Adhesive Hydrogels For Stem Cell Culture, Erin Byrne Rousseau

Nanoscale Science & Engineering (discontinued with class year 2014)

Pluripotent embryonic stem cells (ESCs) offer the unique capacity to differentiate into almost any cell type and have sweeping implications in both basic research and clinical applications. However, unregulated differentiation can cause issues, preventing ESCs from entering clinical research. In order to maximize ESC growth, three dimensional culturing has been utilized in order to have results more similar to in vivo conditions. In the case of alginate scaffolds, cell adhesion sites are missing from the matrix, leading to differentiation. We propose that the inclusion of adhesive polymer to the alginate scaffold will increase cell attachment and maintain pluripotency. The polymer …
