Physiological And Transcriptional Memory In Guard Cells During Repetitive Dehydration Stress, Laetitia Virlouvet, Michael E. Fromm

Arabidopsis plants subjected to a daily dehydration stress and watered recovery cycle display physiological and transcriptional stress memory. Previously stressed plants have stomatal apertures that remain partially closed during a watered recovery period, facilitating reduced transpiration during a subsequent dehydration stress. Guard cells (GCs) display transcriptional memory that is similar to that in leaf tissues for some genes, but display GC-specific transcriptional memory for other genes. The rate-limiting abscisic acid (ABA) biosynthetic genes NINE-CIS-EPOXYCAROTENOID DIOXYGENASE 3 (NCED3) and ALDEHYDE OXIDASE 3 (AAO3) are expressed at much higher levels in GCs, particularly during the watered recovery interval, relative to their low …

Pseudomonas syringae type III effectors are known to suppress plant immunity to promote bacterial virulence. However, the activities and targets of these effectors are not well understood. We used genetic, molecular, and cell biology methods to characterize the activities, localization, and target of the HopD1 type III effector in Arabidopsis. HopD1 contributes to P. syringae virulence in Arabidopsis and reduces effector-triggered immunity (ETI) responses but not pathogen-associated molecular pattern-triggered immunity (PTI) responses. Plants expressing HopD1 supported increased growth of ETI-inducing P. syringae strains compared with wild-type Arabidopsis. Meta-heuristic algorithms play an important role in solving combinatorial optimization problems (COP) in many practical applications. The caveat is that the performance of these meta-heuristic algorithms is highly dependent on their parameter configuration which controls the algorithm behaviour. Selecting the best parameter configuration is often a difficult, tedious and unsatisfying task. This thesis studies the problem of automating the selection of good parameter configurations. Existing approaches to address the challenges of parameter configuration can be classified into one-size-fits-all and instance-specific approaches. n this study, we demonstrate the production of long-chain hydrocarbons (C8+) from 2-methylfuran (2MF) and butanal in a single step reactive process by utilizing a bi-functional catalyst with both acid and metallic sites. Our approach utilizes a solid acid for the hydroalkylation function and as a support as well as a transition metal as hydrodeoxygenation catalyst. A series of solid acids was screened, among which MCM-41 demonstrated the best combination of activity and stability. Platinum nanoparticles were then incorporated into the MCM-41. The Pt/MCM-41 catalyst showed 96% yield for C8+ hydrocarbons and the catalytic performance was stable …

Ultraviolet (UV) irradiation was utilized to gradually modify the chemistry and structure of graphene oxide (GO) flakes, as confirmed by XPS and AFM. Ultrathin GO coatings/membranes, made of UV-irradiated flakes, showed tunable underwater oleophobicity. UV-treated, superoleophobic GO membranes exhibited excellent antifouling capability for oil/water separation.

Land use change for bioenergy feedstocks is likely to intensify as energy demand rises simultaneously with increased pressure to minimize greenhouse gas emissions. Initial assessments of the impact of adopting bioenergy crops as a significant energy source have largely focused on the potential for bioenergy agroecosystems to provide global-scale climate regulating ecosystem services via biogeochemical processes. This thesis presents the design and development of solar powered water purification systems. The systems were designed to meet specific customer requirements that were established from the experiences gained after the installation of five water purification systems in various Haitian communities. The systems are scalable and are able to supply 10,000-20,000 gallons of clean drinking water per day. The design consists of a three-stage filtration system with a disc-type sediment filter, a 0.1 micron ultrafiltration membrane, and an ultraviolet light for disinfection. Assuming homogeneity in alluvial aquifers is convenient, but limits our ability to accurately predict stream-aquifer interactions. Research is needed on (i) identifying the presence of focused, as opposed to diffuse, groundwater discharge/recharge to streams and (ii) the magnitude and role of large-scale bank and transient storage in alluvial floodplains relative to changes in stream stage. The atrazine-degrading bacterial strain Pseudomonas sp. ADP was immobilized by the sol–gel process within thin silica layers coated onto water-retaining carrier materials (expanded clay pellets and scoria). The performance of the obtained biohybrid material has been investigated concerning long-term activity under non-growth conditions. Experiments were run in phosphate buffer containing atrazine (20 mg l-1) as the sole source of carbon and nitrogen. Even after one year of consecutive batch tests, P. Controller Area Network (CAN) bus is used on current agricultural equipment for in-vehicle communication among multiple microcontrollers. In this study, CAN data was collected from the tractor diagnostic port during three field operations: anhydrous ammonia (NH3) application, field cultivation, and planting. Specifically, CAN messages corresponding to engine fuel use rate were filtered, decoded, and analyzed. High fuel use rates were recorded from the tractor when implements were engaged in the soil compared to when the implements were lifted out of the ground at the end of the passes or, when the tractor was turning and idling. The growing global demand for energy will increasingly call upon fusion reactors and Generation IV nuclear fission reactors to supply safe and reliable energy worldwide. Ferritic/martensitic (F/M) alloys are leading candidates for structural components in these reactors because of their high strength, dimensional stability, and low activation. In novel reactor concepts, these materials will be subject to extreme operating conditions, accumulating doses of irradiation up to a few hundred displacements per atom (dpa) at temperatures as high as 600°C. Oxide dispersion strengthened (ODS) F/M alloys containing a dispersion of Y-Ti-0 nanoclusters have been developed to operate at even higher temperatures.

Magnetic field probes are invaluable diagnostics for pulsed inductive plasma devices where field magnitudes on the order of tenths of tesla or larger are common. Typical methods of providing a broadband calibration of Ḃ probes involve either a Helmholtz coil driven by a function generator or a network analyzer. Both calibration methods typically produce field magnitudes of tens of microtesla or less, at least three and as many as six orders of magnitude lower than their intended use. This calibration factor is then assumed constant regardless of magnetic field magnitude and the effects of experimental setup are ignored. This work …

Ligand cross-linking is known to improve the colloidal stability of nanoparticles, particularly in aqueous solutions. However, most cross-linking is performed chemically, in which it is difficult to limit interparticle cross-linking, unless performed at low concentrations. Photochemical cross-linking is a promising approach but usually requires ultraviolet (UV) light to initiate. Using such high-energy photons can be harmful to systems in which the ligand–nanoparticle bond is fairly weak, as is the case for the commonly used semiconductor quantum dots (QDs). Evaluating N-Benzylgalactonoamidines As Putative Transition State Analogs For Β-Galactoside Hydrolysis, Qui-Hua Fan, Susanne Striegler, Rebekah Langston, James Barnett

Experimental evidence is provided for p-methylbenzyl-D-galactonoamidine to function as a true transition state analog for the enzymatic hydrolysis of aryl-β-D-galactopyranosides by β-galactosidase (A. oryzae). The compound exhibits inhibition constants in the low nanomolar concentration range (12–56 nM) for a selection of substrates. Along these lines, a streamlined synthetic method based on phase-transfer catalysis was optimized to afford the required variety of new aryl-β-D-galactopyranosides. Last, the stability of the galactonoamidines under the assay conditions was confirmed.

While integrated circuit (IC) power management has been an eternal topic for chip designers, inductor based DC-DC converters have been dominant in the field for years. However, because of the natures of inductors: large electro-magnetic interference, high coupling noise, and difficult silicon fabrication process, they are not favorable to on-chip solutions. Switched-capacitor (SC) DC-DC converters, which adopt capacitors for their energy storage components, have become increasingly popular among both the academia and the industry, because, apparently, they avoid the drawbacks of the inductor counterparts, and can be directly implemented on-chip without additional fabrication process.

This article discusses explorations made by a research team to find solutions to enable the distance-learning delivery of laboratory courses on embedded microcontroller technology topics. Resonance waveguide grating structures have been a subject of interest for several decades. This optical technique offers unique advantages when compared with other optical components. This stems from their relatively simple structure comprised of layers, as well as their high reflection efficiency within a narrow spectral bandwidth. Typical resonance waveguide grating structures that exhibit these characteristics are cost effective and fabricated with fewer materials than conventional multilayer resonance structures.
