Protein Engineering Of Luciferase To Enhance Bioluminescent Properties, Charles Schwab, Peter Wittig
Bioengineering Senior Theses
Bioluminescence is the ability of naturally occurring organisms to emit light. Many instances of bioluminescence have been identified ranging from fireflies to bacteria. The protein responsible for bioluminescence in all of these organisms is luciferase. The exact function and structure of this protein are still unknown to this day. This project aims at better understanding luciferase as well as enhancing its bioluminescent properties. In order to work with luciferase, we had to isolate all of the genes necessary for bioluminescence. The six genes necessary for bioluminescence are Lux A,B,C,D,E,G. We isolated each of these genes from original bacterial DNA. We …

Designing A Biomimetic Primary Cell-Based 3d Culture System For Neurotoxicity Screening, Teresa Cauvel, Jessca Kosit, Nicolo Mendoza
Bioengineering Senior Theses
Culturing neurons in vitro is a challenging task because they are a highly specialized cell type that reside in a complex and unique environment in the body. The aim of the research presented in the following thesis was to design a biomimetic, three-dimensional scaffold capable of (1) promoting primary neuron maturation and axonal outgrowth and (2) serving as a system for toxicology screening. In the system presented here, neurons were cultured in three-dimensional hydrogels, simulating the physiological environment that these cells experience within the body. In doing so, a biologically relevant response was elicited upon their exposure to acrylamide, a …

It is reported that wearable electronic devices are to be used extensively in the next generation of sensors for sports and health monitoring. The information obtained from sensors on the human body depends on the biological parameters, the measurement rate and the number of sensors. The choice of the wireless protocol depends on the required data rates and on system configurations. The communication link quality is achieved with narrowband technologies such as Bluetooth or Zigbee, provided that the number of sensors is small and data rates are low. However, real-time measurements using wideband channels may also be necessary. This paper …

Simulation And Analysis Of Inhomogeneous Degradation In Large Format Limn2O4/ Carbon Cells, Yiling Dai, Long Cai, Ralph E. White
Faculty Publications
Degradation phenomena are not distributed uniformly in a large-format cell. To better understand the inhomogeneous degradation in large-format cells, a two-dimensional model was developed for a LiMn2O4 (LMO)/Carbon cell. The model includes both the non-uniform porous electrode properties and the electrode mismatch. The simulation results show that when the anode edge is extended over the cathode edge, the LMO particles near the edge suffer larger potential difference, larger charge/discharge depth, and higher insertion-induced stress. Therefore, the predicted loss of LMO is more pronounced near the edge as has been seen experimentally. The influence of different design adjustable …
Ranked Similarity Search Of Scientific Datasets: An Information Retrieval Approach, Veronika Margaret Megler
Ranked Similarity Search Of Scientific Datasets: An Information Retrieval Approach, Veronika Margaret Megler
Dissertations and Theses
In the past decade, the amount of scientific data collected and generated by scientists has grown dramatically. This growth has intensified an existing problem: in large archives consisting of datasets stored in many files, formats and locations, how can scientists find data relevant to their research interests? We approach this problem in a new way: by adapting Information Retrieval techniques, developed for searching text documents, into the world of (primarily numeric) scientific data. We propose an approach that uses a blend of automated and curated methods to extract metadata from large repositories of scientific data. We then perform searches over …

Where To Park? The East Campus Dilemma, Matthew Lewis
Where To Park? The East Campus Dilemma, Matthew Lewis
James Madison Undergraduate Research Journal (JMURJ)
James Madison University's East Campus expansion has created parking problems. Student struggle to find a decent parking spot, sacrificing time to park and then perhaps more time to walk to class. This research project seeks answers to the parking problems on the east side of campus, specifically the Festival and Convocation lots. Should students spend time navigating the Festival lot looking for a spot or should they go directly to the Convocation lot? The data and research explain why the latter solution is much more efficient.

The development of monitoring programs for water quality and habitat assessment in surface waters is an ongoing challenge because of inherent difficulties in determining the effective spatial and temporal distribution of sites and trips. Recent advances in statistical learning theory, in which system characteristics are learned from data, point to the possibility of using the information content of data to shed light on monitoring results that provide sensitive and independent results. One of those techniques, multivariate relevance vector machines (MVRVM), creates as part of its algorithm subsets of a data set, called relevant vectors (RVs), that are most relevant for …

Improving The Complementary Methods To Estimate Evapotranspiration Under Diverse Climatic And Physical Conditions, Fathi Anayah, Jagath J. Kaluarachchi
Improving The Complementary Methods To Estimate Evapotranspiration Under Diverse Climatic And Physical Conditions, Fathi Anayah, Jagath J. Kaluarachchi
Civil and Environmental Engineering Faculty Publications
Reliable estimation of evapotranspiration (ET) is important for the purpose of water resources planning and management. Complementary methods, including complementary relationship areal evapotranspiration (CRAE), advection aridity (AA) and Granger and Gray (GG), have been used to estimate ET because these methods are simple and practical in estimating regional ET using meteorological data only. However, prior studies have found limitations in these methods especially in contrasting climates. This study aims to develop a calibration-free universal method using the complementary relationships to compute regional ET in contrasting climatic and physical conditions with meteorological data only. The proposed methodology consists of a systematic …

Microstructural Factors Of Strain Delocalization In Model Metallic Glass Matrix Composites, Thomas James Hardin
Microstructural Factors Of Strain Delocalization In Model Metallic Glass Matrix Composites, Thomas James Hardin
Theses and Dissertations
Metallic glass matrix composites have enormous potential stemming from the interplay between crystalline and amorphous phases. This work models such a composite using shear transformation zone dynamics (a modified kinetic Monte Carlo method) for the amorphous phase, and a local Taylor dislocation model for the crystalline phase. An N-factorial experiment using the model is presented examining the effects of crystalline volume fraction, microstructure length scale, and yield stress of the crystalline phase. Each replicate is analyzed for maximum stress, maximum strain, strain energy dissipation, strain localization, and strain partitioning between phases. Regression analysis is used to identify statistically-significant trends in …

Toward A Reliable Prediction Of Seasonal Forecast Uncertainty: Addressing Model And Initial Condition Uncertainty With Ensemble Data Assimilation And Sequential Bayesian Combination, Caleb Matthew Dechant, Hamid Moradkhani
Civil and Environmental Engineering Faculty Publications and Presentations
Uncertainties are an unfortunate yet inevitable part of any forecasting system. Within the context of seasonal hydrologic predictions, these uncertainties can be attributed to three causes: imperfect characterization of initial conditions, an incomplete knowledge of future climate and errors within computational models. This study proposes a method to account for all threes sources of uncertainty, providing a framework to reduce uncertainty and accurately convey persistent predictive uncertainty. In currently available forecast products, only a partial accounting of uncertainty is performed, with the focus primarily on meteorological forcing. For example, the Ensemble Streamflow Prediction (ESP) technique uses meteorological climatology to estimate …
