The production of Portland cement-the key ingredient in concrete-generates a significant amount of carbon dioxide. However, because of its incredible versatility, availability, and relatively low cost, concrete is the most consumed manmade material on the planet. One method of reducing concrete's contribution to greenhouse gas emissions is the use of fly ash to replace a significant amount of the cement. an experimental investigation was conducted to compare the bond strength of reinforcing steel in high-volume fly-ash concrete (HVFAC)-concrete with at least 50 % of the cement replaced with fly ash-with conventional concrete (CC). This thesis reports the viability of E. coli cell extracts prepared using equipment that is both common to biotechnology laboratories and able to process small volume samples and expression of proteins containing unnatural amino acids (UAAs) at higher level using PCR amplified linear DNA templates (LETs) in cell-free protein synthesis (CFPS) system. E. coli-based cell extracts are a vital component of inexpensive and high-yielding CFPS reactions. However, effective preparation of E. coli cell extract is limited to high-pressure homogenizers (French press style or impinge-style) or bead mill homogenizers, which all require a significant capital investment. Biodiesel is produced from vegetable oils through transesterification. Triglyceride mixtures extracted from oilseed feedstocks are upgraded by reaction with an alcohol in the presence of a catalyst to produce fatty acid esters. This reaction produces a mixture of esters, glycerin, alcohol, and catalyst. Separation of the fatty acid esters (biodiesel) and glycerin can be accomplished through liquid-liquid extraction by water addition. Designing liquid-liquid extraction with water as the solvent requires ternary liquid-liquid equilibrium data for mixtures of water, glycerin, and fatty acid esters. Ternary mixture LLE data have been experimentally measured for several of these systems. Those measured include mixtures …

There has been an increase in adoption of building information modeling (BIM) by commercial contractors over the past few years but there is not a clear indication of current application or the frequency of each use. This research was undertaken to determine the frequency and best practices of using BIM in commercial construction. Leading commercial contracting firms were contacted and employees were selected to complete a telephone survey that was designed to determine how they are using BIM. In this dissertation we present advances in developing an autonomous air vehicle capable of navigating through unknown, indoor environments. The problem imposes stringent limits on the computational power available onboard the vehicle, but the environment necessitates using 3D sensors such as stereo or RGB-D cameras whose data requires significant processing. We address the problem by proposing and developing key elements of a relative navigation scheme that moves as many processing tasks as possible out of the time-critical functions needed to maintain flight. The usefulness of vibrotactile feedback as a channel to communicate information has been widely recognized. However, most of the recent work on this subject uses actuators that are either too expensive or too large for many practical applications. This thesis explores the generation of vibrotacatile feedback patterns using a simple, inexpensive eccentric-mass motor that is constrained to use a constant voltage and a low on/off switching frequency. In particular, it explores the pulse timing method, which utilizes the slow transient response of the eccentric-mass motor to calculate pulse and rest times for an arbitrary envelope waveform shape. Internal waves exist everywhere in stratified fluids - fluids whose density changes with depth. The two largest bodies of stratified fluid are the atmosphere and ocean. Internal waves are generated from a variety of mechanisms. One common mechanism is wind forcing over repeated sinusoidal topography, like a series of hills. When modeling these waves, linear theory has been employed due to its ease and low computational cost. However, recent research has shown that non-linear effects, such as boundary layer separation, may have a dramatic impact on wave generation. Models (both physical and numerical) of the friction stir (FS) welding process are used to develop a greater understanding of the influence of independent process parameters on dependent process output variables, such as torque, power, specific weld energy, peak temperature, cooling rates and various metallurgical factors (e.g., grain size and precipitates). An understanding of how the independent process parameters influence output variables and ultimately their effect on resultant properties (e.g., strength, hardness, etc..) is desirable. A number of methods for adaptation of existing all-hexahedral grids by localized refinement have been developed; however, none ideally fit all refinement needs. This thesis presents the structure to a method of two-refinement developed for conformal, structured, all-hexahedral grids that offers flexibility beyond what has been offered to date. The method is fundamentally based on pillowing pairs of sheets of hexes. This thesis also suggests an implementation of the method, shows the results of examples refined using it and compares these results to results from implementing three-refinement on the same examples.

Holloman Air Force Base houses the 10 mile long Holloman High Speed Test Track (HHSTT) in New Mexico and can run hypersonic experiments at speeds up to 10,000m/s. Tested objects are loaded onto sleds, which are connected to the track by slippers that slide along the rails. The payload sled is propelled down the track by a series of rocket sleds. The ability to predict the wear that will occur on the slippers during these experiments is important for slipper design and preventing catastrophic failure. Medulloblastoma (MB) is the most common brain cancer diagnosed among children. The cellular pathways that regulate MB invasion in response to environmental cues remain incompletely understood. Herein, we examine the migratory response of human MB-derived Daoy cells to different concentration profiles of Epidermal Growth Factor (EGF) using a microfluidic system. Our findings provide the first quantitative evidence that EGF concentration gradients modulate the chemotaxis of MB-derived cells in a dose-dependent manner via the EGF receptor (EGF-R). Data illustrates that higher concentration gradients caused increased number of cells to migrate. In addition, our results show that EGF-induced receptor phosphorylation triggered the …

The present work demonstrates the application of molecular simulations (MD) in two different areas: proton transport in bulk water and estimation of the dipole moment of polar organic compounds. In both areas, relatively few successful and robust methodologies exist. In the first part, a new polarizable water model is developed for MD simulations of the proton transport process. The model was parametrized from a combination of quantum chemical calculations and experimental water properties. The model was implemented in MD simulation studies of liquid water at room temperature, as well as with excess protons. For pure water the model gave good …

This thesis presents a new design process in which design specifications and task distribution are determined from a parallel multi user prospective. Using this method, projects are more easily decomposed into tasks that can be performed concurrently, thus decreasing the design time. Traditional seismic load resisting systems in buildings are designed to undergo inelastic deformations in order to dissipate energy, resulting in residual displacements. This work explores an approach to eliminate these residual displacements. The systems investigated have low initial stiffness which increases at a predefined displacement, and are therefore called stiffening elastic systems. This thesis begins with an examination of single-degree-of-freedom stiffening elastic systems. A case study is presented which suggests that the benefits from stiffening elastic behavior may be limited to systems which would have long periods if designed traditionally. Dynamic loads can cause severe damage to bridges, and lead to malfunction of transportation networks. A comprehensive understanding of the nature of the dynamic loads and the structural response of bridges can prevent undesired failures while keeping the cost-safety balance. Dissimilar to the static behavior, the dynamic response of bridges depends on several structural parameters such as material properties, damping and mode shapes. Furthermore, dynamic load characteristics can significantly change the structural response. In most cases, complexity and involvement of numerous parameters require the designer to investigate the bridge response via a massive numerical study.

Metallic additives, also known as anticorrosive pigments, can provide sacrificial cathodic protection and complement the barrier protection afforded by heterogeneous organic coatings to metallic substrates. The unique systematic study of the corrosion resistance of an epoxy coating reinforced with different sizes (80nm, 500nm, 10um) and continuous multiple pigment volume concentration (0, 2%, 10%, 20%, 45%) below global critical pigment volume concentration of zinc particles were studied. In this dissertation, we focus on the strategy that places and stabilizes the path of an aerial drogue, which is towed by a mothership aircraft using a long flexible cable, onto a horizontally flat orbit by maneuvering the mothership in the presence of wind. The Consortium of Universities for the Advancement of Hydrologic Science, Inc. (CUAHSI) has developed a Hydrologic Information System (HIS) to provide better access to data by enabling the publication, cataloging, discovery, retrieval, and analysis of hydrologic data using web services. The CUAHSI HIS is an Internet based system comprised of hydrologic databases and servers connected through web services as well as software for data publication, discovery and access. The HIS metadata catalog lists close to 100 web services registered to provide data through this system, ranging from large federal agency data sets to experimental watersheds managed by University investigators. As battery energy storage and solar cell technology improve, solar aircraft are increasingly being considered for High Altitude Long Endurance missions. Although solar vehicles may theoretically remain on-station indefinitely using the sun as a power source, their design and feasibility is sensitive to mission planning details as specific as the time history of the vehicle’s deck orientation relative to the sun; the energy available for capture by the on-board solar array is governed by the solar incidence angle, and at certain orientations, the vehicle may cast shadows on itself and further reduce its energy capture capabilities. The goal of this project was to design and build a compensated pulsed alternator, or compulsator, to power the Cal Poly Electromagnetic Railgun Mark 1.1. This project examines the feasibility of implementing mechanical pulsed power supplies for repeatable use with a railgun load for orbital debris hypervelocity testing. The final system architecture chosen was a passively compensated, iron-core, 2-phase, permanent magnet compulsator. The Cal Poly Compulsator will be capable of storing 45 kJ of mechanical energy with a peak operating speed of 5,000 rpm at 190 V. Research to increase the efficiency of conventional solar cells is constantly underway. The goal of this work is to increase the efficiency of conventional solar cells by incorporating quantum dot (QD) nanoparticles in the absorption mechanism. The strategy is to have the QDs absorb UV and fluoresce photons in the visible region that are more readily absorbed by the cells. The outcome is that the cells have more visible photons to absorb and have increased power output. The QDs, having a CdSe core and a ZnS shell, were applied to the solar cells as follows. BackgroundEvolutionary dynamics of microbial organisms can now be visualized using the Visualizing Evolution in Real Time (VERT) system, in which several isogenic strains expressing different fluorescent proteins compete during adaptive evolution and are tracked using fluorescent cell sorting to construct a population history over time. Mutations conferring enhanced growth rates can be detected by observing changes in the fluorescent population proportions.ResultsUsing data obtained from several VERT experiments, we construct a hidden Markov-derived model to detect these adaptive events in VERT experiments without external intervention beyond initial training. In nanometer-scale CMOS technology, leakage power has become a major component of the total power dissipation due to the downscaling of threshold voltage and gate oxide thickness. The leakage power consumption has received even more attention by increasing demand for mobile devices. Since mobile devices spend a majority of their time in a standby mode, the leakage power savings in standby state is critical to extend battery lifetime. For this reason, low power has become a major factor in designing CMOS circuits.

Transistor scaling following per Moore's Law slows down its pace when entering into nanometer regime where short channel effects (SCEs), including threshold voltage fluctuation, increased leakage current and mobility degradation, become pronounced in the traditional planar silicon MOSFET. In addition, as the demand of diversified functionalities rises, conventional silicon technologies cannot satisfy all non-digital applications requirements because of restrictions that stem from the fundamental material properties. Therefore, novel device materials and structures are desirable to fuel further evolution of semiconductor technologies.
