Imaging is essential in accurately detecting, staging, and treating primary liver cancer (hepatocellular carcinoma [HCC]), one of the most prevalent and lethal malignancies. We developed a novel multifunctional nanoparticle (NP) specifically targeting glypican-3 (GPC3), a proteoglycan implicated in promotion of cell growth that is overexpressed in most HCCs. Quantitative real-time polymerase chain reaction was performed to confirm the differential GPC3 expression in two human HCC cells, Hep G2 (high) and HLF (negligible). These cells were treated with biotin-conjugated GPC3 monoclonal antibody (aGPC3) and subsequently targeted using superparamagnetic iron oxide NPs conjugated to streptavidin and Alexa Fluor 647. Signalized intersections are provided in traffic networks to improve the safety and efficiency of vehicular and pedestrian movement. There are various measures under education, enforcement and engineering headings that are being attempted to improve safety and efficiency of operations at a signalized intersection. This paper examines the effect of information provided to drivers through advance warning flashers (AWFs) on driver’s probability of conflict at the onset of yellow at a high-speed intersection. AWFs are specifically designed to minimize the number of vehicles trapped in their respective dilemma zones at the onset of yellow. A probit modeling technique was used to establish dilemma zone boundaries. Based on the dilemma zone boundaries probability of a perceived conflict curves was computed and compared against actual conflicts observed at each of the studied intersections. Over the years, numerous work-zone, portable sign support systems have been successfully crash tested according to the Test Level 3 safety performance guidelines provided in the National Cooperative Highway Research Program Report 350 and accepted for use along our nation’s highways. For this study, several crashworthy sign support systems were analyzed to predict their safety performance according to the new evaluation criteria provided in the Manual for Assessing Safety Hardware (MASH). Energy and climate change are beginning to dominate the global political agenda and will drive policy formation that will shape the future of agriculture. Energy issues threaten national security and economic stability, as well as access to low-cost nutrient inputs for agriculture. A model was previously developed (Biofuel Energy Systems Simulator; www. bess.unl.edu) to predict greenhouse gas (GHG) emissions and net energy yield when ethanol is produced from corn. The model also predicts feedlot cattle, dairy cattle, and swine performance and feed replacement value of ethanol coproducts. Updated equations that predict performance of feedlot cattle fed 0 to 40% of dietary DM as corn wet (WDGS), modified (MDGS), or dry (DDGS) distillers grains plus solubles replacing dry-rolled and high-moisture corn were developed and incorporated into the model. Fused in Sarcoma (FUS) proteinopathy is a feature of frontotemporal lobar dementia (FTLD), and mutation of the fus gene segregates with FTLD and amyotrophic lateral sclerosis (ALS). To study the consequences of mutation in the fus gene, we created transgenic rats expressing the human fus gene with or without mutation. Overexpression of a mutant (R521C substitution), but not normal, human FUS induced progressive paralysis resembling ALS. Mutant FUS transgenic rats developed progressive paralysis secondary to degeneration of motor axons and displayed a substantial loss of neurons in the cortex and hippocampus. Nanoformulations of crystalline indinavir, ritonavir, atazanavir, and efavirenz were manufactured by wet milling, homogenization or sonication with a variety of excipients. The chemical, biological, immune, virological, and toxicological properties of these formulations were compared using an established monocyte-derived macrophage scoring indicator system. Measurements of drug uptake, retention, release, and antiretroviral activity demonstrated differences amongst preparation methods. Interestingly, for drug cell targeting and antiretroviral responses the most significant difference among the particles was the drug itself. We posit that the choice of drug and formulation composition may ultimately affect clinical utility.

The need to provide students with learning experiences that can help them develop the knowledge and skills relevant to the workplace has been consistently raised by businesses. Project-Assisted Learning (PAL) and Project-Based Learning (PBL) are effective learning methods that closely match workplace situations. Several studies have documented the benefits of PAL and PBL from the perspectives of the educators. This article presents cases of learning experiences in which project assignment, a case of PAL, was used to enhance the learning experiences of the students. Aim: Glucose stimulates insulin secretion from pancreatic islet β cells by altering ion channel activity and membrane potential in the β cells. TRPV1 channel is expressed in the β cells and capsaicin induces insulin secretion similarly to glucose. This study aims to investigate the biophysical properties of the β cells upon stimulation of membrane channels using an atomic force microscopic (AFM) nanoindentation system.

TiO2and Nb-TiO2 were investigated as stable supports for Pt-Ru electrocatalysts towards methanol oxidation. X-ray photo-electron spectroscopy (XPS) data for all these TiO2-based supports show oxidation states of Ti4+, with no Ti3+, suggesting low electronic conductivity. However, the deposition of metal nanoparticles onto the supports at loadings of 60 wt% metal dramatically increased conductivity, making these electrodes (metal particles + support) suitable for electrochemistry even though the supports have low conductivity. An optical cell is described for high-throughput backscattering Raman spectroscopic measurements of hydrogen storagematerials at pressures up to 10 MPa and temperatures up to 823 K. High throughput is obtained by employing a 60 mm diameter × 9 mm thick sapphire window, with a corresponding 50 mm diameter unobstructed optical aperture. To reproducibly seal this relatively large window to the cell body at elevated temperatures and pressures, a gold o-ring is employed. Mechanical properties of polymer electrolyte membrane have important implications in the endurance of many electrochemical energy conversion devices [1]. Mechanical experiments are effective method to reveal discrete damage developed in the membrane by quantifying the change of mechanical strength and toughness. Regular strain measurement methods are overly intrusive to the thin and relatively fragile polymer electrolyte membrane; hence optical-based minimal invasive stress/strain measurement methods are studied.

The single-particle model presented by Santhanagopalan et al. [ J. Power Sources , 156 , 620 (2006)] is extended to include an energy balance. The temperature dependence of the solid phase diffusion coefficient of the lithium in the intercalation particles, the electrochemical reaction rate constants, and the open circuit potentials (OCPs) of the positive and negative electrodes are included in the model. The solution phase polarization is approximated using a nonlinear resistance, which is a function of current and temperature. The electronic conductivity and thermodynamic stability of mixed n-type and p-type doped SrTiO3 have been investigated at anodic solid oxide fuel cell (SOFC) conditions using density functional theory (DFT) calculations. In particular, constrained ab initio thermodynamic calculations have been performed to evaluate the phase stability and reducibility of various Nb- and Ga-doped SrTiO3 at synthesized and anodic SOFC conditions. The density of states (DOS) of these materials was analyzed to study the effects of n- and p-doping on the electronic conductivity. Dilute sulfuric acid was used to pretreat coastal Bermuda grass at high temperature prior to enzymatic hydrolysis. After both pretreatment and enzymatic hydrolysis processes, the highest yield of total sugars (combined xylose and glucose) was 97% of the theoretical value. The prehydrolyzate liquor was analyzed for inhibitory compounds (furfural, hydroxymethylfurfural (HMF)) in order to assess potential risk for inhibition during the following fermentation. Accounting for the formation of the inhibitory compounds, a pretreatment with 1.2% acid at 140 °C for 30 min with a total sugar yield of 94% of the theoretical value may be more favorable for fermentation. Rapid detection of viable virus particles in water and other environmental samples is essential to public health risk assessment, homeland security and environmental protection. Current virus detection methods, especially assessing viral infectivity, are complex and time-consuming, making point-of-care detection a challenge. Faster, more sensitive, highly specific methods are needed to quantify potentially hazardous viral pathogens and to determine if suspected materials contain viable viral particles. Identifying molecular mechanisms or therapeutic targets is typically based on large-scale cellular analysis that measures the abundance of mRNA or protein; however, abundance does not necessarily correlate with activity. We report a method for direct large-scale quantification of active pathways that employs a cellular array with parallel gene delivery of constructs that report pathway activity. The reporter constructs encode luciferase, whose expression is influenced by binding of transcription factors (TFs), which are the downstream targets of signaling pathways. Inefficient gene delivery is a critical factor limiting the use of nonviral methods in therapeutic applications including gene therapy and tissue engineering. There have been few efforts to understand or engineer the molecular signaling pathways that dictate the efficacy of gene transfer. Microarray analysis was used to determine endogenous gene expression profiles modulated during nonviral gene transfer. Nonviral DNA lipoplexes were delivered to HEK 293T cells. There is continued growth and development of outreach programs designed to increase pre-college students’ awareness and understanding of engineering as a profession and as a career. These outreach programs vary in format and in the groups targeted for participation but maintain the same fundamental goal of increasing participant knowledge of engineering. Many of these outreach programs also maintain the implicit goal of increasing the participants' knowledge and attitudes toward college. The additional resources and funding that are commonly allocated to support outreach programs frequently involve documenting accountability which motivates evaluation of program impact. Within the National Weather Service River Forecast System, water supply forecasting is performed through Ensemble Streamflow Prediction (ESP). ESP relies both on the estimation of initial conditions and historically resampled forcing data to produce seasonal volumetric forecasts. In the western US, the accuracy of initial condition estimation is particularly important due to the large quantities of water stored in mountain snowpack. In order to improve the estimation of snow quantities, this study explores the use of ensemble data assimilation. We present measurements which show that the tidal Ems River in Germ any is extremely muddy over a 30 km + turbid zone, with fluid mud o f 1-2 m thickness covering the bed with suspended sediment concentrations (SSC) o f greater than 50 kg.m-3. Moreover, we show that these elevated SSC contain large quantities of organic material which deplete dissolved oxygen (DO) and produce summertime hypoxic zones. Using mathematical modeling, we develop simplified representations o f the estuary physics that reproduce the tidally-averaged circulation, SSC distribution, and oxygen depletion. Low flow forecasting, days or even months in advance, is particularly important to the efficient operation of power plants and freight shipment. This study presents a low flow forecasting model with a lead time of 14 days for the Rhine River. The forecasts inherit uncertainty sources mainly because of model parameterization. Across the state, region, and nation highway safety investigators have developed a wide variety of tools and techniques for highway safety investigation procedures. Analysis techniques can range from systematic evaluation approaches such as the Oregon Department of Transportation’s (ODOT’s) Safety Priority Index System (SPIS) down to specific localized safety assessment strategies. It is important to develop consistent methods for safety evaluations across Oregon to ensure that safety investigations are conducted in a similar manner. Thin manganese films were grown by thermal evaporation in high vacuum. The growth rate was varied to determine how the growth rate affects the continuity of ultra-thin films. The resistance was measured with the two-point method in situ and the data was recorded digitally. The resistance was converted to resistivity and this was plotted against thickness. It was found that resistivity decreases exponentially as the film begins to form in islands. The film reaches a critical thickness as the islands coalesce and the data can then be fitted with Fuchs’ equation. The resistivity continues to decrease but the minimum resistivity …
