The Toxin Complex (Tc) superfamily consists of toxin translocases that contribute to the targeting, delivery, and cytotoxicity of certain pathogenic Gram-negative bacteria. Membrane receptor targeting is driven by the A-subunit (TcA), which comprises IgG-like receptor binding domains (RBDs) at the surface. To better understand XptA2, an insect specific TcA secreted by the symbiont X. nematophilus from the intestine of entomopathogenic nematodes, we determined structures by X-ray crystallography and cryo-EM. Contrary to a previous report, XptA2 is pentameric. RBD-B exhibits an indentation from crystal packing that indicates loose association with the shell and a hotspot for possible receptor binding or a …

Trichloroethylene (TCE) is a prominent groundwater pollutant due to its stability, widespread contamination, and negative health effects upon human exposure; thus, an immense need exists for enhanced environmental remediation techniques. Temperature-responsive domains and catalyst incorporation in membrane domains bring significant advantages for toxic organic decontamination. In this study, hollow fiber membranes (HFMs) were functionalized with stimuli-responsive poly-N-isopropylacrylamide (PNIPAm), poly-methyl methacrylate (PMMA), and catalytic zero- valent iron/palladium (Fe/Pd) for heightened reductive degradation of such pollutants, utilizing methyl orange (MO) as a model compound. The global water crisis growth has led to a tremendous increase in membrane technology research. Membranes are favored over many other technologies for water treatment because, in principle, they require no chemical additives and can be used isothermally at low temperatures. Membranes that can reject contaminants and salts, produce adequate permeate flux values, and require minimal cleaning are highly demanded. However, most synthesized membranes on the market have associated problems, such as membrane fouling; inverse relationships between flux and solute rejection; and the high cost of synthesis, operation, and maintenance. With demand for coal from the Central Appalachian Basin waning, coalitions and organizations throughout the region have sought to diversify the region’s economy. However, the potential exists to use coal and coal-based resources to produce new and innovative products and kick start economic growth in the basin. Opioid use disorder (OUD) is a leading cause of death in the United States placing a tremendous burden on patients, their families, and health care systems. Artificial intelligence (AI) can be harnessed with available healthcare data to produce automated OUD prediction tools. In this retrospective study, we developed AI based models for OUD prediction and showed that AI can predict OUD more effectively than existing clinical tools including the unweighted opioid risk tool (ORT). Data include 474,208 patients’ data over 10 years; 269,748 were females with an average age of 56.78 years. Cases are prescription opioid users with at least …

Electric Vehicles (EVs) are gaining popularity among consumers and are expected to play a significant role in the future of transportation. Within this paper, a reverse auction is formulated through an optimization problem to minimize the utility energy cost using Vehicle-to-Grid (V2G) operation, as well as transition residential communities to dispatchable aggregate constant load profiles for demand response (DR). To improve the charge-carrier transport capabilities of thin-film organic materials, the intermolecular electronic couplings in the material should be maximized. Decreasing intermolecular distance while maintaining proper orbital overlap in highly conjugated aromatic molecules has so far been a successful way to increase electronic coupling. We attempted to decrease the intermolecular distance in this study by synthesizing cocrystals of simple benzoic acid coformers and dipyridyl-2,2′- bithiophene molecules to understand how the coformer identity and pyridine N atom placement affected solid-state properties. We found that with the 5-(3-pyridyl)-5′-(4-pyridyl)-isomer, the 4- pyridyl ring interacted with electrophiles and protons more strongly. Synthesized cocrystal powders …

Smart homes and virtual power plant (VPP) controls are growing fields of research with potential for improved electric power grid operation. A novel testbed for co-simulation of electric power distribution systems and distributed energy resources (DERs), is employed to evaluate VPP scenarios and propose an optimization procedure. DERs of specific interest include behind-the-meter (BTM) solar photovoltaic (PV) systems as well as heating, ventilation, and air-conditioning (HVAC) systems. In this paper, a dual-stator axial flux permanent magnet machine with a spoke-type rotor for electric vehicle applications is investigated. The rotor PMs are positioned in a spoke-type field-concentrating arrangement, which provides high flux concentration. Two stators—one stator configuration with double-layer wound coils and another without winding—are employed. The usage of single-layer wound coils is possible in this topology, and by using this type of winding it is possible to implement advanced in-slot cooling systems. Dynamic wireless charging of electric vehicles (EV) is an emerging technology with the potential to address range anxiety and reduce the size of batteries or provide charge sustaining operation. This report discusses a project undertaken by Kentucky Transportation Center (KTC) researchers to create a comprehensive geotechnical database for highway planning, design, maintenance, and repair. The project consolidated multiple geotechnical data sources into a unified database accessible to Kentucky Transportation Cabinet (KYTC) staff, enabling informed decision making and improving project outcomes. Databases were merged with great attention to detail, ensuring data integrity and coherence. This paper investigates employing a Halbach PM rotor array to increase torque and power density within coreless axial flux permanent magnet (AFPM) machines. Analytical and 2/3-dimensional finite element analysis (FEA) methods are developed to study torque and power capabilities within an example double-rotor, single-stator coreless AFPM machine with a PCB stator. Compared to a surface PM topology of the same mass and volume, employing a Halbach array increases torque density by as much as 30% through increased airgap flux density amplitude. This paper presents a multi-objective design optimization for a novel direct-drive wind turbine generator. The proposed electric machine topology employs an outer rotor of the reluctance type and a special modular stator including three phase-windings and spoke-type permanent magnets (PMs). Each stator module includes a single coil toroidally wound around the ferromagnetic core. Consecutive stator modules are separated by PMs and include coils belonging to a different phase. Environmental conditions of groundwater and surface water greatly vary as a function of location. Factors such as ionic strength, water hardness, and solution pH can change the physical and chemical properties of the nanocomposites used in remediation and the pollutants of interest. In this work, magnetic nanocomposite microparticle (MNM) gels are used as sorbents for remediation of PCB 126 as model organic contaminant. Wastewater-based epidemiology (WBE) has enabled us to describe Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) infections in populations. However, implementation of wastewater monitoring of SARS- CoV-2 is limited due to the need for expert staff, expensive equipment, and prolonged processing times. As WBE increases in scope (beyond SARS-CoV-2) and scale (beyond developed regions), there is a need to make WBE processes simpler, cheaper, and faster. We developed an automated workflow based on a simplified method termed exclusion-based sample preparation (ESP). Transportation planning and design decisions are typically made using point estimates of traffic and safety measures. This report addresses the uncertainty of data used to make these decisions. It focuses on two data elements — project safety ratings and volume estimates from traffic counts. For safety ratings, the Kentucky Transportation Cabinet’s Strategic Highway Investment Formula for Tomorrow (SHIFT) crash history score is used as a case study. Confidence intervals are developed for the Cabinet’s new safety metric, which incorporates estimates of future crashes by severity as well as excess expected crashes above safety goals. The Strategic Highway Investment Formula for Tomorrow (SHIFT) is the Kentucky Transportation Cabinet's data-informed approach for comparing capital improvement projects and prioritizing limited transportation funds. SHIFT 2022 incorporates advancements in methods and flexibility. This project revises the SHIFT crash data safety metric. The crash data safety metric from the previous version of SHIFT was excess expected crashes (EECs). It is computed using the total number of crashes of all severities. In this study, it is found that the lncRNA, DNA damage inducible transcript 4 antisense RNA1 (DDIT4-AS1), is highly expressed in triple-negative breast cancer (TNBC) cell lines and tissues due to H3K27 acetylation in the promoter region, and promotes the proliferation, migration, and invasion of TNBC cells via activating autophagy. Mechanistically, it is shown that DDIT4-AS1 induces autophagy by stabilizing DDIT4 mRNA via recruiting the RNA binding protein AUF1 and promoting the interaction between DDIT4 mRNA and AUF1, thereby inhibiting mTOR signaling pathway. The widespread presence of numerous organic contaminants in water poses a threat to the ecological environment and human health. Magnetic nanocomposites exposed to an alternating magnetic field (AMF) have a unique ability for magnetically mediated energy delivery (MagMED) resulting from the embedded magnetic nanoparticles; this localized energy delivery and associated chemical and thermal effects are a potential method for removing contaminants from water. Slow-moving, chronically destructive landslides are projected to grow in number globally in response to precipitation increases from climate change, and land disturbances from wildfire, mining and construction. In the Cincinnati and northern Kentucky metropolitan area, USA, landslides develop in colluvium that covers the steep slopes along the Ohio River and its tributaries. Here we quantify elevation changes in a slow-moving colluvial landslide over 14 years using county- wide lidar, uncrewed aerial vehicle (UAV) structure-from-motion (SfM) surveys and a UAV lidar survey. This paper aims to address the significant financial, environmental, and social risks posed by climate change to the mining industry, which is responsible for approximately 8% of global greenhouse gas emissions. With 70% of mining projects for the six largest mining companies located in water-stressed regions, the industry is facing increasing pressure to reduce its impact. Our study investigates the applicability of multi-objective optimization to integrate environmental impact considerations into short-term planning for mining operations. Cardiovascular diseases are a leading cause of mortality and morbidity worldwide. Aberrant thrombosis is a common feature of systemic conditions like diabetes and obesity, and chronic inflammatory diseases like atherosclerosis, cancer, and autoimmune diseases. Upon vascular injury, usually the coagulation system, platelets, and endothelium act in an orchestrated manner to prevent bleeding by forming a clot at the site of the injury. Abnormalities in this process lead to either excessive bleeding or uncontrolled thrombosis/insufficient antithrombotic activity, which translates into vessel occlusion and its sequelae. CO2 capture is critical to solving global warming. Amine-based solvents are extensively used to chemically absorb CO2. Thus, it is crucial to study the chemical absorption of CO2 by amine-based solvents to better understand and optimize CO2 capture processes. Here, we use quantum computing algorithms to quantify molecular vibrational energies and reaction pathways between CO2 and a simplified amine-based solvent model—NH3. Molecular vibrational properties are important to understanding kinetics of reactions. However, the molecule size correlates with the strength of anharmonicity effect on vibrational properties, which can be challenging to address using classical computing. Demonstrated to be highly effective for lignocellulosic biomass pretreatment, deep eutectic solvent (DES) has attracted increasing attention owing to its advantages of simple synthesis, relatively low chemical cost, and better biocompatibility as compared to certain ionic liquids. Abdominal aortic aneurysm (AAA) is a common vascular disease associated with significant phenotypic alterations in vascular smooth muscle cells (VSMCs). Gasdermin D (GSDMD) is a pore-forming effector of pyroptosis. In this study, the role of VSMC-specific GSDMD in the phenotypic alteration of VSMCs and AAA formation is determined. Single-cell transcriptome analyses reveal Gsdmd upregulation in aortic VSMCs in angiotensin (Ang) II-induced AAA. VSMC-specific Gsdmd deletion ameliorates Ang II-induced AAA in apolipoprotein E (ApoE)−/− mice. Using untargeted metabolomic analysis, it is found that putrescine is significantly reduced in the plasma and aortic tissues of VSMC-specific GSDMD deficient mice. High putrescine levels …
