Design And Analysis Of Curcumin Conjugated Poly(Beta-Amino Ester) Networks For Controlled Release In Oxidative Stress Environments, Carolyn T. Jordan
Design And Analysis Of Curcumin Conjugated Poly(Beta-Amino Ester) Networks For Controlled Release In Oxidative Stress Environments, Carolyn T. Jordan
Theses and Dissertations--Chemical and Materials Engineering
Oxidative stress, the imbalance of free radical generation with antioxidant defenses, leads to cellular inflammation, apoptosis and cell death. This compromised environment results in debilitating diseases, such as oral mucositis (OM), atherosclerosis, and ischemia/reperfusion injury. Antioxidant therapeutics has been a proposed strategy to ameliorate these imbalances and maintain homeostatic environments. However, the success of these approaches, specifically curcumin, has been limited due to characteristics such as hydrophobicity and high reactivity when released as bolus doses to contest to oxidative stress induced diseases. The development of a controlled release system to aid in protection of the antioxidant capacity of curcumin, as …
The Development Of Microfluidic Devices For The Production Of Safe And Effective Non-Viral Gene Delivery Vectors, Jason Matthew Absher
The Development Of Microfluidic Devices For The Production Of Safe And Effective Non-Viral Gene Delivery Vectors, Jason Matthew Absher
Theses and Dissertations--Chemical and Materials Engineering
Including inherited genetic diseases, like lipoprotein lipase deficiency, and acquired diseases, such as cancer and HIV, gene therapy has the potential to treat or cure afflicted people by driving an affected cell to produce a therapeutic protein. Using primarily viral vectors, gene therapies are involved in a number of ongoing clinical trials and have already been approved by multiple international regulatory drug administrations for several diseases. However, viral vectors suffer from serious disadvantages including poor transduction of many cell types, immunogenicity, direct tissue toxicity and lack of targetability. Non-viral polymeric gene delivery vectors (polyplexes) provide an alternative solution but are …
Combinatorial Screening Approach In Developing Non-Equiatomic High Entropy Alloys, Azin Akbari
Combinatorial Screening Approach In Developing Non-Equiatomic High Entropy Alloys, Azin Akbari
Theses and Dissertations--Chemical and Materials Engineering
High entropy alloys (HEA) are a relatively new group of alloys first introduced in 2004. They usually contain 5 to 6 different principle elements. Each of these elements comprise 5-35 at. % of the chemical composition of the alloy. There is a growing interest in the research community about the development of these alloys as well as their engineering applications. Some HEAs have interesting properties that have made them well suited for higher temperature applications, particularly refractory uses, while some have been shown to maintain their mechanical properties even at cryogenic temperatures.
Initially, the HEA research was focused on developing …
A Microstructure-Based Model Validated Experimentally For Quantification Of Short Fatigue Crack Growth In Three-Dimensions, Pei Cai
Theses and Dissertations--Chemical and Materials Engineering
Built on the recent successes in understanding the crystallographic mechanism for short fatigue crack (SFC) growth across a grain boundary (GB) and developing an experimental method to quantify the GB resistance against short crack growth, a microstructure-based model was developed in this study to simulate the growth behaviors of SFCs in 3-D, by taking into account both the driving force and resistance along at each point along the crack front in an alloy. It was found that the GB resistance was a Weibull function of the minimum twist angle of crack deflection at the boundary in AA2024-T3 Al alloys. In …
Defect Chemistry And Transport Properties Of Solid State Materials For Energy Storage Applications, Xiaowen Zhan
Defect Chemistry And Transport Properties Of Solid State Materials For Energy Storage Applications, Xiaowen Zhan
Theses and Dissertations--Chemical and Materials Engineering
Replacing organic liquid electrolytes with nonflammable solid electrolytes can improve safety, offer higher volumetric and gravimetric energy densities, and lower the cost of lithium-ion batteries. However, today’s all-solid-state batteries suffer from low Li-ion conductivity in the electrolyte, slow Li-ion transport across the electrolyte/electrode interface, and slow solid-state Li-ion diffusion within the electrode. Defect chemistry is critical to understanding ionic conductivity and predicting the charge transport through heterogeneous solid interfaces. The goal of this dissertation is to analyze and improve solid state materials for energy storage applications by understanding their defect structure and transport properties.
I have investigated defect chemistry of …
Mixed Matrix Flat Sheet And Hollow Fiber Membranes For Gas Separation Applications, Nicholas W. Linck
Mixed Matrix Flat Sheet And Hollow Fiber Membranes For Gas Separation Applications, Nicholas W. Linck
Theses and Dissertations--Chemical and Materials Engineering
Mixed matrix membranes (MMM) offer one potential path toward exceeding the Robeson upper bound of selectivity versus permeability for gas separation performance while maintaining the benefits of solution processing. Many inorganic materials, such as zeolites, metal-organic frameworks, or carbon nanotubes, can function as molecular sieves, but as stand-alone membranes are brittle and difficult to manufacture. Incorporating them into a more robust polymeric membrane matrix has the potential to mitigate this issue.
In this work, phase inversion polymer solution processing for the fabrication and testing of asymmetric flat sheet mixed matrix membranes was employed with CVD-derived multiwall carbon nanotubes (MWCNTs) dispersed …
The Formation Mechanism Of Α-Phase Dispersoids And Quantification Of Fatigue Crack Initiation By Experiments And Theoretical Modeling In Modified Aa6061 (Al-Mg-Si-Cu) Alloys, Gongwang Zhang
Theses and Dissertations--Chemical and Materials Engineering
AA6061 Al alloys modified with addition of Mn, Cr and Cu were homogenized at temperatures between 350 ºC and 550 ºC after casting. STEM experiments revealed that the formation of α-Al(MnFeCr)Si dispersoids during homogenization were strongly affected by various factors such as heating rate, concentration of Mn, low temperature pre-nucleation treatment and homogenization temperature. Through analysis of the STEM results using an image software Image-Pro, the size distributions and number densities of the dispersoids formed during different annealing treatments were quantitatively measured. It was revealed that increasing the heating rate or homogenization temperature led to a reduction of …
Synthesis And Characterization Of Magnesium - Titanium Composites By Severe Plastic Deformation, Baleegh Alobaid
Synthesis And Characterization Of Magnesium - Titanium Composites By Severe Plastic Deformation, Baleegh Alobaid
Theses and Dissertations--Chemical and Materials Engineering
Magnesium alloys are widely used in engineering applications, including aerospace and automobile industries, due to their desirable properties, such as lower density, high damping capacity, relatively high thermal conductivity, good machinability, and recyclability. Researchers have, therefore, been developing new magnesium materials. However, mechanical and corrosion properties are still limiting many commercial applications of magnesium alloys. In this Ph.D. thesis research, I developed Mg-Ti composite materials to offer some solutions to further improve the mechanical behavior of magnesium, such as titanium-magnesium (Ti-Mg) claddings, Mg-Ti multilayers, and Ti particle enforced Mg alloys. Low cost manufacturing processes, such as hot roll-bonding (RB) and …

Synthesis Of Biologically-Inspired Nanofiltration Membranes Using Protected, Mutated, And Simulated Aquaporins, Priyesh Ashokrao Wagh
Theses and Dissertations--Chemical and Materials Engineering
Gram-negative bacterial cells are surrounded by a cell membrane which protects the cell and controls the transport of nutrients and waste products in and out of the cells at a fast rate. This rapid transport of nutrients and wastes through the cell membrane is made possible by channel proteins called porins. Various types of porins present in the cell membrane have specific functions depending on their selectivity towards different nutrients, and channel proteins selective towards water are called aquaporins. These proteins restrict the passage of all entities except water molecules and they provide a fast transport rate of water molecules …

Surface Functionalization Via Photoinitiated Radical Polymerization For Rare Cell Isolation And Mechanical Protection, Calvin Frank Cahall
Theses and Dissertations--Chemical and Materials Engineering
Surface functionalization of living cells for cell therapeutics has gained substantial momentum in the last two decades. From encapsulating islets of Langerhans, to cell laden gels for tissue scaffolds, to individual cell encapsulation in thin hydrogels, to surface adhesives and inert surface camouflage, modification of living cell surfaces has a wide array of important applications. Here we use hydrogel encapsulation of individual cells as a mode of protection from mechanical forces for high throughput cell printing, and chemical stimuli for the isolation of rare cells in blood.
In the first study, we review methods of surface functionalization and establish a …
The Physical Behavior And Characterization Of Nanoporous Silicon And Dispenser Cathode Surfaces, Tyler Lucius Corey Maxwell
The Physical Behavior And Characterization Of Nanoporous Silicon And Dispenser Cathode Surfaces, Tyler Lucius Corey Maxwell
Theses and Dissertations--Chemical and Materials Engineering
Nanostructured materials have received a surge of interest in recent years since it has become apparent that reducing the size of a material often leads to heightened mechanical behavior. From a fundamental standpoint, this stems from the confinement of dislocations. Applications in microelectromechanical devices, lithium ion batteries, gas sensing and catalysis are realized by combining the improvements in mechanical behavior from material size reduction with the heightened chemical activity offered by materials with a high surface-area-to-volume ratio. In this study, films of nanoporous Si-Mg were produced through magnetron sputtering, followed by dealloying using an environmentally benign process with distilled water. …
Morphological And Energetic Effects On Charge Transport In Conjugated Polymers And Polymer-Nanowire Composites, Zhiming Liang
Morphological And Energetic Effects On Charge Transport In Conjugated Polymers And Polymer-Nanowire Composites, Zhiming Liang
Theses and Dissertations--Chemistry
Organic semiconductors have wide applications in organic-based light-emitting diodes, field-effect transistors, and thermoelectrics due to the easily modified electrical and optical properties, excellent mechanical flexibility, and solution processability. To fabricate high performance devices, it is important to understand charge transport mechanisms, which are mainly affected by material energetics and material morphology. Currently it is difficult to control the charge transport properties of new organic semiconductors and organic-inorganic nanocomposites due to our incomplete understanding of the large number of influential variables. Molecular doping of π-conjugated polymers and surface modification of nanowires are two means through which charge transport can be manipulated. …

Complex-oxides have seen an enormous amount of attention in the realm of Condensed Matter Physics and Materials Science/Engineering over the last several decades. Their ability to host a wide variety of novel physical properties has even caused them to be exploited commercially as dielectric, metallic and magnetic materials. Indeed, since the discovery of high temperature superconductivity in the “Cuprates” in the late 1980’s there has been an explosion of activity involving complex-oxides. Further, as the experimental techniques and equipment for fabricating thin films and heterostructures of these materials has improved over the last several decades, the search for new and …

Background: Collaboration between humans and computers has become pervasive and ubiquitous, however current computer systems are limited in that they fail to address the emotional component. An accurate understanding of human emotions is necessary for these computers to trigger proper feedback. Among multiple emotional channels, physiological signals are synchronous with emotional responses; therefore, analyzing physiological changes is a recognized way to estimate human emotions. In this paper, a three-stage decision method is proposed to recognize four emotions based on physiological signals in the multi-subject context. Emotion detection is achieved by using a stage-divided strategy in which each stage deals with …

The Role Of Catalytic Residue PKA On The Hydrolysis/Transglycosylation Partition In Family 3 Β-Glucosidases, Inacrist Geronimo, Christina M. Payne, Mats Sandgren
Chemical and Materials Engineering Faculty Publications
β-Glucosidases (βgls) primarily catalyze the hydrolysis of the terminal glycosidic bond at the non-reducing end of β-glucosides, although glycosidic bond synthesis (called transglycosylation) can also occur in the presence of another acceptor. In the final reaction step, the glucose product or another substrate competes with water for transfer to the glycosyl-enzyme intermediate. The factors governing the balance between the two pathways are not fully known; however, the involvement of ionizable residues in binding and catalysis suggests that their pKa may play a role. Through constant pH molecular dynamics simulations of a glycoside hydrolase Family 3 (GH3) βgl, we …

The cytoskeletal architecture directly affects the morphology, motility, and tensional homeostasis of the cell. In addition, the cytoskeleton is important for mitosis, intracellular traffic, organelle motility, and even cellular respiration. The organelle responsible for a majority of the energy conversion for the cell, the mitochondrion, has a dependence on the cytoskeleton for mobility and function. In previous studies, we established that cytoskeletal inhibitors altered the movement of the mitochondria, their morphology, and their respiration in human dermal fibroblasts. Here, we use this protocol to investigate applicability of power law diffusion to describe mitochondrial locomotion, assessment of rates of fission and …

New and advanced opportunities are arising for the synthesis and functionalization of membranes with selective separation, reactivity, and stimuli-responsive behavior. One such advancement is the integration of bio-based channels in membrane technologies. By a layer-by-layer (LbL) assembly of polyelectrolytes, outer membrane protein F trimers (OmpF) or “porins” from Escherichia coli with central pores ∼2 nm in diameter at their opening and ∼0.7 × 1.1 nm at their constricted region are immobilized within the pores of poly(vinylidene fluoride) microfiltration membranes, in contrast to traditional ruptured lipid bilayer or vesicle processes. These OmpF-membranes demonstrate selective rejection of non-charged organics over ionic solutes, …

The present study examines the interaction of hydrogen and nitrogen plasmas with gallium in an effort to gain insights into the mechanisms behind the synergetic effect of plasma and a catalytic metal. Absorption/desorption experiments were performed, accompanied by theoretical-computational calculations. Experiments were carried out in a plasma-enhanced, Ga-packed, batch reactor and entailed monitoring the change in pressure at different temperatures. The results indicated a rapid adsorption/dissolution of the gas into the molten metal when gallium was exposed to plasma, even at a low temperature of 100 °C. The experimental observations, when hydrogen was used, indicate that gallium acts as a …

Background: Patients with repaired tetralogy of Fallot (TOF) have progressive, adverse biventricular remodeling, leading to abnormal contractile mechanics. Defining the mechanisms underlying this dysfunction, such as diffuse myocardial fibrosis, may provide insights into poor long-term outcomes. We hypothesized that left ventricular (LV) diffuse fibrosis is related to impaired LV mechanics.
Methods: Patients with TOF were evaluated with cardiac magnetic resonance in which modified Look-Locker (MOLLI) T1-mapping and spiral cine Displacement encoding (DENSE) sequences were acquired at three LV short-axis positions. Linear mixed modeling was used to define the association between regional LV mechanics from DENSE based on regional T1-derived diffuse …

Chemical and Materials Engineering Faculty Publications
A novel crosslinker [4,4′-dihydroxybiphenyl diacrylate (44BDA)] was developed, and a series of temperature-responsive hydrogels were synthesized through free radical polymerization of N-isopropylacrylamide (NIPAAm) with 44BDA. The temperature-responsive behavior of the resulting gels was characterized by swelling studies, and the lower critical solution temperature (LCST) of the hydrogels was characterized through differential scanning calorimetry. Increased content of 44BDA led to a decreased swelling ratio and shifted the LCST to lower temperatures. These novel hydrogels also displayed resiliency through multiple swelling–deswelling cycles, and their temperature responsiveness was reversible. The successful synthesis of NIPAAm-based hydrogels crosslinked with 44BDA has led to a …

Commercial scale production of biofuels from lignocellulosic feed stocks has been hampered by the resistance of plant cell walls to enzymatic conversion, primarily owing to lignin. This study investigated whether DypB, the lignin-degrading peroxidase from Rodococcus jostii, depolymerizes lignin and reduces recalcitrance in transgenic tobacco (Nicotiana benthamiana). The protein was targeted to the cytosol or the ER using ER-targeting and retention signal peptides. For each construct, five independent transgenic lines were characterized phenotypically and genotypically. Our findings reveal that expression of DypB in the cytosol and ER does not affect plant development. ER-targeting increased protein accumulation, and …

Vapor intrusion (IV) exposure risks are difficult to characterize due to the role of atmospheric, building and subsurface processes. This study presents a three-dimensional VI model that extends the common subsurface fate and transport equations to incorporate wind and stack effects on indoor air pressure, building air exchange rate (AER) and indoor contaminant concentration to improve VI exposure risk estimates. The model incorporates three modeling programs: (1) COMSOL Multiphysics to model subsurface fate and transport processes, (2) CFD0 to model atmospheric air flow around the building, and (3) CONTAM to model indoor air quality. The combined VI model predicts AER …

Solid state hybrid organometal halide perovskite solar cells have become the research hotspots recently. We prepared perovskite solar cells by the simple two-step method in the ambient condition. The solar cell samples were kept in air and vacuum environments, respectively. Jsc shows good stability in vacuum for up to 27 days, while it drops a lot in air. There is a remarkable promotion of FF for the air-stored samples compared that of the vacuum-stored samples. Changes in Voc of vacuum-stored and airstored samples exhibited a similar behavior, both showing a slight increase. We believe the moisture in air …

The Federal Highway Administration (FHWA) requires state transportation agencies to submit a biennial report on local highway finances. The purpose of these reports is to provide FHWA with the data it needs to capture the financing of highway activities at the local level. Based on this information, it can identify trends in revenue, expenditures, investments, and program development, and in turn make decisions about future investments. The report, FHWA-536, asks agencies to report on four areas of local highway finance: 1) disposition of highway-user revenues; 2) revenues used for roads and streets identified by source and funding type; 3) road …

Applicability Of Zipper Merge Versus Early Merge In Kentucky Work Zones, Erin Lammers-Staats, Jerry G. Pigman, Brian K. Howell, Adam J. Kirk
Kentucky Transportation Center Research Report
In an effort to improve work zone safety and streamline traffic flows, a number of state transportation agencies (STAs) have experimented with the zipper merge. The zipper merge differs from a conventional, or early, merge in that vehicles do not merge into the lane that remains open immediately after being notified of a lane closure. Rather, vehicles continue to occupy all lanes until they reach the taper, at which point — and directed by signage — vehicles take turns merging into the open lane, creating a zipper pattern. At the request of the Kentucky Transportation Cabinet (KYTC), Kentucky Transportation Center …

Colloidal perovskite nanocrystals comprised of all inorganic cesium lead halide (CsPbX3, X = Cl, Br, I or a mixture thereof) have potential as optical gain materials due to their high luminescence efficiency. In this work, cesium lead halide nanocrystals are continuously synthesized via a microreactor system consisting of poly(tetrafluoroethylene) (PTFE) capillaries. The synthesized nanocrystals possess excellent optical properties, including a full width at half maximum of 19–35 nm, high fluorescence quantum yield of 47.8–90.55%, and photoluminescence emission in the range of 450–700 nm. For the same precursor concentrations, the photoluminescence emission peak generally increases with increasing reaction temperature, …

Oxides as one of the commonly activating fluxes used in active tungsten inert gas welding (A-TIG welding) can dramatically increase the penetration depth to 2–4 times that of conventional welding. Using the oscillation principle of inviscid fluid, a robust method is developed to measure the average surface tension of molten metal during A-TIG welding for four different oxide activating fluxes of B2O3, TiO2, SiO2, and MgO. The experimental results suggest that the oxygen released from the decomposition of oxides is the dominant factor contributing to the change of the surface tension, which …
Transient Analysis Of Fracture Initiation In A Coupled Thermoelastic Solid, Louis Milton Brock
Transient Analysis Of Fracture Initiation In A Coupled Thermoelastic Solid, Louis Milton Brock
Mechanical Engineering Faculty Publications
An isotropic, thermoelastic solid is initially at rest at uniform (absolute) temperature, and contains a semi-infinite, plane crack. Application of in-plane and normal point forces to each face of the crack causes transient 3D growth. The related problem of discontinuities in temperature and displacement that exist on regions that exhibit dynamic similarity is first considered. Asymptotic expressions, whose inverses are valid near the crack edges for short times, are obtained in integral transform space. These lead to equations of the Wiener–Hopf type for the fracture problem. Analytical solutions are obtained and, upon inversion, subjected to a dynamic energy release rate …
