"Solubility is a critical factor of protein-based drugs during processing and patient administration. This study focused on two aspects of solubility: one was the poly (ethylene) glycol (PEG) precipitation of monoclonal antibodies (mAbs) and another was crystallins aggregation that is associated with cataracts. Protein precipitation by PEG is a common technique for downstream processing. The effects of pH, ionic strength and the exclude volume effect on the protein precipitation by PEG were extensively studied, but the effects of glycosylation on protein precipitation by PEG has not been examined. "Extreme winds impacting civil structures lead to death and destruction in all regions of the world. Specifically, tornadoes and hurricanes impact communities with severe devastation. On average, 1200 tornadoes occur in the United States every year. Tornadoes occur predominantly in the Central and Southeastern United States, accounting for an annual $1 billion in economic losses, 1500 injuries, and 90 deaths. The Joplin, MO Tornado in 2011 killed 161 people, injured more than 1000, destroyed more than 8000 structures, and caused $2.8 billion of property loss. “NASA is developing deployable entry technologies in the form of the Hypersonic Inflatable Aerodynamic Decelerator (HIAD) and the Adaptable Deployable Entry Technology (ADEPT) vehicles to support human exploration of Mars. "This work discusses the effects of radiation damage on organic phase change materials for use in nuclear reactor containments and space craft. The effects of radiation on the latent heat of polyethylene wax (PEW) and a eutectic of methyl palmitate and lauric acid (EMPaLA) are evaluated. These PCMs were irradiated in three locations: the Missouri University of Science and Technology research reactor (MSTR), the University of Missouri research reactor (MURR) and cyclotron (MUC) up to a total dose of 2826, 2895 and 662 Gy, respectively. “Heavy oil resources account for a large portion of the total oil reserves around the world. The target heavy oil reservoir is located on Alaska’s North Slope (ANS). Advantages of low-salinity HPAM polymer (LSP) over high-salinity polymer (HSP) were demonstrated. LSP could recover more oil with 40% less polymer consumption. No additional oil was recovered by HSP after LSP flood. The first-ever polymer flood pilot on ANS showed remarkable success regarding water cut reduction, oil production increase, delayed breakthrough, and projected oil recovery improvement. Polymer alone was insufficient to achieve satisfactory recovery as the reservoirs were highly heterogeneous. Microgels could …

Mine haul roads degrade rapidly due to extremely heavy loads on sub optimal construction materials. This research tests two methods of defect detection to determine limitations to current technology: optical detection and vehicle vibration response. Optical defect detection was found to have the least significant limitations for surface defect detection and vehicle vibration response modeling has the least significant limitations for subsurface defects. This research proposes novel methods to eliminate the critical limitations of both technologies.

The Montauk Bridge deck was assessed based on the portable seismic property analyzer and ground penetrating radar data. Based on the analysis of the portable seismic property analyzer data, it was determined that over 65% of bridge conditions were rated serious to poor condition with an average compressive strength of less than 2500 psi; less than 35% of bridge deck conditions were rated fair to good with an average compressive strength over 2500 psi. “This research investigates the physical and chemical processes that contribute to the detonation synthesis of silicon carbide nanoparticles. Bulk production of SiC nanoparticles through detonation is possible due to pressures achieved over 20 GPa and temperatures over 2000 K as well as quenching rates in excess of 13 billion K/second. These conditions catalyze reaction and bottom-up molecular growth while retaining particles < 100 nm in diameter. In this work, detonation synthesis of SiC was demonstrated by incorporation of polycarbosilane, an SiC precursor material, into an RDX/TNT explosive matrix prior to detonation. Detonation Synthesis of SiC was also accomplished by reacting elemental silicon with carbon liberated by the detonation of negatively oxygen balanced TNT. Hydrodynamic simulation of a 60:40 mass ratio RDX/TNT detonation created conditions thermodynamically suitable to produce cubic silicon carbide within the first 500 nanoseconds after the passage of the detonation wave while carbon remains chemically reactive for molecular formation. Simulations and experimental tests indicated that loading configuration and impedance mismatch of the precursor additives used in detonation synthesis results in conditions in the additives that exceed the accepted detonation pressure of the explosive by greater than three times. Finally, a full factorial experimental design showed increasing silicon concentration, reducing silicon size, and reducing oxygen balance by adjusting the ratio of RDX to TNT decreased the explosives detonation pressure by 20% and increased the soot yield and concentration of SiC observed in the detonation products by 82% and 442% respectively”--Abstract, page iv.

Coal dust explosibility is a health and safety concern that has been a recognized hazard for over 100 years. Initial testing by the Author using a Siwek 20L apparatus recorded a secondary maximum pressure at higher dust concentrations (1,000-7,000g/m3) with Pulverized Pittsburgh Coal (PCC). Higher dust concentrations are beyond the typical ASTM E1226 testing procedure but are possible in mining and processing scenarios. No reference documents have been discovered that show a secondary maximum pressure at higher dust concentrations. ”The fiber-optic extrinsic Fabry-Perot interferometer (EFPI) is one of the simplest sensing configurations and is widely used in various applications due to its prominent features, such as high sensitivity, immunity to electromagnetic interference, and remote operation capability. In this research, a novel one-dimensional wide-range displacement sensor and a three-dimensional displacement sensor based on fiber-optic EFPIs are demonstrated. This creates microstructure and mechanical property inhomogeneities and can stochastically cause defects. "Lead-acid batteries are an established technology with nearly 99% recyclability; however, lead-acid batteries produce only 40% of their theoretical capacity due to poor active mass utilization and PbSO4 pore blockage, and the longevity of the batteries is hampered by secondary reactions during the cycle life such as corrosion and gassing.
Lead-acid batteries were investigated and improved through several different approaches: an alternative electrolyte to mitigate secondary reactions, graphite additives to improve positive active mass (PAM) utilization, and dispersant additives to help the industrial pasting process.

"Dam failures by seepage and internal erosion have reportedly been attributed to up to 40% of all dam failures in the United States. Backwards piping erosion either through the embankment or foundation soils has been reported to be the major contributing method to seepage and internal erosion dam failures. This proposed research is motivated by the lack of understanding of the complex stress regimes associated with internal erosion in cohesionless soils under earthen structures. "Powder based additive manufacturing (AM) exhibits tremendous uncertainties, where variations in build quality is present despite utilizing similar build processing parameters. First, this work reports the features and formation mechanisms of five unique types of spatter during the LPBF process by in-situ high-speed, high-energy x-ray imaging. The unique physical characteristics of spatter are determined. The effect of laser scan speed and laser power on spatter formation, ejection, and mitigation are determined. "Rock bridges were defined as a segment of intact rock that separates from discontinuities. Numerous studies are focused on developing methods to quantify their significance in rock mass strength and deformability behavior. This thesis aimed to enhance understanding of the effect of rock bridges on jointed rock masses in both laboratory and slope scales. For this purpose, the lattice spring base Synthetic Rock Mass (LS-SRM) method was adopted to conduct the numerical models, distinguished in computational efficiency, and simulates the brittle failure and roughness of discontinuities.

"Recursive estimation methodologies, such as Kalman and Bayesian filters, typically require models of some kind to perform the estimation. This filtering process seeks to improve knowledge surrounding some quantities of interest, or states, over time by incorporation of imperfect observations. The models required pertain to the state dynamics, sensors from which measurements are obtained, and probabilistic models of the underlying stochastic processes.
