Due to the coupled electromechanical activity of cardiomyocytes (CM), a comprehensive heart-on-a-chip device as a cell characterizing tool must encompass the capability to quantify cellular contractility, conductivity, excitability, and rhythmicity. Cardiovascular diseases represent the global leading cause of morbidity and mortality. Cardiovascular calcification is the most significant predictor of cardiovascular events, but no therapeutic options exist to prevent or treat mineral formation in the vasculature. The presence of bone-like mineral increases cardiac work required to move blood through systemic circulation and can lead to mechanical stress in atherosclerotic plaques, promoting plaque rupture events that cause heart attacks. Clinical trials correlated bisphosphonates (BiPs), common anti-osteoporosis pharmaceuticals, with contradicting cardiovascular outcomes. Photonic crystal (PhC) hydrogels are a unique class of material that has tremendous promise as biomedical sensors. The underlying crystal structure allows for simple analysis of microstructural properties by assessing the diffraction pattern generated following laser illumination. The hydrogel medium provides elasticity, regenerability, and potential functionalization. Combining these two properties, photonic crystal hydrogels have the potential for sensing physical forces and chemical reagents using a low-cost, reusable platform.

The composition of communication lines and bidirectional smart devices in smart grids makes them greatly vulnerable to cyber-attacks. False data injection attacks (FDIAs) are of the most widespread cyber-attacks severely affecting the performance of smart grids that need to be analyzed. In this context, we analyze the feasibility of launching FDIAs in power systems with high renewables leading to false relay operations (FRO). By modeling the frequency dynamics of power systems, formalizing the FRO attack as a constraint satisfaction problem, our case studies show that power grids with renewables are more susceptible to FRO attacks. This thesis evaluates the impacts of sea-level rise-induced high groundwater table on the structural performance of typical Florida interstate, arterial, and local flexible pavements. Pavement longevity was determined under rising groundwater table level conditions using two analytical methods; the mechanistic-empirical (ME) analysis software and the current Florida empirical analysis practice. Moreover, ABAQUS and KENLAYER software were utilized to analyze the impacts of stress-dependent material nonlinearity on pavement structural performance.

Analysis results estimated that the pavement service life in Florida was reduced by as much as 77 percent with the effects of rising groundwater levels. The empirical method predicted higher rates …

Despite the many governmental and medicinal restrictions created to combat the opioid epidemic in the United States, opioid abuse and overdose rates continue to rise. The development of an aptamer-based voltammetric sensor and biosensor is described in this dissertation. The aim was to develop a low-cost, sensitive, and specific aptamer-based sensor for on-site, label-free determination of codeine and fentanyl in biological fluids. To do this, the surfaces of screen-printed carbon electrodes (SPCE) were modified with gold nanoparticles (AuNPs), followed by the addition of single-stranded DNA aptamers. Consequently, developing and implementing effective flood mitigation strategies is a constant necessity to protect against the damage caused by floods. This dissertation proposes a dynamic water storage management strategy for flood mitigation at the watershed scale. This strategy aims to release water in a coordinated manner from water storage units before a rainfall event, leading to the entire watershed becoming more effective for flood mitigation.

NASA Kennedy Space Center’s technical standard for corrosion protection of space flight hardware provides guidance concerning temperature restrictions and exposure limits for processing unpainted chromate conversion coatings (CCC) for the Orion crew space vehicle as part of the Artemis program. The standard requires that all CCC treated flight hardware components be fully coated within seven days with a maximum storage time of seven days at ambient temperatures (much less time at higher temperatures). This dissertation pursued the definition and evaluation of a processing approach for robust real-time orientation estimation of a miniature Magnetic, Angular-Rate, Gravity (MARG) module for use in a human-computer interaction system that also uses a 3-camera IR-video module for position estimation. The proposed algorithm introduces the novel idea of spatially mapping the level of trustworthiness of the magnetometer-based potential corrections to the orientation estimate. This trustworthiness level is used to reduce the strength of magnetometer-based corrections of the orientation estimate where the magnetic field distortion invalidates the assumptions necessary for those corrections.

Over 95% of radiation therapy (RT) treated breast cancer subjects undergo an adverse skin reaction known as radiation dermatitis (RD). Assessment of severity or grading of RD is clinically visual and hence subjective. Our objective is to determine sub-clinical tissue oxygenation (StO2) changes in response to RT treatment in breast cancer subjects using near-infrared spectroscopic imaging and correlate these changes to RD grading. Slab beam superstructures have been used for short-span bridges since prestressed concrete began to be used in the US. Poor joint performance of these superstructures led to recent revisions to design standards; one example being the Florida Slab Beam (FSB). The FSB and other similar slab beam systems include a composite cast-in-place (CIP) reinforced concrete deck and joint between adjacent beams. An optimized joint design has been developed to accelerate the construction times and eliminate the need for a CIP deck by using ultra-high performance concrete (UHPC) in the longitudinal closure pour to connect the precast members.

The prevalence of underactive bladder (UAB) increases with age, suggesting a link between age-related processes and lower urinary tract (LUT) symptoms; however, the underlying mechanisms of age-related UAB are poorly understood. UAB is characterized by inefficient voiding and bladder overdistension. Due to the unknown etiology, current therapeutic options are insufficient. Thus, a detailed understanding of its mechanism will facilitate the discovery of new treatments.

In Aim 1, we investigated the relationship between age and systems-level function of the LUT reflexes in three age groups of rats, testing the hypothesis that aging causes voiding reflexes to weaken. We systematically investigated the …
