The most complex heart valve is the mitral valve (MV). Many pathologies can affect the MV, including stenosis, regurgitation, prolapse, and mitral annulus calcification (MAC). MAC is chronic degeneration of the annulus, which is the fibrous, saddle-shaped “ring” that can contract and relax with the myocardium. The prevalence of MAC is around 15% but increases in patients with other cardiovascular diseases and risk factors. It is also thought to increase in patients with type 2 diabetes, but MAC has not been properly characterized within this population because of confounding factors such as cardiac disease and kidney disease. This thesis project describes the modification and evaluation of a new affinity membrane for rapid chromatographic purification of non-antibody proteins. The affinity membrane utilizes Im7/CL7 coupling technology developed by Dr. Vassylyev's lab at the University of Alabama Birmingham (UAB), licensed by TriAltus Bioscience. The behavior of the membrane was evaluated using purified CL7-tagged Cas9 as my model protein for static and dynamic binding capacity analysis.

Subwavelength gratings are a particularly promising option for creating flat optical devices; however, the implementation of subwavelength grating-based optics is limited by fabrication constraints. This system is based off a Particle micro-controller, Boron LTE CAT-M1 which allows for cellular connectivity for real-time data transmission. Natural language processing techniques have made leaps and bounds as far as usability and application domain, although it does not work for every task. Cardiovascular diseases (CVDs) are the leading cause of death worldwide in males (XY) and females (XX). Prior to menopause, females have a relative protection against serious cardiac pathologies compared to age-matched males. This phenomenon is widely attributed to the ovarian hormone estrogen. Unfortunately, hormone replacement therapy to maintain estrogen levels in postmenopausal females has overall adverse effects, and it is not recommended for long-term use or as a preventative measure for eCVDs. Microwave dielectric sensing has become a popular technique in biological cell sensing for its potential in online, label-free, and real-time sensing. At microwave frequencies probing signals are sensitive to intracellular properties since they are able to penetrate cell membranes, making microwave flow cytometry a promising technology for label-free biosensing. In this dissertation a microwave flow cytometer is designed and used to measure single biological cells and micro particles. This study developed an analytical tool for understanding spine tissues’ behavior in response to vertebral kinematics and spine pathology over a range of body postures. It proposed a novel pipeline of computational models based on predicting individual vertebral kinematics from measurable body-level motions, using musculoskeletal dynamics simulations to drive the vertebrae in corresponding spine FEMs.
A reformulated elastic surface node (ESN) lumbar model was developed for use in MSD simulations. Using computational modeling, we focus on the phase behavior of multicomponent systems incorporating enzyme and bottlebrush polymers where the agglomeration of multiple components occurs. We start with all-atom molecular dynamics (MD) simulations of lysozyme and polyethylene glycol (PEG) based polymer mixture to understand the mechanisms of preservation of lysozyme bioactivity at high temperatures with addition of PEG-derived bottlebrushes. We show that the PEG part of bottlebrushes phase separates at high temperature and shells the access of water to lysozyme, resulting in the preservation of lysozyme secondary structures. A semi-analytic trial function model is applied to simulate matrix diffusion in systems of parallel fractures, 2-D/3-D discrete fracture networks, and in 3-D heterogeneous media. The trial function model gives low normalized root mean square error (NRMSE) when compared to the parallel fracture analytical solution over a range of fracture spacing, with considerations for retardation and decay. The semi-analytic trial function model is efficient in time and maintains less than 6% NRMSE when it simulates matrix diffusive transport in 2-D/3-D discrete fracture networks (DFN). The objective of this study was to characterize the variability of dynamic modulus of asphalt mixes in South Carolina on the basis of geometry and anisotropy.
