Changes in the biomechanical properties of cells accompanying the development of various pathological conditions have been increasingly reported as biomarkers for various diseases and as a predictor of disease progression stages. For instance, cancer cells have been found to be less stiff compared to their healthy counterparts due to the proteomic and lipidomic dysregulations conferred by the underlying pathology. The separation and selective recovery of cells or extracellular vesicles secreted from such cells that have undergone these changes have been suggested to be of diagnostic and prognostic value.

The purpose of this thesis was to develop microscale fracture toughness tests to be performed in situ based off previously used macroscale fracture toughness tests. The thesis also was to use these tests to perform in situ analysis and imaging of reinforced adhesives during crack propagation. Two different fracture toughness tests were developed for this thesis through developing fixtures and sample geometry. A microscale double cantilever beam (DCB) test was developed for mode I fracture (opening mode). A microscale end notch flexure (ENF) test was developed for mode II fracture (sliding mode).

Magnetoelectric (ME) nanoparticles (MENs) intrinsically couple magnetic and electric fields. Using them as nuclear magnetic resonance (NMR) sensitive nanoprobes adds another dimension for NMR detection of biological cells based on the cell type and corresponding particle association with the cell. Based on ME property, for the first time we show that MENs can distinguish different cancer cells among themselves as well as from their normal counterparts. The core-shell nanoparticles are 30 nm in size and were not superparamagnetic. Past nuclear weapon production activities have left a significant legacy of uranium (U) contamination in the vadose zone (VZ) of the Department of Energy (DOE) Hanford Site. This U is a source of groundwater (GW) contamination. There is a concern that elevated U concentration would slowly infiltrate through the VZ, reach the GW water table, and then end up in nearby rivers and lakes. The development of a robotic slit-lamp for remote ophthalmology is the primary purpose of this work. In addition to novel mechanical designs and implementation, it was also a goal to develop a control system that was flexible enough to be adapted with minimal user adjustment to various styles and configurations of slit-lamps. The system was developed with intentions of commercialization, so common hardware was used for all components to minimize the costs. In regard to cancer therapy, magnetoelectric nanoparticles (MENs) have proven to be in a class of its own when compared to any other nanoparticle type. Like conventional magnetic nanoparticles, they can be used for externally controlled drug delivery via application of a magnetic field gradient and image-guided delivery. The current practice of the decision-making process does not consider all construction impacts in an integrated decision-making process. Despite the widespread recycling of Reclaimed Asphalt Pavement (RAP), a large portion of it is still wasted. One of the main reasons is the concern with the performance of high RAP mixtures. Asphalt binder aging and subsequent rejuvenation is one source of uncertainty. Rejuvenators are frequently added to high RAP mixes to enhance the properties of the binder. This enhancement is often perceived as simply lowering the viscosity. Three-dimensional graphene foam (3D GrF) is an interconnected, porous structure of graphene sheets with excellent mechanical, electrical and thermal properties, making it a candidate reinforcement for polymer matrices. GrF’s 3D structure eliminates nanoparticle agglomeration and provides seamless pathways for electron travel. The objective of this work is to fabricate low density GrF reinforced epoxy composites with superior mechanical and electrical properties and study the underlying deformation mechanisms. Coatings are widely used to mitigate corrosion of structural steel in aggressive humid environments. However, the service life is often diminished in aggressive environments. Repair of coatings can be costly due to materials, labor and environmental controls. So, in search for novel coatings, Chemically Bonded Phosphate Ceramic (CBPC) coating was investigated for marine bridge application. The research on CBPC coating considered various exposure environments such as inland, beach, salt-fog, wet and alternate wet and dry exposure to identify the degradation mechanism. As a proof of concept, copper (Cu) and Graphene Nanoplatelets (GNP) were chosen as the representative metal and 2D nanoplatelets, respectively. Densification and submergence of floating crude oil is proposed as a novel oil spills treatment method. Surface application of dry granular materials (e.g., quartz sand, limestone) on top of a floating oil layer increases the density of the floating oil phase/granule mixture and leads to formation of relatively large and stable aggregates with significant amounts of captured oil. The aggregates separate from the floating hydrophobic phase and settle by gravity. Implementation of this method will reduce the impact radius of a spill and its mobility, preventing direct contamination of beaches, coastal flora and fauna.

Capacitive coupling and direct shuttling of charges in nanoscale plasmonic components across a dielectric spacer and through a conductive junction lead to excitation of significantly different dipolar and charge transfer plasmon (CTP) resonances, respectively. Here, we demonstrate the excitation of dipolar and CTP resonant modes in metallic nanodimers bridged by phase-change material (PCM) sections, material and electrical characteristics of which can be controlled by external stimuli. Non-glucose biomarker-DNA oxidative damage biomarker 8-hydroxy-2?-deoxyguanosine (8-OHdG) has been successfully detected using a smartphone-enabled glucose meter. Through a series of immune reactions and enzymatic reactions on a solid lateral flow platform, 8-OHdG concentration has been converted to a relative amount of glucose, and therefore can be detected by conventional glucose meter directly. The device was able to detect 8-OHdG concentrations in phosphate buffer saline as low as 1.73 ng mL?1 with a dynamic range of 1?200 ng mL?1. This study examines the role of Graphene nanoplatelets (GNPs) as a solid lubricant additive to aluminum. Pure Al and Al-2 vol % GNP pellets are sintered by Spark Plasma Sintering (SPS). Their tribological properties are evaluated by a ball-on-disk tribometer at room temperature (RT) and high temperature (200 °C). Al-2 vol % GNP composite displayed poor densification (91%) and low hardness, resulting in poor wear resistance as compared to pure Al. However GNP addition resulted in a lower coefficient of friction (COF) as compared to pure aluminum at both temperatures. The results demonstrated that GNPs contribute to reducing COF by …
