The State of Wisconsin’s Department of Natural Resources is responsible for operating one of the largest state park systems in the United States. Potawatomi State Park, located on the Door County peninsula, consists of about 1,200 acres of flat to gently rolling upland terrain bordered by steep slopes and rugged limestone cliffs along Lake Michigan’s shoreline. A 75-ft observation tower sits atop a 150-ft bluff overlooking Lake Michigan. The USDA Forest Service, Forest Products Laboratory, was asked to conduct an assessment of the main support timbers of the tower. This report summarizes the results obtained from the inspection and assessment. …

Biologically inspired design is a fairly new field with ongoing research. There are different tools to assist designers and biologists in bio-inspired design. Biofiltration without pre-ozonation has the capability to remove natural organic matter (NOM) fractions that serve as precursors of disinfection byproducts (DBPs), which include the four regulated trihalomethanes (THMs) and dichloroacetonitrile (DCAN). Rapid small-scale column tests (RSSCTs) and Pilot Plant filters operated at empty-bed contact times (EBCTs) of 4, 8, and 16 minutes were used to evaluate the performance of nutrient-amended (free ammonia and phosphorus) biofiltration for THM and DCAN precursor removal, as measured using formation potential (FP) tests. Many printing inks use volatile solvents in the formulation, which are hazardous to the environment from emission of VOC’s and at the same time, synthetic resins in these inks are not biodegradable. These problems with the fluctuating and rising price of petroleum are main reasons to look for new resources for making more environmentally friendly printing inks. Metastatic melanoma is an aggressive skin cancer with a relatively low survival rate. Immune-based therapies have shown promise in the treatment of melanoma, but overall complete response rates are still low. Previous studies have demonstrated the potential of plasmid IL-12 (pIL-12) delivered by gene electrotransfer (GET) to be an effective immunotherapy for melanoma. However, events occurring in the tumor microenvironment following delivery have not been delineated. Therefore, utilizing a B16F10 mouse melanoma model, we evaluated changes in the tumor microenvironment following delivery of pIL-12 using different GET parameters or injection of plasmid alone. Electrolytic flocculation using non-sacrificial electrodes with flocculants added was studied on harvesting Scenedesmus sp. In order to optimize the operating conditions of the electrolytic flocculation process and to quantify the amount of flocculants added, aluminum electrodes were first used in the process. It was found that under optimal conditions, the microalgae removal efficiency using aluminum electrodes could reach 98.5%, while 34.2 mg L-1 of aluminum ions were released during the process. Different metal electrodes were also studied, but high microalgae removal efficiency was witnessed only using aluminum electrodes, indicating the influence of the aluminum ion in flocculation. Organic products that have historically been viewed as waste products may improve soil health by adding carbon (C) and nutrients to soil. Two such products are woodchips, generated from forest or rangeland management activities, and livestock manure. In Nebraska, eastern redcedar (Juniperus virginiana L.) is a native but invasive tree species inhibiting rangeland productivity. Livestock manure that is underutilized while inorganic fertilizers are imported for crop production presents a water quality risk by contributing to local- and regional-scale nutrient imbalances. This study evaluates the impact of protein acetylation on breast cancer gene expression and the regulation of metabolism. Acetylation is the second abundant post-translational modification after phosphorylation, regulating protein activity and function. The alterations in acetylation of both histone and non-histone proteins is known to be related to many human diseases, including cancer. Acetylation and deacetylation of histones is closely associated with the regulation of gene expression, while acetylation of non-histone proteins may have a broad effect on major cellular processes, such as proliferation, metabolism, cell cycle and apoptosis, imbalanced regulation of which is essential for cancer development. This work classifies examples of infrastructure interdependencies found in the food and agriculture critical infrastructure sector. Interdependencies are identified through an examination of rice and poultry agriculture throughout the state of Arkansas. The subtleties of interdependence examples in the food and agriculture sector are inadequately captured by the well-studied interdependence classification taxonomies. Through 39 interviews, we develop an understanding of the subtle temporal, geographic, and productivity scales of interdependence in over 100 examples and present five new, distinct classifications of interdependence: (1) dynamic physical, (2) dynamic geographic, (3) deadline, (4) delay, and (5) human, economic, and natural resource interdependencies. Microalgae are one of the most promising feedstocks for biodiesel production due to their high lipid content and easy farming. However, the extraction of lipids from microalgae is energy intensive and costly and involves the use of toxic organic solvents. Compared with organic solvent extraction, supercritical CO2 (SCCO2) has demonstrated advantages through lower toxicity and no solvent-liquid separation. Background: Effective enzymatic degradation of crystalline polysaccharides requires a synergistic cocktail of hydrolytic enzymes tailored to the wide-ranging degree of substrate crystallinity. To accomplish this type of targeted carbohydrate recognition, nature produces multi-modular enzymes, having at least one catalytic domain appended to one or more carbohydrate binding modules (CBMs). The Type B CBM categorization encompasses several families (i.e., protein folds) of CBMs that are generally thought to selectively bind oligomeric polysaccharides; however, a subset of cellulose-specific CBM families (17 and 28) appear to bind non-crystalline cellulose more tightly than oligomers and in a manner that discriminates between surface topology.

Quantification of evapotranspiration (ET) is crucial for understanding the water balance and for efficient water resources planning. Agricultural settings have received most attention regarding ET measurements while less knowledge is available for actual ET (ETA) in natural ecosystems, many of which have soils containing significant amounts of stones. This study is focused on modelling ETA from stony soil, particularly in montane ecosystems where we estimate the contribution of stone content on water retention properties in soil. Taking into consideration two different types of surfaces, this dissertation proposes techniques to coat the surface of natural fibers and oil-water interfaces by spontaneous self-assembly of halloysite clay nanotubes. The material of choice, halloysite clay, is a widely available natural resource and can be used after minimal processing. In regards to chemical composition, halloysite clay resembles aluminosilicate mineral kaolin; physically, a defect in the crystal structure ordering causes the sheets of kaolinite to roll into halloysite nanotubes. Introduction: Polymeric nanoparticles (NPs) have been utilized as drug delivery vehicles for a variety of applications. However, achieving sustained-release of small hydrophilic agents is a primary challenge for their use in prolonged delivery applications.
Objective: This study investigates how novel lipid-polymer hybrid particle architectures can be used to improve the release profile of small hydrophilic encapsulants. Here, PLGA NPs were produced via electrospraying and emulsions. Particles with a core-shell architecture were produced via coaxial electrospraying and the ability of this architecture to sustain release was examined. Physiology and Disease is a Biology elective at IMSA that has been developed to be mostly student-centered. Some examples of student projects include modeling heart structure to reflect function and creating LED arduino monitors to measure heart rate. Students also measure their lung capacity and blood pressure to demonstrate correlation of these values with heart rate, and trace the correlation back to neuronal controls. Projects such as these integrate other disciplines such as engineering and conform to NGSS Science and Engineering standards and NGSS Cross cutting Concepts standards. Students take responsibility for their own learning and articulate better on tests.

Background: Lignin is a promising source of building blocks for upgrading to valuable aromatic chemicals and materials. Endocarp biomass represents a non-edible crop residue in an existing agricultural setting which cannot be used as animal feed nor soil amendment. With significantly higher lignin content and bulk energy density, endo-carps have significant advantages to be converted into both biofuel and bioproducts as compared to other biomass resources. Deep eutectic solvent (DES) is highly effective in fractionating lignin from a variety of biomass feedstocks with high yield and purity while at lower cost comparing to certain ionic liquids.

Whilst the phenomenon of an electrical resting membrane potential (RMP) is a central tenet of biology, it is nearly always discussed as a phenomenon that facilitates the propagation of action potentials in excitable tissue, muscle, and nerve. However, as ion channel research shifts beyond these tissues, it became clear that the RMP is a feature of virtually all cells studied. The RMP is maintained by the cell's compliment of ion channels. Transcriptome sequencing is increasingly revealing that equally rich compliments of ion channels exist in both excitable and non-excitable tissue. Agro-industrial waste is highly nutritious in nature and facilitates microbial growth. Most agricultural wastes are lignocellulosic in nature; a large fraction of it is composed of carbohydrates. Agricultural residues can thus be used for the production of various value-added products, such as industrially important enzymes. Agro-industrial wastes, such as sugar cane bagasse, corn cob and rice bran, have been widely investigated via different fermentation strategies for the production of enzymes. Solid-state fermentation holds much potential compared with submerged fermentation methods for the utilization of agro-based wastes for enzyme production. Several catastrophes could block the sun, including asteroid/comet impact, super volcanic eruption, and nuclear war with the burning of cities (nuclear winter). Previous work has analyzed alternate food supplies (e.g., mushrooms growing on dead trees, bacteria growing on natural gas). This was shown to be technically capable of feeding everyone with macronutrients (protein, carbohydrates, and lipids) and minerals, although economics and politics remain uncertain. The present work analyzes vitamin availability in such alternative food scenarios. A new type III polyketide synthase gene (Ssars) was discovered from the genome of Shiraia sp. Slf14, an endophytic fungal strain from Huperzia serrata. The intron-free gene was cloned from the cDNA and ligated to two expression vectors pET28a and YEpADH2p-URA3 for expression in Escherichia coli BL21(DE3) and Saccharomyces cerevisiae BJ5464, respectively. SsARS was efficiently expressed in E. coli BL21(DE3), leading to the synthesis of a series of polyketide products. Six major products were isolated from the engineered E. coli and characterized as 1,3-dihydroxyphenyl-5-undecane, 1,3-dihydroxyphenyl-5-cis-6'-tridecene,1,3-dihydroxyphenyl-5-tridecane, 1,3-dihydroxyphenyl-5-cis-8'-pentadecene, 1,3-dihydroxyphenyl-5-pentadecane and 1,3-dihydroxyphenyl-5-cis-10'-heptadecene, respectively, …
